Objective To optimize supercritical extraction technique from four medicinal materials, which are the part components in the recipe of Keganliyan Oral Liquor and are extracted traditionally for essential oils. Methods Extraction ratio and menthol extraction quantity were taken as evaluated indexes. Supercritical extraction technique was researched with orthogonal tests, gas chromatography, and SAS statistic. Results Within the test levels, temperature and time showed evident effect on extraction ratio and menthol extraction quantity, while pressure did not show any evident effect on them、 The preferable technique to extraction ratio is temperature at 55 ℃, time for 120 min, and extracted pressure at 27 MPa; and the preferable technique to menthol extraction quantity is temperature at 45 ℃, time for 120 min, and extracted pressure at 22 MPa. Conclusion The optimized supercritical extraction technique for Keganliyan Oral Liquor is feasible.

Objective To investigate the prevalence features of helicobacter pylori (Hp) infection, anemia and iron deficiency in a po-pulation of Wuhan children with 2 to 6 years old,and the relationship between Hp infection and anemia, iron deficiency in the children. Methods Randomly taking 95 children who had taken tests in our hospital's check-up centre in 2008 as the study objects,2 kinds of exa-mination were employed to detect Hp infection.Serum levels of Hp-IgG were measured by enzyme linked immunosorbent assay (ELISA) methods to evaluate past infection. The 14C urea breath test (14C-UBT) was conducted to obtain information of the presence of current/active Hp infection. In the morning 3 mL fasting venous blood was collected to determine the serum levels of Hp-IgG antibodies and ferritin.Hemoglobin values were determined with a hemoglobinometer.Serum levels of C-reactive protein (CRP) were tested in order to determine whether the children had evidence of current inflammation or infection. In addition, demographic information such as age and gender of the children and information about their use of antibiotics within the prior month were recorded. All cases were divided into 2 groups including the Hp infection group and non-Hp infection group according to laboratory examinations, then the Logistic regression was applied to analyze the relationship between Hp infection and anemia, iron deficiency. The Kappa identity test was taked to compare the 2 measures. Results Of the 95 children,18.9% were anemic and 36.8% were iron deficient.Forty percent of the cohort had Hp-IgG antibodies,74.4% tested positive by the UBT.Presence of Hp-IgG emerged as a significant risk factor for anemia, iron deficiency in adjusted analysis controlling for demographic factors, current inflammation, and antibiotic use. Conclusions Findings from different measure of Hp may reflect different stages of infection, with UBT results reflecting an earlier stage of infection, and presence of Hp-IgG reflecting established Hp infection associated with anemia, iron deficiency.

Based on the conserved region nucleotide sequences of five potato viruses/viroid(Alfalfa Mosaic Virus,ALMV;Cucumber mosaic virus,CMV;Cucumber mosaic virus-satellite,CMV-sat;Potato virus Y,PVY;Potato spindle tuber viroid,PSTVd) and one inner control(18S rRNA),the microarray containing specific oligonucleotide probes and PCR probes were designed and fabricated. The effects of probe concentration, hybridization time, hybridization temperature and spotting solutions on microarray hybridiazation were evaluated. Finally the specificity of optimized plant virus detection array was validated. No significant effect on hybridization signal intensity was observed when the concentration of the oligonucleotide probes ranged from 5 to 20 ?mol/L,there was a linear relationship between the concentration of PCR probe and hybridization signal intensity. The greatest signal intensity were obtained when hybridized at 45℃ for 4 h,and the oligonucleotide probes and PCR probes had a similar effect on microarray hybrization.Among the different spotting solutions,DMSO produced a good reproducibility. The plant virus could be detected specifically by oligonucleotide probe microarray and PCR probe microarray after optimization.

Objective To determine the variation of bone mineral density(BMD)and bone mineral content (BMC)in cadaveric lumbar vertebrae and investigate their significance in determining fracture modes and biomecha- nical properties. Methods The lumbar motion segments of the spines of 19 cadavers(mean age:56 years)were compressed to failure. The BMD and BMC of the cranial and caudal endplates were determined, along with the BMD and BMC of the upper and lower 1/3 transverse sub-endplate layers and the middle 1/3 transverse layer. The anteri- or,middle and posterior 1/3 vertical parts, anterior and posterior 1/2 vertical parts and the whole BMD and BMC were measured by using dual energy X-ray absorptiometry as well. All the data were analyzed statistically. Results Among the 19 segments, macrography after dissection revealed that 16 had been fractured, about 84.2%.The frac- ture always occurred in the centre or in the anterior part of the endplate.Within a vertebral body, the BMD and BMC of the cranial endplate were less than those of the caudal endplate, and the cranial 1/3 transverse layer and middle layer had lower BMD and BMC readings than the caudal layer. For the vertical region, from the anterior 1/3 to the posterior 1/3,or from the anterior 1/2 to the posterior 1/2,the BMD and BMC increased. For the endplate around the disc, the BMD of upper endplate was higher than that of the lower one,but the BMCs were equal. In one motion segment, the failure load correlated positively with BMD,and with the BMC of the endplate and the sub-endplate bone. Conclusion The variations in BMD and BMC within a vertebra and around a disc can explain why vertebral fractures are concentrated on the upper endplate, and why compressive fractures are always wedge-shaped.This un- derstanding can assist in the placement of inter-vertebral spacers.

【Objective】 To establish a new method for the determination of fibrinogen content in cryoprecipitated antihemophilic factor. 【Methods】 Fibrinogen (Fib) could bind with sheep anti-human fibrinogen (anti-Fib) specifically and further form antigen-antibody complex. When the Fib was present in the solution, the fluorescence of fluorescein isothiocyanate (FITC) labeled on the anti-Fib (FITC-anti-Fib) was quenched due to the formation of immune complex. The fluorescence quenching degree of FITC-anti-Fib was positively correlated with Fib concentration (cFib) in a certain concentration range. 【Results】 The linear relationship between fluorescence quenching degree [(I

OBJECTIVETo investigate the relationship of virological breakthrough and production of neutralizing anti-interferon antibody (NAb) in chronic hepatitis B patients treated with recombinant interferon-alpha (rIFN-alpha).
METHODFour hundred eighty-five patients with histological proven chronic hepatitis B were treated with 5 MU recombinant interferon-alpha 1b (rIFN-alpha1b) thrice weekly for 6-37 months (median 10). Serum HBV DNA, HBeAg and NAb levels of the patients were detected by fluorescent-quantitative PCR, enzymoimmunoassay and antiviral neutralizing biological assay respectively during the therapy.
RESULTSVirological breakthrough occurred in 66 patients (13.6%), and NAb was found in 98 patients (20.2%) of the total 485 patients. The rate of NAb positivity was higher in patients with viral breakthrough than those without it (68.2%, 45/66, vs 12.6%, 53/419, chi(2)=109.06, P < 0.01), and viral breakthrough occurred more in patients with positive NAb than with negative NAb (45.9%, 45/98, vs 5.4%, 21/387, chi(2)=109.06, P < 0.01). The time of the viral breakthrough occurrence and the time of NAb production had a significant correlation (P < 0.01). The occurrence of viral breakthrough was also influenced by the age of patients (P < 0.05) and HBeAg status (P < 0.01) before they were treated.
CONCLUSIONViral breakthrough occurred in 13.6% of our 485 chronic hepatitis B patients treated with recombinant interferon-alpha. Their viral breakthrough and production of NAb production had a significant correlation.
