Objective:To perform a preliminary proteomic analysis of mouse ovaries and to study theprotein's function in mouse ovary.Methods:The two-dimensional gel electrophoresis(2-DE)and matrixassisted laser desorption/ionization-time of flight mass spectrometry( MALDI-TOF MS)were used to ana-lyze mouse ovarian proteome.A 12.5% sodium dodecyl sulfate(SDS)reference gel was generated byimmobilized pH gradient isoelectric focusing of mouse ovary proteins in a non-linear gradient(pH 3-10).And GRP78 was selected to perform with immunohistochemistry within mouse ovaries.Results:Based onpeptide mass fingerprinting,52 proteins were identified and classified into seven functional groups:Cell/organism defense and antioxidant,cell signaling/communications proteins,cell structure/motility pro-teins,metabolism proteins,RNA synthesis processing,Protein synthesis and processing,and unclassifiedproteins.The immunoreactivity of GRP78 was detected in GCs in the follicular,and with during GCs Lu-teinizing in the menstrual cycle,the protein expression(brown)increased continually and came to ahead when ovulation happened.Conclusion:This work provides a first step toward the establishment of asystematic ovary protein database and stands as a valuable resource for molecular analyses of normal andpathologic conditions affecting mouse ovaries.

As a most common algal toxin in eutrophic freshwater body, microcystins can be produced by the bloom-forming Cyanophyta microcystis, it has become a potential hazardous substance in aquatic environments for its toxic, distribution and stability. With the increasing recognition of microcystins, China had added microcystin-LR into the related water quality standards. It is very important to detect and control the microcystins in the aquatic environment. This paper systematically introduced the current situation of researches about the determination and pollution control technique of microcystins in drinking water at home and abroad in resent years and then some issues worth of paying more attention to and doing further study in pollution control in future were presented.

BACKGROUND: Vascular endothelial growth factor receptor plays an important role in the blood vessel growth and prognosis of many solid tumors. However, there are few reports regarding the relationship between vascular growth factor receptor and prognosis of lung cancer.OBJECTIVE: To probe into the relationship between the expressions of vascular endothelial growth factor receptor F1t1, KDR and the metastasis and prognosis of lung cancer.DESIGN: Single sample study based on pathological samples SETTING: Department of pathology in a university.MATERIALS: Totally 75 lung cancer specimens were randomly selected from the specimens removed by Department of Thoracic Surgery of Qingdao University from January 1985 to December 1990. The study was conducted in the Department of Pathology of Qingdao University from September to November 2002.METHODS: Immunohistochemical method PowerVision TM-9000 (PV-9000)was used to assay the Flt1 and KDR expression in 75 lung cancer specimens.MAIN OUTCOME MEASURES: Expression of vascular endothelial growth factor F1t1 and KDR in lung cancer and its relationship with age, sex,pathological type, histological grading, tumor size, lymph node metastasis and prognosis, as well as the expression of F1t1, KDR in tumor cells and fibroblasts.RESULTS: There is extensive expression of F1t1 and KDR in lung cancer tissues, especially in cytoplasm and cell membrane of tumor cells, also in cytoplasm of fibroblasts and vascular endothelial cells. The positive rates of F1t1 and KDR in tumor cells are much higher than those iu matrix fibroblasts (x2 = 6.07,5.88, P ＜ 0.05). There is no difference in the positive rates of these two receptors in tumor cells and fibroblast between cases with different age, sex, pathological types, pathological grading( P＞0.05). There is significant difference in the positive expression rates of Flt1 and KDR in tumor cells among three groups with different tumor size(x2 = 10. 35, 7.29, P ＜ 0.05)while there is no difference in fibroblasts(x2=2.86,2.56, P ＞ 0. 05). There is also significant difference in positive rates of F1t1 and KDR in tumor cells and fibroblasts between groups with and without lymph node metastasis (x2 =4.72 -9.32, P＜0.05) and among three groups with different survival time(x2 = 8.81 - 19. 19, P ＜ 0. 05) . The expression of Flt1 and KDR in tumor cells has greatly positive correlation( r =0.44, P＜0.01).CONCLUSION: The expression of Flt1 and KDR in tumor cells is much higher than that in matrix fibroblasts which suggests that the growth of lung cancer is mainly dependent on autocrine metabolism. The expressions of these two receptors in tumor cells relate to lymph node metastasis, survival time after operation and cooperate with each other. It suggests that it is of great value to assess the metastasis and prognosis of lung cancer by jointly testing the expression of F1t1 and KDR.

