1.Optimizing Preparation Technics of Sodium Carboxylmethylcellulose Mucilage by Orthogonal Test

OBJECTIVE:To study the preparation technique for CMC-Na mucilage.METHODS:The factors including kinds of solvents,the prescription dosage of CMC-Na and its adding way,the preferable prescription of and preparation for the above were inspected using orthogonal test of3factors and levels.RESULTS:The preferable technique was that2.5%CMC-Na was spreaded into the hot purified water solution under its stirring.CONCLUSIONS:The prescription and preparation for CMC-Na mucilage selected by the orthogonal test is consistent with Chinese Hospital's Preparation Standards.
2.Clinical significance of thrombospondin 4 expression in gastric carcinoma

Objective The aim of this study was to measure quantitatively the TSP-4 mRNA expression and its significance in gastric carcinoma was evaluated by correlating its expression with clinicopathological features, microvessel density(MVD) and MMP-9.Methods Eighty-two patients with gastric carcinoma were recruited in this study. TSP-4 mRNA expression in gastric carcinoma and adjacent tissue was detected by real-time PCR. CD34 and MMP-9 protein expression were examined by immunohistochemistry. MVD was determined according to CD34-positive tubular structures. Results The expression level of TSP-4mRNA in gastric carcinoma was obviously higher than those of adjacent tissue(P=0.03) and it was associated with tumor size, histological type, lymph node metastasis, MVD and MMP-9(P=0.002, P=0.031, P=0.014,r=0.67 P<0.01, P=0.008),but not sex, age and depth of invasion. (P=0.53,P=0.57,P=0.15).Conclusion TSP-4 may play an important role in occurrence and progression of gastric carcinoma and that the effect of TSP-4 on tumor growth and metastasis may be exerted through regulation of angiogenesis and MMP-9 expression in gastric carcinoma.
3.The protective effect of EPO on Alzheimer disease rat learning and memory injury and its effect on Bcl-xl protein expression in hippocampal CA1 subregion

Objective To explore the protecting mechanism of erythropoietin (EPO) on learning and memory of Alzheimer disease (AD) rats. Methods The AD model was made by injected into rat hippocampal CA1 subregion with amyloid beta-protein(Aβ). Male Spraque-Dawley rats were as the experimental objects, which were randomly separated into 3 groups including Sham, Saline control and EPO treatment. After Aβ was injected into rats hippocampal CA1 subregion ,saline or EPO was respectively injected into the lateral ventricle of rats,with help of stereotaxic coordinates, upon the designed conditions. Hippocampal CA1 subregion Bcl-xl expression changes were observed 24 hours after the operation, and learning and memory abilities were checked 4 weeks after the operation. Results 24 hours after the operation Bcl-xl expression in the EPO group and the Saline group was less than the Sham control ,while Bcl-xl positive cells( 100.42 ± 12.43/field) in the EPO group were more than in the Saline group( 82.06 ± 19.68/field ) (P < 0. 05 ). 4 weeks after the operation learning ability in the EPO group ( 20.38 ± 5.88 ) was better than Saline group ( 25.50 - 3.25 ) (P < 0. 05 ), and memory ability in the EPO group (4.75 ± 1.75 ) was better than the Saline group(2.88 ± 1.55 )(P < 0.05 ). Conclusion EPO could improve the learning and memory abilities in the model rats,and it could be related with EPO restraining Bcl-xl expression decreasing.
4.Comparison of Angioseal closure device with manual compression

Objective To assess the safety and efficiency of Angioseal device in patients undergoing percutaneous femoral artery puncture in brain angiograph or interventional therapy. Methods A prospective trial was carried out in 128 patients undergoing brain angiograph and interventional therapy,in which 93undergoing brain angiography and 35 interventionsl therapy. All patients were divided into pure compression group by manual compression (83 cases) and vascular blocking group by Angioseal device (45 cases)according to different hemostatic measures. Results In angiography, hemostasis time of vascular blocking group and pure compression group was (1.8 ±0.8) min and(20.2 ±9.4) min (P<0.01 ),ambulation time was (3.8 ± 0.8) h and (19.4 ± 2.2) h (P < 0.01). In interventional therapy,hemostasis time of vascular blocking group and pure compression group was (2.0 ± 1.1) min and (24.5 ± 10.3) min (P < 0.01), ambulation time was (4.3 ± 1.5) h and (24.3 ± 3.2) h (P< 0.01). Pure compression group had higher complication rate (such as bleeding,hematoma,skin ulceration and blister) than vascular blocking group (P<0.01).Conclusion Angioseal closure device is safe, efficient and easy to use.
5.Clinical study of minimally invasive punctural evacuation in the treatment of basal ganglion region cerebral hemorrhage

ObjectiveTo study the clinical effect of YL-1 type hematoma puncture needle in the treatment of basal ganglion region cerebral hemorrhage.MethodsSixty-two patients with hypertensive basal ganglion region cerebral hemorrhage were treated by YL-1 type hematoma puncture needle from January 2007 to May 2011 (minimally invasive punctural evacuation group),of which,60 patients were treated by conservative treatment(conservative treatment group) as control,compared two groups of neural function defect score,hematoma clearance rate on admission,after 3 weeks treatment,and quality of life after 6 months.ResultsNeural function defect score on admission of minimally invasive punctural evacuation group was (23.6 ± 18.4) scores,while (23.4 ± 17.8) scores in conservative treatment group,the difference was not statistically significant(P > 0.05).After 3 weeks' treatment,neural function defect score and hematoma clearance rate of minimally invasive punctural evacuation group was superior to conservative treatment group [ (14.6 ± 12.4) scores vs.(20.1 ± 18.4) scores,(92.3 ± 5.4)% vs.(79.5 ± 13.8)% ] (P <0.05 ).After 6 months' treatment,the good rate of quality of life in minimally invasive punctural evacuation group was 81.7%(49/60),which was significantly increased compared with that of conservative treatment group [67.2% (39/58)] (P < 0.05).ConclusionsThe minimally invasive punctural evacuation in the treatment of basal ganglion region cerebral hemorrhage has small invasion,better prognosis,effective and fast decompression of intracranial hematoma,reducing disability rates,improvement of the quality of life,which could be a beneficial complement for traditional therapies.
6.Modulatory effect of Rho kinase on the cerebral vasospasm following subarachnoid hemorrhage

