5. Expression of zinc finger protein 262 mRNA in patients with autosomal-dominant polycystic kidney disease and its significance
Academic Journal of Second Military Medical University 2006;27(6):581-584
Objective: To investigate the expression of zinc finger protein 262 (ZNF262) mRNA in normal kidney tissues and kidney tissues of patients with autosomal-dominant polycystic kidney disease (ADPKD) at different stages, and to explore the role of ZNF262 in pathogenesis of ADPKD. Methods: Patients with ADPKD were staged according to glomerular filter rate (GFR). Imaging observation and routine pathological examination were performed. The expression of ZNF262 mRNA and proliferating cell nuclear antigen (PCNA) mRNA in normal kidney tissues (n=8), early stage ADPKD kidney tissues (n=4) and advanced stage ADPKD kidney tissues (n=4) was examined by semi-quantitative RT-PCR. The correlation between the expressions of the 2 genes was investigated in all tissue specimens. Results: Expression of ZNF262 mRNA and PCNA mRNA in early and advanced ADPKD kidney tissues was significantly higher than that in normal renal tissues (both P<0.01), and that in the advanced stage ADPKD was significantly higher than that in early stage ADPKD (both P<0.05). The expression of ZNF262 and PCNA mRNA was highly correlated in the early, advanced ADPKD and normal renal tissues(r1 =0.842 6, r2=0.902 1 and r3=0.883 5, respectively, all P<0.05). Conclusion: The ZNF262 mRNA lev el is higher in ADPKD kidney tissue than that in normal control and increases with the advancement of ADPKD. The expression of ZNF262 is significantly correlated with the expression of PCNA in the same renal tissues. The expression of ZNF262 mRNA may serve as an indicator in diagnosis of ADPKD and may be used for clinical staging of ADPKD patients.
7.Nitric oxide signal transmission pathway and its regulatory effect on skeletal muscle glucose uptake during exercise
Chinese Journal of Tissue Engineering Research 2007;0(15):-
BACKGROUND: Nitric oxide (NO) can influence glucose transportation. Although its signal transmission pathway is not certain, it has been confirmed that the pathway is different from insulin. OBJECTIVE: To explore NO signal transmission pathway and the regulatory mechanism for the glucose uptake of skeletal muscle during exercise. RETRIEVAL STRATEGY: Using the keywords of "nitric oxide, signal, glucose", we searched the Pubmed database for the relevant articles about the transmission pathway and its regulation of glucose uptake in skeletal muscle published from January 1996 to November 2007. Meanwhile, the related foreign language books were retrieved manually from national library. Articles about the regulation of NO to muscular blood flow, signal transmission and glucose under exercise stress in human and gnawer were selected. Pathologically related basic studies were excluded. LITERATURE EVALUATION: Sixty related literatures (books) were collected, and 30 were accorded with the inclusive criteria, of which 5 were review articles, 23 were basic researches and 2 were related books. Meanwhile, 20 articles were related to the effects of NO on blood flow regulation and vasodilation, 4 articles and 2 books were related to signal transduction and 4 were about its signaling effect during exercise. DATA SYNTHESIS: NO is a signal molecule. It can mediate various biological phenomenon and displays strong vasodilation effect. The production of NO is increased in vivo during exercise, and NO can stimulate glucose uptake in skeletal muscle through its transduction. CONCLUSION: The generation of NO during exercise has positive effect on glucose uptake in skeletal muscle. The contraction and glucose uptake are closely correlated to the generation and transmission of NO, but the mechanism is still unclear.

Objective To evaluate the effect of single hand pipe method on venous indwelling needle puncture. Methods Literature about the impact of single hand pipe method on venous indwelling needle puncture was retrieved from digital databases of PubMed, Medline, EMbase, The Cochrane Library, SinoMed, VIP database, China National Knowledge Infrastructure(CNKI), and WanFang Data. The quality of literature was evaluated by the Cochrane Handbook (5.1.0). The Meta data was analyzed by RevMan 5.3. Results Ten random control trials were included . Ten studies indicated that single hand pipe method increased success rate of puncture[related to the risk(RR)=1.16, 95%confidence interval(CI) 1.11-1.22]. And, it decreased the complication rate (3 studies) (RR=0.32, 95%CI 0.13-0.79) and pain rate(2 studies) (RR=0.25, 95%CI 0.13-0.47) as well as shorten the time of tube(2 studies) (WMD:-1.68, 95%CI-2.44--0.93). Conclusions Single hand pipe method can improve a puncture success rate and reduce complications and pain rate and shorten the time of tube, so that get more time to rescue the patient, and improve the level of the working efficiency and nursing for the nursing staff. In addition, to some extent, save manpower. It is worth being popularized.

ObjectiveTo study the effects of intermittent subglottic secretion drainage(SSD)with an endotracheal tube on incidience of ventilator- associated pneumonia(VAP). MethodsWe searched computerized databases, including Cochrane library,Pubmed,EMbase,CBM,CNKI,Wanfang and some relevant databases from initializing to March 2012,extracting data about subglottic secretion drainage(SSD)with an endotracheal tube on incidence of VAP using RCT.The data had Meta- analysis by software RevMan5,and to those data which could not be conversed or merged, they underwent descriptive analysis. ResultsWeidentifled 5 randomized clinical trials that met the inclusion criteria with a total of 683 randomized patients. In Meta- analysis, the overall risk ratio for ventilator- associated pneumonia was decreased significantly by intermittent subglottic secretion drainage with an endotracheal tube. There was no significant difference on adverse events or on hospital or intensive care unit mortality.Databases, including ICU and hospital LOS,duration of endotracheal catheterization, bacteria concentration, which were difficult to be conversed and obtained full data, they underwent a descriptive research. ConclusionsCompared with the common endotracheal tubes, the use of endotracheal tubes with subglottic secretion drainage is effective for prevention of ventilatorassociated pneumonia. But it has no effect on adverse events or on hospital or intensive care unit mortality. So large sample RCTs with multiply- center should be carried out to assess the effect of the use of SSD.
