3.Expression and significance of miR-193b in cervical cancer

AIM:To investigate the expression of microRNA-193b(miR-193b) in the cervical tissues, and fur-ther to explore the effect of silencing miR-193b on diamminedichloroplatinum ( DDP)-treated HeLa cell viability.METH-ODS:The expression levels of miR-193b in different cervical tissues were examined by qPCR.After transfection of miR-193b-inhibitor, the cell migration was determined by Transwell assay, the sensitivity of HeLa cells to DDP was measured by MTT assay, the protein levels of phosphate and tension homology deleted on chromsome ten ( PTEN), protein kinase B (Akt), p-Akt and p-glycoprotein(P-gp) were determined by Western blot.RESULTS:The mRNA level of miR-193b was significantly increased in the cervical cancer tissues compared with normal cervical tissues ( P<0.05) .Knockdown of miR-193b obviously inhibited migration and enhanced sensitivity to DDP of HeLa cells (P<0.05).Additionally, after transfec-tion of miR-193b-inhibitor, the expression of PTEN was increased, whereas the protein levels of p-Akt and P-gp were de-creased (P<0.05).CONCLUSION:miR-193b is highly expressed in the cervical cancer tissues.Inhibition of miR-193b augments the sensitivity to DDP of HeLa cells, at least in part, through PTEN-PI3K/Akt signaling pathway.
4.Application and Progress of Helper Phage in Phage Display

Phage display is a widely used gene engineering high technology.Through the display of exogenous peptides or proteins fused specific coat protein on the surface of phages,it is possible to construct proteins or peptides libraries and screen interesting proteins,peptides and antibodies successfully.Most commonly used phage display technology is phagemid/helper phage system,in which helper phages are essential for the replication and assembly of phagemid particles.In this review,in combination with the newest research dynamic status,we summarize phagemid/helper phage double-genome system.We mainly emphasized the features and mutation mechanisms of different helper phages.We also made some prospects for the future directions,in the meanwhile,we also expect that our experience can provide some help for further maturity of the technology.

Aim To investigate the effect of sulfur dioxide (SO2) on blood pressure and aortic structure of the spontaneous hypertensive rat(SHR).Methods Sixteen male SHRs at the age of 4 weeks were randomly divided into SHR control group and SHR+Na2SO3/NaHSO3 group,respectively.Another eight male WKY rats served as WKY control group.Five weeks later, the blood pressure was determined.The rat aortas were dyed with Hart's Method. The morphometric parameters including outer radius, lumen radius and the wall thickness were calculated by Leica workstation. 6.Screening and identification of antioxidant endophytes from Lycium barbarum of Ningxia.

In this paper, 29 endophytes were isolated from different organs and tissues of Lycium barbarum of Ningxia by tablet coating method, 18 of them was fungi, and 11 of them was actinomycetes. The endophytes quantity in the different tissues were leaves > flowers > roots >fruits; The hydroxyl radical scavenging activities of 11 endophytes were investigated by Fenton reaction, and total antioxidant capacities of them were examined by a. total antioxidant capacity test kit; culture features and strain-specific sequence analysis were employed to explore the diversity of the 11 endophytes. The result showed that 5 fungi and 6 actinomycetes that having antioxidant activity could be phylogenetically classified into 3 genera, 3 genera and 3 families, respectively. The total antioxidant capacity and hydroxyl radical scavenging activity of the 11 endophytes showed distinct difference. The antioxidant activity of Aspergillus were stronger, among which total antioxidant capacity of fL1 was (188.5 ± 0.549) U · mL⁻¹ and the IC₅₀ was 0.3 mg · L⁻¹; the IC₅₀ of strain fL1 was 0.42 mg · L⁻¹ and the total antioxidant capacity of fL9 was (113.63 ± 1.021) U · mL⁻¹, all of them were stronger than the positive control Vit C. The experimental results indicated that endophytic fungi of L. barbarum of Ningxia have a great developing and application prospect for the development of antioxidant agent.

