1.A study on the life quality of patients with condyloma acuminatum and correlated factors

Objective To evaluate the quality of life and correlated factors in patients with condyloma acuminatum (CA). Methods 101 patients with CA and 50 healthy people as control were assessed by using the questionnaire of QOL-35. Results Compared with control group, the total score and general, physical, inde-pendence, psychological, social, environment domains scores of QOL were lower significantly in the CA group ( P <0.05). The QOL score has negative correlation with course of disease( r= -0.233, P<0.05), recurrence times(r= -0.556, P<0.05). Nevertheless, it has no noticeable correlation with gender, age, marital status, education, career (P0.05). Conclusion Quality of life was lower in patients with CA, and it was closely related with the course of disease and recurrence times.
2.Surgical therapy for malignancy patients complicated with hyperthyroidism

Objective To summarize the therapeutic experience on malignancy patients complicated with hyperthyroidism.Methods Clinical date of 10 cases of surgical malignancy complicated with hyperthyroidism admitted from May 2004 to May 2010 in Tianjin Medical University General Hospital were analyzed retrospectively.Results Four cases were treated by subtotal thyroidectomy before radical operation for cancer.Radical operation for cancer was performed on 6 patients after clinical symptoms of hyperthyroidism were controlled by perioperative antithyroid agents.Postoperatively 3 patients complicated with clinical manifestations similar to thyroid crisis.There was no postoperative mortality.Chemotherapy was given to 10 patients,and 1 patient was discontinued for chemotherapy caused leucopenia.Conclusions Hyperthyroidism should be controlled by surgery or antithyroid agents before patients of malignant diseases could proceed with radical surgery.Proper preoperative medication and effective postoperative management can reduce operation risk and help the patients get through the perioperative period safely.
3.Spiral CT Evaluation in Patients with Obstructive Sleep Apnea

Objective To investigate the upper airway caliber and the corresponding pharyngeal wall in obstructive sleep apnea syndrome(OSAS) by spiral CT.Methods Seven patients with OSAS confirmed by polysomnography(PSG) underwent spiral CT scan.The thickness of posterior pharyngeal wall of retropalatal region(RP),retroglossal region(RG) and epiglottal region(EPG) were measured.Results ①Among 7 patients,5 had only RP obstruction,and 2 had RP and RG regions obstruction;②The length and width of soft palate were 44.28 mm?1.16 mm(normally 41.90 mm?1.95 mm) and 9.42 mm?0.46 mm(normally 5.84 mm?0.24 mm) respectively;③In OSAS patients,the thickness of posterior pharyngeal wall of RP,RG and EPG region were statistically larger than that of the normal group.Conclusion Spiral CT is of practical value in evaluating the pharyngeal airway obstruction of OSAS.
4.Prolongation of cardiac allograft survival in mice by Decitabine-induced upregulation of Foxp3 expression

Objective To investigate the effect of Foxp3 gene expression induced with the DNA methyltransferase inhibitor 5-aza-2'-deoxycytidine (Aza) in recipients on ameliorating allograft rejection and prolonging allograft survival and the possible mechanisms.Method BALB/c and C57BL/6 splenocytes treated with mitomycin (MMC) served as the stimulators,those not treated with MMC as responders,and they were subjected to one-way mixed lymphocyte culture (MLC).In the presence or absence of Aza,the percentage of CD4+ CD25+ Tregs was detected by flow cytometry.Abdominal heterotopic heart transplantation model was established by using inbred male Balb/c mice as donors and C57BL/6 as recipients respectively.In experimental group,recipients were intravenously administrated with decitabine (1.5 mg/kg/day) on the day 1 to day 3 post-transplantation.The control mice were treated with normal saline.CD4+ CD25+ Tregs proportion in the blood and Foxp3 mRNA in cardiac grafts and spleen of recipients were measured respectively,and the survival and the histopathologic changes of the cardiac grafts were also observed.Result Decitabine enhanced the proliferation CD4 + CD25 + Tregs of MLC in vitro.The median survival time of the cardiac grafts in experimental group (11 days) was longer than that in control group (7 days) (P<0.01).As compared with control group,Foxp3 mRNA expression in the cardiac grafts and spleen in recipient mice was significantly up-regulated,and CD4+ CD25 + Tregs proportion was increased (P<0.01) and the proliferation of lymphocytes and monocytes infiltration was inhibited.Conclusion Up-regulation of Foxp3 gene expression induced with decitabine in recipients could alleviate cardiac allograft rejection and prolong cardiac allograft survival via the induction of proliferation and differentiation of CD4+ CD25 + Foxp3 + Tregs.
5.A dosimetric comparison of three plans in simultaneous integrated boost intensity-modulated radiotherapy in cervical esophageal carcinoma undergoing prophylactic lymphatic irradiation

