1.The study of diffusion weighted imaging and MR spectroscopy in auditory cortex and related area of prelingual hearing-loss patients

Objective To study the time-dependent changes and mechanisms of auditory cortex in prelingual sensorineural hearing loss. Methods Two groups (9-12, 19-22-year-old) of hearing-loss patients and matched normal hearing subjects received MR diffusion and MRS examination. The ROI for ADC measurement were placed on the Heschl gyrus (HG), and for NAA/Cr, Cho/Cr on the superior temporal gyrus(STG). Results There was no statistical difference among groups both in the NAA/Cr and Cho/Cr (P>0.05), Left side NAA/Cr 2.46±0.51, 2.49±0.52, 2.26±0.33;Right side NAA/Cr 2.26±0. 46, 2. 44±0. 45, 2. 27±0. 46. Left side Cho/Cr 0. 88±0. 21,0. 92±0. 18, 0. 87±0. 13 ; Right side Cho/Cr 0. 88 ± 0. 18, 0. 87 ± 0. 22, 0. 81 ± 0. 16. There was statistical difference of ADC value among normal hearing subjects and 9-12, 19-22-year-old hearing-loss patients(F = 4. 42, P < 0. 05, F = 11.37,P<0. 05). There was significant difference of ADC value between normal hearing and 19-22 year-oldhearing-loss (q = - 44. 89, P = 0. 008, q = - 54. 28, P = 0. 027) on both left and right HG, and there wassignificant difference between normal hearing and 9-12 year-old hearing-loss only on right HG (q =- 32. 54, P = 0. 000), Left side (860 ± 103), (885 ± 48), (905±77) mm<'2>/s, Right side (848 ± 73),(880 ± 61), (902 ± 52) mm2/s. Conclusion Compared with the normal hearing, the ADC value increased in 20-year-length of deafness. The NAA/Cr and Cho/Cr of STG showed no changes among the three groups.
2.Significance of measuring roundness of gastric gland lumens in differential diagnosis of well-differentiated gastric adenocarcinoma

Objective To investigate the diagnostic value of computer image analysis assisted aroundness measurement of gastric gland lumens in well-differentiated gastric adenocarcinoma.Methods From January 2016 to July 2016,10 hematoxylin-eosin (H-E) stained slides of pathologically diagnosed well-differentiated gastric adenocarcinoma were collected.At the same period,H-E stained slides of gastric epithelial hyperplasia and normal gastric mucosa,six slides of each,were collected as controls.All of them were scanned into high resolution.Digital data roundness,long diameter and short diameter were measured and compared.Kruskal-Wallis rank sum test was used for statistical analysis.Results The median values of lumen roundness of normal gastric mucosa,gastric epithelial hyperplasia and welldifferentiated gastric adenocarcinoma were 79.7°(10.6°),73.0°(12.3°) and 58.6°(15.2°),respectively,and the difference among three groups was statistically significant (H=494.827,P<0.01).The values of gland roundness of normal gastric mucosa were higher than those of gastric mucosal hyperplasia and welldifferentiated gastric adenocarcinoma,and the differences were statistically significant (T=156.007 and -508.579,both P<0.01).The value of gland roundness of gastric mucosal hyperplasia was higher than that of well-differentiated gastric adenocarcinoma,and the difference was statistically significant (T=-352.572,P<0.01).The values of long diameter of gland lumens in normal gastric mucosa,gastric mucosal hyperplasia and well-differentiated gastric adenocarcinoma were 24.1 μm(11.8 μm),43.1 μm (28.8 μm),and 96.0 μm(79.3 μm),respectively,and the difference among three groups was statistically significant (H=744.987,P<0.01).The values of short diameter of gland lumens in normal gastric mucosa,gastric mucosal hyperplasia and well-differentiated gastric adenocarcinoma were 16.0 μm (8.7 μm),22.7 μm (13.1 μm) and 33.5 μm (38.0 μm),respectively,and the difference among three groups was statistically significant(H=248.170,P<0.01).Conclusion The roundness of gastric gland lumens can be used as an effective parameter for the differential diagnosis of normal gastric mucosa,gastric mucosal hyperplasia and well-differentiated gastric adenocarcinoma.
3.Leonurine combined with bone marrow mesenchymal stem cell transplantation in the treatment of intrauterine adhesions

BACKGROUND:Increasing evidence has demonstrated that bone marrow mesenchymal stem cel (BMSC) transplantation can promote skin, liver and lung repair in animal models; andLeonurus sibiricus L. has the ability of promoting blood circulation and regulating menstruation.
OBJECTIVE: To investigate the therapeutic effect of BMSC transplantation with leonurine on intrauterine adhesions (IUA) in rabbits and the relevant mechanism of action.
METHODS: After modeled using dual injury method, rabbit models of IUA were randomly divided into five groups: sham-operated group (sham), model group (IUA), BMSC transplantation group, leonurine treatment group and combined treatment group (BMSCs+leonurine). Rabbits in the sham group were only given normal saline rinsing after hysterotomy, while those in the latter three groups were correspondingly given intrauterine BMSC transplantation or/and intragastric administration of 4 mg/kg leonurine for 14 days. Morphological changes of the endometrium were observed using hematoxylin-eosin staining, and expression levels of transforming growth factor β protein, Smad3 protein and interferon-γ mRNA were detected using immunohistochemical staining and real-time fluorescence quantitative PCR, respectively.
RESULTS AND CONCLUSION:Compared with the model group, the degree of IUA was al significantly improved in the other groups, especialy in the combined treatment group. Moreover, BMSC transplantation, leonurine treatment and their combined use al could inhibit IUA-induced increase of transforming growth factorβ and Smad3 protein expression and IUA-induced decrease of interferon-γ mRNA level. Importantly, al these alternations were much more pronounced in the combined treatment group. Our results show that the combined use of BMSC transplantation and leonurine treatment can exert a synergistic effect in the improvement of IUA through the transforming growth factor β/Smad3 pathway.
4.Expression and significance of miR-193b in cervical cancer

