1.Sources of sugar in flowers and fruits of Macrocarpium officinale during fruiting

Object To study the source of sugar in flowers and fruits of Macrocarpium officinalis (Sieb et Zucc.) Nakai during fruiting.Methods The sugar contents were analyzed by HPLC.Results The contents of glucose, fructose and total sugar decreased slowly in the third or the forth branch after the barks were cyclically cut and there were significant differences between test group and control group, the changes of sugar contents in the first or the second branch were similar to control group.Conclusion These sugars in flowers and fruits were provided by the first or the second branch during fruiting.
2.Regulation of dry substance and active components accumulation in fruit of Forsythia suspensa

Objective To study the regulation of dry substance and active components accumulation in the fruit of Forsythia suspensa. Methods The dry substance of the fruit was analyzed by the method of hundred grain mass. The contents of the active components in leaves, shucks, and seeds were analyzed by HPLC. Results The period for F. suspensa to accumulate the dry substance was mainly May and July. During the whole growth phase, the active components, such as forsythiaside, forsythin, and rutin in different parts, were decreasing. From the last ten-day of August to the first ten-day of September, the accumulation of the dry substance came to stability and the contents of active components in shucks and seed were stable. The contents of active components in shucks decreased rapidly from September to October, and then came to stability and the contents in shucks is the least during the last ten-day of October. So the contents of active components in younger F. suspensa are higher than that in elder one, and the proportion among forsythiaside, forsythins, and lution is also significantly different from elder seeds. Conclusion It is more preferable to pick younger F. suspensa from the last ten-day of August to the first ten-day of September and to pick elder F. suspensa during last ten-day of October.
4.Physiological and Biochemical Changes During Embryo After-Ripening of American Ginseng(Panax quinquefolius)and Effect of Hormones on Such Changes

In natural condition, two years were needed for Panax quinquefolius seeds to break dormancy. In the first year, the embryo developed slowly, and the activities of amylase and peroxidase were low. In the second year the embryo developed rapidly, and the activities of both enzymes also increased gradually. But with the treatment of hormones enzymatic activ ities accelarated remarkably and speeded up the decomposition of stored nutrients in endos perm, so that starch and non-reducing sugar decreased while reducing sugar incrcased rapidly, resulting in earlier germination.
6.Fusion Immunotoxin Anti-HER-2-scFv-SEC2 Expressed in E.coli With an Improved Expression Vector pASK75-EX: Its Construction and Functioning

A tumor-targeting recombinant fusion immunotoxin B-L-SEC2 was constructed by fusing staphylococcal enterotoxin C2 (SEC2) and an anti-HER-2 single-chain Fv B1 through a peptide linker, and expressed in E. coli strain BL21 (DE3) with an improved expression vector pASK75-EX as inclusion body. The denatured inclusion body was purified with Ni-NTA chelate agarose, and then re-natured by dialysis. FACS and MTT assays indicated that the re-natured fusion immunotoxin B-L-SEC2 could target the HER-2 over-expressing breast tumor cell SK-Br-3 in vitro, and inhibit the growth of SK-Br-3.
7.Multiplication of virus-free seedlings of Rehmannia glutinosa CV. 85-5 in vitro

Object To develop a method of multiplying virus free clonal seedlings of CV.85 5 of Rehmannia glutinosa (Gaert.) Libosch. ex Fisch. et Mey.. Methods The leaf cut segments with or without tips of plantlets were grown in different media (mg/L) (1.MS+BA 0.5; 2 MS+BA 0.5+NAA 0.02;3 MS+BA 0.5+NAA 0.02+GA 3 0.1; 4 1/2 MS+BA 0.1; 5 1/2 MS+BA 0.1+GA 3 0.1; 6.1/2 MS+GA 3 0.1) to select a suitable medium, and the height and the numbers of newly formed leaves and roots were recorded 30 days later. To screen out a favorable condition, similar segments were transferred to No.2 medium at different temperatures in different illuminations, and the height together with the leaf number and the fresh weight of the plantlets was recorded.Results In the former three media, the segments with tips were flourishing and one to three axillary buds were formed at its basal stem; while in those without tips almost every axillary bud developed with the main tip length 0.5~1.5 cm. In the later three media, thd elongation of the segments was usually overstimulated, and the slim plantlets with fewer leaves and more roots formed. When the segments cultured in No.2 medium, all records at 28 ℃ were higher than those at 23 ℃ in the same illuminations. But the leaf size and the height of the plantlets were inversely proportional to the intensity of illumination at the same temperature. In addition, antagonism between BA and GA 3 was found.Conclusion No.2 medium is more suitable for the multiplication of the virus free seedlings of 85 5 when cultured at 28 ℃ in the illumination of 1 000~2 000 lx.
8.Pharmacodynamic differences between hangmaidong and chuanmaidong based on metabonomics.

