1.THE INFLUENCE OF Fe~(3+) ON Ca~(2+) AND THE RELATIONSHIP BETWEEN Ca~(3+) AND THE CELL APOPTOSIS IN Caco-2 CELLS

Objective To study the influence of ion concentration change out of cells on calcium transportation and the relationship between the rising of calcium concentration in the Caco-2 cells and its apoptosis to offer the theoretical and experimental bases for clinical study and digestive tract physiology and patholoogy. Methods Confocal laser scanning microscope(CLSM) and flow cytometry(FCM) were used in this study. Results (1) Diversion of Ca 2+ into cells was increased with the decrease of Fe 3+ concentration out of Caco-2 cells(the final consistence of DFO was 100-300??mol/L),but was hindered with the increase of Fe 3+ concentration out of them(the final consistence fo FAC was 10-100??mol/L) as observed under CLSM; (2) The cell state was fine and its viability was more than 90% as observed under CLSM after treated by A 23187 and Fluo-3/AM(examined with FDA);(3) The Ca 2+ concentration in the Caco-2 cells was increased by A 23187 and this function was dose depended.The Caco-2 cell apoptosis was induced by the increase of Ca 2+ in cells,which was examined with FCM.Conclusion The Ca 2+ transportation was increased with the decrease of Fe 3+ concentration out of Caco-2 cells but was hindered with the increase of Fe 2+ concentration out of them.The Caco-2 cell apoptosis was induced by the increase of Ca 2+ concentration in them.
2.Hepcidin and iron homeostasis

The understanding of iron metabolism in humans, especially of its mechanism involved in controlling iron absorption in the proximal small intestine, is of great importance since diseases associated with iron deficiency or overload are very common worldwide. Recent study on hepcidin which is senthesized by liver has showed that this originally identified as a circulating antimicrobial peptide is a putative iron regulatory hormone. It plays a central role in the regulation of small intestine iron absorption and body iron homeostasis. The increased expression of hepcidin in the liver, induced by inflammation, might be an initial cause of the anemia of infection or chronic diseases and the iron-overload diseases might be tightly associated with the decreased hepcidin expression in the liver.
3.An effect of cerebral blood flow on Broca aphasia patient of cerebrovascular disease by early rehabilitative intervention

Objective To observe the effect of cerebral blood flow on Broca aphasia patient of cerebrovascular disease by early rehabilitative intervention, to evaluate the result of early rehabilitative intervention, to ques the marker evaluating early effect. Methods 41 Broca aphasia patients were divided to rehabilitation groups (23 patients) and control groups(18 patients) according to the sequence of hospitalization, and meanwhile 37 patients of health examination were chosen as health control groups, the patients of rehabilitation groups were treated with drug and rehabilitative intervention but the patients of control group were treated only with drug, the average cerebral blood flow of left middle cerebral artery of both groups and health control group was examined with transcranium doppler before and after treatment. Results The average cerebral blood flow of left middle cerebral artery of rehabilitation group and control group was not different before treatment, the rehabilitation group and control group was less than health control group; the average cerebral blood flow of left middle cerebral artery of rehabilitation group and control group was increased obviously after treatment than that before treatment; the average cerebral blood flow of left middle cerebral artery was not different between rehabilitation group and health control group after treatment, it was different between control group and health control group; the average cerebral blood flow of left middle cerebral artery was increased obviously on rehabilitation group than that on control group after treatment. Conclusions the early rehabilitative intervention can increase the average cerebral blood flow of left middle cerebral artery of Broca aphasia patient accelerate the functional restoration; TCD can be regarded as an early marker to evaluate early effect.
4.Stimulator of Fe transport and brain iron metabolism

