2.The result of the antibiotics susceptibility testing of Neisseria gonorrhoeae

Objective To analyse the antibiotics susceptibility result of Neisseria gonorrhoeae(N.g) isolates.Method 95 strains of N.g were performed the in-vitro susceptibility test.MICs of penicillin, tetracycline, ciprofloxacin, levofloxacin, ceftriaxone, cefotaxime, cefepime, and spectinomycin were determined by agar dilution.Results Almost all of the N.g strains were susceptible to the first-line antibiotics used for treating the infection. The resistance rate to ciprofloxacin, tetracycline and penicillin were 100%, 78.9% and 68.4% respectively.Conclusion Although N.g is susceptible to first-line antibiotics used for treating the infections at the clinics, the situation of resistance to penicillin, tetracycline and ciprofloxacin is very serious.
4.Effects of Bupi-Qufeng decoction on serum TIgE and EOS of paitents with atopic dermatitis belonging to spleen deficiency syndrome

Objective To study the effect of Bupi-Qufeng decoction on total serum IgE (TIgE) and EOS counts of atopic dermatitis (AD).Methods A total of 101 cases were randomly recruited into a treatment group and a control group.During the treatment both groups were give Danggui-Bohe ointment for external use,and based on this,oral Bupi-Qufeng decoction was added in the treatment group and loratadine in the control group.TIgE level and EOS count were detected both before the treatment and four weeks after the treatment.Results TIgE in the treatment group decreased significantly after the treatment (t=8.0063,P<0.001),the decreased value of which was larger than the control group (t=3.6434,P<0.001).EOS count in the treatment group also obviously decreased after the treatment (t=3.0314,P<0.01) ; the decreased value of which was also larger than the control group (t=3.3331,P<0.01).Conclnsion Bupi-Qufeng decoction could decrease TIgE and EOS level of AD patient,while the therapeutic mechanism needed further research.
5.High-resolution CT findings of otosclerosis

Objective To evaluate HRCT features of otosclerosis.Methods HRCT findings of 61 ears with the diagnosis of otosclerosis based on clinical diagnostic criteria in 34 patients were evaluated retrospectively.Results Hypodense regions in the bony otic capsule were found on HRCT in 55 ears and no abnormality was identified on HRCT in 6 ears.In 55 ears with abnormal HRCT findings, HRCT demonstrated the hypodense region of bony otic capsule anterior to oval window alone in 6 ears, the hypodense region anterior to oval window associated with thicked stapedial footplate and pericochlear hypodensity in 6 ears, the hypodense region anterior to oval window associated with thicked stapedial footplate and hypodensity posterior to oval window in 11 ears, the hypodense region anterior to oval window associated with thicked stapedial footplate in 20 ears, the hypodense region anterior to oval window associated with pericochlear hypodensity in 10 ears, and pericochlear hypodensity in the bony otic capsule alone in 2 ears.Conclusion HRCT can detect abnormalities in the bony otic capsule and the stapedial footplate,contributing to confirming diagnosis of otosclerosis.
6.A mouse model of acute lung inflammation induced by lipopolysaccharide inhalation

Objective:To develop a convenient,economical and stable model of acute lung inflammation in mice.Methods:BALB/c mice were inhaled intranasally with 50 ?L of LPS(1 g/L) or sterile PBS,and sacrificed at different time points after being anaesthetized.The bronchoalveolar lavage fluid(BALF) was collected,and the lungs were separated and homogenated or embedded and sliced to 5 ?m sections,which were then stained by HE to determine the severity of inflammation.The inflammatory cell infiltration in bronchoalveolar lavage was counted and IL-1?,the pro-inflammatory cytokine,measured by ELISA in lung homogenate and BALF.Results:The data showed that administration with 50 ?g of LPS(1 g/L) for 2 h resulted in significant inflammation in the lung.LPS mainly stimulated the recruitment of neutrophils within 24 h.And LPS was a quick revulsant of IL-1? production in BALF and in lung tissue between 4 and 24 h.Macrophages and lymphocytes recruited after 1 day,and sustained for at least 3 days.Conclusion:The results indicate that intranasal administration of LPS can induce a rapid and stable acute inflammatory model in mice.
7.Determination of tricin in malt by HPLC

Objective To develop an HPLC method for the quantitative analysis of tricin in malt. Methods HPLC method was used. Analysis was performed on Spherigel C_ 18 column (150 mm?4.6 mm, 5 ?m) with acetonitrile-water-tetrahydrofuran-methanoic acid (26∶74∶10∶1) as mobile phase. The detection wavelength was 350 nm, flow rate was 1.0 mL/min, and the analysic volumn was set at 20 ?L. Results The calibration curve of tricin was linear in the range of 0.22-2.15 mg/L (r=0.999 8). The average recovery rate of tricin was 97.4% (RSD=1.7%) (n=9). Conclusion The method can be used to determine tricin in raw malt quantitatively.
8.Relationship between the increase of hepatic D-bifunctional protein activity and bile acid biosynthesis in rats.

OBJECTIVETo determine the physiological role of D-bifunctional protein (DBP) in bile acid biosynthesis through investigating the effect of increasing activity of DBP on bile acid biosynthesis.
METHODSTwenty male Wistar rats were divided into two groups: diethylhexyl phthalate (DEHP) group (n = 10) and control group (n = 10). Serum triglyceride, total cholesterol, hepatic DBP activity, and fecal bile acids were assayed. The mRNA levels of hepatic peroxisome proliferator-activated receptor alpha (PPARalpha), DBP, and cholesterol 7alpha-hydroxylase (CYP7A1) were detected by RT-PCR.
RESULTSCompared with control group, serum triglyceride level was decreased significantly and PPARalphamRNA level was increased significantly in DEHP group (P < 0.01). Together with a sharp induction of DBP mRNA expression and DBP activity in DEHP group (P < 0.01), the levels of CYP7A1 mRNA and fecal bile acids were significantly increased by 1.9 times and 1.6 times respectively compared to control group (P < 0.01). There was a significantly positive correlation between DBP mRNA level or DBP activity and CYP7A1 mRNA level (r = 0.89, P < 0.01; r = 0.95, P < 0.01).
CONCLUSIONThe up-regulation of DBP mRNA and activity in liver can result in the increase in CYP7A1 mRNA expression and bile acid biosynthesis, suggesting that DBP may be involved in bile acid biosynthesis together with CYP7A1.

9.The molecular mechanism between baicalin metal complexes and bovin serum album.

In this paper, the new carbon nanotube modified glassy carbon electrode (F-CNTs/GCE) was prepared to establish a new method for studying the molecular interaction mechanism between baicalin metal complexes (BMC) and bovine serum album (BSA), and the principle of this method was discussed deeply. Under the physiological condition, the thermodynamics and kinetics properties of interaction between BMC and BSA were studied by cyclic voltammetry (CV) to inference their molecular effective mechanism. The results show that the presence of F-CNTs can accelerate the electron transfer, and better response signal was showed in the BMC/BMC-BSA system. The detection of interaction of BMC-BSA used new method show that BMC-BSA generates stable thermodynamically non-covalent compounds, and the obtained average binding sites of BMC-BSA were 1.7; the number of electron transfer in BMC/BMC-BSA reaction process was 2, and non electroactive supramolecular compounds of BMC-BSA were generated by this interacting reaction. The relevant research work provides a new way to study the molecular mechanism for the interaction of drugs with protein, and with a certain reference value for discussion on the non covalent interactions.
