1.Effects of RGDS tetrapeptide on the actin and apoptosis in human glomerular mesangial cells

Observing the effects of Arg-Gly-Asp-Ser(RGDS) peptides on the actin organization and apoptosis in human glomerular mesangial cells, in order to explore the interaction mechanism of extracellular matrix (ECM)-integrin receptor-cy-toskeleton. Methods The alterations of actin microfilaments in glomerular mesangial cells were observed under laser scanning confocal microscope. And DNA electrophoresis and flowcytometry were used for assessing apoptosis. Results After two hours of incubation with RGDS peptides, some of the attached glomerular mesangial cells began to round up, with a prominent marginaliza-tion of the actin microfilaments which were bundled in thick aggergates to one edge of the cell. Later these cells detached and floated in the culture medium.The above sequently-occuring changes showed a time-dependent pattern. After ten hours of incubation with RGDS peptide, most of the cells had detached, and features of apoptosis appeared in them, such as inter-nucleo-somal DNA fragmentation and decreased DNA content. Conclusion RGDS tetrapeptide can disrupt the organization of cytoplasmic actin mi-crofilament, inhibit the attachment of the cultured glomerular mesangial cells and cause apoptosis in them.
2.Role of type I plasminogen activator inhibitor in mediating renal tubulointerstitial injury of patients with IgA nephropathy

Objective To explore the role of type 1 plasminogen activator inhibitor (PAI-1 ) in mediating renal tubulointerstitial injury of patients with IgA nephropathy. Methods The mRNA and protein production of PAI-l in renal tubulointerstitium were defected using in stiu hybridization and immunohistochemistry. Detections of antigens of ?-smooth muscle actin(?-SMA) and proliferating cell nucleus antigen (PCNA) were also performed. Results PAI-l was normally expressed in the walls of vessels and distal tubules, but significantly increased in lesions of IgA nephropathy, including crescents, Bowman capsules and tubulointerstitial infiltrating cells. 3.A STUDY ON THE MECHANISM OF BENAZIPRIL IN PREVENTING THE PROGERSSION OF GLOMERULOSCLEROSIS

nephrectomy in Wistar rats were used to replicate the animal model of focal and segmental glomerular sclerosis.Then a group of 5/6 nephrectomized rats were given Benzaipril for 20 weeks , in order to evaluate the effects of Benazipril on degradation of extracellular matrix. The results showed that Benzipril not only reduced renal pathological changes, but also significantly decreased the type Ⅳ collagen and MMP 9 deposition and down regulated the mRNA expressions of MMP 9 and TIMP 1 in the remnant kidneys of the 5/6 nephrectomized rats. It suggested that Benzipril might ameliorate glomerulosclerosis through the mechanism of modulating extracellular matrix degradation in the remnant kidney tissue.
4.EXPRESSIONS OF ALPHA-SMA, PAI-1, MMP-9 AND TIMP-1 IN RENAL VESSELS OF PATIENTS WITH IGA NEPHROPATHY

To explore expressions of ? smooth muscle actin (? SMA), plasminogen activator inhibitor 1 (PAI 1), matrix metalloproteinase 9 (MMP 9)and tissue type inhibitor of metalloproteinase 1 (TIMP 1) in renal vessels of 38 patients with immunoglobulin A nephropathy (IgAN), immunohistochemistry method was used to detect abundances of the above proteins. The results showed that ? SMA staining was found mainly in vascular smooth muscle cells (SMCs); apparent PAI 1 staining was also shown by vascular SMCs; MMP 9 expressed considerably in vascular both SMCs and endothelial cells; TIMP 1 was located only in vascular endothelial cells. Objective To observe the expression of neuron apoptotic protein caused by whole body gyperthermis (WBH) in rats anesthetized using propofol and chloral hydrate,and to explore the pathway of any hippocampal neuron apoptosis induced by WBH. Methods Sixty-three male Wistar rats were randomly divided into 3groups,21 in each group.One guoup received neither anesthesia nor WBH treatment sa a control (group A);group B were anesthetized with an intraperitoneal injection of 100 mg/kg of propofol;group C receivde a similar injection of chloral hydrate. This was followed by WBH (keeping the rats' core temperature at 42°for 30 min) for all three groups. The rats'brains were then removed 24 h after WBH to separate the ghpplcampal CA1,CA2 zones and the dentate gyrus regions.Any neuron apoptosis was detected using the TUNEL methol;Bax/Bcl-2/caspase-3 protein expression was measured with a SABC immmunogistochemical technique;and the changes in ultrastructure were observed tith an electron microscope. Results Compared with group A,the changes in the ultrastructure of neurons in the hippocampus changed after WBh were the most severe in group C.The changes included edema of the organelles,vacuolization of mitochondria,rarefaction of the endocytoplasmic reticulum,incopleasmic reticulum,incomplete synaptic membranes and autophagy.Both the number of apoptotic neurons and the expression of Bax and capase-3 protein in the hippocampus increased in the following order: group A<group B<group C.Expression of Bcl-2 protein in the hippocampus increased in the following order: group A<group C<group B. Conclusion Hippocampal apoptosis induced by WBh involves upregulating the expression of Bax and caspase-3 and downregulating the expression of Bcl-2.
8.PrP 106-126 Altered PrP mRNA Gene Expression in Mouse Microglia BV-2 Cells'

Prion diseases are infectious and fatal neurodegenerative diseases. The pathogenic agent is an abnormal prion protein aggregate. Microglial activation in the centre nervous system is a characteristic feature of prion disease. In this study, we examined the effect of PrP 106-126 on PrP mRNA gene expression in Mouse microglia cells BV-2 by real-time quantitative PCR. PrP mRNA expression level was found to be significantly increased after 18 h exposure of BV-2 cells to PrP 106-126, with 3-fold increase after 18 h and 4.5-fold increase after 24 h and BV-2 cells proliferating occurred correspondingly. Our results provide the first in vitro evidence of the increase of PrP mRNA levels in microglial cells exposed to PrP 106-126, and indicate that microglial cells might play a critical role in prion pathogenesis.

10.30 Children with Hand Foot and Mouth Disease following Acute Flaccid Paralysis

ObjectiveTo investigate the clinical characteristics and epidemic analysis in children with hand foot and mouth disease(HFMD) following acute flaccid paralysis(AFP).Methods30 children with HFMD following AFP in Hunan Province Children's Hospital were surveyed retrospectively.ResultsAll the patients were below 5 years old, 83.34% were 1~3 years old, 90% from countryside. Besides AFP of limbs, 86.67% complicated with encephalitis. The major clinical characteristics as followed: fever(100.00%), skin rash(100.00%), startle and skip(80.00%), emesis(63.33%), neurogenic pulmonary edema(13.33%) and urinary retention(16.67%). The most palsy occurred in 3~5 days after onset, and the palsy in lower limbs was major, only a few had palsy in upper extremities or tetraplegia. 26 cases were infected with enterovirus type 71(EV 71), 1 case infected other EV, 3 cases were negatively. The EMG showed that the amplitude of active potential lowered in the femoral nervus, tibial nerve, peroneal nerve, axillay nerve and musculocutaneous nerve, or/and the motor conduction velocity slowed down, the damage of proximate nerve was the most common.ConclusionThe most children with HFMD following AFP were infected with EV 71. Encephalitis was the common complication. The symptom of EV 71 was more severious than distribute EV.
