Since Bence-Jones protein was discovered and used in multiple myeloma, many tumor markers have been used in diagnosis and treatment monitoring of various cancers. However the lack of specificity and sensitivity of most tumor markers is always limiting their application in research and clinical diagnosis currently. It is predicted that the future development of tumor marker research may mainly focus on the innovation of research strategies, such as combined detection of a set of related tumors marker and comprehensive interpretation of testing results.

Leptin,a 16 kD protein encoded by ob gene,has been shown to play a pivotal role in satiaty control and adipose metabolism.However,numemus recent studies indicme that leptin may also take part in mammary cells transformation,proliferation and angiogensis.Those evidences suggest that leptin has an important role in the mammary cell tumorigenesis and progession,but the undedine mechanism needs further exploration.Our aim is to review the current andvances on leptin's role in tumourigenesis and progression of breast cancer from perspectives of in vitro,aniamal model and breast cancer biopsy studies.

As an increasing threat to women's health worldwide, breast cancer has many well-established risk factors, including genetic suscebility, high concentration of serum estrogen, early-age menarche, late-age menopause and postmenopansal obesity. However, these risk factors only account for about half of the in-cidence of breast cancer. Recent studies provided some clues that three primary hormone responsive virues-human papilloma virus (HPV), mouse mammary tumor virus (MMTV) and Epstein Barr virus (EBV)may act play roles in pathogenesis of human breast cancer. This review aims to have a close look at advances on the potential role, probable mechanism and relating hypothesis of Human papillomavirus in human breast cancer.

Objective To investigate inhibition effect of leptin on apoptosis of MCF-7 cells and its potential mechanism.Methods MCF-7 cells were cultured and devided into 5 groups:control group,tamoxifen (10-5 mol/L) group,tamoxifen(10-5 mol/L)-leptin(102 ng/mL)group,tamoxifen (10-5 mol/L)-leptin(103 ng/mL) group,tamoxifen(10-5 mol/L)-leptin(104 ng/mL)group.Fourty-eight hours after being treated with corresponding concentration of leptin and tamoxifen respectively,all groups of cells were quantatively tested for apoptosis by ELISA and were semi-quantatively detected for survivin,Bcl-2,Bax mRNA change by real-time PCR.Results Tamoxifen induced MCF-7 apoptosis were inhibited by leptin at a serial concentration of 103 ng/mL,104 ng/mL.Meanwhile,leptin largely enhanced survivin mRNA expression in MCF-7 cells,compared with control group.However,no significant increase of BCL-2,BAX mRNA in MCF-7 cells was observed after leptin treatment in either group.Conclusions Leptin could effectively inhibit tamoxifen induced MCF-7 apoptosis in a dose dependent manner.The potential mechanisrn of leptin' apoptosis inbition in MCF-7 cells may involve upregulation of survivin mRNA.

Mannose-binding lectin(MBL) is a key component of the innate immune system that plays a pluripotent role in natural defense.MBL genetic variants are associated with increased susceptibility to and the severity and prognosis of various diseases.The association between low MBL-producing allelic variants and disease risk and/or severity is particularly strong in infectious and autoimmune diseases.Enhanced risk for disease by low MBL producers is particularly the case for children and immunocompromised patients,especially when primary and secondary immune deficiencies coexist.Therefore the use of MBL testing and replacement therapy has reached the threshold of personalized medicine.The author presents a review of the role of MBL in health and diseases,the advances in MBL testing methodology and MBL related therapies.

BACKGROUND: The angiotensin-converting enzyme (ACE) is the important component of the renin-angiotensin-aldosterone system (RAS). The ACE gene has, in fact,insertion/deletion polymorphism in intron 16, consisting of a 287-base pair Alu repeat sequence. ACE gene heterozygotes insertion/deletion (I/D) polymorphism is correlated with cardiovascular disease and IgA nephropathy and other diseases. OBJECTIVE: To investigate the distribution of ACE gene I/D polymor-phism in Chinese Han population in comparison with other known ethnic populations. DESIGN: Observation study on healthy individuals of Han nationality. SETTING: Key Laboratory of Clinical Immunology of Jiangsu Province; Department of Laboratory Medicine, First Hospital Affiliated to Soochow University; Department of aboratory Medicine, College of Medical Technology of Jiangsu University PARTICIPANTS: Totally 241 healthy individuals who received the healthy examination in the First Hospital of Soochow University between December 2005 and January 2006 were recruited in the experiment. They were 152 male and 89 female , with mean age of (27±8)years. All the participants without blood relationship were Han nationality from Suzhou region in China, free from disorder of hepatic, renal, endocrine and cardio- cerebrovascular diseases which were confirmed by clinical and experimen- tal examination. METHODS: Genotype of ACE gene I/D polymorphism allele of 241 healthy individuals of Han nationality was detected with polymerase chain reaction (PCR). PCR purified products with genotype of deletion/deletion (DD) and insertion/insertion (Ⅱ) polymorphism were performed DNA sequencing with fluorescence-labeled end termination method. MAIN OUTCOME MEASURES: Genotype and allele frequency of ACE gene I/D, as well as the comparison between them and those of other ethnic population. RESULTS: All the 241 subjects participated in final result analysis. ① The genotypes of ACE were DD, Ⅱ and ID. Compared with allele Ⅰ, allele D lost 287-base pair Alu repeat sequence. ②The frequencies of genotype Ⅱ, ID and DD were 46.1％, 41.5％ and 12.4％ respectively, with an allelic frequency of 66.8％ for allele Ⅰ and 33.2％ for allele D. ③The distribution of ACE genotype was similar between Japanese and Han nationality crowd, both presenting that type Ⅱ was commonly seen and type DD was the least; ID was mostly found in European and American crowd, but Ⅱ was little found. There was racial diversify of frequency of the distribution of ACE genotype among individuals of Han nationality and Japanese as well as Europeans and Americans. Compared with other nationalities, allele Ⅰ of individuals of Han nationality was significantly higher than that of above nationalities (χ2=105.55,P ＜ 0.01), but allele D was obviously lower (χ2=87.54,P ＜ 0.01). CONCLUSION: ACE gene polymorphism has racial diversify. To know genetic features of ACE gene polymorphism of individuals of different na tionalities is the basis and prerequisite to study the correlation of ACE gene I/D polymorphism with diseases.