OBJECTIVE: In order to understand the effect of stimulator of Fe transport(SFT) on Fe metabolism and its abnormality(absence or overloading),this study reviews the research development of SFT at home and abroad and focused on the relationship among expression,structure,physiological function,expressing controlling and expressing abnormality with brain iron metabolism.DATA SOURCES: Electronic literature search of NCBI related to SFT was performed using the terms "stimulator of iron transport" or "stimulator of Fe transport",and the language was restricted in English. And simultaneously CNKI database was searched with the word "brain iron metabolism" and"stimulator of Fe transport" in Chinese from January 1997 to October 2004.STUDY SELECTION: Articles that reported the structure,expression regulation of SFT and its relationship to brain iron metabolism diseases were included.DATA EXTRACTION: Twenty pieces of SFT-related literatures and 1300pieces of literatures related to brain iron metabolism were found,among which 21 pieces were included.DATA SYNTHESIS: From the 21 pieces of literatures,the structure,distribution,biological function,expression regulation of SFT and its relationship with brain iron metabolism were mainly discussed.CONCLUSION: SFT can stimulate both transferrin- and nontransferrin-bound iron uptake. The expression of SFT can be regulated transcriptionally and post-transcriptionally,mainly regulated in response to different cellular iron levels. So SFT plays an important role in brain iron metabolism.

Results The shape and ultrastructure of myocytes became complicated gradually with development. Conclusion All the changes are related to function of blood supply of human fetal heart.
6.Effect of lipopolysaccharide on the iron metabolism in macrophages

Objective To study the effect of lipopolysaccharide ( LPS ) on the activity of primary cultured macrophages and the distribution of divalent metal transporter 1 ( DMT1 ) and ferroportin 1 ( FPN1 ) .Methods Primary cell culture , MTT chromotest , cytochemistry chromotest and cell immunofluorescence techniques were used in this work . Results DMT1 was mainly distributed in the cytoplasm , which illuminates that DMT1 mediates the macrophage intracellular transit of iron from phagolysosome to cytoplasm .FPN1 was distributed in the cytoplasm and membrane , and the cytoplasm was the main site of FPN 1 distribution in macrophages .Conclusion Iron liberation from heme inside the phagolysosome occurs after erythrophagocytosis and it is possible that FPN 1 mediates intracellular transit of iron released by heme catabolism .The study found that LPS promoted the cell growth and this effect was reached to the peak in the 10 -5μg/L LPS group, but the cell growth was blocked with the increase of LPS concentration .
7.Effects of lipopolysaccharide on mice iron metabolism related genes mRNA expression

ObjectiveTo investigate the effects of lipopolysaccharide (LPS) on mRNA expression of iron metabolism related genes. Methods Ten male mice (2 months) were injected intraperitoneally with lipopolysaccharide(0.5 μg/g). After 6 hours, mice were sacrificed and then sera, liver and spleen were collected. The mice blood routine was measured. The serum iron and total iron binding capacity (TIBC) were determined with reagent kit. The quasi-quantitative reverse transcription polymerase chain reaction (RT-PCR) was performed for mRNA of hepatic hepcidin(HP), ferroportin1(Fpn1), transferrin receptor 1(TfR1) and spleenic HP, Fpn1 and interleukin-6(IL-6). Results The serum iron and TIBC were reduced in mice injected LPS, which exhibited mild anemia(P<0.05) . LPS can increase the expression of hepatic hepcidin and decrease Fpn1 and TfR1 in liver after LPS administration 6 hours(P<0.05). In spleen, IL-6 was upregulated and Fpn1 downregulated(P<0.05). Conclusion LPS can influence serum iron through regulating the mRNA expression of hepatic and spleenic iron metabolism related genes, such as HP, Fpn1 and TfR1.