8.The value of detection pre-S1 antigen in patients with chronicity type B hepatitis (CHB)

Objective To evaluate the value of pre-S1 Ag in combined detection of hepatitis B virus. Methods 288 patients' serums with chronicity type B hepatitis (CHB)were collected when they had been detected with HBV-DNA,HBVM or ALT/AST,and then ELISA assay were applied to detect pre-S1Ag. Results A total of 288 patients ,including 175 males and 113 females( their meso-age is 36.89 years old) were recruited into this study. 161 patients were pre-S1 Ag positive(68.4%), 197 patients' HBV DNA contents exceeded 5.0E + 2 copy/ml(55.9%) ,71patients were H BeAg positive (24.6%), 186 patients were HBeAg negative (64.58%). The rate of 64 patients' ALT and AST exceeded 0. 84, and their' ALT exceeded 40 u/ml ( 22. 2% ). 60 patients' AST exceeded 50 u/ml (20. 8% ). These patients were divided into pre-S1 Ag positive group and pre-S1 Ag group. In pre-S1 Ag positive group, 124 patients' HBV DNA contents exceeded 5.0E + 2 copy/ml( 62.9% ), 118 patients were HBeAg negative (59.8%). When HBV DNA content was less than 5.0E+2 copy/ml,pre-S1 Ag positive rate was 35.2% and pre-S1 Ag negative rate was 63.6%. when HBV DNA content was less than 5.0E + 3 copy/ml, pre-S1 Ag positive rate was 56.9% and pre-S1 Ag negative rate was 81.7%. When HBV DNA content exceeded 5.0E + 5 copy/ml but less than 5.0E + 8 copy/ml ,pre-S1 Ag positive rate was 33% and pre-S1 Ag negative rate was 14.3% ,so two groups had significant difference. Conclusion Pre-S1 Ag positive and HBeAg negative in the CHB patients serums signify virus replication,and it can help us decide prognosis.
9.Mechanism of protective effect of sodium nitroprusside against endotoxin- induced acute lung injury in rats

Objective To investigate the effect of sodium nitroprusside (SNP) , an exogenous NO donor, on expression of hemeoxygenase-1 (HO-1) and inducible NO synthase (iNOS) in the lung tissue in a rat model of lpopolysaccharide (LPS)-induced acute lung injury and the underlying mechanism.Methods Thirty-two Wistar rats of either sex weighing 190-210 g were randomly divided into 4 groups ( n = 8 each) : Ⅰ sham operation (S) ;Ⅱ LPS;Ⅲ hemin + LPS and Ⅳ SNP + LPS. The animals were anesthetized with intraperitoneal (IP) 7% chloralhydrate 5 ml?kg-1. LPS 750 ?g?kg-1 in 300 ?l was instilled into the lungs via trachea in group Ⅱ . In group Ⅲ hemin (a HO-1 inducer) 40 ?mol?kg-1 was injected IP 12 h before LPS. In group Ⅳ SNP 25 ?g?kg-1 + LPS 750 ?g?kg-1 in 300 ?l was instilled into the lungs via trachea. In group S normal saline 300 ?l was instilled via trachea instead of LPS. The animals were killed by (1) exsanguination from carotid artery 8h (2) after LPS instillation and lungs were removed for broncho-alveolar lavage (BAL) and detennination of protein concentration in BALF and the expression of HO-1 and iNOS protein in the lung tissue by immune-histochemical technique and HPIAS-2000 image analysis system and MDA content in lung tissue and (3) light microscopic examination (4) wet/ dry lung weight ratio.Results The 4 groups were comparable with respect to body weight and sex (M/F ratio) . Objective To observe and evaluate the clinical efficiency and safety of topiramate for treatment of patients with various types of epilepsy Methods 142 patients suffering from epilepsy were underwent the treatment of topiramate, of them 93 cases received a to piramate add on therapy and 49 cases a monotherapy The efficiency and side effects were evaluated after 8 weeks titration period and 12 weeks stabilization period Results The total effective rate was 79% and 84% in add on group and monotherapy group respectively The seizure free rate was 28% and 41% in two groups respectively The optimal dosages were 100~200 mg/d in adults and 3~5 mg/d in children Side effects were generally mild Conclusion Topiramate is a highly effective, safe and widespread first line antiepileptic drug which can be used as a monotherapy or add on treatment
