2.Effect of RNA Interference Targeting HIF-1αGene on the Proliferation of Human Tongue Cancer Cells

Objective To explore the effect of RNA interference targeting hypoxia-inducible factor-1α(HIF-1α) gene on the proliferation of human tongue cancer cells (Tca8113 and Tscca). Methods HIF-1α-specific short hairpin RNA (shRNA) expression vector and empty shRNA vector were respectively transferred into two kinds of cultured human tongue cancer cells. The expression of HIF-1αgene was detected by RT-PCR and Western blot method. The clone forma-tion assay was used to observe the cell proliferation. The cell counting was used to monitor the number of living cells. Flow cytometry was applied to detect cell cycle distribution. Additionally, the cells were injected into nude mice, and tumor form-ing condition was observed. Results After the shRNA HIF-1αplasmid was transfected into tongue cancer cells, the expres-sions of both HIF-1αmRNA and protein were significantly decreased in tongue cancer cells under hypoxia condition. The number of colony formation was significantly lower in HIF-1αsmall interfering RNA (siRNA) group than that in empty and negative control group. The cell counting revealed that the number of living cells was remarkably lower in HIF-1αsiRNA group than that in negative control group. Also, the cell cycle in HIF-1α siRNA group was arrested at G0/G1 phase after transfection and 48 hours hypoxia culture. Besides, the tumor volume was remarkably smaller in HIF-1αsiRNA group than that in negative control group after the tongue cancer cells transplanted into nude mice for several days. The statistical analy-sis indicated that the difference was significant (P<0.05). Conclusion RNA interference targeting HIF-1α gene could suppress the proliferation of human tongue cancer cells effectively. HIF-1αgene could be researched as a new therapeutic target of tongue cancer in the future.
3.Effect of Yinaotongluo Capsule on diffuse weighting image and energy metabolism of ischemia-reperfused rats

ObjectiveTo explore the effect of Yinaotongluo Capsule on diffuse weighting image and enenery metabolism of ischemia-reperfuing rat.MethodsMagnetic resonance diffusion-weighted imaging(DWI) and 1H and magnetic resonance spectroscopy(MRS) were performed in different brain regions in focal cerebral ischemic-reperfusion injury model rats. ResultsYinaotongluo Capsule could significantly reduce the area and the intensity of infarction of rats by DWI 3 h and 5 d after treatment compared with the model group.The result of 1H MRS examination demonstrated Lac peak was lower and NAA peak higher than that of the model group. ConclusionYinaotongluo Capsule can effectually improve enenery metabolism during cerebral ischemia and reperfuing, so that to reduce the neuronal dysfunction and death in infarcts at the late stage.
4.Reform of Medical Foundation Courses for Rehabilitation Therapy in Medical College

5.The Gene Polymorphism of VMAT2 Is Associated with Risk of Schizophrenia in Male Han Chinese

Objective:
To investigate the association between gene polymorphism of vesicular monoamine transporter type 2(VMAT2) and schizophrenia in Han Chinese population.
Methods:
430 patients with schizophrenia and 470 age-sex matched controls were recruited from four mental health centers. All patients were diagnosed by two psychiatrists based on the Structured Clinical Interview for DSM Disorders (SCID). The ligase detection reactions (LDR) method was used to assess the polymorphism of the two SNPs (rs363371 and rs363324) of VMAT2.
Results:
No associations of two SNPs with schizophrenia was found. When we stratified males and females for the analysis, we found that that in the recessive model of rs363371, there was an obvious significant association between rs363371 and schizophrenia in males (OR=0.564, 95% CI=0.357–0.892, p=0.014) but not females. For the association between rs363324 and schizophrenia, no association was found in either males or females. No association was found when stratifying early-onset schizophrenia and late-onset schizophrenia.
Conclusion
Our findings indicate that both rs363371 and rs363324 were not associated with schizophrenia, while it seemed that the AA genotype of rs363371 plays a protective effect in male Chinese in developing schizophrenia.
6.One case of Creutzfeldt-Jakob disease with restless leg syndrome as the first symptom and literature analysis

Objective:To analyze the clinical features and auxiliary examination results of sporadic Creutzfeldt-Jakob disease (sCJD) with restless leg syndrome (RLS) as the first symptom.Methods:The clinical features and auxiliary examination results of one case of sCJD who received treatment in Sichuan Mianyang 404 Hospital were analyzed based on relevant literature.Results:A 59-year-old woman of Han nationality who had sCJD with restless leg-like manifestation of the left lower limb for 18 days was included in this study. The patient was first treated in orthopedic department, but her symptom did not improve after treatment. Twenty days later, she was transferred to neurology department for further treatment. Her daily life and activities were not affected. Head magnetic resonance imaging, electroencephalography, cerebrospinal fluid routine examination and biochemical test results were normal. Five days later, the patient had mild left-sided ataxia, which then progressed rapidly, followed by right-sided ataxia, left-leg spasticity and adduction, involuntary movement, myoclonia, cognitive decline, akinetic mutism, repeated hyperthermia, repeated complex partial seizures. Two weeks later, head magnetic resonance imaging examination revealed hyperintense signal of the cingulate gyrus, frontal cortex and right island cortex on DWI, with cerebellar atrophy and three-phase electroencephalography wave. Four weeks later, CSF14-3-3 protein was positive, and no related genetic mutation in the prion protein gene was found. The duration from onset to death was about 8 months.Conclusion:sCJD is a common subtype of prion protein disease, and the condition can be stabilized for more than 1 month after the onset of RLS. There is no specificity in early clinical and auxiliary examinations, and neither dobutazine treatment nor neurotrophic treatment is effective. The disease progresses rapidly after 1 month, head MRI and EEG reexamination can reveal clues, and CSF14-3-3 protein can assist clinical diagnosis.
