2.Molecular basis for losartan regulating transient outward potassium current

Objective To investigate the molecular basis for anti-ventricular arrhythmic effects by losartan through measuring alteration in mRNA and protein levels of key K+ α channel-and-β subunits (Kv4.2,Kv4.3 and KChIP2) in ventricular myocytes in spontaneously hypertensive rats (SHRs).Methods SHRs were randomly assigned to losartan[10mg·kg-1·d-1,n=12] or placebo (n= 12) with age-and weight-matched WKY rats (n = 12) as control.After 8 weeks of treatment,cardiomyocytes were isolated by enzymolysis.Action potential of cardiomyocytes Ito was recorded,mRNA and protein levels of Kv4.2,Kv4.3 and KChIP2 were assessed by reverse transcriptase polymerase chain reaction and Western blot.Results The action potential duration (APD) measured at 50% and 90% repolarization was shorter in losartan group [(16.82 ± 3.79) ms and (68.49±13.25) ms] than in SHR control group [(24.56±4.59) ms and (73.26±15.47) ms,all P<0.01].Losartan increased Ito current density associated with significant increases in the mean levels of mRNA and protein of Kv4.2 and Ky4.3,and with significant decreases in the mean levels of mRNA and protein of KChIP2 compared with those in placebo SHR (all P<0.01).Conclusions Chronic blockade of AT1 receptors with losartan reverses cardiomyocytes electrical remodeling in SHR,resulting in the shortening of APD,which is associated with increasing Ito density by increasing mRNA and protein expression of Kv4.2,Kv4.3 and by decreasing mRNA and protein expression of KChIP2.
3.Nosocomial infections: an investigation of 27 129 inpatients

ObjectiveTo explore the effective measures to prevent and control the hospital infections.MethodsThe hospital infections'data of 27 129 inpatients were retrospectively studied and retrospectively analyzed.ResultsThe nosocomial infection rate was 3.40%. The major sites of the infection were lower respiratory tract (50.05% ). The most common pathogens were mainly Gram-negative bacteria (41.91% ). The infection mainly occurs in old peple(75.34% ). The highest infection rate is in organ transplantation unit( 14.62% ). The factors of high risk included antibiotics(53.47% ), immunocompromise and invasive therapies. ConclusionTo improve the pathogen detective rate and the monitoring of antibiotic resistance ,to decrease the use of invasive therapies and to keep high risk areas under surveillance were the effective measures to control hospital infection and to reduce the incidence rate of hospital infection.
4.CLINICAL EVALUATION OF SPECIFIC IMMUNOLOGICAL ASSAYS IN AUTOIMMUNE THYROID DISEASES

After purifying thyroglobulin (TG) and thyromicrosomal (TM) antigens,the ELISA (enzyme-linked immunosorbent assay) for detecting anti-TG,anti-TM,serum TG level,TG-antithyroglobulin immune complex (TG-IC) and IgG-,IgM-,IgA-specfic antithyroglobulin,antithyromicrosome were established.These specific immunplogical assays were sensitive,reproducible and did not have the shortcomings associated with the impurity of the TM antigen.The results of clinical studies in patients with Hashimoto's thyroiditis (n=35),hyperthyroidism (n=86),primary hypothyroidism (n=7) suggested that these specific assays were helpful to improve the diagnosis and investigation into pathogenesis for aformetioned autoimmune thyroid diseases (AITD).
5.Gene regulation of renal cell apoptosis and effect of anisodamine on overtrained rat by exhausting swimming

