2.Correlation of p53 expression with triple negative breast cancer and its prognosis:a meta analysis

Objective To evaluate the expression of p53 in triple negative breast cancer( TNBC) and its impact on the prognosis of TNBC systematically. Methods Documents published on PubMed, Medline,CNKI,CNKI,Wanfang data,and VIP data were screened for relative researches. Two reviewers filtered,extracted and assessed the literatures according to the inclusion criteria and exclusion criteria. The meta analysis was conducted by using RevMan5. 0 software. Result A total 13 trails were included,con-taining 11 Chinese documents and 2 English documents. The meta analysis showed that p53 is significantly expressed in TNBC[OR=2. 02,95%CI(1. 28,3. 19)]. TNBC patients with positive p53 expression had higher rates in lymph node metastasis[OR=2. 02,95%CI(1. 28,3. 19)],recurrence[OR=3. 39,95%CI(1.45,7. 90)],metastasis[OR =2. 65(1. 36,5. 18)],disease-free survival[OR =0. 29(0. 18, 0. 47)]and overall survival[OR=0. 29(0. 18,0. 47)]than patients with negative p53 expression. Con-clusion p53 is overexpressed in TNBC,which plays a certain role in guiding the clinical prognosis of TN-BC. It can be regarded as an independent factor to evaluate the prognosis of TNBC,and a therapeutic tar-get in further studies.
3.Determination of camphor,menthol,synthetic bornel and the wintergreen oil in Shexiang Zhuanggu Plaster by GC

AIM:To study the method of determing camphor, menthol, synthetic bornel and the wintergreen oil contents in Shexiang Zhunggu Plaster. METHODS: Gas phase chromatogram was used to determine contents of camphor, menthol, synthetic bornel and the wintergreen oil in Shexiang Zhuanggu Plaster. The chromatogram condition consisted of the capillary vessel column(PEG-20M, 30 m?0.32 mm?0.25 ?m). The temperature programing rose from 100 ?C to 130 ?C by 5 ?C /minute, then 30 ?C /minute to 200 ?C , lasting 2 minutes in the end. The temperature of the entrance of the capillary vessel column: at 220 ?C , temperature of the detector was 240 ?C ; N_2: 5 mL/minute, H_2: 45 mL/minute, Air: 450 mL/minute; sample quantity was 0.2 ?L. RESULTS: Under certain chromatogram condition, there was linear relation among the camphor, menthol, synthetic bornel and the wintergreen oil and the peak area in the scope of 0.027 38 mg/mL-0.438 08 mg/mL, 0.041 15 mg/mL-0.658 4 mg/mL, 0.033 38 mg/mL-0.534 1 mg/mL, 0.042 15 mg/mL-0.674 4 mg/mL, respectively. The average recoveries were 98.37%, 99.76%, 98.80% and 97.61%, respectively. RSD were 1.7%, 1.8%, 0.96% and 1.5%, respectively. CONCLUSION: This experiment provides an accurate and reliable method to control the quality of Shexiang Zhuanggu Plaster.

Staphylococcus aureus is one of the common cause of respiratory tract infections in children.Methi-cillin -resistant staphylococcusaureus was reported in 1 960s,then the resistance of other drugs were found in recent years,which leading to many problems in the clinical treatment.Clinician have to master the resistance trend and mechanism of this bacteria and decrease the speed of resistance as much as possible.
6.Fingerprint of Zedoary turmeric oil by GC

AIM: To establish the method of fingerprinting analysis on Zedoary turmeric oil by GC.Zedoary turmeric oil for the quality control of reference. METHODS: GC method was used to determine the fingerprint and its similarity of Zedoary turmeric oil calculated on Excel and cluster analysis. RESULTS: Seventeen samples of Zedoary turmeric oil indicated that there was a little similarity among them.Based on the cluster analysis,all the Zedoary turmeric oils could be divided into two categories,self-made and market purchasing oil,the latter oils purchased from Liaoning province and Jiangsu province varied considerably.Every category had a common fingerprint mode with clear resemblance. CONCLUSION: When finding the difference between the self-made and the market purchasing Zedoary turmeric oil,quality control requires serious consideration.
7.Simultaneous determination of 11 opiates in hair by liquid chromatography-tandem mass spectrometry.

The paper reports the establishment of a liquid chromatography-tandem mass spectrometry (LC-MS/MS) for simultaneous analysis of 11 opiates in hair samples, and the study of presence of opiates in the hair of active heroin addicts. About 20 mg of decontaminated and pulverized hair sample was hydrolyzed with buffer solution for 30 min, in the presence of morphine-d3 and acetylmorphine-d6 used as internal standards, and then extracted with the mixture of dichlormethane and isopropanol, separated by the Allure PFP propyl column with a mobile phase consisting of acetonitrile and 20 mmol L(-1) ammonium acetate buffer, and then analyzed by LC-MS/MS. Multiple reaction monitoring (MRM) mode was used to analyze 11 opiates. Eleven opiates showed a fairly good linearity over the corresponding range (r > 0.996 0). The detection limits were less than 0.05 ng mg(-1). The recoveries were between 47.2% and 110%, and the deviations of intra- and inter-day precision were less than 14%. Heroin, acetylmorphine, morphine, codeine, acetylcodeine and hydrocodone were detected in hair samples of 21 herion addicts. The developed method shows high sensitivity and selectivity, and is suitable for the simultaneous analysis of 11 opiates in hair samples and identify legal and illegal use of opiates.
8.Effect of ketamine on spinal astrocytes in mice tolerant to morphine

