1. The Effect of Radix Ranuncoli Ternati on the Expression of Granule Lytic Peptide and Bactericidal Ability of T Lymphocyte in Peripheral Blood Lymphocytes of Patients with Pulmonary Tuberculosis

OBJECTIVE: To explore the effect of Radix Ranuncoli Ternati on the expression of granule lytic peptide and bactericidal ability of T lymphocyte in peripheral blood lymphocytes of patients with pulmonary tuberculosis. METHODS: Peripheral blood lymphocytes were treated with Ranunculus. MTT assay was used to detect cell proliferation. Real-time PCR was used to detect the mRNA level of GLS in patients. RESULTS: Radix Ranuncoli Ternati could enhance expression of Tubercle bacillus. The RESULTS of MTT assay indicated that the rate of peripheral blood lymphocytes apoptosis inducted by the Radix Ranuncoli Ternat. The Western blotting RESULTS showed that Radix Ranuncoli Ternati made the expression levels of GLS in peripheral blood lymphocyte increasing. When the concentration of the drug is 200 mg·L-1, it can effectively induce the activation of tuberculosis resting bacteria. The expression of GLS and the level of mRNA were increased in peripheral blood lymphocytes. CONCLUSION: Radix Ranuncoli Ternati may increase the expression of GLS and enhance the body's killing effect on pathogenic bacteria to achieve the therapeutic effect.
2. Metabolites of tannins in Punica granatum L. husk in rat urine

OBJECTIVE: To study the in vivo metabolism of granatum tannins by liquid chromatography-mass spectrometry and clarify their metabolic pathways. METHODS: The rat urine samples were collected after intragastric administration of tannins preparation of Punica granatum L. husk. The analysis was performed on a Venusil MP-C18 column with gradient elution system of acetonitrile and water containing 0.1% formic acid. Using electrospray ionization (ESI) in negative ion mode with scanning range from m/z 100 to 1000, the metabolites were identified by comparing the MS spectra and MS data with the literatures. RESULTS: Seven metabolites were detected in the rat urine, among which six were identified according to their second mass spectra to be U-1 (urolithin B), U-2 (urolithin A methyl ether), U-3 (urolithin A), U-4 (urolithin A glucuronide), U-5 (ellagic acid) and U-6 (urolithin C). CONCLUSION: Tannins in Punica granatum L. husk are metabolized after oral administration mainly through hydrolysis, glucuronidation gly-cosidation, decomposition and methylation reaction to generate a series of derivatives of urolithin. These results preliminarily indicate the metabolic pathway and material basis of the pharmacological activity of tannins in Punica granatum L. husk in rats.
3. Study on effect of tannins in Granati Pericarpium on renal fibrosis in rats

Objective: To study the improvement of tannins from Granati Pericarpium on renal fibrosis in rat. Methods: SD rats were randomly divided into five groups, Sham operation, model, Valsartan, high-dose (60 mg/kg), and low-dose (20 mg/kg) tannins groups. Renal fibrosis model of rats was constructed using left ureter ligation method except the rats in the Sham operation group in which the left side ureter of rats was not ligated or cut. The ig administration started at the day of operation, Urinary protein of 24 h, the concentration of Ca2+ and P3+ in urine, and the levels of Scr and BUN in serum were detected in 1-3 week after the operation. The staining of HE and Masson to the kidney tissue was carried out to investigate how tannins from Granati Pericarpium could impact the indexes of renal fibrosis in rats on the morphological basis. Results: The results of urine and blood test showed that the levels of urine protein, Scr, and BUN were deviated from the normal level. The concentration of Ca2+ was increased, while the concentration of P3+ was reduced. The staining of HE and Masson showed that the renal tissue was damaged, the left side of kidney was found renal fibrosis, tannins from Granati Pericarpium could improve the renal fibrosis in rats in some extent. Conclusion: Tannins from Granati Pericarpium have the great improvement on the renal fibrosis in rats and are able to adjust the disorders of Ca2+ and P3+ in rats.
4. Determination of ropivacaine in rat plasma with liquid chromatography-tandem mass spectrometry

Objective: To develop an LC-MS/MS method for the determination of ropivacaine in rat plasma. Methods: The chromatographic separation was achieved on an Agela MP C18(2.1 mm×50 mm,3 µm)analytical column. The multiple reaction monitoring(MRM) mode of the positive ion was adopted in the MS detection, and the precursors to the product ion transitions of m/z 275.2/126.1 and 326.0/291 were used to measure ropivacaine and internal standard (midazolam). Results: The method was linear over the ropivacaine concentration range of 0.5-250 ng/ml with the correlation coefficients r of 0.9946. The average inter-day precision values (RSD) were 1.32%-6.74%, the matrix effect values were 95.5%-102.6% and the average recovery values were 89.8%-104.7%. Conclusion: The analysis methods for ropivacaine are simple, accurate and sensitive, thus can be used for the pharmacokinetic research of ropivacaine incorporated cubic liquid crystal in rat.
5.Effect of Bixie deacidification fang on hyperuricemia mouse model and its effect on the expression of renal urate transporter

