International Journal of Cerebrovascular Diseases 2008;16(12):921-925
Long-term prognosis of stroke rehabilitation is associated with a number of factors, such as age, sex, race, lesion sites and sides, lesion nature and various complications. Under-standing these related factors not only contribute to clinical judgement of prognosis, but also provide the basis for the targeted rehabilitation therapy.

OBJECTIVE:To discuss the effect of anti-tuberculosis therapy on plasma concentration of calcineurin phosphatase inhibitors in renal transplant recipients.METHODS:The change in plasma concentration of calcineurin phosphatase inhibitors in 2 renal transplant recipients who were complicated with tuberculosis infection before or after renal transplantation was analyzed. The patients were routinely treated with cyclosporine(CsA)or tacrolimus(FK506)+ mycophenolate mofetil(MMF)+ prednisolone(Pred).Monoclone fluorescence polarization immunoassay(FPIA)was employed to determine the concentration of CsA or FK506 in different time point before and after administration of antituberculosis drugs.RESULTS:Plasma concentration of CsA was down-regulated significantly by Rifampicin, and the daily dose of CsA has to be increased to more than 3 times the previous concentration before meeting the target concentration. The concentration of FK506 was slightly down-regulated by rifapentine.CONCLUSION:The concentration of CsA or FK506 and the adverse effect of antituberculotic drugs should be closely monitored for renal transplant recipients complicated with tuberculosis infection.

To evaluate species specificity of 20 STRs loci used in our lab. Human DNA and 10 different kinds of common animals' DNA were amplified separately. The PCR products were analysed with PAGE. There are amplified products in DYS388, DYS389, DYS390, D7S809, D13S631 loci of human 'DNA, but not of 10 different kinds of animal DNA tested. There were amplified products in TH01, vWA, AR, CD4, FABP loci for human and animals' DNA, but the lengths of amplified fragment were distinctly different between human and animals. There were products of FES, D12S391, CSF1PO, D19S253, D21S11, FGA, DYS19 loci for human and animals DNA, but the product lengths were not different between human and animals. There were PCR products in SE33 and D11S554 loci for human and animals, their amplified fragment lengths were within the same electrophoretic field. 10 STR loci, such as TH01 etc, were highly specific for human being and the amplified fragments lengths of 8 STR loci, such as FES etc, were different between human and animal tested.

By analysing the 300 cases of paternity test in our lab in the recent year, we have demonstrated that mutation rates in STR loci are very high. The experimental steps include extracting DNA by chelex 100, amplification of DNA, polyacrylamide gel electrophoresis, silver staining, DNA typing by standard samples. The STR mutations in 11 cases among 300 paternity test cases were found, induding D11S554 locus6 cases, D19S253 locus2 cases, SE33 locuslcase, D12S391 locus 1 case, D13S631 locus 1 case. The results indicated that high mutation rates in STR loci should be considered when STR genotyping are applied to paternity test.

Chinese Journal of Forensic Medicine 2000;0(S1):-
To establish a calculation method of paternity probability in cases of absence of mother. Calculating the accumulated non-father exclusion probability of multiple polymorphic DNA loci was performed. The results showed that in cases of inheritance with Mendelian Law, through testing 8 or more polymorphic DNA loci, the paternity probability may reach 0. 9990 or more. In cases of paternity exclusion, exclusion 3 or more than 3 loci were required. For the purpose of identifying paternity in cases of absence of mother, more than 8 polymorphic DNA loci must be tested. The paternity probability must be more than 0. 9990 in cases of paternity inclusion and in cases of paternity exclusion, 3 or more loci of exclusion is needed.

Objective To design a set of long bone and bone-mode auto-matching system which is based on computer image processing technology. Methods Through the analysis on clinical pictures and image data,we got the exact data.Based on these data, the system searched out the bone-mode from the bone-mode database which matched with the skeletal. Results It can find a suitable bone-mode which can match with the bone in a very short time. Conclusion The system improves the efficiency of matching,as well as the success rate.It reduces the surgery time and the suffering of patents.

Acta Anatomica Sinica 1954;0(02):-
Objective To study the influence of ion concentration change out of cells on calcium transportation and the relationship between the rising of calcium concentration in the Caco-2 cells and its apoptosis to offer the theoretical and experimental bases for clinical study and digestive tract physiology and patholoogy. Methods Confocal laser scanning microscope(CLSM) and flow cytometry(FCM) were used in this study. Results (1) Diversion of Ca 2+ into cells was increased with the decrease of Fe 3+ concentration out of Caco-2 cells(the final consistence of DFO was 100-300??mol/L),but was hindered with the increase of Fe 3+ concentration out of them(the final consistence fo FAC was 10-100??mol/L) as observed under CLSM; (2) The cell state was fine and its viability was more than 90% as observed under CLSM after treated by A 23187 and Fluo-3/AM(examined with FDA);(3) The Ca 2+ concentration in the Caco-2 cells was increased by A 23187 and this function was dose depended. The Caco-2 cell apoptosis was induced by the increase of Ca 2+ in cells, which was examined with FCM.Conclusion The Ca 2+ transportation was increased with the decrease of Fe 3+ concentration out of Caco-2 cells but was hindered with the increase of Fe 2+ concentration out of them. The Caco-2 cell apoptosis was induced by the increase of Ca 2+ concentration in them.

Objective To research the significance of the levels of neuron-specific enolase(NSE), S100B and myelin basic protein(MBP) in predicting the severity and prognosis in patients with subarachnoid hemorrhage (SAH). Methods The serum levels ofNSE, SI00B and MBPwere measured within 72 hours after the injury in 36 patients with SAH(SAH group), then the severity of illness and prognosis was evaluated by world federation of neurosurgical societies(WFNS) grade and Glasgow outcome scale(GOS) respectively. Twenty healthy persons were selected as controls(control group). Results After the injury, the serum levels of NSE,SI00B and MBP in SAH group increased significantly compared with those in control group. Moreover, there were significant difference in the serum levels of NSE, S100B and MBP among different groups in different severity of illness and prognosis. Assessed the severity and prognosis by serum levels of NSE 25 μg/L, S100B 1.2 μg/L and MBP 10.0 μg/L after 72 hours of SAH, the specificity of assessing the severity by NSE, SI00B and MBP was 71.43%,61.90%, 66.67%, and the sensitivity was 73.33%,86.62% and 73.33%. At assessment of prognosis, the specificity of NSE, S100B, MBP was 69.57%, 60.87%,65.22%, and the sensitivity was 61.54%,76.92% and 69.23%. Conclusion There are higher specificity and sensitivity in evaluating the severity of illnoss and prognosis in patients with SAH by serum levels of NSE, S100B and MBP.
