6.Research progress of stem cell transplantation in treating spinal cord injury
International Journal of Surgery 2012;39(1):60-63
Spinal cord injury is a traumatic disorder resulting in a functional deficit that usually leads to severe and permanent paralysis. Now several strategies including pharmacological and rehabilitation therapies are not effective treatments for spinal cord injury.Recently stem cell transplantion caused extensive concerns from scholars because of its feasibility in theories and definite effectiveness in experiments.Stem cell types used in spinal cord injury therapy include embryonic stem cells, neural stem cells, mesenchymal stem cells and induced pluripotent stem cells. We extensively review the related articles in recent years, discusse and analyze current situations and perspectives of different stem cells transplantion for treating spinal cord injury.
7. Preparation of folic acid-modified dextran-5-fluorouracil and its cytotoxicity
Chinese Pharmaceutical Journal 2013;48(12):1030-1033
OBJECTIVE: To synthesize folic acid-modified dextran-5-fluorouracil (FA-DEX-5-Fu) conjugates and investigate its cytotoxicity on cells with folate receptors. METHODS: Dextran (DEX) was selected as the macromolecular carrier and the anti-cancer drug 5-fluorouracil (5-Fu) was selected as a model drug to synthesize 5-Fu-DEX, and then folic acid (FA) was linked into 5-Fu-DEX as the targeting ligand to obtain the conjugate, FA-DEX-5-Fu. UV spectra, IR and ESI-MS were used to identify the structure. UV spectrophotometry was used to determine the contents of FA and 5-Fu in the conjugate. MTT assay was applied to examine the cytotoxicity of free 5-Fu, 5-Fu-DEX and FA-DEX-5-Fu on the Human cervical carcinoma cell line Hela (overexpressing folate receptors) and human lung carcinoma cell line A549 (folate receptor deficient, as negative group). RESULTS: 5-Fu and FA were successfully linked with 5-Fu-DEX as confirmed by UV, IR and ESI-MS spectra. The molar ratio of 5-Fu and FA in the conjutate was 2.7:1 as determined by UV spectrophotometry. MTT assay results showed the inhibition rate of FA-DEX-5-Fu on Hela cells was much higher than that on A549 cells. And the inhibition rate of FA-DEX-5-Fu on Hela cells was much higher than those of 5-Fu-DEX and free 5-Fu at the same concentration. CONCLUSION: FA-DEX-5-Fu is successfully synthesized and the conjugate shows significant targeting effect against tumor cells overexpressing folate receptors.
8. Anesthesia selection in the establishment of the model of middle cerebral artery occlusion in rats
Chinese Journal of Cerebrovascular Diseases 2006;3(9):407-411
Objective: To study the effect of intraperitoneal injection anesthesia and endotracheal anesthesia during creating a rat model of middle cerebral artery occlusion (MCAO). Methods: Rats were anesthetized and maintained respectively with chloral hydrate by intraperitoneal injection and inbreathing 2% halothane and 30% O2 via tracheal intubation. MCAO model was created by method of Zea Longa. The time of induction, maintenance, recovery and physiological indexes were monitored, and the result of blood gas analyses, blood glucose, brain temperatures, rectal temperatures, neurological deficit, 48 hours' survival rate and the volume of brain infarction were also studied. Results: Intraperitoneal injection anesthesia inhibited the respiration significantly. Blood gas analysis indicated serious respiratory acidosis (pH 7.29 ± 0.03, PCO2 56.8 ± 4.2 mm Hg, PO2 119.1 ± 15.6 mm Hg). But it appeared approximately normal in rats with halothane anesthesia (pH 7.48 ± 0.06, PCO2 36.2 ± 0.3 mm Hg, PO2 219.1 ± 27.4 mm Hg). There was significant difference between two kinds of anesthesia in pH, PCO2, PO2 (all P < 0.01). Anesthesia with chloral hydrate by intraperitoneal injection can reduce brain and rectal temperature significantly (cerebral cortex in ischemia area 33.3 ± 0.8 °C, striatum 33.6 ± 0.3 °C, rectum 34.7 ± 0.5 °C), However, endotracheal anesthesia affected little on brain and rectal temperature (cerebral cortex in ischemia area 35.1 ± 0.39 °C, striatum 36.2 ± 0.3 °C, rectum 35.1 ± 0.3 °C). There were significant difference between two kinds of anesthesia in brain temperature (P < 0.01), but in rectal temperature they had not significant difference (P > 0.05). There was no significant difference of survival rate at 48 hours and brain infract volume between the two kinds of anesthesia (P > 0.05). Conclusion: This study suggestes that endotracheal anesthesia with 2% halothane and 30% O2 is better than intraperitoneal injection of chloral hydrate. It is effective and less influence on the brain temperature and other relative indexes during creating middle cerebral artery ocelusion rat model.

Journal of Practical Radiology 2001;0(09):-
Objective To research the special manifestations of magnetic resonance cholangiopancreatography(MRCP) in carcinoma of pancreatic head. Methods MRCP in 22 cases with pancreatic head carcinoma confirmed by operation and pathology (including SE sequences with T1WI and fat saturation,FSE with T2WI and fat saturation and MRCP in coronal sectior) were performed before operations.Additional CE MRA on abdominal vessels including abdominal ateries, portal veins and superior mesenteric arteries and their veins in 16 cases also examined before operations. Results Carcinomas of the pancreatic head were diagnosed correctly by MRCP in 21 cases, and 1 case was misdiagnosed as chronic pancreatitis. MRCP all demonstrated abrupt interruption at pancreatic head segments of dilated left common bile duct and main pancreatic duct at the pancreatic head ones, their left segments were dilate with higher signal and both didn't cross each other, because they were destroyed by carcinomas of pancreatic head. Destruction and no cross each other of pancreatic head segments of choledochus and main pancreas duct,it was called "no cross sign". CE MRA demonstrated destruction of abdominal ateries in 3 cases, portal vein in 3 cases and superior mesenteric vein in 4 cases and not being resected. Conclusion "No cross sign"on MRCP examination is of important value in diagnosis of carcinoma of the pancreatic head.
