1.Magnesium lithospermate B inhibits intracellular calcium elevation induced by ATP and KCl in rat vascular smooth muscle cells

Aim To investigate the effects of magnesium lithospermate B(MLB) on vasoconstriction of denuded aortic rings in vitro,as well as the effects of MLB on cytosolic free calcium concentration([Ca~(2+)]_i) in aortic vascular smooth muscle cells(AVSMCs).Methods Isotonic tension was measured in the rat thoracic aortic rings without endothelium and quantitative changes of [Ca~(2+)]_i were directly monitored in AVSMCs loaded with the fluorescent calcium indicator Fluo-3 using intracellular cation measurement system.Results MLB had no effects on resting ring tension in the presence or absence of extracellular Ca~(2+).In rings exposed to 50~200 ?mol?L~(-1) MLB,the isotonic contractions induced by 1 ?mol?L~(-1) PE were decreased in calcium-free K-H solution.However,in the presence of extracellular Ca~(2+),KCl 60 mmol?L~(-1) produced isotonic contractions that were abolished by verapamil 10 ?mol?L~(-1) or inhibited by pretreatment of 50~200 ?mol?L~(-1) MLB in a concentration-dependent manner.When administered to the rings precontracted with 1 ?mol?L~(-1) PE in the absence of extracellular Ca~(2+),a second extracelluar-calcium-dependent vasoconstriction induced by restoration of extracellular Ca~(2+) was also reduced by pretreatment of 50~200 ?mol?L~(-1) MLB.Moreover,MLB,at concentration of 50~200 ?mol?L~(-1),produced little effect on AVSMCs [Ca~(2+)]_i in the presence or absence of extracellular Ca~(2+).In AVSMCs exposed to 50~200 ?mol?L~(-1) MLB,the inhibitory effects of MLB on [Ca~(2+)]_i induced by 20 ?mol?L~(-1) ATP were concentration-dependent,with decreases ranging from 17.4% to 61.1% in the absence of extracellular Ca~(2+).In the presence of extracellular Ca~(2+) and thapsigargin,the increases in AVSMCs [Ca~(2+)]_i evoked by 60 mmol?L~(-1) KCl were inhibited by pretreatment with MLB(50~200 ?mol?L~(-1)) or abolished by pretreatment with verapamil(10 ?mol?L~(-1)).Conclusions MLB inhibits vasoconstriction induced by PE,high-K~+,and re-Ca~(2+).MLB also has inhibitory effects on increase of [Ca~(2+)]_i induced by ATP and high K~+ in AVSMCs.These results indicate that both the blocking effects on calcium release and voltage-dependent calcium channels may be responsible for the effects of MLB on [Ca~(2+)]_i in vascular smooth muscle cells.

4.Roles of IL-8 and eotaxin in airway inflammation and lung function injury of chronic obstruction pulmonary disease

The clinical data of 25 patients with acute exacerbations of chronic obstruction pulmonary disease (AECOPD) admitted from March 2009 to October 2010 were analyzed. The sputum samples were collected and the levels of inflammatory cell, interleukin-8 (IL-8) and eotaxin in sputum were determined. The lung function was measured. Twenty healthy subjects served as controls. The counts of neutrophils and eosinophils, and the levels of IL-8 and eotaxin in sputum of AECOPD were higher than those of health subjects. There was a positive correlation between the levels of IL-8 and the counts of neutrophils (P < 0. 01), and between the levels of eotaxin and the counts of eosinophils in sputum ( P < 0.01). The levels of IL-8 and eotaxin were negatively correlated with the lung function (FEV1% ).

