1.The expression and significance of high mobility group protein box1 in ovarian endometriosis tissue

Objective To investigate the expression of high mobility group protein box1 (HMGB1)in ovarian endometriosis tissue and to provide new idea for early diagnosis,treatment and prognosis of endometriosis.Methods The levels of HMGB1 in ectopic endometrium and eutopic endometrium of 69 patients with ovarian endometriosis and endometrium of 69 cases with non-endometrioses were detected by reverse transcription-polymerase chain reaction (RT-PCR) and Western-blotting.Results HMGB1 was expressed in all the three groups (ectopic endometrium eutopic endometrium endometrium with non-endometrioses).The relative expression level of HMGB1 mRNA was 0.54383 ± 0.0565,0.3016 ± 0.0614,and 0.1536 ±0.0444,respectively (F =377.923,P < 0.0001).The relative of HMGB1 protein was 0.83990 ±0.12869,0.40100 ±0.16425,and 0.19986 ± 0.08162,respectively (F =185.625,P =0.0000).The expression level of HMGB1 was related to clinical stages (t =10.28,P <0.01) ; however,it was not obviously related to age,the diameter of lump,and course of disease (P > 0.05).Conclusions Over expression of HMGB1 in ovary endometriosis indicates that HMGB1 plays a significant role in occurrence and development of ovary endometriosis.The expression of HMGB1 in endometriosis eutopic endometrium was much higher than that of non-endometrioses endometrium,which supports the eutopic endometrium determinism.The overexpression of HMGB1 was consistent with clinical stage,which proves that HMGB1 maybe be tightly associated with the severity of the disease and to likely become biomarker.
2.Effect evaluation of scene-style education mode in endoscopic ultrasonography examination

Objective To investigate the effect of scene-style education mode in endoscopic ultrasonography examination.Methods 170 patients undergoing endoscopic ultrasonography from May 2009to December 2011 in our hospital were chosen as the research object.They were randomly divided into the control group(conventional education mode group)and the observation group (scene-style education mode) with 85 cases in each group.The repeated rate of insertion,rate of complication,compliance degree,the POMS scale score,blood pressure and heart rate for the two groups of patients were compared.Results Compared with the control group,the repeated rate of insertion,rate of complication were lower,blood pressure and heart rate were lower,and compliance degree and POMS mood scale score were better in the observation group,there were significant differences.Conclusions The scene-style education mode can improve negative emotions of patients undergoing endoscopic ultrasonography significantly,it also plays a positive role in increasing their compliance degree and easing stress response.
3.Real-time fluorescent quantitative PCR assay for detection of human Bocavirus

Objective To establish the method for detecting human Bocavirus (HBoV) by a real-time fluorescent quantitative PCR (FQ-PCR) and to assay HBoV from nasopharyngeal samples of children with acute respiratory tract infections.Methods The specific primers and Taqman probes of targeted HBoV NP1 gene were designed.The aimed fragment of NP1 gene was amplified with PCR and ligated into a pCEM-T Easy vector to set up the standards.The method for detection of NP1 gene using FQ-PCR was established.The sensitivity and specificity of the method had been tested.Then 195 clinical specimens were subsequently tested.Results The specificity of this method was excellent.The linear amplification of the assay ranges from 10 copies/μl to 10~8 copies/μl.Twelve specimens were tested positive for HBoV in 195 clinical specimens (6.2%).Conclusions The method for detection of HBoV NP1 geneby real-time fluorescent quantitative PCR was established successfully.It can offer fast and reliable diagnosis for HBoV infection.

