Objective To observe the effect and safety of helicobacter pylori eradication by doxycycline, levofloxacin, bismuth potassium citrate combined with omeprazole. Methods 240 patients who infected helicobacter pylori were selected, and were randomly divided into four groups:treatment group(doxycycline, levofloxacin, bismuth potassium citrate, omeprazole),control group A(doxycycline, levofloxacin, omeprazole),control group B(amoxicillin, clarithromycin, bismuth potassium citrate, omeprazole),control group C (amoxicillin, clarithromycin, omeprazole), 60 patients in each group.Helicobacter pylori eradication treatment for 7 days, patients with gastritis stop after the course of the treatment, peptic ulcer patients continue taking omeprazole 5 weeks after 7 days treatment. All patients stopped medicine 2 weeks accepted 14 carbon breath test. And adverse events were investigated. Results In the treatment group and the control group A,B,C,the helicobacter pylori eradication rates were 93.3%,80.0%,91.7%, 75.0%.Compared with group A,the treatment group helicobacter pylori eradication rate increased significantly(χ2 =4.61,P <0.05 ).Compared with group C,the treatment group helicobacter pylori eradication rate increased significantly(χ2 =7.57,P <0.05).But compared with group B,the treatment group helicobacter pylori eradication rate had no significant difference(χ2 =0.12,P >0.05).Compared with the treatment group, group B had high inci-dence of oral odor(χ2 =6.56,P <0.05).Compared with the treatment group, group C had high incidence of oral odor (χ2 =5.46,P <0.05).Conclusion Doxycycline, levofloxacin, bismuth potassium citrate combined with omeprazole can improve the helicobacter pylori eradication rate significantly, and with low price, has little side effect,no need of penicillin skin test, can used for the patients poverty or allergic to penicillin.

To clone and sequence the human tissue kallikrein gene of Chinese, and to construct eukaryotic expression recombinant of KK, total RNA was extracted from human pancreas and human tissue kallikrein gene cDNA was amplified by PCR after reverse-transcription by using Oligo(dT) primer. The original kallikrein cDNA was recovered and filled with Klenow enzyme and inserted into KS plasmid. After restriction endonuclease digestion, KK cDNA was sequenced by ABI377 analyzer. Then the KK gene was amplified from pBluescript KSKK and inserted into pcDNA3. A sequence comparison showed that the cloned kallikrein gene was only one nucleotide different from that reported in the Genbank. The coding amino acid was Asp in the Genbank gene, while the coding amino acid of Chinese kallikrein gene was Asn. The KK cDNA fragment was inserted into the eukaryotic expression vector pcDNA3. The cloned kallikrein gene and the pcDNA3KK can be used for further study in gene therapy.. .

Objective To study the effects of lucentis preventing vitreous hemorrhage after vitrectomy for diabetic retinopathy combined with iris neovascularization. Methods The clinical data of 70 patients (70 eyes) underwent vitrectomy for diabetic retinopathy combined with iris neovascularization during 2009 to 2015 in our hospital were retrospectively analyzed. The control group (30 eyes) accepted panretinal photocoagulation (PRP),re-vitrectomy,cyclocryotherapy,and the study group (40 eyes) had the 0.5mg lucentis in addition. The follow-up time was 3 months, and the visual acuity,IOP,vitreous hemorrhage,INV regression and complication were observed. Results At 1 month,2 months in the follow-up,the visual acuity of study group was better than the control group, but there was no significant difference along with the follow up.The average preoperative IOP was (26.312 ±4.566) mmHg (1 kPa =7.5 mmHg) in the study group and (24.586 ±5.783) mmHg in the control group, and at the end of the follow up,IOP was (18.576 ±4.762) mmHg in the study group and (28.587 ±7.786) mmHg in the control group, there was statistical difference between the two groups (P =0.041).The intraoperative and postoperative anterior chamber or vitreous hemorrhage occurred in 15 cases,5 cases of the control group, and 3 cases,1 case of the control group, there were significant differences (all P =0.000).At the end of the follow up,3 eyes (7.5%) developed to NVG in the study group and 10 eyes (33.3%) in the control group, there was statistical difference (P < 0.05).Conclusion Lucentis can effectively eliminate the new vessels, reduce the incidence of neovascularization glaucoma and vitreous hemorrhage for patients after vitrectomy for diabetic retinopathy combined with iris neovascularization, and improve the visual acuity in a short time.

BACKGROUND:Recently, there are many documents al over the world reporting hypotoxicity infection after fracture internal fixation surgery, but reports are different on whether it is necessary for chronic hypotoxicity infection internal fixation removal post surgery. There are no fixed judgment criteria of curative effects, which leads to inexact conclusion of treatment method. <br> OBJECTIVE:To observe the curative effects of taking rifampicin and ciprofloxacin with transfer of skin flap in the treatment of chronic hypotoxicity infection with sinus formation after fracture surgery on tibial plateau and ankle. METHODS:A total of 56 cases of chronic hypotoxicity infection after fracture surgery of tibial plateau and ankle were col ected from September 2005 to December 2012. 30 cases in the therapy group were treated with <br> levofloxacin and rifampicin with transfer of skin flap. 26 cases in the control group were treated with conventional intravenous antibiotics with local debridement to remove internal fixation. In both groups, the course of disease was 3 to 6 months. The erythrocyte sedimentation rate, kidney function and radiographic indices were reviewed every 1 to 3 months, and curative effects were evaluated. <br> RESULTS AND CONCLUSION:During the fol ow-up visit of 6-24 months, no recurrence happened to the cured and improved patients. In the therapy group, 24 cases were cured, 4 cases were improved, and 2 cases were invalid, with a total effective rate of 93%. In the control group, 11 cases were cured, 6 cases were improved and 9 cases were invalid with the total effective rate of 65%.χ2 test showed that therapeutic effects were significantly better in the therapy group than in the control group (P<0.05). These data indicated that rifampicin and ciprofloxacin with transfer of skin flap for chronic hypotoxicity infection with sinus formation after fracture surgery of tibial plateau and ankle showed good curative effects.

OBJECTIVE:
To explore the genetic basis for 4 patients with globozoospermia.
METHODS:
Semen and blood samples were collected from the patients for the determination of sperm concentration, viability, survival rate, morphology and acrosome antigen CD46. Meanwhile, DNA was extracted for whole exome sequencing (WES), and candidate variants were validated by Sanger sequencing.
RESULTS:
All of the four patients were found to harbor variants of the DPY19L2 gene. Patients 1 ~ 3 had homozygous deletions of the DPY19L2 gene. Sanger sequencing confirmed that the DPY19L2 gene in patient 3 was disrupted at a recombination breakpoint area BP2, resulting in nonallelic homologous recombination and complete deletion of the DPY19L2 gene. Patients 2 and 3 respectively harbored novel homozygous deletions of exons 2 ~ 22 and exons 14 ~ 15. Patient 4 harbored heterozygous deletion of the DPY19L2 gene, in addition with a rare homozygous deletion of the 3' UTR region.
CONCLUSION
DPY19L2 gene variants probably underlay the globozoospermia in the four patients, which has fit an autosomal recessive pattern of inheritance and the characteristics of genomic diseases.