6.Experimentla study on the expression of albumin in blood-retinal barrier injury of oxygen-induced retinopathy

Objective To investigate expressions of albumin during retinal neovascularization in a murine model of oxygen -in-duced retinopathy.Methods C57BL/6J mice were exposed to (75 ±2)%oxygen for 5 days and returned to room air to induce retinal neovascularization .Retinal neovascularization was observed by fluorescence angiography , and was quantified by counting the endotheli-al nuclei protruding into the vitreous cavity after hematoxylin-eosin staining.The blood-retina barrier was evaluated by Western blot a-nalysis of extravasated albumin in the retina .Results In fluorescence angiograms , irregular neovascularization and fluorescein leakage was observed surrounding the unperfused areas in the hypoxic group .Compared to the normoxic group , there was a significant increase in both-side retinal albumin protein levels in the hypoxic group at P 13, P15 , and P17.Conclusions Albumin expression was in-creased in murine retina under hypoxia , and might be used to detect blood-retinal barrier damage of oxygen-induced retinopathy .
7.Experimental Study of the Survival Mechanism of Non-physiological Blood Perfusion of Arterialized Venous Flap

Objective To investigate the microcirculative route of the arterialized venous flap.Methods Selecting 15 Chinese white rabbits designed the model of arterialized venous flap by anastomosis with from the central artery to central vein.The reversed blood flow perfusion was directly observed by the "ear windows" under the microscope and,simultaneous recording the changes in the microcirculation of the flap.Results All of the flaps were alived.The structure of the capillary net in the flap was a stereo-net .In the early arterialized stage,the upstream blood flows quickly in the venule;there was no blood flowing in the capillary net with the opening of the capillary net,the reconstructed procedure of the non-physilogical microcirculation was accomplished:the arterial blood mainly flow through small vein and venule,finally arrived the capillary net.Conclusions Keep up the drainage vein free,the arterialized venousflap could be alived by reconstructed effective microcirculation through the venous system.
9.Effect of high-flux hemodialysis on advanced oxidation protein products in maintenance hemodialysis patients

Objective To investigate the effect of high-flux hemodialysis (HFHD) on advanced oxidation protein products (AOPPs) in maintenance hemodialysis (MHD) patients.Methods 40 MHD patients (MHD group),50 non-dialytic chronic kidney disease (CKD) patients (CKD group) and 31 healthy subjects undergoing health examination survey received AOPPs detection.After lowflux hemodialysis (LFHD) for 3 months,HFHD was performed in MHD group for 3 months.Plasma AOPPs and other biochemical indicators were tested at the end of 0,3 and 6 months.AOPPs levels were detected in HFHD and LFHD dialysates.Results Plasma AOPPs level was higher in MHD group than in CKD and control groups,and plasma AOPPs level was higher in CKD group than in control group.Compared with before LFHD in MHD group,AOPPs and triglyceride (TG) levels had no significant differences after LFHD for 3 months(t=0.831,-0.191,both P>0.05),while plasma cholesterol (CHO) and β2 MG levels were increased in MHD group after LFHD for 3 months(t=-2.697,10.39,P<0.05 or 0.001).There were significant differences in plasma AOPPs,TG,β2-MG and CHO levels before versus after HFHD for 3 months (t=4.292,2.271,8.297 and 2.295,P <0.001 or 0.05).No AOPPs were detected in HFHD and LFHD dialysates.Conclusions Plasma AOPPs levels are increased in CKD patients.LFHD can not effectively eliminate AOPPs,while long term HFHD can decrease the plasma levels of AOPPs,TG and CHO,and reduce β2-MG accumulation.
10.Effect of activated kupffer cells on immune tolerance after liver transplantation

Until now, the exactly effect of Kupffer cells (KCs) on inducing immune tolerance or aggravating acute rejection is still unknown. Activated by various way after liver transplantation, have the ability of phagucytosis the apoptositic T cells, up-regulated expression of FasL and many Th2/Th3 cytokines, such as interleukin-10 and transforming growth factor-lB. These up-regulated cytokines could induce the apoptosis of Th1 cells and enhance the proliferation and differentiation of the Th2 cells, finally induce the immune tolerance, However, the activated KCs also have the ability of expression many cytokine-dependent molecules,such as class Ⅱ major histocompatibility antigens, adhesion molecule and costimulatory molecules which could enhance the function of the antigen presentation, increase the expression of Thl cytokine and aggravate the acute rejection after liver transplantation. It maybe relate to the ratio of theTh1/Th2 cells determined by the complicated net of the cytokine produced by the activated Kupffer cells: the predominance of Th2 cells could induce the immune tolerance, on the contrary, the acute rejection proceed.