Objective To evaluate the optimized simultaneous integrated boost intensity-modulated radiotherapy (SIB-IMRT) plans in cervical esophageal carcinoma underwent prophylactic lymphatic irradiation.MethodsSix patients with cervical esophageal carcinoma were studied.The gross tumor volume (GTV) and clinical target volume (CTV) (including bilateral mid-lower neck and supraclavicular lymph drainage regions,upper mediastinal lymph drainage regions) were delineated on sim-CT images,GTV and CTV were uniformly expanded by 5 mm to create PGTV and PTV.Five fields(5F) ,7F and 9F SIB-IMRT plans were designed.The prescribed doses to PGTV and PTV were 66 Gy and 60 Gy in 30 fractions respectively.The parameters of dose-volume histograms in three groups planning were compared.Results The values of conformity index(CI) of PGTV were 0.56,0.62 and 0.69 (F =6.22,P <0.01 ),the V66 with 88.24% ,95.03% and 94.91% ( F = 4.39,P < 0.05 ) and D95 with 6539.67,6601.83 and 6602.00 cGy (F=4.46,P <0.05) in 5F,7F and 9F SIB-IMRT plans.The values of CI,V66 and D95 of PTV were 0.80,0.85 and 0.87 (F=11.29,P<0.01),with 91.64%,94.05% and 95.06% (F=4.10,P<0.05) and with 5934.00,5987.17 and 6006.33 cGy (F =4.01,P <0.05).The values of maximal dose of spinal cord were 4707.17,4497.83 and 4357.00 cGy( F = 11.26 ,P <0.011 ) in 5F ,7F and 9F SIB-IMRT plans.The values of all dosimetic parameters about PGTV and PTV were same in 7F and 9F SIB-IMRT plans ( all P > 0.05 ).Conclusion 7F-SIB-IMRT plan is best one scheme for cervical esophageal carcinoma underwent prophylactic lymphatic irradiation.
6.The inhibiting effect on rat C Ⅱ TA and MHC Ⅱ gene expression by plasmid vector-based RNAi technulogy

Objective To investigate the inhibitory effect on rat MHC Ⅱ transactivator (C Ⅱ TA) and MHC Ⅱ gene expression by plasmid vector-based RNAi technology. Methods According to C Ⅱ TA ge-netic information, three short hairpin RNA (shRNA) sequences were designed and the corresponding plas- mid vectors were constructed. Rat dendritic cell (DC) was transfected in vitro and rat spleen was transfected in vivo. Real time RT-PCR and flow cytometry were used to detect the expression of C Ⅱ TA and MHC Ⅱ in DC and spleen after transfection. Results C Ⅱ TA-shRNA plasmids were successfully constructed. Com-pared with control groups, the mRNA transcription levels of C Ⅱ TA and MHC Ⅱ and the expression level of MHC H were significantly inhibited in all three pC Ⅱ TA-shRNA experimental groups ( P < 0.01 ). There was positive correlation between the expression of C Ⅱ TA and MHC Ⅱ. Among the three shRNA groups, the first one showed the strongest inhibitory effect. Conclusion The expression of rat C Ⅱ TA and MHC Ⅱ can be obviously inhibited by plasmid vector of shRNA targeting C Ⅱ TA gene, which can be used for further investi-gation of gene therapy.

