1.The measure of chlamydia pneumoniae in the peripheral blood mononuclear cell of coronary heart disease

Objective To determine chlamydia pneumoniae(Cpn) DNA and inclusion bodies in peripheral blood mononuclear cell(PBMC) of coronary heart disease.Methods The PBMC were isolated by low permeate hemolysis method.The presence of Cpn-DNA and inclusion bodies were determined by nest-PCR and directed immunofluorescence(DIF) in 150 cases with CHD and 55 non CHD(control group,CG).Results Cpn DNA positive rates were 32.7%(49/150) in CHD and 1.8%(1/55) in CG,simultaneity inclusion bodies were detected 13.3%(20/150) in CHD and 1.8%(1/55) in CG.The positivity rates of Cpn-DNA and inclusion bodies in CHD groups were significantly higher than those in control group.Conclusion Chlamydia pneumoniae DNA and inclusion bodies are present in PBMC among the significant proportion of CHD patients.PBMC sample can be collected easily and trustworthily.
2.Effect of immunity function in condyloma acuminatum patients treated with differences of bacilli phlei injection after fulgurized

Objective To observe the curative effect of condyloma acuminatum patients treated with differences of bacilli phlei injection after fulfufized and the effect of immunity function. Methods We compared curative effect and peripheral blood T lymphocyte subpopulation in the treatment group( 55 cases, 8 weeks, prior and posttreatment with differences of bacilli phlei only) and the control group (50 cases, 8 weeks, prior and post-treatment with recombinant human interferon α-2b only). Results Compared with the control group (24.0 %/41.7 % ), the treatment group(47.3 %/11.5 % ) had advantage in the terms of cure rate and relapse rate( P<0.05 ). CD+3, CD+4 ,CD4+/CD+8 increased and CD+8 decreased in two groups after treatment. CD+3 , CD+4, CD+4/CDs+ were significantly increased in treatment group compared with the control group after treatment(P<0.05), but CD+8 had no obvious difference in two groups(P>0.05). Conclusion Differences of bacilli phlei injection can prevent relapse of condyloma acuminatum and increase immunity function of patients.
3.Pulmonary endarterectomy in 50 patients with chronic thromboembolic pulmonary hypertension: experience and doubts

Objective To evaluate the feasibility,validity and safety of pulmonary endarterectomy for patients with chronic thromboembolic pulmonary hypertension.Methods 50 patients undertook pulmonary endarterectomy operations were enrolled in this study.Of them,38 were males (76%),the average age was (43.35 ± 12.51) years,23 patients had deep venous thrombosis.Obvious pulmonary hypertension and hypoxemia were observed in all patients.Bilateral pulmonary endarterectomy was performed under cardiopulmonary bypass with profound hypothermic circulatory arrest.Preoperative systolic pulmonary artery pressure was(104.16 ± 16.95) mmHg,pulmonary vascular resistance was(129.68 ± 50.88) kPa · s · L-1,central venous pressure was (14.91 ± 4.88) mmHg,and cardiac output was (3.28 ± 1.04) L/min.Results The average time of cardiopulmonary bypass was(294.37 ± 94.01) min,aortic cross clamp time was(127.93 ± 35.57) min,circulatory arrest time was(34.30 ±21.74) min.Post-operative mechanical ventilation time was (97.24 ±70.53) hours,and the ICU stay was (9.52 ± 12.96) days.There were 4 patients that died after PEA surgery for postoperative residual pulmonary hypertension of reperfusion pulmonary edema.Post-operation,all patients had significant decrease in systolic pulmonary artery pressure (54.11 ± 16.86) mmHg and pulmonary vascular resistance(20.55 ± 15.17) kPa · s · L-1,and central venous pressure (9.00 ± 3.09) mmHg,and great improvement in cardiac output (5.75 ± 1.48) L/min.6-months follow-up showed that the cardiac function of 44 (95.7 %) cases returned to NYHA class Ⅰ or Ⅱ,with great improvement in computed tomography pulmonary angiography.All patients go back to normal work and physical exercise at 1-year follow-up.Conclusion Conclusions According the successful experience of surgery for CTEPH patients,pulmonary endarterectomy for patients with CTEPH results in significant pulmonary hemodynamic improvement,with favorable outcomes of heart and lung function in short and middle time follow-up.
4.Analysis on Medicine Using Regularity of Astragali Radix in Drug Standard of Ministry of Public Health of the People’s Republic of China-TCM Prescription Preparation

Objective To analyze the composing principles of Astragali Radix in Drug Standard of Ministry of Public Health of the People’s Republic of China-TCM Prescription Preparation;To provide references for clinical application and research and development of new medicine. Methods Traditional Chinese Medicine Inheritance Support System (V2.0) was used to establish database. Data mining methods, such as association rules, improved mutual information method, were used to analyze the diseases, syndrome rules, and common composing combinations of prescriptions containing Astragali Radix. Results Through analysis on the 446 cases of prescriptions containing Astragali Radix, 16 core medicine combinations and 18 diseases were discovered. Then, based on the 18 diseases, insomnia, consumptive disease and vertigo were chosen to make comparative analysis, the medicine using regularity and compatibility features of Astragali Radix in different diseases were disclosed. Conclusion Herbal medicines used frequently in prescriptions containing Astragali Radix could tonify qi and nourish blood. The medicine using is relatively concentrated, and the composition is clear. The composing principles reflect the compatibility features of Astragali Radix in treating different diseases, which are in accordance with the clinical application of multiple-efficacy Chinese medicine.
5.Restriction landmark genomic scanning for screening aberrant CpG methylations in prostate cancer.