OBJECTIVE: Intraocular lens are applied in trigeminal operations of cataract extraction and comeal transplantation, and the selection of artificial lens is modified. This paper evaluated the application of colodzed polymethyl methacrylate (PMMA) intraocular lens in interlaced non-phacoemulsification cataract extraction with intraocular lens implantation and keratoplasty (trigeminal operation).METHODS: A computer-based online search of CNKI (200312007) and Medline database (1974/2004) was performed for related articles with the key words "colorized intraocular lens, corneal transplantation, cataract, combination operation" in Chinese and "PMMA intraocular lens, cataract extraction, corneal transplantation" in English. Articles related to PMMA intraocular lens in cataract and corneal transplantation trigeminal operation was selected. Meta analysis and repetitive articles were excluded. A total of 11 articles were included and summarized.RESULTS: A total of 21 cases (21 eyes) of corneal leukoma and cataract caused by various trauma or ophthalmopathy underwent colorized PMMA intraocular lens implantation. During the follow-up for 10 months, visual acuities of naked eyes in 92% cases were significantly improved. Two weeks after operations, visual acuities of 9 cases (42.9%) were over 4.7, 10 cases over 4.5, and 2 cases (9.5%) over 4.0. At days 7 to 14 after operation, corneal grafts of 21 cases were completely transparent; 10 months after operation, corneal grafts of 16 cases were completely transparent, 3 cases were semitransparent, and 2 cases were turbid, who underwent second penetrable corneal transplantation.CONCLUSION: The application of colodzed PMMA intraocular lens in penetrable corneal transplantation combined cataract extraction can cure the refracting media turbidity of forepart eye. Compared with traditional operations, trigeminal operation reduces complications, increases safety and improves visual acuity.

Objective To translate the Venous Leg Ulcer Quality of life Questionnaire (VLU-QoL) into Chinese and evaluate the reliability and validity of Chinese version.Methods VLU-QoL was translated into Chinese according to the standards setted by ISPOR.Totally 232 patients with venous leg ulcer were recruited and investigated by the Chinese version of VLU-QoL.Results The Chinese version of VLU-QoL comprised 34 items,Cronbach α was 0.940,inter-rater reliability was 0.841;test-retest reliability was 0.825.I-CVI was 0.83-1.00,S-CVI/Ave was 0.955;Criterion-related validity were 0.714 and 0.541;factor analysis got 3 factors,which explained 54.367％ of the total variance.Conclusions The Chinese version of VLU-QoL has been proved to be reliable and valid.It can be used as a valid tool for the measurement of quality of life for patients with venous leg ulcer.

Objective To investigate the expression of IL-10 and IL-6 in tissue and plasma of subtypes lymphoma and its significance. Methods Immunohistochemical technique and enzyme-linked immunosorbent assay (ELISA) were used to assess the expression of IL-10 and IL-6 in 97 cases paraffin tissue and plasma samples from different subtypes of lymphoma.Results Expression of IL-10 and IL-6 was positive in all lymphoma tissue samples, but there is no significant difference among different subtypes lymphoma (x2 =0.815, x2 =0.542, P ＞ 0.05). Tumor samples were found positive IL-10 and IL-6 expression in macrophages and vascular endothelial cells by immunohistochemistry respectively. Expression of IL-10 and IL-6 in plasma of lymphoma were higher than that in control respectively [(232.57±191.59) pg/ml vs (59.12±68.35) pg/ml,(80.70±89.68) pg/ml vs (45.68±33.66) pg/ml],with significant difference (t =6.968,t =2.896,P ＜0.05).IL-10 and IL-6 plasma levels increased with enhanced expression in lymphoma (x2 =0.815,x2 =0.542,P ＜ 0.05).Which was found to be correlated with each other (rs =0.394,P ＜ 0.05). Conclusion The expression of IL-10 and IL-6 had been found at various degrees in tissues and plasma of lymphoma.IL-10 and IL-6 in plasma of lymphoma were higher than that in controls. These results showed that IL-10 and IL-6 may cooperate in vivo with lymphoma cell proliferation and infiltrate. IL-6 and IL-10 maybe served as useful indicators for diagnosis of lymphoma.
