BACKGROUND:Increasing evidence has demonstrated that bone marrow mesenchymal stem cel (BMSC) transplantation can promote skin, liver and lung repair in animal models; andLeonurus sibiricus L. has the ability of promoting blood circulation and regulating menstruation.
OBJECTIVE: To investigate the therapeutic effect of BMSC transplantation with leonurine on intrauterine adhesions (IUA) in rabbits and the relevant mechanism of action.
METHODS: After modeled using dual injury method, rabbit models of IUA were randomly divided into five groups: sham-operated group (sham), model group (IUA), BMSC transplantation group, leonurine treatment group and combined treatment group (BMSCs+leonurine). Rabbits in the sham group were only given normal saline rinsing after hysterotomy, while those in the latter three groups were correspondingly given intrauterine BMSC transplantation or/and intragastric administration of 4 mg/kg leonurine for 14 days. Morphological changes of the endometrium were observed using hematoxylin-eosin staining, and expression levels of transforming growth factor β protein, Smad3 protein and interferon-γ mRNA were detected using immunohistochemical staining and real-time fluorescence quantitative PCR, respectively.
RESULTS AND CONCLUSION:Compared with the model group, the degree of IUA was al significantly improved in the other groups, especialy in the combined treatment group. Moreover, BMSC transplantation, leonurine treatment and their combined use al could inhibit IUA-induced increase of transforming growth factorβ and Smad3 protein expression and IUA-induced decrease of interferon-γ mRNA level. Importantly, al these alternations were much more pronounced in the combined treatment group. Our results show that the combined use of BMSC transplantation and leonurine treatment can exert a synergistic effect in the improvement of IUA through the transforming growth factor β/Smad3 pathway.

Objective To study the time-dependent changes and mechanisms of auditory cortex in prelingual sensorineural hearing loss. Methods Two groups (9-12, 19-22-year-old) of hearing-loss patients and matched normal hearing subjects received MR diffusion and MRS examination. The ROI for ADC measurement were placed on the Heschl gyrus (HG), and for NAA/Cr, Cho/Cr on the superior temporal gyrus(STG). Results There was no statistical difference among groups both in the NAA/Cr and Cho/Cr (P>0.05), Left side NAA/Cr 2.46±0.51, 2.49±0.52, 2.26±0.33;Right side NAA/Cr 2.26±0. 46, 2. 44±0. 45, 2. 27±0. 46. Left side Cho/Cr 0. 88±0. 21,0. 92±0. 18, 0. 87±0. 13 ; Right side Cho/Cr 0. 88 ± 0. 18, 0. 87 ± 0. 22, 0. 81 ± 0. 16. There was statistical difference of ADC value among normal hearing subjects and 9-12, 19-22-year-old hearing-loss patients(F = 4. 42, P < 0. 05, F = 11.37,P<0. 05). There was significant difference of ADC value between normal hearing and 19-22 year-oldhearing-loss (q = - 44. 89, P = 0. 008, q = - 54. 28, P = 0. 027) on both left and right HG, and there wassignificant difference between normal hearing and 9-12 year-old hearing-loss only on right HG (q =- 32. 54, P = 0. 000), Left side (860 ± 103), (885 ± 48), (905±77) mm<'2>/s, Right side (848 ± 73),(880 ± 61), (902 ± 52) mm2/s. Conclusion Compared with the normal hearing, the ADC value increased in 20-year-length of deafness. The NAA/Cr and Cho/Cr of STG showed no changes among the three groups.