Objective To investigate the anti-hyperuricemia effects of Bixie deacidification fang on hyperuricemia mice and its mechanism of renal protein transport. Methods The effects of Bixie deacidification fang were investigated on hyperuricemia mice induced by potassium oxonate. Bixie deacidification fang was administered to hyperuricemia mice daily at doses of 220, 440 and 880 mg/kg for 10 days, and allopurinol (5mg/kg) was given as positive control. Serum and urine levels of uric acid and creatinine were determined by colorimetric method. Simultaneously, protein levels of urate transporter 1 (URAT1) and organic anion transporter 1 (OAT1) in the kidney were analyzed by Western blot. Results Compared with the model group, high-dose of Bixie deacidification fang inhibited xanthine oxidase (XOD) activities in serum (18.12±1.33 u/L) and that in liver (70.15±5.20 u/g protein) (P<0.05), decrease levels of serum uric acid (2.04 ± 0.64mg/L) (P<0.05) and serum creatinine (0.35±0.18µmol/L) and blood urea nitrogen (BUN)(8.83±0.71mmol/L) (P<0.05), ncreased levels of urine uric acid (38.34±8.23mg/L), urine creatinine (34.38±1.98mmol/L), down-regulated of URAT1 and up-regulated of OAT1 protein expressions (P<0.05) in the renal tissue of hyperuricemia mice. Conclusion Bixie deacidification fang recipe may promote the excretion of uric acid in the kidney by up-regulating the expression of OAT1 protein to promote the excretion of uric acid, and down-regulating the expression of URAT1 protein to inhibit the reabsorption of uric acid.
6.Golgi Transport 1A Promotes Cell Proliferation and Metastasis of Papillary Thyroid Carcinoma

ObjectiveTo investigate the expression level of Golgi transport 1A (GOLT1A) in thyroid carcinoma and its effects on the proliferation, migration, invasion, and epithelial-mesenchymal transition (EMT) of thyroid carcinoma cells. MethodsThe expression of GOLT1A in thyroid carcinoma was analyzed online by tumor immune estimation resource (TIMER), the University of Alabama at Birmingham cancer data analysis portal (UALCAN), gene expression profiling interactive analysis 2 (GEPIA2). The expression level of GOLT1A in thyroid carcinoma cells was detected by real-time fluorescence quantitative polymerase chain reaction (RTFQ-PCR) and western blot. Cell counting kit-8 (CCK-8) assay, colony formation assay, and transwell assay were used to detect the effects of GOLT1A expression on the proliferation, migration, and invasion of thyroid carcinoma cells. Western blot assay was used to detect the effect of GOLT1A on the expression of EMT-related genes including E-cadherin, vimentin, and N-cadherin. ResultsThe online analysis of GEPIA2, TIMER, and UALCAN showed that the expression of GOLT1A was higher in thyroid carcinoma than in normal tissues, and the expression of GOLT1A in thyroid carcinoma cells was significantly higher than in normal control cells. Knockdown of GOLT1A inhibited TPC1 cell proliferation, migration, and invasion. The expression of E-cadherin increased and the expressions of N-cadherin and vimentin decreased in GOLT1A knockdown TPC1 cells. Overexpression of GOLT1A promoted BCPAP cell proliferation, migration, and invasion. The expression of E-cadherin decreased and the expressions of N-cadherin and vimentin increased in GOLT1A overexpression BCPAP cells. ConclusionGOLT1A is highly expressed in thyroid carcinoma and can promote the proliferation, migration, and invasion of thyroid carcinoma cells.
7.Epidemic characteristics and prevalence trends of syphilis in Wuhan in 2010-2021

Objective To understand the characteristics and epidemic trend of syphilis in Wuhan from 2010 to 2021, and to provide a reference for the formulation of further prevention and control measures. Methods Data on Wuhan syphilis epidemics from 2010 to 2021 was collected from the China Disease Prevention and Control Information System. The epidemic characteristics and trends of syphilis were analyzed. Results From 2010 to 2021, a total of 36 628 confirmed cases of syphilis were reported in Wuhan, and the reported incidence rate decreased from 29.70/100 000 to 29.62/100 000, with an average annual decrease of 0.33% (P=0.843). Among them, the reported incidence of stage 1 syphilis decreased by 14.37% annually (P<0.001), stage 2 syphilis decreased by 18.59% annually (P<0.001), stage 3 syphilis decreased by 2.02% annually (P=0.523), and fetal syphilis decreased by 22.90% annually (P<0.001), while recessive syphilis increased by 9.19% annually (P<0.001). The ratio of male to female increased from 1.04:1 to 1.36:1 (P<0.001). The proportion of people aged 60 and above increased from 18.44% to 31.17% (P<0.001), and the 35-59 age group had the highest number of reported cases of syphilis in both men and women. Recessive syphilis was most common in men and women aged over 18 years old, and the proportion of recessive syphilis increased with age. Conclusion The prevention and control of syphilis has achieved phased results, especially in mother-to-child interruption and non-recessive syphilis control. On the basis of continuing to consolidate and strengthen the achievements of the current stage, the future syphilis prevention and control work should focus on and respond to the increasing trend of recessive syphilis cases, and take more targeted interventions for people over 60 years old, so as to further curb the spread of syphilis.
8.Centralized blood screening across the regions of Hubei province: a retrospective survey