Objective To investigate the dynamical changes of Rho kinase in the cerebrospinal fluid (CSF) and its relationship with cerebral vascular spasm CVS. Methods CSF were collected on the ist, 3rd, 7th, 10th and 14th day after subarachnoid hemorrhage. The expression of Rho-kinase mRNA in CSF was determined by RT-PCR. The expression of endothelin-1 in CSF was determined by radioimmuno-assay. TCD was used to measure the velocity of the cerebral artery. Results The levels of ET-1 and Rho-kinase mRNA in CSF were re-markably increased on the 3rd day, and reached at the peak on the 7th day after subarachnoid hemorrhage, which were significantly higher than those without CVS. Conclusion There is a positive correlation between the level of Rho-kinase mRNA and ET-1 in CSF. Rho-kinase may participate in the development of CVS.
7.Tranexamic acid reduces blood loss in total knee arthroplasty:effectiveness and safety

BACKGROUND:Tranexamic acid is a synthetic anti-fibrinolytic drug, and can effectively control blood loss after total knee arthroplasty through vein. OBJECTIVE:To evaluate the effectiveness and safety of tranexamic acid in reducing blood loss after primary unilateral total knee arthroplasty. METHODS:From 2014 to 2015, 100 patients from the Department of Lower Limb, Sichuan Orthopaedic Hospital underwent primary unilateral total knee arthroplasty, and randomly divided into test group which used tranexamic acid and control group which used tranexamic acid, with 50 cases in each group. 1 g tranexamic acid was infused into the vein at 10 minutes before total knee arthroplasty in the test group, but tranexamic acid was not given in the control group. RESULTS AND CONCLUSION:The amount of total blood loss, postoperative wound drainage in 24 hours, and the amount of blood transfusion were significantly less in the test group than in the control group (P < 0.05). No significant differences in intraoperative blood loss and the number of patients receiving blood transfusion were found between the test and control groups. No significant difference in preoperative hemoglobin levels was detected before surgery, but hemoglobin levels were decreased after surgery between the two groups. Postoperative hemoglobin levels were apparently higher in the test group than in the control group after surgery. At 24 hours after surgery, no significant difference in coagulation was detectable between the two groups. At 6 days, double lower limb vein color Doppler ultrasound examination did not reveal deep vein thrombosis in both groups. These findings verify that treatment with tranexamic acid in the vein before primary unilateral total knee arthroplasty can evidently diminish the amount of perioperative blood loss and blood transfusion, reduce the risk and cost of blood transfusion, which is good for rehabilitation and does not increase the risk of deep vein thrombosis.
8.Comparision of Partially and Totally Depolarization of Peripheral Nerve in Total Knee Arthroplasty

Objective To compare the effects of peripatellar peripheral nerve partially and totally depolarized in total knee arthroplasty (TKA). Methods 66 TKA patients were divided into partially depolarized group (A, 42 knees) and totally depolarized group (B, 38 knees). The scores of knee-joint, Feller and Visual Analogue Scale were recorded. Results There was no significant difference in all the scores between 2 groups before operation (P>0.05). All the scores improved after operation (P<0.05), however, there was still no significant difference between 2 groups (P>0.05). Conclusion Patellar peripheral nerve partially depolarization technique can reduce postoperative knee pain, which has the same effect with totally depolarization.
9.Methylation of Runx3 promoter in different breast lesions.

OBJECTIVETo investigate the methylation status of Runx3 promoter and Runx3 expression in breast lesion tissues.
METHODSOne hundred and fourteen breast lesions, including 35 cases of fibroadenoma, 39 cases of intraductal carcinoma, 40 cases of invasive ductal carcinoma, and 33 cases of normal breast tissue from Fabruary 2010 to August 2012 were included in this study. Runx3 protein expression was assessed by immunohistochemical SP method; whereas methylation of Runx3 promoter was assessed by high resolution melting (HRM) analysis.
RESULTSRunx3 protein was mainly expressed in the cytoplasm of ductal epithelial cells. The expression rates of Runx3 in normal breast tissue, fibroadenoma, ductal carcinoma in situ, invasive ductal carcinoma were 87.9% (29/33), 85.7% (30/35), 53.8% (21/39), and 40.0% (16/40) respectively. The methylation rates of Runx3 promoter were 12.1% (4/33), 20.0% (7/35), 46.2% (18/39), and 57.5% (23/40), respectively. Correlation analysis between promoter methylation and protein expression of Runx3 in different breast tissue showed the r value in normal breast tissue, fibroadenoma, ductal carcinoma in situ and invasive ductal carcinoma was -0.431 (P = 0.012), -0.408 (P = 0.015), -0.589 (P = 0.000) and -0.743 (P = 0.000) respectively.
CONCLUSIONSRunx3 protein expression shows a downward trend in ductal carcinoma in situ and invasive ductal carcinoma, meanwhile its promoter methylation increases significantly. The methylation of Runx3 promoter may be one of the important factors in the occurrence and development of breast cancer.

To investigate the expression of long non-coding RNA (lncRNA) SNHG8 in EB virus related gastric cancer and their correlation prognosis.