Objective To study the expressions of apoptosis related genes Bcl-2 and Bax proteins in rat's renal tissue and the relationship between the ratio of Bcl-2/Bax and renal cell apoptosis induced by overtraining,and observe the effect of anisodamine on the expression of genes Bcl-2 and Bax in exhausted rats.Methods The animal model of acute kidney injury induced by exhausting swimming was reproduced.Forty eight male Wistar rats were randomly assigned into sedentary control group(CN,n=8),exhausting swimming group(immediate,6 hours and 24 hours after exhausting swimming,ESI,ES6h and ES24h,8 each),Anisodamine group(6 hours and 24 hours after exhausting swimming,AD6h and AD24h,8 each).The expressions of Bcl-2 and Bax were detected by immunohistochemical staining and image analyzer.The correlation between the ratio of Bcl-2/Bax and renal cell apoptosis was analyzed.Results The analysis showed that the expression of Bax increased,and of Bcl-2 decreased,the ratio of Bax/Bcl-2 increased remarkably in ESI group compared with CN group(P
6.Significance of ERK/MAPK signal transduction pathway in the targeting treatment of gastrointestinal cancer

Extracellular signal-regulated kinase/mitogen-activated protein kinase(ERK/MAPK) signaling pathway plays an important role in gastrointestinal tumorigenesis and development.ERK/MAPK signal transduction pathway has three important molecular targets: the small G protein Ras,Raf kinase and MEK1/2 and ERK1/2.At present,there are three kinds of approach which can inhibit ERK / MAPK signal transduction pathway:⑴ destroying the structure and(or) function of the target protein,⑵adopting the deficit strategy,⑶damaging the interaction between proteins.These approaches can provide new ideas for the treatment of gastrointestinal cancer.
7.Evaluation of intraoperative touch imprint cytology diagnosis of sentinel lymph nodes in breast cancer

The surgery of the breast cancer has dramatically changed,sentinel lymph node(SLN) is one of the aspects.SLN biopsy can precisely predict the status of axillary basin.If the SLN is negative,the patients can spare the unnecessary axillary lymph node dissection(ALND) and reduce the morbidity of the surgery.One of the most important issues with sentinel node biopsy is the intraoperative pathological evaluation of the sentinel nodes.Accurate intraoperative pathological examination of sentinel nodes would enable the selection of candidates for ALND during the initial operation,eliminating the need for second surgery.This article will review the two techniques(touch imprint cytology and frozen section) employed for intraoperative examination of SLN and compare these two methods.And evaluate the role of touch imprint cytology and existing problems in introperative diagnosis of the sentinel lymph nodes and raise the probably ways to improve the sensitivity of touch imprint cytology initially.
8.Protective effect of medicinal serum of Jiangtang compound recipe on oxidative-injured endothelial cells

AIM: To study the protective effect of medicinal serum of Jiangtang compound recipe on endothelial cells which were damaged by H2O2. METHODS: The effects of medicinal serum of Jiangtang compound recipe on cell viability, concentration of malondialdehyde (MDA), NO, endothelin-1 (ET-1) and activity of superoxide dismutase (SOD) were determined for human umbilical vein endothelial cells (ECV304) damaged by H2O2. RESULTS: The medicinal serum of Jiangtang compound recipe significantly improved the structural change of ECV304 cells damaged by H2O2, suppressed the production of MDA and the content of ET-1, increased activity of SOD and the rate of animate cells, and promoted secretion of NO. CONCLUSION: The medicinal serum of Jiangtang compound recipe can resist the injury of ECV304 caused by H2O2, playing a role in the protective effect.
9.Attempt to Improvement of the Teaching Quality of the Chemical Experiment

10.Pneumonia in acute stroke patients.

Objective To study the risk factors of pneumonia in the acute stroke patients,and provide support for nursing. Methods 391 acute stroke patients were analysed to investigat the rate of pneumonia and its relation with age,the type and the severity of stroke, the basic disease and the prognosis. Results It was showed that the rate increased in the patients of old age, with severe stroke, and having some basic diseases such as diabetes, heart disease, tumor, immunity disease,old stroke. The mortality in the acute stroke patients with pneumonia was higher than the control. Conclusion Controlling the rate of pneumonia can help us to succeed in treating acute stroke patients.
