2.Morphometric Quantitative Analysis of Common Thyroid Benign and Malignant Tumors

Objective To explore the value of morphometric quantitative parameters in differential diagnosis of common thyroid benign and malignant tumors, and establish a data bank as a basis for diagnosing diseases with the help of computer. Methods Using PIPS-2020 image analysis system to make a morphometric analysis in 32 cases of thyroid benign papillary lesion (TBPL), 32 cases of thyroid papillary adenocarcinoma(TPAC), 27 cases of thyroid follicular adenoma(TFA) and 24 cases of thyroid follicular adenocarcinoma(TFAC). Results Among many morphometric quantitative parameters of nuclei, eight including area, circumferential length, diameter,volume,long diameter, short diameter, largest to smallest diameter ratio, and circular rate were selected to analyze. The eight morphometric quantitative parameters between TFA and TFAC had a highly significant difference(P0.01). Conclusion These nuclear parameters have some value in differentiation of common thyroid benign and malignant tumors.
3.Expressions of GLP-1R and CK17 in papillary thyroid cancer and clinical significance

Objective To investigate the expressions of glucagon-like peptide-1 receptors (GLP-1R) and cytokeratin17 (CK17) in papillary thyroid cancer tissue and clinicopathological significance.Methods The immunohistochemistry and immunoblotting method were employed to detect the expressions of GLP-1R and CK17 in normal thyroid tissues (control group,50 cases) and papillary thyroid cancer tissue (observation group,80 cases).Then the correlation between the expressions of GLP-1R and CK17 with pathological factors of thyroid cancer was analyzed.Results Low expression of GLP-1R was found in 22 cases (44.00 %) of the control group,while GLP-1R was expressed in 77 cases(96.25 %) of the observation group,and the difference between both groups was statistically significant(P<0.01).The expression of CK17 was positive in 8 cases(16.00%) of the control group,while 77 cases(96.25 %) in the observation group were CK17 expression positive,and the difference between both groups was statistically significant(P<0.01).The difference between GLP-1 positive expression and clinicopathologic factors had no statistical significance(P>0.05).The difference between CK17 expression with sex and age had no statistical significance (P>0.05),and was obviously related with the tumor diameter,clinical stages and cervical node metastasis of thyroid neoplasms(P<0.05).Conclusion GLP-1R and CK17 all are highly expressed in papillary thyroid cancer tissue and CK17 expression is obviously related with the tumor diameter,clinical stages and cervical node metastasis of papillary thyroid cancer.
4.Myocardial injury in the long-term follow-up after catheter radiofrequency ablation in children

AVNRT, (3)There was no evidence of new arrhythmia occurring for the late electrophysiologic sequelae of RFCA lesions. Conclusion The technique of RFCA is safe, it causes significant minor myocardial injury and takes 3 months to develop from initially resembling coagulation necrosis to fibrosis. There is no evidence of new arrhythmia occurring for late electrophysiologic sequelae of RFC lesions.
5.Hydrophidae identification through analysis on Cyt b gene barcode.

Hydrophidae, one of the precious traditional Chinese medicines, is generally drily preserved to prevent corruption, but it is hard to identify the species of Hydrophidae through the appearance because of the change due to the drying process. The identification through analysis on gene barcode, a new technique in species identification, can avoid the problem. The gene barcodes of the 6 species of Hydrophidae like Lapemis hardwickii were aquired through DNA extraction and gene sequencing. These barcodes were then in sequence alignment and test the identification efficency by BLAST. Our results revealed that the barcode sequences performed high identification efficiency, and had obvious difference between intra- and inter-species. These all indicated that Cyt b DNA barcoding can confirm the Hydrophidae identification.

6. Analysis of sinomenine synthesis pathway based on transcriptome sequencing in Sinomenii Caulis

Objective: In order to explore the expression of sinomenine content control genes, synthetic control sites and expression pathway. Methods: In this study, high performance liquid chromatography (HPLC) was used to determine the content of sinomenine in the roots and stems of 49 Sinomenii Caulis in six populations. Two populations with large multiple differences in sinomenine content were selected, namely Shanxi Baoji and Guizhou Zunyi. The most representative of them were selected, and their roots and stems were taken for transcriptome sequencing and named as HR/LR and HS/LS. Results: Sequencing results showed that 355 201 transcripts were obtained by splicing clean reads, including 275 491 Unigene transcripts. There were 23 562 and 37 143 differentially expressed genes in HR/LR and HS/LS, respectively. GO database analysis showed that the functions of these differentially expressed genes were significantly enriched in aspartic-type endopeptidase activity and aspartic-type peptidase activity, it is speculated that these two enzymes might be encoded. The results of KEGG enrichment explained that the differentially expressed genes were involved in carbohydrate metabolism, protein binding to cell membrane and vitamin C synthesis. The results of qRT-PCR verified the expression of upstream key genes of the isoquinoline alkaloid synthesis pathway and found that it was positively correlated with the accumulation of sinomenine. Conclusion: This study provided a preliminary understanding of the molecular mechanism that caused the difference in sinomenine content, and provided a reference for further understanding of the accumulation rules and synthesis pathways of sinomenine.
7.Effects of different concentrations of sevoflurane on adhesion and expression of CD 44 in human breast cancer cell line MCF-7