Objective To investigate the effects of erythropoietin(EPO) on extracellular matrix remodeling and the expressions of matrix metalloproteinase(MMPs) and tissue inhibitor of metalloproteinase-4(TIMP-4) after myocardial infarction in rats.Methods Eighty male SD rats were randomly divided into four groups:sham operation(SH) group,sham operation and drug(SHE) group,myocardial infarction(MI) group,and myocardial inferct and erythropoietin(MIE) group.Myocardial infarction in MI group and MIE group was reproduced by ligation of a branch of left anterior descending coronary artery.Hematocrit,echocardiography,capillary density,infarct area,fibrosis and collagen volume fraction were tested,and MMP-2,MMP-9 and TIMP-4 were measured by immunohistochemistry at 1-week and 6-week after myocardial infraction.Results One week after myocardial infraction,the protein expressions of MMP-2 and MMP-9 in MI and MIE groups were significantly higher than that in SH and SHE group(P0.05).Six weeks after myocardial infraction,echocardiography revealed that the left ventricular end-diastolic dimension(LVDd),left ventricular end-systolic dimension(LVDs),left ventricular end-diastolic volume(LVEDV) and left ventricular end-systolic volume(LVESV) were significantly increased,and left ventricular ejection fraction(LVEF) and left ventricular fractional shortening(LVFS) were significantly decreased in MI and MIE groups compared with those in SH and SHE groups(P

Objective To explore the correlation of positive psychological capital and time management disposition in medical college students.Methods A total of 880 students from medical university were surveyed by using positive psychological capital scale and time management disposition scale.Results For gender variables,the scores of hope and optimism of males were significantly lower than those of females(P ＜ 0.05);there was no significant difference in the scores of time value sense,time monitoring sense,time efficacy sense,self-confidence and toughness between males and females (P ＞ 0.05).For birthplace variables,the rural students' scores of time value sense,hope and optimism were significantly higher than those of urban students (P ＜ 0.05);there was no significant difference in the scores of time monitoring sense,time efficacy sense,self-confidence and toughness between rural students and urban students (P ＞ 0.05).For grade variables,the score of time value sense of seniors was significantly higher than that of freshman(P ＜ 0.05);the score of time value sense of the fifth grader students was significantly higher than that of freshmen and sophomore (P ＜ 0.05);the score of time efficacy sense of the fifth grader students was significantly higher than that of sophomore (P ＜ 0.05);the scores of hope and optimism of the fifth grader students were significantly higher than those of sophomore (P ＜ 0.05);the score of optimism of freshman was significantly higher than that of sophomore(P ＜ 0.05);there was no significant difference in the scores of another dimensions in the other grades (P ＞ 0.05).Positive psychological capital and its dimensions were positively correlated with time management disposition and its dimensions (P ＜ 0.01).Positive psychological capital has good positive predictive ability on the four dimensions of time management disposition.Conclusion There is a positive correlation between positive psychological capital and time management disposition.The improvement of the level of psychological capital of college students can help to improve the ability of time management.

Usher syndrome (USH) is an autosomal recessive hereditary disease,characterized as retinitis pigmentosa and deafness.According to the severity of hearing loss,presence or absence of vestibular dysfunction,Usher syndrome is divided into 3 clinical subtypes..USH1,USH2 and USH3.Due to the genetically heterogeneous,it is important and valuable to find out the gene mutations in USH patients,which will be helpful to prenatal diagnosis,early intervention and gene therapy.Till now,the following 13 USH-related chromosomal loci were reported in the literature:USH1B,USH1C,USH1D (CDH23 gene),USH1F (PCDH15 gene),USH1G (SANS gene),USH1E,USH1H,USH1J and USH1K,USH2A,USH2C,USH2D and USH3 (CLRN1 gene).Ten out of all 13 loci have been located and identified.But more mechanisms should be further investigated,such as the relationship between the locus of gene mutations and clinical symptoms,how the modified protein structures and functions trigger clinical symptoms.
