Acta Medicinae Universitatis Scientiae et Technologiae Huazhong 2000;29(5):380-382
To investigate whether low density lipoprotein (LDL), very low density lipoprotein (VLDL), oxidized LDL (OX-LDL) and VLDL (OX-VLDL) could induce the cultured human endothelial cells (ECs) to express high levels of macrophage inflammatory protein lα (MIP-lα) and vascular cell adhesion molecule 1 (VCAM-1) mRNA. LDL and VLDL were isolated from normal human blood donors by density gradient ultracentrifugation and oxidized by Cu2+. The cultured human umbilical vein ECs were incubated with native LDL, VLDL, OX- LDL and OX-VLDL respectively, for 24 h, and in the control group no lipoproteins were added. Total cellular RNA was extracted from the cultured ECs by guanidinium isothiocyanate method. The expression of MIP-lα and VCAM-1 mRNA was detected by dot blotting analysis with two probes of digoxigenin-labeled MIP-lα and VCAM-1 cDNA fragments. The results showed that the cultured ECs could express MIP-lα and VCAM-1 mRNA. Incubation of ECs with LDL and VLDL led to a slightly increased expression of MIP-1α and VCAM-1 mRNA, whereas exposure of ECs to OX-LDL and OX-VLDL resulted in a significant increase ofMIP-1α mRNA expression that was 3. 2- and 2. 5-fold as much as that of the control group, respectively, and the VCAM-1 mRNA expression was increased as well, which was 2. 6- and 2. 17-fold as much as that of the control group respectively. It was suggested that lipoproteins especially oxidized lipoproteins could induce the expression of MIP-1α and VCAM-1 mRNA in ECs, which might play an important role in the recruitment of monocytes into the intima.

OBJECTIVE: To investigate the protective effects of Chaiqin Chengqi Decoction (CQCQD), a compound traditional Chinese herbal medicine, on acinar cells and the changes of intracellular calcium concentration ([Ca(2+)]i) in acute pancreatitis (AP) rats. METHODS: CQCQ serum (CQCQS) was prepared by intragastric administration of CQCQD in SD rats, and AP was induced in SD rats by biliary-pancreatic duct ligation method. Pancreatic acinar cells were isolated by collagenase digestion and co-incubated with the drug serum containing CQCQD in vitro, then cell viability was determined by methyl thiazolyl tetrazolium (MTT) method and intracellular fluorescent intensity (FI) was observed and analyzed to investigate the changes of intracellular calcium concentration ([Ca(2+)]i). RESULTS: Both 5% and 10% CQCQS could improve the viability of pancreatic acinar cells in AP rats (P<0.05), and 10% CQCQS was more effective than 5% CQCQS (P<0.05). [Ca(2+)]i was elevated in AP rats (P<0.05), while the level of [Ca(2+)]i elevation was reduced after CQCQS treatment (P<0.05). CONCLUSION: CQCQD has a protective effect on pancreatic acinar cells of AP rats, which may be associated with its inhibition effect on intracellular calcium overload.

Objective To investigate whether tumor necrosis factor (TNF)a, TNFb and TNFc microsatellite polymorphisms correlate with chronic atrophic gastritis and gastric adenocarcinoma in Chinese Han population. Methods TNFa, TNFb and TNFc microsatellite alleles in 164 healthy subjects, 53 patients with chronic atrophic gastritis and 56 patients with gastric adenocarcinoma were typed using PCR technique combined with High Voltage denaturing PAGE and silver staining. At the same time, the PCR products were cloned and sequenced. Results The frequency of TNFa10 allele was significantly higher in patients with chronic atrophic gastritis than in healthy individuals ( 19.81% vs. 11.89% , P = 0.04 ). However it was not related to age, gender, degree of atrophy or intestinal metaplasia in patients with chronic atrophic gastritis. The frequency of TNFa6b5c1 haplotype homozygote was significantly lower in patients with gastric adenocarcinoma than in healthy individuals ( 1.79% vs. 15.85% , P = 0.006 ). The sequence result revealed that the copy number of dinucleiotide repeating within the same TNFa allele was not consistent with that in the reports from Western countries. Conclusions It should be more accurate and clear to define TNFa alleles. TNFa10 allele is associated with the susceptibility to chronic atrophic gastritis. TNFa6b5c1 haplotype homozygote is negatively associated with gastric adenocarcinoma and thus may play a resistant role in the shifting process from chronic atrophic gastritis to gastric adenocarcinoma.

Objective To observe the effects of marzulene(L-glutamine and sodium gualenate)on cellular immune function in patients with severe acute pancreatitis(SAP). Methods A prospective study method was conducted in which 80 patients with SAP were divided into trial group and control group(each 40 cases)according to the table of random number methods, and in the mean time 20 healthy volunteers were enrolled in the healthy control group. Both SAP groups received standard total parenteral nutrition(TPN)with same quantities of heat and nitrogen, and the trial group additionally accepted intravenous drip of marzulene 0.4 g?kg-1?d-1(once a day)for 7 days, while the control group received the same amount of normal saline intravenous drip for 7 days. One day before treatment and on the 7th day after treatment, the peripheral venous blood samples were collected. The T-lymphocyte proliferation activity was tested by methyl thiazolyl tetrazolium(MTT)assay, and the interleukin-2(IL-2)and interleukin-6(IL-6) secretion levels were detected by enzyme-linked immunosorbent assay(ELISA). Results Compared to the healthy control group, the levels of T lymphocyte proliferation activity〔absorbance(A)value〕and IL-2(ng/L)were markedly decreased and IL-6(ng/L)secretion level was obviously increased before treatment in the two SAP groups(P<0.05 or P<0.01). Compared to prior treatment, the levels of T lymphocyte proliferation activity and IL-2 were significantly increased after 7 days of treatment in the trial group(both P<0.05),and basically restored to normal levels, while in the control group they were progressively decreased(both P<0.05),the levels in trial group being significantly higher than those of the control group(T lymphocyte proliferation activity:1.08±0.27 vs. 0.43±0.25,IL-2:16.5±1.4 vs. 9.4±2.9,both P<0.05),but compared to prior treatment, the level of IL-6 was significantly decreased in the two SAP groups(both P<0.05),the level of IL-6 in control group was recovered to the level of healthy control group, and the level in trial group being significantly lower than that of the control group(18.8±4.5 vs. 22.3±3.1, P<0.05). Conclusions The patients with SAP have cellular immune dysfunction, the manifestations being the suppression of T lymphocyte proliferation and IL-2 and increase of IL-6 release. Early application of marzulene can help to improve the immune function of lymphocytes in patients with SAP.