AIM:To investigate the expression of microRNA-193b(miR-193b) in the cervical tissues, and fur-ther to explore the effect of silencing miR-193b on diamminedichloroplatinum ( DDP)-treated HeLa cell viability.METH-ODS:The expression levels of miR-193b in different cervical tissues were examined by qPCR.After transfection of miR-193b-inhibitor, the cell migration was determined by Transwell assay, the sensitivity of HeLa cells to DDP was measured by MTT assay, the protein levels of phosphate and tension homology deleted on chromsome ten ( PTEN), protein kinase B (Akt), p-Akt and p-glycoprotein(P-gp) were determined by Western blot.RESULTS:The mRNA level of miR-193b was significantly increased in the cervical cancer tissues compared with normal cervical tissues ( P<0.05) .Knockdown of miR-193b obviously inhibited migration and enhanced sensitivity to DDP of HeLa cells (P<0.05).Additionally, after transfec-tion of miR-193b-inhibitor, the expression of PTEN was increased, whereas the protein levels of p-Akt and P-gp were de-creased (P<0.05).CONCLUSION:miR-193b is highly expressed in the cervical cancer tissues.Inhibition of miR-193b augments the sensitivity to DDP of HeLa cells, at least in part, through PTEN-PI3K/Akt signaling pathway.

9.Expression and significance of survivin and CD44v6 protein in non-small cell lung cancer

Objective To investigate the expression and significance of survivin and CD44v6 in non-small cell lung cancer (NSCLC) and their correlation. Methods SP immunohistochemical technique was used to detect the expression of survivin and CD44V6 protein in 53 cases of NSCLC and 13 cases of para-eaneer nor-mal tissues. Results The positive rate of survivin and CD44v6 in NSCLC was 60. 38% and 69. 81% respec-tively,which was higher than that of normal pulmonary tissues adjacent to carcinoma(P <0. 01). The expres-sion of survivin and CD44v6 in NSCLC was related to lymph node metastasis (P < 0. 05), but not to patients' age and gender(P>0.05). The expression of survivin was related to TNM stages and cell differentiation (P <0. 05) ,but not to histology type(P >0. 05). The expression of CD44v6 in squamous carcinoma was significantly higher than that of adenocareinoma (P <0. 05),but the expression of CD44v6 was not related to TNM stages and cell differentiation (P >0. 05). There was no correlation between the expression of survivin and C1)44v6 (r = -0. 058, P >0. 05). Conclusion Survivin might be used to evaluate NSCLC development;CD44v6 might be used for the differential diagnosis of squamous carcinoma in NSCLC;both of them might be helpful to predict the metastasis of NSCLC. They might be two independent events in the process of NSCLC genesis and develop-ment.
10.The MR imaging of osmotic myelinolysis ha patients after liver transplantation

Objective To study the imaging of central pentine myelinolysis (CPM) and extrapontine myelinolysis(EPM) after liver transplantation and value of DWI. Methods Eight patients after liver transplantation with CPM and EPM were included in our study, 4 cases of female and 4 cases of male.MR examinations were performed on Philips 1.5 T MRI system. The sequences included SE T1 WI, FSE T2Wl,axial SE-EPI DWI,TR/TE 6225/118.7 ms, ETL 128, FLIP 90°, thickness 5 nun, matrix 128 ×128,b=1000 mm2/s. Results Four of B type hepatitis and cirrhosis and 1 case combined with hepatic carcinoma, 1 of C type hepatitis with cirrhosis from schistosome,2 of C type hepatitis and cirrhosis, 1 of cholangiogenic cirrhosis. MR demonstrated prolongation of T1 and T2 with a shape of butterfly, round or quadrangle on the brain stem sparing of the corticospinal tract and peripontine tissue. The lesion manifested high signal intensity both on DWI and ADC map indicating T2 effect and vasogenic edema. Follow up MR in 2 cases 2 months later showed more prolongation of T1 and T2 than the first time and the lesion manifested iso-intensity signal on DWI and high signal intensity on ADC map suggesting increased diffusivity. Abnormal signal intensities were detected on the bilateral caudate head, globus pallidus, putamen and cerebral cortex in 3 cases. Conclusion CPM should be considered when prolongation of T1 and T2 with a shape of butterfly, round or quadrangle on the brain stem, sparing of the corticospinal tract and peripontine tissue was detected for patients of pest-transplantation. When combined with other area abnormality, EPM and CPM should be considered. DWI can early and sensitively show the lesion.