Objective To investigate the effects of different concentrations of sevoflurane on adhesion and expression of CD 44 in human breast cancer cell line MCF‐7 .Methods The cells were randomly divided into 4 groups :control group and 3 sevoflurane groups exposed to 1 .7% ,3 .4% and 5 .1% sevollurane for 2 ,4 and 6 h respectively .Cell adhesion rate was detected by adhesion test and the expression of CD44 mRNA and protein was determined by qRT‐PCR .Results in Treat 1 ,2 ,3 ,the adhesion rate at 2 ,4 ,6 h were lower than that before treat ,mRNA expression of CD44 reduced (P<0 .05);the adhesion rate of Treat 2 and Treat 3 at 4 and 6 h were lower than that of 2 h ,and the mRNA expression of CD44 reduced (P<0 .05);the adhesion rate of Treat 2 and Treat 3 at 6 h were lower than that of 4 h ,and the mRNA expression of CD44 reduced (P<0 .05);compared with Control group ,the adhesion rate of Treat 1 ,2 and 3 at 2 ,4 ,6 h were all lower ,mRNA expression of CD44 reduced (P<0 .05);the differences between each group were statisyicaly significant(P<0 .05) .Conclusion Sevoflurane can inhibit cell adhesion in a concentration and duration of exposure dependent manner ,and its mechanism could be related with the down regulation of CD44 expression .
8.Pancreatic trauma: an analysis of 148 cases

Objective To explore the early diagnosis, option of the surgical procedures, preven-tion and treatment of the complications in patients with pancreatic trauma.Methods All patients with pancreatic trauma during the past 20 years were studied retrospectively in respect of sex, age, cause of injury, grade by AAST, style of operation, therapeutic efficacy, complications and factors for death etc.Statistical analysis was made with Chi-square test.Results In all 148 cases of the present series, 132 underwent surgical interventions including simple suture or external drainage alone, distal pancre-atectomy, distal pancreaticojejunostomy or other internal drainage, diverticularization, Whipple's pro-cedure, and Damage Control Surgery (DCS) etc.Postoperative morbidity was 27.83% with a signifi-cant difference of the incidence of pancreatic fistula between pancreas grade Ⅲ-Ⅴ injuries and grade Ⅰ-Ⅱ(P<0.01).The mortality rate was 11.49%.The cause of the deaths was mainly massive bleed-ing due to severe associated injuries(76.47%).Among various grades, the difference of the mortality was not significant (P>0.05).Conclusion To improve the survival rate, it is important to control massive hemorrhage from associated injuries precedes dealing with pancreas trauma.Selection of surgi-cal procedures should be based on whether the main duct is injured.The removing of devitalized tis-sue, adequate external and internal drainage are essential for treatment of pancreatic injuries.Early recognition of pancreatic injury and correct choice of surgical procedures may obviously decrease the in-cidence of postoperative complications.
9.Analysis on characteristics and drug resistance of pathogens in ICU elderly patients with respiratory tract infection

Objective To investigate the distribution characteristic and drug resistance of respiratory tract pathogens in ICU eld‐erly patients to provide the basis for clinical medication and control of nosocomial infection .Methods The isolation situation and drug resistance of pathogens in ICU elderly patients with respiratory tract infection from January 2012 to December 2014 were ret‐rospectively analyzed .Results Among 501 cases of respiratory tract infection ,350 cases were Gram‐negative bacilli infection ,which were mainly P .aeruginosa and A .baumannii;50 cases were Gram‐positive coccus infection ,which was mainly S .aureus ;101 cases were complicating fungal infection ,which was mainly C .albicans .The resistance of P .aeruginosa to imipenem showed upward trend (P<0 .05) ,A .baumannii had higher resistance to commonly used antimicrobial drugs ,but the drug resistance trend had no obvious change(P>0 .05) .Imipenem‐resistant A .bauman(IRAB) ,ESBLs‐producing E .coli and methicillin‐resistant S .aureus (MRSA) in the elderly patients with respiratory tract infection all exceeded 50% of each constitution ratio .Conclusion Multi‐drug resistant bacteria are usually isolated from ICU elderly patients ,their drug resistance rates maintain a higher level .Therefore clinicians should rationally select antibacterial drugs by combining with the laboratory reports ,increase the prevention and management of multi‐drug resistant bacteria and reduce the nosocomial infection occurrence .
10.Expression of Aquaporin-4 in White Matter of Spinal Cord in Rats after Spinal Cord Contusion

Objective To explore the expression of aquaporin (AQP)-4 in white matter of spinal cord after spinal cord contusion (SCC). Methods 88 adult Sprague-Dawley rats were assigned randomly to sham operation group and SCC group. The model was established by Al-len's method. BBB sore was used to assess the motor function of rats. The relative expression of AQP-4 mRNA was determined by Q-PCR technique. The localization of AQP-4 was observed by immunohistochemistry. Results BBB score showed motor dysfunction in SCC group, and it increased 7 and 14 days after SCC (t>5.061, P<0.001). The level of AQP-4 mRNA decreased on the 1st and 3rd days (t>50.44, P<0.001), and increased on the 5th day (t=-3.968, P=0.001), and lasted until the 28th day (t=-4.227, P=0.001) compared with that on the 3rd day. The immunohistochemistry showed AQP-4 was located on the process of glial cell and vascular endothelial cells in white matter of spi-nal cord. Conclusion AQP-4 may play various roles at different stages in SCC.
