1.The Simultaneous Determination of Gatifloxacin and Tinidazole by HPLC.

Objective To establish a reversed HPLC method to determine the contents of gatifloxacin and tinidazole simultaneously. Methods ODS C_ 18 column (4.6mm?250mm,5?m) was used as an analytical column. The mobile phase consisted of 15% acetonitrile, 25% of methanol and 60% of 0.05mol/L KH_2PO_4 buffer,which contained 0.5% triethylamine and was adjusted to pH 3.5 by phosphoric acid. The flow rate was 1ml/min. The detection wavelength was 318nm. The column temperature was 30℃. 20?l of diluted sample solution was injected to the sampler. Results The calibration curve of gatifloxacin was linear in the range of 2~20?g /ml (r=0.999 6) ,and that of tinidazole was 2~20 ?g /ml (r=0.999 9). The recovery rates of them were 100.1% and 100.4%,respectively. RSDs of both drugs were less than 1.1 %. Conclusions The method is simple,accurate and sensitive and can be used to determine the two components simultaneously.
2.Research of health education needs for parents of children with attention deficit hyperactivity disorder

Objective In order to assist the medical staff to take appropriate contents and ways of health education,and to promote the recovery of physical and mental health of children with attention deficit hyperactivity disorder (ADHD),the needs of the parents about how to help them acquire the related knowledge were investigated.Methods A survey on the degree of familiarity of nursing knowledge,the requirements and the ways for getting the knowledge about the health education was carried out.Results The rate of 226 parents who knew about nursing knowledge was 10.62%-87.61% (24/226-198/226).More than half of the parents needed health education.The rates of concerns were as follows,92.48% (209/226) of all the parents concerned on the factors affecting prognosis,94.69%(214/226) on the comorbidities,92.04% (208/226) on the training methods of attention,and 91.15% (206/226) on psychological and behavioral interventions.Conclusions Parents could acquire related knowledge and educational methods about ADHD from the medical staff and afford a good care,which may improve behavior problems and mental health of the children with ADHD.
3.The biology and potential clinical application of bone marrow-derived stem cells

Bone marrow-derived stem cells, mainly including hematopoietic stem cells, mesenchymal stem cells, multipotent adult progenitor cells, etc., are the most densely studied adult stem cells up to now. Recent findings indicate that adult stem cells have the potential to differentiate across lineage barriers and to adopt the expression profiles and functional phenotypes of cells unique to other tissues. This flexibility is termed "plasticity",which has posed challenges to conventional concepts. Meanwhile, it brings new hopes for the therapeutic application of cell engineering and tissue engineering. This review focuses on the biological features, the plasticity and its possible mechanisms, research method and potential clinical application of bone marrow-derived adult stem cells
5.Detection of the qnr gene responsible for plasmid-mediated quinolone resistance

Objective To explore the prevalence of qnr gene in gram-negative bacteria isolated from Renmin Hospital of Wuhan University. Methods qnr gene was detected by PCR in non-duplicate E.coli (n=129), E.cloacae (n=10) and K.pneumoniae (n=29) isolates.PCR technique was used to amplify the gene encoding type I integrons.Kirby-Bauer disk diffusion method was used to test the in vitro suscepti- bility of the isolates to 16 antimicrobial agents.ESBLs were investigated according to phenotypie screening and confirmatory test recom- mended by NCCLS.MIC values of ciprofloxacin were determined for qnr gene positive strains.Results qnr gene were identified in 6 clinical isolates including 5 strains of E.coli and I E.cloacae, qnr gene was not found in K.pneumoniae isolates.The 6 qnr-positive clinical iso- lates were positive for type I integrons.All the 6 strains were susceptible to imipenem but resistant to many other drugs tested.Only 2 of the E.coli isolates were susceptible to ciprofloxacin.Conclusions In Hubei Province, the prevalence of qnr is confirmed.The serious situa- tion of multidrug-resistance in clinical isolates with qnr gene requires careful monitoring of qnr-carrying isolates.