To evaluate the differences of Ophiopogonjaponicus from different cultivations, the metabolomics based method was conducted to compare the effects of Hangmaidong and Chuanmaidong (Chinese name) on plasma endogenous metabolites of normal rats. Data were collected by ultra performance liquid chromatography quadrupole time of flight mass spectrometry (UPLC-Q-TOF/MS), and were analyzed by multivariate statistical method, such as Principal Component Analysis and Orthogonal Signal Correction Partial Least Square Discriminant Analysis. Results revealed that the plasma metabolites profiling of low and middle dose group of Chuanmaidong were similar to the control group, but different from the high dose group obviously. Meanwhile the high, middle and low dose groups of Hangmaidong were different from control group notably, and the difference is dose dependent. Lysophosphatidylcholines, the possible endogenous metabolites which contribute to the classification most significantly, are closely related to cardiovascular system diseases. Compared with the group of Chuanmaidong, Hangmaidong has greater impact on the plasma metabolic profiling of normal rats. Hangmaidong and Chuanmaidong showed significant differences pharmacodynamically.
9.Clinical efficacy of interventional therapy on children with congenital heart diseases

Objective To summarize clinical outcomes of interventional therapy on children with common congenital heart diseases(CHD).Methods A retrospective study was conducted.One hundred and fourteen patients with CHD were selected as our subjects,who underwent catheter interventional therapy in the Second Affiliated Hospital of Zhengzhou University from Jan.2004 and Dec.2012.The size of occluder was chose according to intraoperative echocardiography or cardiac imaging measurements,and occluder was released under assisted monitoring by subtraction angiography or cardiac ultrasound.Results There are all together 112 patients got the therapy successfully,2 cases failed(occluder detachment),and the success rate of operation was 98.2%.After the success of interventional,echocardiography examination showed that 11 cases were with star point across shunt,but the function of the around valve was not affected.One months after operation,echocardiography examination showed star point across shunt of 11 cases were disappeared,and no occluder was shifted as well as no thrombosis formed.Three months after operation,chest radiograph showed pulmonary congestion decreases and heart shadow was shrink.Thirty-eight cases were with three tricuspid regurgitation before operation and 32 cases were without reflux at 3 months after operation,and 6 cases relieved significantly.The patients were followed up for 6 months or 3 years,activity endurance was significantly improved than that before operation.No occluder was shiftand hemolysis and arrhythmia occurred.Meanwhile,No thrombosis or embolism occurred.Conclusion Interventional treatment for children with congenital heart disease is proved as a safe,effective methods and it have broad prospects in clinical application.
10.Endovascular repair of Stanford type B aortic dissection: initial experience in 85 cases

Objective To explore the technology and curative effect of thoracic endovascular aortic repair (TEVAR) for Stanford type B aortic dissection.Methods The clinical data of 85 patients with Stanford type B aortic dissection,who were admitted to authors' hospital during the period from January 2010 to April 2016 to receive TEVAR,were retrospectively analyzed.Conventional left brachial artery puncture and straight incision of right femoral artery were employed in all 85 patients,and DSA of ascending aorta was performed to find out the position of rupture,the position of the true and false lumens,and their relationship with the vascular openings of important organs.Endovascular covered stent was implanted to seal off the primary rupture;reexamination of ascending aorta angiography was adopted to check the sealing-off condition of the proximal rupture and the changes of blood flow in the aortic branches as well as in the true and false lumens.Results Successful TEVAR was accomplished in 84 patients.One patient died of sudden rupture of aortic dissection during preoperative anaesthesia.The technical success rate was 100%.In 9 patients the covered stent partially overlapped the left subclavian artery,in one patient the left subclavian artery “chimney” stem completely obstructed both the left common carotid artery and the left subclavian artery,and bypass surgery between left common carotid artery and left subclavian artery was carried out in 2 patients.After the treatment,internal leakage of type Ⅰ was detected in 2 patients.No death occurred during hospitalization period.After the surgery the patients were followed up for 3 months to 3 years,and all patients survived.New rupture at the distal site occurred in 2 patients.Conclusion For the treatment of Stanford type B aortic dissection,TEVAR is safe and effective.Strict observance of surgical indications,careful operative manipulation,and strengthening postoperative management after discharge from hospital are the key points to ensure a successful surgery as well as to improve the long-term survival rate.
