1.Interleukin-1β promotes the expression of pSTAT3 in rat retinal Müller cells

Objective To observe the influence of interleukin-1β (IL-1β) on the expression of phosphorylated signal transducers and activators of transcription 3 (pSTAT 3) in rat retinal MOiler cells. Methods For in vitro study cultured Mailer cells were treated with IL-1β of different concentrations (0, 0.1, 1, 5 and 10 ng/ml) for 24 hours. For in vivo study, 32 Sprague-Dawley(SD)rats were divided into 4 groups randomly (control group, 100,500 and 1000 ng/ml group) with 8 rats in each group. After 24 hours of injection with phosphate buffered solution (PBS), or 100, 500, 1000 ng/ml IL-1β into the vitreous treated retinas were evaluated by indirect immunofluorescence and western blotting. Results After 24 hours of incubation without IL-1β, pSTAT3 has little expression in cultured Muller cells, but was up-regulated by 1 ng/ml or higher IL-1β in a dosage-dependent manner (F=46.64, 43.78; P<0.01). pSTAT3 was not expressed in adult rat retina, but was up-regulated by vitreous injection of 100 ng/ml or higher IL-1β in a dosage-dependent manner (F=73.53, 43.70; P<0.01). pSTAT3 expressed mainly in inner nuclear layer and ganglion cell layer. Double-labeling showed that there was no co-staining of pSTAT3 and glial fibrillary treated with IL-1β. Conclusions Expression of pSTAT3 in MUller cells could be activated by IL-1β which may represent one pathway link to reactive gliosis.
2.The regulation of Kir4.1 by pigment epithelium-derived factor in Müller cells under high glucose conditions

ObjectiveTo investigate Kir4.1 expressions in Müller cells under high glucose conditions and treatment of pigment epithelium derived factor (PEDF).Methods Cultured rat Müller cells were divided into control group (5 mmol/L glucose),high glucose group (25 mmol/L glucose),PEDF treatment group (25 mmol/L glucose+ 100 ng/ml PEDF) and intervention control group(25 mmol/L glucose +phosphate buffer solution). Kir4.1 expressions were measured by Western blot and real-time reverse transcription polymerase chain reaction (RT-PCR). Reactive oxygen species (ROS) productions were measured using 2′7′-dichlorofluorescin diacetate and glutathione peroxidase (GPx)expressions were studied by real-time RT-PCR.ResultsBy Western blot and real-time RT-PCR,it was found the expressions of Kir4.1 decreased obviously under high glucose conditions (real-time RT-PCR:t=4.12,P<0.05; Western blot:t=3.53,P<0.05) ; simultaneously,ROS generation was increased (t=3.76,P<0.05) and GPx level was decreased (t=3.18,P<0.05).PEDF treatment inhibited the high glucose-induced Kir4.1 down regulation (real-time RT-PCR:t =3.66,P<0.05 ; Western blot:t =6.43,P<0,01 ) and decreased ROS generations (t=4.11,P<0.05) and increased GPx levels (t=5.12,P<0.01).ConclusionsThe high glucose can supress Kir4.1 expressions in Müller cells by oxidative stress,and PEDF can ameliorate these effects.
3.Network of multiple factors and cascades: hotspots of mechanism studies for diabetic retinopathy

The pathogenesis of diabetic retinopathy is complicated.The vast network of multiple factors including unifying mechanism,inflammatory reaction,neuron degeneration and metabolic memory of glucose,and the four established pathogenic molecular pathways are hotspots of mechanism research for diabetic retinopathy.Nevertheless,these researches may be only one corner of the iceberg of DR mechanism,and we still face enormous challenges in DR mechanism research.Collaboration with multiple disciplines to study the relationship between DR and diabetes and other systemic diseases,search novel therapy targets may increase the result in an unexpected windfall for DR basic research.
4.Application of breast fiberoptic ductoscopy with abnormal nipple secretion(an analysis of 1000 cases)

Background and purpose:Nipple secretion is one of the frequent symptoms in mammary gland disease.Clinically,about 1% breast cancer patients demonstrate with nipple secretion as initial symptom.It could be used as a screening tool for the patients with nipple secretion but no palpated mass and increase the possibility of early diagnosis for breast cancer.In 1997,our country developed a new technique——breast fiberoptic ductoscopy(FDS). It uses a ?(0.7) millimeter optic scope to insert into the nipple discharge canal orifice,and depends on introscope technology to observe and record the normal or abnormal changes in mammary gland ductus.Its advantages are not only in highly diagnostic sensitivity(above 90%),but also in accurate location of tumor,so it can be used to diagnose early breast cancer and mammary gland ductal diseases.This study evaluated the clinical effect of breast FDS in diagnosis and treatment of abnormal nipple secretion.Methods:1 000 patients had been under examination by FDS.Results:Of the 1 000 cases,there were 33 cases((3.3%)) of early breast cancer;412 cases(41.2%) of papilloma and papillomatosis;and 480 cases(48%) of either ductal ectasia and or chronic inflammation.Duct endoscoptic biopsy was successfully performed on 30 cases,220 cases(55%) with chromic inflammation were cured by perfusion.453 cases were received operation.The accordance rate with pathologic eiagnosis was 90.7%.Conclusions:FDS is a very important and new method to diagnose breast.FDS could be inserted into 5th ducta lumen of breast and significantly increase the positive detection of breast cancer.Because the position and nature of the lesions can be determined accurately by FDS,tt is used as intervening treatment for some intraductal papilloma and chronic inflammatory disease.FDS will be of great influence on early diagnoses and treatment of the breast cancer.
5.57 cases of exudative retinal detachment treated with vitrectomy