7.Immunodiagnostic value of recombinant membrane protein Tetraspanin 2 of Schistosoma japonicum

Objective To study the immunodiagnostic potential of recombinant membrane protein Tetraspanin 2 of Schistosoma japonicum(rSjTsp2) as a diagnostic antigen. Methods The rSjTsp2 was expressed and purified as an antigen used in indirect ELISA to detect specific serum antibody against Schistosoma japonicum of rabbits that at different stages of infection, and the results were compared with those of ELISA using natural antigen (AWA) as antigen. Results The sensitivities of rSjTsp2-ELISA were 75.4% (46/61),77.6% (45/58) and 81.5% (44/54), respectively, 2, 4, 6 weeks after rabbits were infected with Schistosoma japonicum, meanwhile the sensitivities of AWA-ELISA were 68.9% (42/61),86.2% (50/58) and 88.9% (48/54), respectively. The sensitivity difference of two methods was not significant statistically (x2 = 0.652, 1.454,1.174, all P > 0.05).Conclusions The rSjTsp2 is expressed successfully in vitro, and rSjTsp2 recombinant protein expressed in prokaryotic system has a certain application potential in immunodiagnosis.
8.OPTIMISATION AND VALIDATION OF AN IN VITRO EXTRACELLULAR ELECTROSTIMULATING DEVICE

In order to study the mechanisms of high frequency stimulation ( HFS), we designed an in vitro device to deliver extracellular electrical stimulation to live cells in culture, using the same parameters as in the surgical treatment of Parkinson's disease (PD). The system was made of a 24 wells culture plate electrified with titanium wire bridges or two titanium wires placed parallel in a culture dish of 10 centimetres diameter. The extremities of the series of wells were connected to a constant current generator. The result showed that this system had no toxicity on GH3 and PC12 cell cultures. Preliminary experiences demonstrate that this system is suitable for the study with DNA and protein chip technologies.

Objective To observe the effect of Fuyuan Capsule (FC) on vascular endothelial cell function of rats with qi deficiency and blood stasis.Method Forty-eight 16-month-old SD rats were equally randomized to 5 groups:FC group,positive control group (Qishen Capsule),model group and blank control group.Rat models of qi deficiency and blood stasis were induced by exertion,starvation and cold.The mice were given the drugs by gastric perfusion for 4 weeks.After treatment,endothelin (ET),6-keto-prostaglandin F_(1a) and nitric oxide (NO) levels were mea- sured.Results NO and 6-keto-PGF_(1a) levels were lower in the model group than those in the blank control group (P
10.Analysis on iron metabolism indicators in children with various genotypes of thalassemia

Objective To study the value of iron metabolism indicatorsin thalassemia .Methods 218 cases of thalassemia ,57 ca‐ses of iron deficiency anemia (IDA) children and 70 healthy children(control group) were enrolled in the study ,and serum iron (SI) ,total iron binding capacity(TIBC) ,transferrin(Tf) ,serum ferritin (SF) were detected for them .Results β‐thalassemia double heterozygous(β0 ) or homozygous group :SI levels were significantly higher than the other groups(P<0 .05) ,while TIBC ,Tf levels were significantly lower than the other groups(P<0 .05);α‐thalassemia group (static α‐thalassemia ,standard α‐thalassemia):SI , TIBC levels compared with control group showed no significant difference(P>0 .05) ,but Tf level was higher than control group(P<0 .05);hemoglobin H disease group:when TIBC ,Tf levels compared with the control group ,the difference was statistically signif‐icant(P<0 .05) ,when SI compared with the control group ,the difference was not statistically significant(P>0 .05) .Conclusion Compared with Tf ,SI and TIBC are better indicators for monitoring iron loading in children with thalassemia .The increased SI level and decreased TIBC level are two indicators for the diagnosis of β0 thalassemia in children with cellule anaemia .
