1.Secondary structure of target RNA impacts on RNA interference efficiency

Objective To study the effects of the RNA secondary structure on the RNA interference efficacy and to optimize the RNAi target sequence selection.Methods N-Ras mRNA in K562 human chronic erythro-leukemia cell line was selected as the target for RNAi experiments.Four siRNAs were designed aiming at secondary structure region and none-secondary structure region separately.The N-Ras expression change and the cell growth was tested by semi-quantitative reverse transcription polymerase chain reaction(RT-PCR),light microscopy,MTT,and flow cytometry.Results Knock-down efficiency was recorded in all of the 4 experiment groups,but the degree varied a lot.The siRNAs targeting none-secondary structure region presented higher silencing efficiency than those targeting secondary structure region.Conclusion The secondary structure of mRNA closely relate to the RNA interference efficiency,which should be considered in the processing of the target sequence selection.

3.Rhinoplasty with autogenous nasal septal cartilage for saddle nose caused by nasal trauma.

OBJECTIVE:
To observe the efficacy of rhinoplasty with autogenous nasal septal cartilage for saddle nose caused by nasal bone and nasal septum fracture after trauma.
METHOD:
During the treatment process of 30 cases of nasal trauma, simple nasal bone fracture reposition and nasal septum correction was done, and rhinoplasty was done using the nasal septal cartilage. All the patients were followed up to evaluate subsequent effect.
RESULT:
All the patients were satisfied with the result of one stage repairment. After following-up for 3-42 months, 28 cases (93.3%) were of long-term satisfaction.
CONCLUSION
Rhinoplasty with autogenous nasal septal cartilage for saddle nose was an effective method to repair the nasal trauma.

4.The significance of PPARγexpression in lung tissue of rats with hypoxic pulmonary hypertension

Objective To investigate the significance of peroxisome proliferator-activated receptor (PPAR)γexpression in the lung tissue of rats with chronic hypoxic pulmonary hypertension (HPAH). Methods Forty male Sprague-Dawley rats were randomly divided into four groups (n=10 for each group):normal control group (NC), hypoxia control group-one-week (HC-1w), hypoxia control group-two-week (HC-2w) and hypoxia control group-three-week (HC-3w). Normal control group was raised under normal oxygen condition in ventilated animal cage for three weeks. The other HC groups were placed in a low oxygen chamber (O2 concentration of 10%) from 9:00 AM-5:00 PM (8 h/d) everyday by one week, two weeks and three weeks. The mean pulmonary arterial pressure (mPAP), right ventricular systolic pressure (RVSP) were detected. The index of right ventricular hypertrophy RV/(LV+S) was measured by dissecting rat heart. The morphological changes of the small pul-monary arteries were observed by HE staining, and the percentage of vascular wall thickness (WT%) was calculated. The ex-pression level of PPARγprotein was detected by Westren blot assay. Results The mPAP, RVSP and RV/(LV+S) were sig-nificantly higher in HC groups than those of NC group (P<0.05). The morphology of pulmonary arteries showed vessel wall thickening and vessel lumina stenosis in HC groups compared with that of NC group. The PPARγexpression in lung tissue was significantly lower in HC groups than that of NC group, and the downward trend was more obvious with the extension of time. Conclusion PPARγplays an important role in the occurrence and development of chronic hypoxic pulmonary hyper-tension.
5.Clinical Observation on 50 Cases Hyperplasis of Mammary Glands Treated with Self-made RuPi Decoction

Objective To observe the clinical efficacy of RuPi decoction in the treatment of breast hyperplasia. Methods 100 cases with breast hyperplasia were randomly recruited into a treatment group, which was treated with the RuPi decoction, and a control group, which was treated with RuNing tablet. Three months constituted one course of treatment for both groups. Observe the clinic curative effects, and improvement of major symptoms and signs. Results The total effective rate of the treatment group and control group was 96% and 84% respectively, showing significant difference between the tow groups(X2=5.01, P<0.05). Conclusion Modified RuPi decoction was effective in the treatment of Breast Hyperplasia.

7.Comparison of the Therapeutic Effect of Ultrasound Guided and Freehand Botulinum Toxin Type of the Treatment in Children with Cerebral Palsy

Objective To study the comparison of the therapeutic effect of Ultrasound guided and freehand botulinum toxin type of the treatment in children with cerebral palsy.Methods From March 2014 to September 2016,98 cases of children with cerebral palsy in our hospital by injection of botulinum toxin were treated.According to the way of injection,the children were divided into ultrasound group and free group(n =49).They were respectively gave botulinum toxin injection by guided Ultrasound guided and freehand ultrasound injection of hotulinum toxin injection.The amount of two groups of children was 3U/kg.Children muscle spasm degree of before and after treatment was assessed by modified Ashworth spasm scale.And exercise capacity of children before and after treatment was assessed by the gross motor function rating scale.The follow-up time was 3 months,the clinical curative effect of two groups of children was observed.Results The total efficiency of ultrasound guided group (98.0%) was significantly higher than the free group (79.6%),with statistically significant (P < 0.05).Both groups before and after treatment in children with Ashworth score and GMFM score were significantly improved,with statistical significance (P < 0.05).Ashworth score and GMFM scores of ultrasound guided group were significantly better than the free group,with statistically significant (P < 0.05).Conclusion Botulinum toxin injection in the treatment of cerebral palsy children can effectively solve the spastic problem.Treatment of children with cerebral palsy by injecting botulinum toxin with ultrasound guided injection of botulinurn toxin is better than manual injection of botulinum toxin.

10.Level of Plasma Stromal Derived Factor-1 in Essential Hypertension Patient and Effect of Enalapril on It

ObjectiveTo investigate the level of plasma stromal cell-derived factor-1 (SDF-1) in essential hypertension and the effect of angiotensin-converting enzyme inhibitor Enalapril on it. MethodsThe levels of plasma SDF-1 in 34 patients with hypertension and 28 normal healthy people were detected with ELISA. Hypertension patients were detected again after 2-week Enalapril treatment. ResultsThe level of SDF-1 in hypertension patients was (4.211±1.132) mmol/L, higher than control (3.568±0.936) mmol/L (P<0.05), and became (4.950±1.050) mmol/L after treatment, higher than pre-treatment (P<0.05). ConclusionThe level of SDF-1 increases in patients with hypertension, and Enalapril can further increase the level.