【Objective】 To retrospectively analyse the quality status and annual trend of centralized blood screening across the regions of Hubei province, and discuss the effect and advantages of the application of centralized blood screening across blood establishment. 【Methods】 Blood screening results of five blood establishment across Hubei province from 2005 to 2019 were statistically analyzed. The sample size, overall rate of qualified blood donations, overall and annual rate of positive testing items were compared among regions. 【Results】 There were differences in blood quality among different regions of Hubei province. In Wuhan, Ezhou, Xiantao, Tianmen and Qianjiang, the overall rate of qualified blood donations was 97.1%(2 466 396/2 538 871), 96.5%(85 405/88 547), 95.2%(59 087/62 047), 95.5%(68 920/72 146) and 94.7%(34 274/36 188), respectively; the overall deferral rate of ALT was 1.472% (37 374/2 538 871), 2.024% (1 792/88 547), 2.840% (1 762/62 047), 2.476% (1 786/72 146) and 3.777% (1 367/36 188), with significant differences across regions; the overall positive rate of HBsAg was 0.587% (14 897/2 538 871), 0.664% (588/88 547), 1.094% (679/62 047), 1.282% (925/72 146) and 0.862% (312/36 188), with significant differences across regions. The deferral rate of ALT showed a downward trend by year in these five regions from 2005 to 2019.An decreasing trend in the seroprevalence of HBV was noticed in Wuhan, Tianmen, and Qianjiang, HCV in Wuhan, Ezhou, and Tianmen, as well as syphilis infections in Wuhan. 【Conclusion】 Centralized blood screening can optimize the allocation of testing resources, reveal problems during blood collection and supply and improve the ability to guarantee blood safety. Future efforts are needed to expand the scope of centralized blood screening and seek for a centralized screening mode suitable for local situation.
9.Influencing factors of stroke symptoms in middle-aged and elderly patients with hypertension in the community

Objective To understand the influencing factors of stroke symptoms in middle-aged and elderly patients with hypertension in the community, and to provide reference for the prevention and control of stroke. Methods A total of 410 cases of middle-aged and elderly hypertensive patients registered in community health service centers (or health centers) were randomly selected from 2 administrative regions of Wuhan to conduct a unified questionnaire survey. Chi-square test, analysis of variance and multivariate logistic regression were used to analyze the influencing factors of stroke symptoms in hypertensive patients. Results Of 31.83% the study subjects had stroke symptoms. The analysis of the results showed that high monthly per capital household income, adequate fruit intake and high level of dietary knowledge were protective factors for stroke symptoms in middle-aged and elderly patients with hypertension. Hypertension complications, annual medical check-ups in the hospital and a history of stroke / heart disease in immediate family members were risk factors for stroke symptoms. Conclusion Improving the health awareness of middle-aged and elderly people, vigorously promoting health education and raising the level of social security for low-income groups can reduce the risk of stroke in middle-aged and elderly patients with hypertension.
10.Establishment and application of a method for the determination of plasma concentration of delamanid

OBJECTIVE To establish and apply a method for the determination of plasma concentration of delamanid (DLM). METHODS After plasma samples were treated with methanol precipitation, LC-MS/MS was adopted to determine the plasma concentration of DLM. The chromatographic column was Phenomenex SynergiTM Fusion-RP with mobile phase of methanol-0.1% formic acid solution (gradient elution). The column temperature was 40 ℃ , the flow rate was 0.3 mL/min, and the sample size was 1 μL. The ion source was electrospray ion source, and positive ion scanning was carried out in multi-reaction monitoring mode. The DLM ion pair used for quantitative analysis was m/z 535.0→352.0. The plasma concentration of DLM in 6 multidrug resistant tuberculosis (MDR-TB) patients were determined by the LC-MS/MS method. RESULTS The linear range of DLM was 0.05-8 μg/mL (r=0.999 5), and the lowest limit of quantitation was 0.05 μg/mL. RSDs of intra-batch and inter-batch precision were all less than 10%. The accuracy ranged 92.7%-104.9%. Average matrix effect was 94.3%-107.5%. Average recoveries were 93.2%-98.1%. The plasma concentration of DLM in 6 MDR-TB patients ranged from 0.61-2.76 μg/mL, with an average of (1.67± 0.74) μg/mL. CONCLUSIONS The established LC-MS/MS analysis method has good specificity, high sensitivity, accuracy and precision, and can be used to determine DLM plasma concentration in MDR-TB patients.