Objective To study the therapeutic effect on murine allergic asthma with recombinant Bla g 2(rBla g 2) allergen and its possible mechanism.Methods Eighteen BALB/C mice were randomly divided into three groups:normal control group(group A) , asthma model group(group B) , and recombinant protein rBlag2 treatment group(group C).Mice in groups B and C were subcutaneously immunized weekly with rBla g 2(50 mg) formulated in Al(OH)3 adjuvant for three weeks.Group A received only adjuvant emulsified with normal saline.Two weeks after the last inoculation, mice in group C were administered each with rBla g 2(100 mg) /dose, and groups A and B were given PBS.All the mice received eight doses at 2-day intervals.One week after the last immunotherapy, mice in groups B and C were intranasally challenged with 50 mg rBla g 2 daily for seven days, while mice in group A received PBS.Twenty-four hours after the challenge, the following items were examined:airway hyperresponsiveness of mice, total cellular score and cell classification in bronchoalveolar lavage fluid(BALF) , level of rBla g 2-specific IgE and IgG2a in serum, lung inflammation by HE stain, and Bcl-2 expression of eosinophils of lung by immunohistochemistry.Results Compared with group B, group C showed a decreased Penh value of airway hyperresponsiveness(P
9.Construction and identification of wild-type PTEN eukaryotic expression vector

Objective To construct the recombinant plasmid that highly expressed human subcellular PTEN,in order to provide a basis for further study on its anti-tumor effect. Methods PTEN cDNA was amplified by RT-PCR based on mRNA of placenta.The PCR product was ligated into T-vector,and transfected into E.coli;the obtained T-PTEN plasmid was identified with restrictive digestion and sequencing.PCR was used to incorporte nuclear signal of localization(NSL) into PTEN when T-PTEN was used as template.Then the PCR product was ligated into T-vector,and transfected into E.coli,and T-NSL-PTEN plasmid was obtained.pcDNA3.1 and T-NSL-PTEN were ligated after digested with EcoRⅠand BamHⅠ,and transfected into E.coli,the recombinant vector pcDNA3.1-NSL-PTEN was obtained,and identified with digestion and sequcncing.Results The recombinant expression vector DUM-PTEN and PUM-NSL PTEN were identified by restrictive digestion and DNA sequencing.As expected,by EcoRⅠ and BamHⅠ digestion,it showed the band of 1 200 bp.The sequencing result showed the NSL was incorporated successfully.The recombinant pcDNA3.1-PTEN was obtained with 1 200 bp,the sequencing result showed that its sequence was same as target gene;the recombinant pcDNA3.1-NSL-PTEN was comfirmed by restrictive digestion and sequencing,and the NSL was incorporated successfully. Conclusion The recombinant expression plasmid pcDNA3.1-NSL-PTEN is constructed successfully which can highly express human subcellular PTEN.
10.Mediating effect of insomnia in mobile phone addiction, aggressive behaviors and self-perceived identity among university students

Objective:
To investigate the mediating effect of insomnia among mobile phone addiction, aggressive behaviors and self-perceived identity among university students.
Methods:
A total of 740 university students were sampled from five universities in Binjiang District, Hangzhou City using a cluster random sampling method. The mobile phone addiction, aggressive behaviors and self-perceived identity were assessed using the Mobile Phone Addiction Index Scale, the Athens Insomnia Scale, the Chinese college students' version of the Buss-Perry Aggression Questionnaire, and the Self-Perceived Identity Scale, and the mediating effect of insomnia among mobile phone addiction, aggressive behaviors and self-perceived identity was examined using Process macro program and Bootstrap method.
Results:
A total of 740 questionnaires were allocated, and 700 valid questionnaires were recovered, with an effective recovery rate of 94.59%. The respondents included 221 men (31.57%) and 479 women (68.43%), and there were 607 respondents with a specialty of medicine (86.71%). There were 331 participants detected with mobile phone addiction (47.29%), 90 with aggressive behaviors (12.86%) and 289 with low-level self-perceived identity (41.29%), and the prevalence rates of mild, moderate, severe and extremely severe insomnia were 28.00%, 26.14%, 26.43% and 19.43% among respondents, respectively. Mobile phone addiction had a direct predictive effect on aggressive behaviors (β=0.301, P<0.001) and self-perceived identity (β=-0.129, P<0.001), and presented an indirect predictive effect on aggressive behaviors (effect of mobile phone addiction on insomnia: β=0.083, P<0.001; effect of insomnia on aggressive behaviors: β=0.999, P<0.001; effect of insomnia on self-perceived identity: β=-0.698, P<0.001). The contributions of mediating effects caused by insomnia were 21.61% and 31.02% to total effects.
Conclusions
Insomnia presents partial mediating effects among mobile phone addiction, aggressive behaviors and self-perceived identity among university students. Mobile phone addiction may directly affect aggressive behaviors and self-perceived identity, and indirectly affect aggressive behaviors and self-perceived identity via insomnia.
