1.Expression of eIF4E in acute myeloid leukemia

Objectives To analysis the expression difference of eIF4E in bone marrow between the acute myeloid leukemia (AML) and the non-tumor patient,before and after induction therapy,and the different subtypes of AML,and to explore the relation between eIF4E and other molecular biology abnormalities in AML.Methods The bone marrow specimens of newly diagnosed AML and non-tumor control patients were collected.The expression level of eIF4E was detected by RT-PCR.Results The positive rate of eIF4E was 65.2 % (101/155) in AML.eIF4E turned to negative in 12 patients (17.6 %) who got complete remission after induction therapy.eIF4E was negatively expressed in bone marrow of the nontumor control patients.The positive rates of eIF4E were 75.0 % (15/20) and 80.8 % (21/26) in M4 and M5 type AML,respectively,and was 59.6 % (65/109) in other subtype AML (P > 0.05).FLT3/ITD gene mutation was found in 26 cases of newly diagnosed AML.The eIF4E expression and FLT3-ITD gene mutation were independent each other (P > 0.05).Conclusions eIF4E is positive in most of the newly diagnosed AML and turns to negative in part of AML achieving complete remission.The expression of eIF4E is no difference among different subtype of AMLs.eIF4E and FLT3-ITD gene mutation are independent each other.
2.The Study of Establishment of Virtual Three-dimensional Cerebral Aneurysm Models with the Three-dimensional Moulding Software

Objective To study the establishment methods of virtual three-dimensional cerebral aneurysm models by using three-dimensional moulding software.Methods The virtual model including the cerebral arteries and the aneurysms were established by using the three-dimensional moulding software of 3D Studio MAX R3 in 20 cases of normal cerebral DSA image and 216 cases of cerebral aneurysms DSA image as the original.Results ①In comparison with DSA image,the virtual cerebral arteries possessed similarity in appearance,and had a benefit for understanding the vascular three-dimensional spatial relation in visual sense.The virtual cerebral arteries could display the virtual guide wire and catheter in semitransparent.The several models of different variant anatomy could be established easily based on the copy files of the virtual cerebral arteries model;②In comparison with DSA image,virtual cerebral aneurysm could display the location of development,direction of growth and feature of appearance correctly.The several aneurysms models of different location,direction of growth and appearance could be established easily based on the copy files of the virtual cerebral aneurysms model.Conclusion The virtual three-dimensional cerebral aneurysm models that were established by the software method can display the three-dimensional spatial relation of the cerebral arterial system and aneurysmal shape distinctly,and would be useful in assisting morphology study and guiding clinical work.
3.Oxidative stress and inflammatory changes in the lung caused by cigarette smoking exposure in mice and the effect of smoking cessation

Objective To observe the changes of pulmonary oxidative stress after cigarette smoking exposure,its re-lationship with inflammatory cytokines,and the effects of smoking cessation. Methods Fifty male BALB / c mice were randomly divided into the smoke exposure group,smoke cessation group,and the controls. Mice in smoke cessation group were exposed to cigarette smoking for 16 weeks. On 4,8,and 12 week after smoking cessation mice were executed and the bronchoalveolar lavage fluid(BALF)and lung tissue were collected. The morphologi-cal alternations of lung tissue were observed. Mean length of interval and mean alveolar number were measured. Total cell numbers in BALF were counted. Superoxide dismutase(SOD)activity was measured with hydroxylamine method,malondialdehyde(MDA)level was measured with TBA method. The levels of pulmonary interleukin-8 (IL-8)and tumor necrosis factor-α(TNF-α)in BALF and lung tissue homogenate were measured with ELISA. Re-sults Compared with the mice in the controls,emphysematous changes were remarkable in the lung of cigarette ex-posed mice,the total cell numbers in BALF were increased significantly(P < 0. 05)and reduced gradually after smoking cessation(P < 0. 05). SOD and MDA levels increased remarkably in the cigarette exposure group(P <0. 05),and declined gradually after smoking cessation. The levels of IL-8 and TNF-α in BALF and lung tissue ho-mogenate in the smoke exposure group increased significantly( P < 0. 05),and lowered time-dependently after smoking cessation,but not reached to normal level even 12 weeks after smoking cessation. SOD and MDA levels were positively correlated with the cytokine changes. Conclusion Abnormal oxidative stress in the airways caused by cigarette smoking exposure was merely partially reversed after smoking cessation. And the inflammation remains persistent concomitantly.
4.Expression and significance of LBP mRNA in early liver tissue after burn combined with endotoxemia in rats