Objective Recent studies have shown that activation of spinal astrocytes (ASTs) may be involved in the development of morphine tolerance. The purpose of this study was to investigate the effect of ketamine (K) on spinal ASTs in mice tolerant to morphine (M) .Methods Thirty Kun-Ming mice of both sexes weighing 18-22 g were randomly divided into 5 groups of six animals each : (A) control group received only subcutaneous (s.c.) and intraperitoneal (i.p.) normal saline (NS); (B) chronic M-tolerance group received M 10 mg?kg-1 s.c. followed after 30 min by NS 10 ml?kg-1 i.p. twice a day (at 8:00 and 17:00) for 9 days;(C), (D), (E) K group received M 10 mg?kg-1 s.c. followed after 30 min by K 5 mg? kg-1(C), 10 mg?kg-1 (D) or 20 mg?kg-1 (E) i.p. twice a day for 9 days. Pain threshold was estimated by measuring paw withdrawal response to Von Frey filament stimulation every other day (1st, 3rd, 5th, 7th, 9th) In after second administration of drugs. The percentage of maximal possible effect (MPE% ) was calculated : MPE% = [ (test group PWTV - control group PWTV) / (15 - control group PWTV)] ? 100% (PWTV = paw withdrawal threshold value). On the 9th day after pain threshold was measured the animals were sacrificed and lumbosacral segment of spinal cord was removed. The changes in spinal ASTs were detected by immunohistochemistry. The average areas of GFAP immuno-reactive cells in the dorsal horn were measured to show the degree of spinal AST activation. Results 1. MPE% was 0 at all time points in group A. In group B MPE% was 42.8% on the 1st and 3rd day and gradually decreasing on the 5th and 7th day and became 0 on the 9th day signifying full development of morphine tolerance. In group C the change in MPE% was almost the same as in group B. In group D and E MPE % tended to decrease but was still above 30% at all time points signifying that ketamine 10 and 20 mg?kg-1 could partly antagonize the development of morphine tolerance. 2. 9. Hydrogen-containing preservation solution relieves oxidative stress and inflammatory damage of rat donor heart

Objective: To study the effect of hydrogen-containing preservation solution against oxidative stress and inflammatory damage of rat donor heart. Methods: Thirty-two SD rats were evenly randomized into four groups(n = 8) : control group (the hearts were protected by HTK solution), H1 group (hydrogen concentration was about 0.2 mmol/L in the HTK solution), H2 group (hydrogen concentration was about 0.4 mmol/L in the HTK solution) and H3 group (hydrogen concentration was about 0.8 mmol/L in the HTK solution). The rat hearts were harvested in all groups and were mounted on the Langendorff apparatus to estimate baseline hemodynamic values. All the hearts underwent hypothermic (4°C) storage for 6 h in the corresponding cardioprotective solutions. Then, superoxide dismutase (SOD) activities and malondiadehyd (MDA) contents in myocardium tissues were measured after reperfusion by xanthine oxidase method and TBA method, respectively. The levels of 8-hydroxydeoxygunosine, TNF-α, and IL-6 in the myocardium tissues were measured by enzyme-linked immunosorbent assay (ELISA). Results: The MDA levels in H1, H2, and H3 groups were lower than that in the control group 6 h after preservation (P<0.01 or P<0.05), and the SOD activities in H2, H3 groups were higher than that in the control group (P<0.01 or P<0.05). The levels of 8-hydroxydeoxygunosine, TNF-α, and IL-6 in the control group were higher than those in H1, H2, and H3 groups (P<0.01 or P<0.05). Conclusion: Hydrogen-containing preservation solution can relieve the oxidative damage and reduce production of inflammation factors during preservation of rat donor heart.
10.Evolutionary Trace and Molecular Dynamics Simulation of the Beta2 Adrenergic Receptor Mutation-induced Activation

Beta2 adrenergic receptor (β2 AR) is one member of G protein coupled receptors (GPCRs), which is a key pharmaceutical target in the treatment of asthma. Evolutionary trace (ET) method was employed to analyze AR sequences and 44 conserved residues were identified. Then molecular dynamics (MD) simulation of the β2 wild-type receptor, D130N active mutant and D79N inactive mutant were carried out and tried to explore the structural/dynamic features characterizing functionally different states of the receptor,by means of investigating ET identified conserved basic residues in the wild-type receptor and its two mutants. Particularly, it was found that the departing of D130 from R131 of DRY motif and approaching to K149 are highly correlated with the receptor activation,and the movement of helix 2, 4 and 6 upon receptor activation is inferred from the observation that R151 and K270 interact with other residues in the receptor active state on the basis of little change of the side chain orientations. The results might provide further insights into the activating mechanism of β2 AR mutants, as well as the molecular bases of the diseases induced by the mutations of the receptor.
