1.Influence of tirofiban to percutaneous coronary intervention

Objective To evaluate the influence of tirofiban to percutaneous coronary intervention.Methods 66 elderly patients with coronary artery disease were treated,they were divided into the study group of 33 cases and the control group of 33 cases.The control group was treated with simple percutaneous coronary intervention treatment, the study group was treated with percutaneous coronary intervention combined with tirofiban.The changes of LVEF, cTnT,and TIMI,symptoms in the treatmeat and the symptoms after 2 years were observed and compared.Results After treatment,the patients'platelet count was to the basic line,the difference was not statistically significant(t =0.353 3,P >0.05).The cTnT,LVEF levels of the study group were significantly better than the control group,the differences were statistically significant(t =5.138 1,8.658 1,all P <0.05).After treatment,the incidence rate of TIM in both two groups were improved compared with before treatment.After treatment,the incidence rate of TIMI 3 of the study group(93.9%) was significantly higher than the control group(69.7%),the difference was statistically significant(χ2 =6.518 5,P <0.05).After treatment,the remission rate of angina of the study group(87.9%) was significantly higher than the control group(60.6%),the difference was statistically significant (χ2 =6.417 8,P <0.05).After 2 years of follow -up , the remission rate of angina and recurrent angina in the study group were significantly better than the control group,the differences were statistically significant (χ2 =5.076 9,3.995 2,all P <0.05).Conclusion The efficacy of percutaneous coronary intervention combined with tirofiban is significant,the prognosis is good,has higher remission,it is suitable for long -term clinical application.
2.Experimental study of the biological activities of several alpha1-adrenoceptor antagonists in vitro.

OBJECTIVETo study the biological activities of several alpha1-adrenoceptor antagonists in vitro.
METHODSThe anococcygeal muscle from male Sprague-Dawley rats (300-350 g) was isolated, removed and suspended in a 20-ml organ chamber containing Krebs solution at 37 degrees C, pH 7.4. The muscle preparations were set at a resting tension of 1.0 g and allowed to equilibrate for 1 h in the Krebs solution. After thorough washing, the anococcygeal muscle preparations were examined for the effects of the tested compounds with increased concentrations on its contractile/relaxant responses and the pA2 of antagonistic activity was assessed.
RESULTSSome of the target compounds displayed blocking activity to alpha1-adrenoceptor.
CONCLUSIONCompounds WB IV-1 and WB IV-3 showed good inhibiting activity, and were worth further studying.

5.Effect of mild-warm moxibustion on fatigue, immune state and quality of life in sub-health population

Objective: To observe the effect of mild-warm moxa stick moxibustion on fatigue, immune state and quality of life (QOL) in sub-health population. Methods: A total of 120 cases were randomly allocated into a moxibustion group (n=60) and a Chinese patent medicine (CPM) group (n=60). People in the moxibustion group were intervened with mild-warm moxa stick moxibustion, once a day; while people in the control group took oral Bu Zhong Yi Qi Wan (Spleen/stomach-supplementing and Qi-boosting Pill), 3 doses a day. Two months made up a course of treatment. The fatigue, immune state and QOL were then observed after one treatment course, followed by a 12-month follow-up. Results: A total of 115 cases completed the clinical trial. After intervention, the fatigue scores, immune parameters and QOL scores were significantly increased in both groups, showing statistical significances (P<0.05). There were between-group significant differences in fatigue score, levels of immunoglobulin M (IgM) and IgG, natural killer (NK) cell activity and QOL score (P<0.05). After the 12-month follow-up, there were still significant between-group differences in fatigue score, physical health, psychological, social relationships and overall perception of QOL and overall perception of health (P<0.05). Conclusion: Mild-warm moxibustion can markedly improve the fatigue, immune state and QOL in sub-health population.
6.Application of array-CGH and MLPA for detection of 4 cryptic unbalanced translocations.

OBJECTIVETo use array comparative genomic hybridization (array-CGH) and multiplex ligation-dependent probe amplification (MLPA) to detect unbalanced rearrangements in 4 cases suspected to have chromosome disease but were undetected with conventional karyotype analysis, and to assess the applicability of array-CGH and MLPA for detection of unbalanced translocation.
METHODSGenomic DNA was extracted with standard procedures. All cases were analyzed by array-CGH and subtelomeric MLPA.
RESULTSAll of the cases were identified to have unbalanced translocations by array-CGH analysis, among which 3 were consistent with subtelomeric MLPA analysis. For the remaining one, its chromosomal abnormality was not detected by MLPA as the imbalance has occurred outside of target regions.
CONCLUSIONBoth array-CGH and MLPA techniques can complement conventional karyotyping for detecting unbalanced translocations. The combination features both high resolution and efficiency for clinical use.

7.Mutation analysis of the androgen receptor gene in a complete androgen insensitivity syndrome family.

OBJECTIVETo study the mutation of the androgen receptor gene in a family with complete androgen insensitivity syndrome and to explore the pathogenicity of the mutation.
METHODSPCR and DNA sequencing were performed to study the AR gene mutation; Mbo I restriction endonuclease was used to detect existence of the mutation in normal controls; conservation of the mutation site was analyzed by comparison of the sequence of amino acid among different species.
RESULTSThe DNA sequence of the three patients contained the same substitution of a single nucleotide on codon 681 GAG to GAT of exon 4, which located in the ligand binding domain of the AR receptor and led to substitution of glutamic acid to aspartic acid in the AR receptor. Their mother was heterozygote of E681D. E681D was not observed in the normal controls. The E681 site was extremely conservative in different species.
CONCLUSIONThe E681D mutation of the AR gene is a novel mutation of leading to complete androgen insensitivity syndrome.

8.Combined G-banded karyotyping and multiplex ligation-dependent probe amplification for the detection of chromosomal abnormalities in fetuses with congenital heart defects.

OBJECTIVETo assess the value of G-banded karyotyping in combination with multiplex ligation-dependent probe amplification (MLPA) as a tool for the detection of chromosomal abnormalities in fetuses with congenital heart defects.
METHODSThe combined method was used to analyze 104 fetuses with heart malformations identified by ultrasonography. Abnormal findings were confirmed with chromosomal microarray analysis (CMA).
RESULTSNineteen (18%) fetuses were found to harbor chromosomal aberrations by G-banded karyotyping and MLPA. For 93 cases, CMA has detected abnormalities in 14 cases including 10 pathogenic copy number variations (CNVs) and 4 CNVs of uncertain significance (VOUS). MLPA was able to detect all of the pathogenic CNVs and 1 VOUS CNV.
CONCLUSIONCombined use of G-banded karyotyping and MLPA is a rapid, low-cost and effective method to detect chromosomal abnormalities in fetuses with various heart malformations.
