Objective To evaluate the therapeutic effects of combining acupuncture and physical therapy with drugs on treating Duchenne muscular dystrophy (DMD).Methods Sixty infants with DMD were randomly divided into a treatment group and a control group (n =30 in each). Both groups were injected with Bozhi glycopeptide and given levocamitine and fructose sodium diphosphate orally. Moreover, the treatment group was additionally given acupuncture and physical therapy. Before and after 90 days of treatment, the time to walk 10 metres and to climb stairs of 2 metres high was measured,as were the serum creatine phosphate kinase (CPK),lactate dehydrogenase (LDH) and aspertate aminotransferase (AST) levels of all participants. Results After treatment, the average walking and stair climbing times of both groups were significantly shorter, and those of the treatment group were significantly shorter than those of the control group. Moreover, the average CPK,LDH and AST levels in the treatment group were significantly lower than before treatment and than those of the control group after treatment. There was a significant difference in the overall response rates, with 93% observed in the treatment group and 73% in the control group. Conclusion Combining acupuncture and physical therapy with drugs can significantly increase the mobility and improve the serum CPK,LDH and AST levels of children with DMD.

AIM:Sodium butyrate has antitumor effects on colon cancer cells such as inhibiting cell growth and promoting differentiation and apoptosis. The aim of this study is to investigate whether sodium butyrate induces apoptosis in human colon cancer cell line HT-29 and to examine the intracellular mechanisms involved, especially the role of caspase activation in the process.METHODS:HT-29 cells were cultured to logarithmic phase before treatment with sodium butyrate at concentration of 5.0 mmol/L and caspase inhibitors at the concentration of 20 ?mol/L.The latter were added in the medium ahead of sodium butyrate for 1 h.Then, the staining of Annexin V-FITC and PI were used to analyze HT-29 apoptosis and the dye JC-1 was applied to detect mitochondrial membrane potential by flow cytometry.Caspase activity within the cells was measured respectively using a specific caspase activity assay kit and a microplate reader.RESULTS:Preincubation of HT-29 cells with sodium butyrate significantly increased apoptosis [(35.40?0.70)%] and decreased mitochondrial membrane potential(5.53?0.91).This effect was blocked when pretreatments were enforced with z-VAD-fmk,z-DEVD-fmk and z-LEHD-fmk.The apoptosis percentages were(1.33?0.59)%,(1.40?0.53)% and(1.27?0.91)%,respectively and mitochondrial membrane potentials were 9.80?1.15,10.23?0.50 and 10.33?1.02,respectively.However,the role of reduction by z-IETD-fmk,which presented the apoptosis percentage of(32.10?2.33)% and mitochondrial membrane potential of 5.93?1.31,was not observed. An enhancement of caspase-3 and-9 activities(2-3-fold)but no change of caspase-8 activity was confirmed.CONCLUSION:Apoptosis of HT-29 colon carcinoma cells induced by sodium butyrate is tightly linked to caspase activation via mitochondrial pathway other than tumor necrosis factor-alpha and has the potential to inhibit proliferation and thereby may contribute to the progression of colon cancer.

OBJECTIVE:To establish a method for the determination of related substances in Thioctic acid injection. METH-ODS:HPLC was performed on the column of Agilent Eclipse Plus C18 with mobile phase A of 0.005 mol/L potassium dehydroge-nate phosphate solution(adjusted with phosphoric acid to pH 3.0 to 3.1)and B of methanol:aoetonitrile(1∶1,V/V)(gradient elu-tion) at a flow rate of 1.2 ml/min;detection wavelength was 215 nm,column temperature was 35 ℃,and the injection volume was 20 μl. RESULTS:Under the established chromatographic conditions, thioctic acid with impurities and its degradation products were well separated;the linear range of thioctic acid was 0.828-52.992 μg/ml(r=0.999 9);RSDs of precision and stability tests were lower than 3%;recovery of impurity A was 98.45%-105.73%(RSD=2.41%,n=9);the largest single impurity content in mass fraction was 0.152%-0.189%,the total impurity content in mass fraction was 0.487%-0.516%. CONCLUSIONS:The meth-od is simple, accurate, specific and sensitivity, and can be used to determine the related substances of the Thioctic acid injection.

Objective To investigate the role of soluble intracellular adhesion molecule-1(sICAM-1), corticotropin-releasing hormone(CRH)and interleukin-6(IL-6) in the pathogenesis of pregnancy-induced hypertension (PIH). Methods Maternal plasma sICAM-1, CRH and IL-6 level were determined in 81women at 26~37 gestational weeks among which 23 were normal pregnant women and 58 with PIH. The later were grouped into 21 women with mild, 19 with moderate, 18 with severe PIH. sICAM-1 and IL-6 were measured by ELISA and CRH by RIA. Results The levels of plasma sICAM, CRH and IL-6 were significantly elevated in women with moderate and severe PIH compared with control subjects(P

Objective To investigate the protective effects of propofol in clinical relevant concentration on TNF-induced apoptosis in endothelial cells derived from human umbilical vein (HUVECs) .Methods The third and fourth passages of the cultured endothelial cells were divided into 5 groups. 0, 12.5, 25, 50 or 100 ?mol/L propofol was added to the cultured cells respectively and the cells were incubated for 30 min. Then TNF-? was added to the cultured cells (the final concentration of TNF-? was 2000 U.ml-1 ) which was incubated for 24h. Apoptosis was detected by using "terminal deoxynucleotidyl transferase (tdt )-mediated deoxyuridine triphosphate (dUTP )-biotin nick end-labelling" (TUNED . Morphologic changes were examined by means of electron microscope. Results Apoptosis expression was very high in control group (no propofol was added) . The process can be inhibited by pre-treatment with propofol (P

Clinical Medicine of China 2011;27(8):874-876
Objective To investigate the role of subtype 1 autoantibody against angiotensin Ⅱ receptor in the pathogenesis of preeclampsia by detecting its expression in the peripheral blood of preeclampsia patients,Methods Thirty patients with preeclampsia were assigned to preeclampsia group. Twenty normal pregnant women at the late stage and twenty non-pregnant healthy women as controls were investigated. The level of type 1 antoantibody against angiotensin Ⅱ in the peripheral blood was detected by indirect SA-ELISA assay with the produced ATR-1 as the antigen. Results The level of subtype 1 antoantibody against angiotensin Ⅱ (65 ±4. 7) % in the peripheral blood of preeclampsia patients is significantly higher than that of normal late pregnant (26 ±2. 8)% and non-pregnant women(7.8 ±2. 2)% groups (t1 =24. 97 ;t2 =38.56;P <0. 01 for both) ;The angiotensin Ⅱ receptor subtype 1 autoantibody in the group of normal late pregnancy (26 ± 2. 8 )% was significantly higher than that of healthy non-pregnant women group ( 7. 8 ± 2. 2 ) % ( t = 4. 58, P < 0. 05 ).Conclusion Compared with the normal pregnant women and the healthy non-pregant women, the autoantibody against AT1 receptor in sera of preeclamptic patients is elevated ata high frequency. These results suggest that overproduction of AT1-AA may play an important role during the development of preeclamptic patients. AT1-AA is a novel risk factor in pregnant women. Immune mechanisms may be involved in the process of pregnancy.