OBJECTIVETo screen methylations of CpG islands in prostate cancer using restriction landmark genomic scanning (RLGS).
METHODSThe DNA was extracted from homogeneous cells captured by laser capture microdissection in 20 prostate cancer and 18 benign prostatic hyperplasia (BPH) tissues for scanning the CpG islands using RLGS. The methylation status of each CpG island was compared between the cancer and BPH samples to screen the genes involved in prostate cancer development. The screened genes were uploaded to DAVID database for GO analysis, and the genes with the most significant methylation were analyzed by pyrosequencing.
RESULTS AND CONCLUSIONAmong all the tested CpG islands, 10245 (37.2%) in prostate cancer and 8658 (30.3%) in BPH samples were found to be abnormally methylated, and >60% of the methylated CpG islands were in the promoter region. Compared with BPH samples, the prostate cancer samples showed differential methyation in 735 CpG islands, including 458 hepermethyated and 256 hypomethelated ones. Seven genes (DPYS, P16, APC, GSTP1, TMEM122, RARB, and ARHGAP20) in prostate cancer were identified to have distinct methylations. Bioinformatics analysis suggested that these genes were associated with several biomolecular and biological processes, and among them DPYS gene was involved in 13 GO anotated biologic functions, development of 50 diseases and 47 protein interactions. Pyrosequencing of 7 sites of the CPG island in DPYS gene showed a methylation frequency of 32.7%, suggesting the importance of DPYS gene in the carcinogenesis and progression of prostate cancer.

6.Clinical efficacy of transperitoneal verus retroperitoneal laparoscopic partial nephrectomy for renal tumors with R. E. N. A. L score over 7.

OBJECTIVETo compare the safety, feasibility and efficacy of transperitoneal and retroperitoneal laparoscopic partial nephrectomy (LPN) in the treatment of renal tumors with R. E. N. A. L score more than 7.
METHODSThe clinical data were collected from 62 patients undergoing transperitoneal LPN (32 cases) and retroperitoneal LPN (30 cases) for a complex renal mass (R.E.N.A.L. score≥7) between January 2012 and March 2014. The surgical and early postoperative outcomes and complications were analyzed to evaluate the efficacy of the treatments. The mean operative time, estimated blood loss, warm ischemia time, surgical complications, blood transfusion rate, tolerating regular diet time, postoperative hospital stay and surgical margin were compared between the two groups.
RESULTSThe operations were completed successfully in all cases except for 1 case in transperitoneal group and 3 in retroperitoneal group that required conversion to open surgery. No significant differences were found in age, body mass index, ASA score, Charlson comorbidity index, tumor size or R.E.N.A.L. nephrometry score (P>0.05), nor in estimated blood loss, warm ischemia time, intraoperative complication, blood transfusion rate or surgical margin between the two groups (P>0.05, respectively). The transperitoneal LPN group had a shorter mean operative time than retroperitoneal LPN group (210.4∓59.2 vs 252∓58.3 min, P<0.05) but showed longer tolerating regular diet time (47∓10 h vs 23∓6 h, P<0.05) and postoperative hospital stay time (8.4∓1.9 days vs 6.5∓1.6 days, P<0.05).
CONCLUSIONBoth transperitoneal LPN and retroperitoneal LPN are safe, feasible and effective for surgical management of complex localized tumors, but the transperitoneal procedure offers larger operative space with better exposure; the retroperitoneal procedure better promotes postoperative recovery of the patients.

7.Hair Growth Promoting Effects of 650 nm Red Light Stimulation on Human Hair Follicles and Study of Its Mechanisms via RNA Sequencing Transcriptome Analysis

Background:
Androgenetic alopecia (AGA) leads to thinning of scalp hair and affects 60%~70% of the adult population worldwide. Developing more effective treatments and studying its mechanism are of great significance. Previous clinical studies have revealed that hair growth is stimulated by 650-nm red light.
Objective:
This study aimed to explore the effect and mechanism of 650-nm red light on the treatment of AGA by using ex vivo hair follicle culture.
Methods:
Human hair follicles were obtained from hair transplant patients with AGA. Hair follicles were cultured in Williams E medium and treated with or without 650-nm red light.Real-time RT-PCR and immunofluorescence staining were used to detect the expression level of genes and proteins in hair follicles, respectively. RNA-sequencing analysis was carried out to reveal the distinct gene signatures upon 650 nm treatment.
Results:
Low-level 650 nm red light promoted the proliferation of human hair follicles in the experimental cultured-tissue model. Consistently, 650 nm red light significantly delayed the transition of hair cycle from anagen to catagen in vitro. RNA-seq analysis and gene clustering for the differentially expressed genes suggests that leukocyte transendothelial migration, metabolism, adherens junction and other biological process maybe involved in stimulation of hair follicles by 650-nm red light treatment.
Conclusion
The effect of 650-nm red light on ex vivo hair follicles and the transcriptome set which implicates the role of red light in promoting hair growth and reversing of miniaturization process of AGA were identified.
8.Research progress on sex-hormone-like effects of epimedium

Epimedii folium is a commonly used Traditional Chinese Medicine (TCM) for warming the kidney and strengthening the yang qi. It has androgen-estrogen-like effect. It can not only directly act on sexual organs to regulate hormone levels, but also exert sex-hormone-like effect through hypothalamus-pituitary-gonadal axis. Its regulation of hormone levels is similar to that of plant hormones. At present, Epimedii folium is commonly used with other TCMs to treat diseases caused by sex-hormone deficiency, such as male spermatopenia, asthenospermia, benign prostatic hyperplasia, functional erectile dysfunction, female premature ovarian failure, perimenopausal syndrome, dysfunctional infertility during ovulation, hyperandrogenemia of PCOS patients, etc.