Objective To investigate the change of eNOS,the spermatogenic cell proliferation,apoptosis in mouse testis exerted by alcohol. Methods The immunohistochemical staining method for detecting of eNOS,PCNA,TUNEL method for detecting of apoptotic cells and the satistics analysis were used in the present study. Results With the increase of alcohol concentration,the structure of seminiferous tubule changed,the diameter of seminiferous tubule decreased,the surface density of postive eNOS cells increased gradually,and the number of positve PCNA cells and apoptosis cells also increased.There were significant difference in 15% alcohol concentration group compared with the other groups(P
9.Analysis of the epidemic situation of inter-human brucellosis in Zhongwei City of Ningxia from 2005 to 2017

Objective To analyze the epidemic situation of inter-human brucellosis in Zhongwei City of Ningxia from 2005 to 2017,and to provide basis for making scientific and effective prevention and control measures of the disease.Methods Incidence data of brucellosis were collected in Zhongwei City from 2005 to 2017 and prevalence trends and distribution characteristics (place,time,and population distribution) were analyzed by descriptive epidemiological method.Results A total of 1 880 human brucellosis cases were reported in Zhongwei City from 2005 to 2017.The incidence of brucellosis in 2012 and 2015 was 5.03/100 000 and 61.80/100 000,which showed a dramatic and rapid ascent (x2trend =681.40,P < 0.05),while after 2016,it showed a downward trend (xtrend =324.85,P < 0.05),and in 2017 the reported incidence dropped to 25.13/100 000.Zhongning and Haiyuan Counties showed a high incidence from 2005 to 2017,and the incidence was 39.79%(748/1 880) and 36.06% (678/1 880),respectively;April to September was the peak season;the sex ratio was 2.72:1.00 (1 375/505);1 379 cases (73.35%)occurred between 30 and 64 years old,and 1 616 cases (85.96%) occurred among farmer.Conclusion After 2016,the incidence of human outbreaks in Zhongwei City has declined;Zhongning and Haiyuan Counties are the key areas of morbidity;the key population for prevention and control is farmer.
10.Epidemiological analysis of human brucellosis in Ningxia Hui Autonomous Region from 2004 to 2019

Objective:To learn about the epidemiological characteristics and trends of brucellosis in Ningxia Hui Autonomous Region (Ningxia), and to provide reference for formulation of brucellosis prevention strategies and identification of key areas.Methods:The case data from 2004 to 2019 were collected from the "China Disease Prevention and Control Information System Infectious Disease Monitoring and Reporting System", and descriptive epidemiological methods were used to analyze the distribution of cases (time, population, and regional distribution).Results:From 2004 to 2019, a total of 15 337 human brucellosis cases and 1 death case were reported in Ningxia. The average annual incidence rate was 15.21/100 000, ranging from 0.02/100 000 to 44.83/100 000. The difference in incidence rate in different years was statistically significant (χ 2 = 14 731.60, P < 0.001). Among them, from 2004 to 2010, the incidence rate was low, with an average annual incidence rate of 1.32/100 000; from 2011 to 2019, it entered a high incidence stage, with an average annual incidence rate of 24.83/100 000. Brucellosis occured in every month of the year, and the onset time was mainly in March to September, accounting for 71.60% (10 981/15 337); seasonal indexes from April to August was all > 100%, with obvious seasonality. The age of onset mainly concentrated in 20 - < 70 years old, accounting for 89.60% (13 742/15 337); males were significantly more than females, with a sex ratio of 2.66 ∶ 1.00 (11 141/4 196); the occupations were mainly farmers and herdsmen, accounting for 86.03% (13 194/15 337). Since 2014, cases had been reported in 22 counties (districts, cities) in the region, all of which were all severely endemic areas; the top three counties (districts) with incidence rate were Yanchi County (103.73/100 000), Hongsibao District (56.01/100 000) and Tongxin County (29.18/ 100 000), respectively, the difference between different regions was statistically significant (χ 2 = 3.80, P < 0.001). Conclusions:The incidence rate of brucellosis in Ningxia is on the rise, and the epidemic situation is severe. It is recommended to carry out active and effective intervention measures in areas with high incidence of brucellosis to curb the further spread of brucellosis.