Objective To review the causes of exudative retinal detachment(ERD),indications of vitrectomy for ERD and its outcomes.Design Retrospective case series.Participants 57 patients with ERD received vitrectomy in EENT Hospital,Fudan Uni- versity.Methods A retrospective analysis of clinical records were conducted.Main Outcome Measures Etiology,vitrectomy indica- tions and its outcomes.Results 57 cases(62 eyes)with ERD received vitrectomy.The disease distribution of vitrectomy were Coats dis- ease in 23 cases(45.2%),familial exudative vitreoretinopathy in 8 cases(12.9%),uveitis in 7 cases(11.3%),retinal hemangioma in 7 cases(11.3%),bullous retinal detachment in 6 cases(9.7%),endophthalmitis in 4 cases(6.5%)and pit of optic nerve in 2 cases(3.2%). Severe ERD and proliferative change were major indication for treatment with vitrectomy.During follow-up period,most patients gained useful vision in early stage.Follow up rate of 6 months or more was 64.9%.The recurrence of ERD was 5.4% and most patients also gained useful vision.Condusion Coats disease is the prominent cause of being treated with vitrectomy in ERD.When proliferation or vitreous hemorrhage happens or macula is involved,vitrectomy should be performed.Most patients can gain useful vision after vitrecto- my.(Ophthalmol CHN,2008,17:52-55)

Objective To investigate the effect of amniotic membrane on the proliferation of YAC-1 in vitro. Methods The YAC-1 cells were cultured with amniotic membrane and their proliferation were observed by MTT automated colorimetry on the lst day, the 3rd day and the 7th day respectively.Results Amniotic membrane could significantly enhance the proliferation of YAC-1 on the lst day, the 3rd day and the 7th day respectively after having been seeded (P<0.01).Conclusion Amniotic membrane can enhance the proliferation of tumor cells (YAC-1), which should be paid attention to during amniotic membrane transplantation for treating ocular surface lesion caused by epibulbar tumors. 9.Inhibitory effects of pigment epithelium derived factor on oxygen-induced retinal neovascularization

Objective To study the inhibitory effects of pigment epithelium derived factor (PEDF) on oxygen-induced retinal neovascularization in mice,and to investigate the possible involvement of interleukin-1β (IL-1β) in the neovascular-inhibitory function of PEDF.Methods A total of 140 postnatal day (P)7 C57BL/6 mice were randomly divided into normal control group,oxygen-induced retinopathy (OIR) model group,PEDF treatment group and PBS treatment control group.All mice except normal control group with their mothers were exposed to (75 ± 2)% oxygen environment for 5 days and then kept in room air for another 5 days to establish the OIR model.Mice in normal control group were kept in room air only.At P12 and P14,respectively,mice in PEDF treatment group received intravitreous injections of 1 μl PEDF (2 μg/μl),while PBS treatment control group received the same volume of PBS (10 mmol/L,pH7.4).All mice were euthanized at P17 and eyes were isolated.The changes of retinal vessels were observed on retinal flat mounts and cryosections by fluorescence microscopy.Retinal specimens were prepared for IL-1β protein and mRNA analysis by Western blot and real time fluorescence quantitative reverse transcription-polymerase chain reaction (Real-time RT-PCR).Results Changes of retinal vessels had been viewed by fluorescence microscopy on flat-mounted retina,the relative retinal neovascularization areas were significantly increased in OIR model group compared with normal control group (t =15.02,P<0.01),and the relative retinal neovascularization areas were obviously smaller in PEDF treatment group than those in PBS treatment control group (t=5.96,P<0.01).Fluorescence staining revealed that retinal vascular tufts were extending from outer plexiform layer (OPL) to ganglion cell layer (GCL) of the retina along with multiple interconnections; Neovascular tufts in OIR model group and PBS treatment control group were presenting distinctly more than those of normal control group and PEDF treatment group.The specific expression levels of IL-1β protein in retinas of OIR mice by Western blot analysis were higher than those of normal control group(t=3.35,P<0.05),While these of PEDF treatment group showed a considerable decline in comparison with PBS treatment control group (P<0.01),and there were no difference in normal control group and PEDF-treated group (F=11.764,P>0.05).Similarly,expression levels of IL-1β mRNA tested by Real-time RT-PCR were obviously increased in the OIR model group when compared to normal control group(t =4.43,P < 0.01).After treated with PEDF,expression levels of IL-1β mRNA showed a considerable decrease when compared to PBS treatment control group (P < 0.01),and there were no difference in normal control group and PEDF-treated group (F=11.15,P>0.05).Conclusions PEDF can inhibit oxygen-induced retinal neovascularization.The mechanism may be related to that PEDF can downregulate the expression of IL 1β in retina.