Objective To explore the pathogenesis of early liver damage after burn combined with endotoxemia in rats. Methods A total of 156 healthy Wistar rats were randomized into burn combined with injection of endotoxin group (BCEG), single burn group (SBG), single endotoxin injection group (SEG), and normal control group (NCG). An animal model of rats inflicted by 20% TBSA Ⅲ degree burn combined with intraperitoneal injection of lipopolysaccharide (LPS) was used for this experiment. The changes of liver morphology, the hepatic functions, and the localization of liver LBP mRNA were determined by light microscopy and electron microscopy, quantitative analysis, and in situ hybridization(ISH). 5.Determination of dynamic parameters of interaction between methotrexate enantiomer and dihydrofolate reductase by affinity capillary electrophoresis

Objective To develop a method for the determination of the dynamic parameters of interaction between methotrexate(MTX)enantiomer and dihydrofolate reductase(DHFR).Methods An affinity capillary electrophoresis(ACE)method was adopted.Using bare fused-silica capillary,the electrophoresis buffer was 50 mmoL/L sodium tetraborate with 0.2%Brij-35,pH 9.50.The temperature of separation was controlled at 25℃ and a voltage of 25 kV was applied.The separation of the reaction mixture was performed at a wavelength of 254 nm.The difference of peak areas about the product was used to calculate the inhibitory rate and IC50 values.Results We establish the detection method for the dynamic parameters of interaction between MTX enantioner and DHFR.The separation of the reaction mixture could be achieved within 30 min.The IC50 value of D-(-)-MTX and L-(+)-MTX were 3.17×10-7 and 2.48×10-8mol/L,respective.The IC50 value of the D-(-)-MTX was 31.67×10-8mol/L,the L-(+)-MTX was2.48×10-8mol/L.The IC50 value of the D-(-)-MTX was about 13 times higher than that of the L-(+)-MTX.Conclusions ACE is a rapid.simple and accurate method that can be used to monitor DHFRdynamic reaction.The IC50 values of MTX enantiomer were quite different.The result first indicated that reaction between MTX enantiomer and DHFR had three-dimensional selection.
6.Analysis of Plasma Levels of Omentum-1 and Vaspin in Patients With Acute Coronary Syndrome

Objective: To observe the changes of plasma levels of omentin-1 and vaspin in patients with acute coronary syndrome (ACS). Methods: Our research included in 2 groups: ACS group, the patients with conifrmed diagnosis in our hospital from 2013-05 to 2013-09,n=100 including 52 male, 48 female and Control group, the patients with simultaneous coronary angiography excluded CAD,n=88. Plasma levels of omentin-1 and vaspin were examined by ELISA and the differences were compared between 2 groups. Results: Plasma levels of omentin-1 and vaspin in ACS group were lower than Control group (t=2.718,P<0.05) and (t=2.416,P<0.05). Correlation analysis presented that in ACS group, omentin-1 level was negatively related to total cholesterol (TC) (r=-0.702,P<0.05) and C-reactive protein (CRP) (r=-0.714,P<0.05); vaspin was negatively related to TC (r=-0.655, P<0.05) and CRP (r=-0.587,P<0.05); Omentin-1 was positively related to vaspin (r=0.643,P<0.05). Multiple linear stepwise regression analysis showed that plasma levels of omentin-1 and vaspin might be affected by CRP, TC and LDL-C. Conclusion: ACS patients had decreased plasma levels of omentin-1 and vaspin; in addition, omentin-1 and vaspin might be involved in lipid metabolism.