Objective To investigate the diagnostic value of computer image analysis assisted aroundness measurement of gastric gland lumens in well-differentiated gastric adenocarcinoma.Methods From January 2016 to July 2016,10 hematoxylin-eosin (H-E) stained slides of pathologically diagnosed well-differentiated gastric adenocarcinoma were collected.At the same period,H-E stained slides of gastric epithelial hyperplasia and normal gastric mucosa,six slides of each,were collected as controls.All of them were scanned into high resolution.Digital data roundness,long diameter and short diameter were measured and compared.Kruskal-Wallis rank sum test was used for statistical analysis.Results The median values of lumen roundness of normal gastric mucosa,gastric epithelial hyperplasia and welldifferentiated gastric adenocarcinoma were 79.7°(10.6°),73.0°(12.3°) and 58.6°(15.2°),respectively,and the difference among three groups was statistically significant (H=494.827,P＜0.01).The values of gland roundness of normal gastric mucosa were higher than those of gastric mucosal hyperplasia and welldifferentiated gastric adenocarcinoma,and the differences were statistically significant (T=156.007 and -508.579,both P＜0.01).The value of gland roundness of gastric mucosal hyperplasia was higher than that of well-differentiated gastric adenocarcinoma,and the difference was statistically significant (T=-352.572,P＜0.01).The values of long diameter of gland lumens in normal gastric mucosa,gastric mucosal hyperplasia and well-differentiated gastric adenocarcinoma were 24.1 μm(11.8 μm),43.1 μm (28.8 μm),and 96.0 μm(79.3 μm),respectively,and the difference among three groups was statistically significant (H=744.987,P＜0.01).The values of short diameter of gland lumens in normal gastric mucosa,gastric mucosal hyperplasia and well-differentiated gastric adenocarcinoma were 16.0 μm (8.7 μm),22.7 μm (13.1 μm) and 33.5 μm (38.0 μm),respectively,and the difference among three groups was statistically significant(H=248.170,P＜0.01).Conclusion The roundness of gastric gland lumens can be used as an effective parameter for the differential diagnosis of normal gastric mucosa,gastric mucosal hyperplasia and well-differentiated gastric adenocarcinoma.

AIM:To investigate the expression of microRNA-193b(miR-193b) in the cervical tissues, and fur-ther to explore the effect of silencing miR-193b on diamminedichloroplatinum ( DDP)-treated HeLa cell viability.METH-ODS:The expression levels of miR-193b in different cervical tissues were examined by qPCR.After transfection of miR-193b-inhibitor, the cell migration was determined by Transwell assay, the sensitivity of HeLa cells to DDP was measured by MTT assay, the protein levels of phosphate and tension homology deleted on chromsome ten ( PTEN), protein kinase B (Akt), p-Akt and p-glycoprotein(P-gp) were determined by Western blot.RESULTS:The mRNA level of miR-193b was significantly increased in the cervical cancer tissues compared with normal cervical tissues ( P<0.05) .Knockdown of miR-193b obviously inhibited migration and enhanced sensitivity to DDP of HeLa cells (P<0.05).Additionally, after transfec-tion of miR-193b-inhibitor, the expression of PTEN was increased, whereas the protein levels of p-Akt and P-gp were de-creased (P<0.05).CONCLUSION:miR-193b is highly expressed in the cervical cancer tissues.Inhibition of miR-193b augments the sensitivity to DDP of HeLa cells, at least in part, through PTEN-PI3K/Akt signaling pathway.