5.Effect of the levonorgestrel-releasing intrauterine device (Mirena) in the treatment of dysfunctional uterine bleeding

Objective To evaluate the efficacy and side-effects of the levonorgestrel-releasing intrauterine device (Mirena) in the treatment of dysfunctional uterine bleeding .Methods 45 cases of dysfunctional uterine bleeding diagnosed by pathology were installed with Mirena.The endometrial thickness by ultrasound hemoglobulin increase and pathologic changes of endometria were recorded before and 24 months after the installation.ResultsThe endometrial thickness before and after the insertion of Mirena were(15.6 ±4.2) mm and (3.2 ± 1.3)mmrespectively (P < 0.01) .Six months after the insertion of Mirena, patients' hemoglobulin was significantly increased from (97.6 ± 8.2)g/L to (123.7 ± 8.1)g/L (U = 2.26, P < 0.05) .After the installation of Mirena, patient experienced metrorrhagia.Conclusions Mirena is a device providing effective and continual therapy for dysfunctional uterine bleeding .It can not only significantly reduce the menstrual blood loss but also reverse the complex hyperplasia effectively.
6.Efficacy of sertraline hydrochloride in the treatment of ulcerative colitis complicated with depression

Objective To investigate the efficacy of sertraline hydrochloride combined with mesalazine in the treatment of patients with ulcerative colitis complicated with depression .Methods From March 2014 to March 2015 ,eighty patients with mild active ulcerative colitis ,self‐rating depression scale (SDS) score between > 53 and 70 points ,were enrolled .Eighty patients were divided into sertraline hydrochloride combined with mesalazine group and mesalazine group (control group) ,40 patients in each group .SDS questionnaire and gastrointestinal endoscopy were applied at the beginning of the study and 12 weeks after treatment to evaluate the efficacy .Chi square test or t test was performed for statistical analysis .Results After 12 weeks treatment ,the efficacy of mesalazine combined with sertraline group and mesalazine group was 75% (30/40) and 65% (26/40) , respectively ,and the difference was not statistically significant ( P > 0 .05 ) .Before the treatment ,ulcerative colitis activity index (UCAI) of mesalazine combined with sertraline group and mesalazine group was 13 .13 ± 4 .41 and 12 .41 ± 3 .96 , respectively ;12 weeks after treatment ,the UCAI of the two groups were 7 .29 ± 5 .78 and 8 .60 ± 4 .56 , respectively .The UCAI of both the two groups were lower than those before treatment , and the differences were statistically significant (t= 6 .106 7 and 5 .189 3 ,both P < 0 .01) .The difference was not statistically significant in UCAI between the two groups after treatment (P > 0 .05) .Before treatment ,SDS score of mesalazine combined with sertraline group and mesalazine group was 55 .98 ± 9 .61 and 56 .27 ± 8 .45 , respectively ;12 weeks after treatment ,SDS scores of the two groups were 49 .36 ± 4 .51 and 52 .81 ± 6 .11 ,respectively .Compared with those before treatment , after 12 weeks treatment , SDS scores of mesalazine combined with sertraline group and mesalazine group both decreased (t= 3 .944 0 ,P= 0 .000 2 ;t=2 .098 6 ,P= 0 .039 1) .After treatment ,SDS score of mesalazine combined with sertraline group was lower than that of mesalazine group (t = 2 .873 2 , P = 0 .005 2) .Conclusion Routine ulcerative colitis treatment combined with sertraline hydrochloride in ulcerative colitis can significantly improve depression of ulcerative colitis patients and improve the quality of patients′ life .
7.Clinical application of high resolution melting curve assay in bacteria identification and antimicrobial susceptibility testing

High resolution melting ( HRM) analysis is a real-time PCR based technique which can detect DNA sequence polymorphism .Immediately after routine PCR with saturated fluorescent dye , melting transition of the amplicon was monitored and fluorescence was collected .According to the different melting curve, single base differences can be distinguished .HRM is a low -cost, single -step, closed -tube, accurate and rapid method , which has aroused general concern .Rapid and sensitive detection of the clinical pathogens is critical to diagnosis of infectious diseases .However , clinical research on application of HRM in infectious disease pathogen identification and drug resistance , especially melting curve analysis based bedside technology integration will provide strong support for the diagnosis and treatment of infectious diseases.
8.Adjuvant chemotherapy of breast cancer

Clinical studies confirm that postoperative adjuvant chemotherapy could significantly reduce mortality rate of patients with the operable breast cancer and improve surviral rate.