Objective To examine the effect of endotoxin on myocardial lipid metabolism and determine it′s possible mechanism Methods Sixty nine male Wistar rats (350 450g) were randomly divided into 2 groups: in vivo group and in vitro group In in vivo group endotoxin 0 1?g?kg -1 (small dose)or 1mg?kg -1 (large dose) was injected intrabdominally 3h after endotoxin administration the animals were anesthetized Heart was excised and passively perfused in a Laugendorff apparatus with oxygenated Krebs Heuseleit buffer at 37℃ Non esterified fatty acids (NEFA)and triacylglycerols(TAG) were used as energy supplying substance when the isolated heart had been stabilized for 15min NEFA or TAG was slowly added to the perfusate circuit 1 ml of perfusate was drawn every 10 min until the end of experiment (80 min ) for determination of the effects of endotoxin on NEFA or TAG oxidation and utilization by isolated heart At the end of experiment heart was harvested and frozen for measurement of myocardial lipids and lipoprotein lipase activity In in vitro group endotoxin was added to perfusate (1mg/ml) after the heart was excised and passively perfused In control group endotoxin was replaced by normal saline Results Large dose of endotoxin administered in vivo decreased NEFA oxidation rate by 30% in perfused isolated heart and caused increased accumulation of lipid in myocardial tissue By contrast endotoxin administered in vitro increased oxidation rate of TAG accompanied by an increase in myocardial lipoprotein lipase activity Administration of endotoxin in vivo and in vitro also caused accumulation of various exogenous lipids in myocardium in both NEFA and TAG perfused heart Conclusions This study shows that endotoxin has a significant effect on myocardial lipid metabolism The effect of endotoxin could be either direct or indirect according to the different lipids available
9.Changes of interleukin-17A and interleukin-22 in the lung of cigarette smoke exposed mice and the effect of N-acetylcysteine

Objective To investigate the changes of interleukin-17A(IL-17A)and interleukin-22(IL-22)in the lung of cigarette smoke exposed mice and the effect of smoking cessation and N -acetylcysteine (NAC )treatment . Methods BALB /c mice in experimental groups were exposed to cigarette smoke.Then the smoke-exposure was stopped and mice were treated with NAC gavage.The mice were executed 1,2,and 3 months after smoking cessa-tion.Lung tissue sample and bronchoalveolar lavage fluid(BALF)were collected.HE staining was used to observe the pathologic changes of the lung and ELISA was used to measure the level of IL-17A and IL-22.Results Con-siderable emphysematous changes was found in the lung of mice exposed to cigarette smoke.Compared with the controls,the level of IL-17A and IL-22 elevated remarkably in pulmonary tissue and BALF after smoking exposure and declined gradually after smoking cessation.Additional NAC gavage treatment enhanced the decline tendency. Conclusion IL-17A and IL-22 might play a complex role in the chronic inflammatory changes of lung in mice ex-posed to cigarette smoke.
10.Effect of melatonin by nasal feeding on sleep in tracheotomised patients in ICU

Objective To discuss the effect of melatonin on sleep and agitation in patients who had undergone tracheotomy in ICU.Methods Sixty-six tracheotomisd patients of ASA Ⅱ or Ⅲ in ICU were included,age 18-60 years,weaning from mechanical ventilation not receiving continuous sedation.We conducted a double-blind, placebo-controlled randomized pilot study comparing melatonin with placebo in such patients.They were allocated into two groups by compute randomiza-tion-melatonin group(n =32)and control group(n = 34).We administered either oral melatonin (3 mg)or placebo at 20∶00 for 7 days.We collected pre-and post-dosage blood samples on days 1 and 3 to confirm drug delivery.The hours of sleep at day and night were assessed by the bedside nurse as the primary outcome.And the secondary outcome included comparison of the incidence of agitation, assessed by score on the Riker Sedation Agitation Scale,and requirement for sedatives or haloperidol to settle agitation.Results Pre-treatment melatonin levels in the two groups were similarly low.Post-treatment,melatonin levels increased significantly in the melatonin group compared with the placebo group (3 233±1 961 )pg/ml versus (3.8 ± 2.2 )pg/ml(P < 0.05 ).And subsequently the observed sleep time in the two groups was (1 1.9±2.0)hours for melatonin vs (7.8±1.6)hours for placebo, respectively (P < 0.01 ).The incidence of agitation was significantly lower in the melatonin group (0% vs 8.8%).Conclusion Melatonin is well absorbed,and a standard dose increases blood levels approximately 1000-fold.Furthermore it would help the tracheotomisd patients sleep and ease agitation in ICU.