BACKGROUND:Recently, there is more and more attention on uric acid (UA), especially the correlation with cardiovascular disease in the filed of epidemiology; however, researches of the effect of UA on endothelial cells are lack based on cytology.OBJECTIVE:To explore the effects of UA in different concentrations on human umbilical vein endothelial cells line (ECV304), malondialdehyde (MDA), nitric oxide (NO) and 3-(4,5-dimethyl thiazole-2)-2, 5-diphenyl thiazolyl blue (MTT)in vitro.DESIGN: Randomized controlled study based on ECV304.5ETTING: Department of Endocrine, Xijing Hospital, the Fourth Military Medical University of Chinese PLA.MATERIALS:ECV304 was provided by Department of Immunology, the Fourth Military Medical University of Chinese PLA; DMEM Iow-glucose medium by Gibco Company, USA; bovine serum by Beijing Yuanheng Shengma Biotechnology Researching Institute; trypsin by Gibco Company, USA; MTT by Huamei Company; dimethyl sulfoxide (DMSO) analytical pure by Tianjin Bodi Chemical Engineering Co. Ltd.; UA by Sigma Company; NO kit and MDA kit by Nanjing Jiancheng Company; ordinary invert microscope and enzyme-linked immune detector IX70 invert microscope by Olympus, Japan;enzyme-linked immune detector by Eastern China Electron Tube Factory.METHODS: The experiment was carried out in Department of Endocrine and Burning, the Fourth Military Medical University of Chinese PLA from December 2003 to April 2004. Resuscitation, culture, regeneration and inoculation of endothelia cells were undertaken Cell Culture published by Situ et al. Endothelia cells were cultured with non-serum DMEM for 24hours so as to maintain synchronization at the phase of G0/G1 and divided into 4 groups, including control group, low-concentration UA group, moderate-concentration UA group and high-concentration UA group. Each group was divided into 3time points, including 24 hours, 48 hours and 72 hours with 8 samples in each time point. Samples were added with 5％serum medium containing 0 mmol/L, 0.1 mmol/L, 0.2 mmol/L and 0.4 mmol/L UA in control group, low-concentration UA group, moderate-concentration UA group and high-concentration UA group, respectively, and incubated in box with the volume fraction of 0.05 CO2 at 37 ℃. Twenty-four hours later, MDA and MTT were detected; additionally, MTT was detected once more after 48 and 72 hours.MAIN OUTCOME MEASURES: Proliferation of ECV304 and content of NO and MDA.RESULTS:With the increasing concentration of UA, content of MDA was decreased to (2.97±0.05),(2.89±0.09),(2.78±0.10) and (2.44±0.03) μmol/L, respectively; content of NO was (6.86±1.41), (12.5±2.7), (18.9±1.8) and (21.1±1.4) μ mol/L. Absorbencies of NO and MTT were increased and proliferation was increased remarkably at 48 hour.CONCLUSION: A which is characterized by anti-oxidation may promote proliferation of vascular endothelia cells and release of NO.

Objective To evaluate the value of MSCT in differentiate cystic lesions in different cervical region. Methods Forty cases with cyst lesions in the neck which was confirmed by pathology were collected in our study. The following diseases were excluded: cystic lesions from thyroid gland and thymus or bronchus, cystic changes of solid lesions ( necrotic lymph node) and cystic or solid lesions in the parotid gland. All patients underwent MSCT scan on GE Lightspeed 16. The parameters were following: 120 kV,260 mA, thickness 5 mm, pitch 1. 375. The delay time for contrast enhanced CT was 25 seconds for arterial phase and 60 seconds for vein phase. Results Lesions were analyzed in 4 different cervical regions including parotid gland, midline, lateral cervix and posterior cervix. Among the lesions in parotid area,2 were first brachial cyst and the other 2 were epidermoid cyst. First brachial cyst occurred intraparotid gland and epidermoid cyst occurred intraparotid or extraparotid gland. Among the lesions in midline area, 17 cases were thyroglossal cysts,81.0% were in the level hyoid bone, 12 cases of thyroglossal cysts protruded at the preepiglottic space or oral base. Another kind of lesions in midline included 3 cases of epiglottic cysts and 1 epidermoid cyst in the lower level of the neck which involved infrasternal level. The 12 cases of second brachial cysts occurred in the lateral neck. Ten cases were in the left side. Type Ⅱ was the most common type and account for 5 cases among all of second brachial cysts. Type Ⅰ and type Ⅱ were running up,accounting 3 cases respectively. Type Ⅳ account for 1 case. Two cases with lymphangioma and 1 case with third brachial cyst located at the posterior neck. Conclusion According to the location and typical imaging appearance, MSCT can do the differentiation of different cystic lesions in the different area of neck.

Nasopharyngeal carcinoma (NPC) is prevalent in Southeast Asia, especially in the area of southern China. It is acknowl-edged that NPC is absolutely related to EBV infection. Nevertheless, there are just a few researches about the relationship between the level of EBV antibodies and the tumor stages and it hasn't come to conclusion. This review makes a summary on present research progress and would be valuable for the better understanding of the contribution of EBV antibodies to the tumor stages in NPC.
