1.Application of drug intervention and cognitive intervention in patients with chronic obstructive pulmonary disease (COPD)

Objective To explore the application of cognitive intervention and drug intervention in patients with chronic obstructive pulmonary disease(COPD).Methods A total of 86 patients with chronic obstructive pulmonary disease(COPD)admitted from February 2016 to July 2016 were included in this study.The patients were divided into experimental group and control group according to the order of admission.43 patients in the control group received routine measures,In the experimental group,43 patients received cognitive intervention and drug intervention,compared the clinical treatment effect and treatment compliance of the two groups after intervention.Results The compliance rate of disease cognition,smoking,medication,pulmonary function,family oxygen therapy,respiration exercise,rehabilitation therapy,nutritional support and influenza vaccination were significantly better in the experimental group than in the control group(P<0.05)(1.2±0.7)times,the number of acute attack was(2.0±0.8)times and the quality of life was(4.4±0.9)points,which was significantly better than that of the control group(P<0.05).Conclusion Cognitive intervention and drug intervention have a positive effect on the treatment compliance of patients with chronic obstructive pulmonary disease,improve the quality of life of patients,and have significant clinical application value.
2.Clinical observation on treatment of diabetic peripheral neuropathy bytraditional Chinese medicine foot bath combined with Xueshuantong

Objective To observe the clinical efficacy of traditional Chinese medicine foot bath combined with Xueshuantong injection in the treatment of diabetic peripheral neuropathy.Methods Sixty patients with diabetic peripheral neuropathy(DPN)were randomly divided into two groups.According to the patients' condition,oral hypoglycemic drugs or insulin were used to control blood glucose.The control group was treated with Xueshuantong injection on routine nutrition nerve program.In the conventional nutrition nerve program based on the use of traditional Chinese medicine foot bath combined with Xueshuantong injection treatment,2 groups were treated after 10 days to evaluate the efficacy.Results The total effective rate of the observation group and the control group was 90%and 66.7%respectively.The sensory and motor function of the observation group were significantly improved after the treatment group,and the blood rheology and serum endothelin level were significantly decreased.Conclusion Chinese medicine foot bath combined with Xueshuantong injection for the treatment of diabetic peripheral neuropathy.
3.Association of pre-treatment platelet-to-lymphocyte ratios with re-sponse to neoadjuvant chemotherapy and clinical outcomes of cer-vical cancer patients

Objective:To evaluate the predictive value of pretreatment platelet-to-lymphocyte ratios (PLRs) in response to neoad-juvant chemotherapy and prognostic outcome in patients with International Federation of Gynecologists and Obstetricians (FIGO) Stag-es IB2-IIB cervical cancer. Methods:An investigation was conducted from January 2010 to December 2012 on 75 patients with FIGO Stages IB2-IIB cervical cancer, who underwent neoadjuvant chemotherapy and radical surgery in Changhai Hospital, Shanghai. A re-ceiver operating characteristic (ROC) curve was used to determine the best PLR cut-off value in predicting the response to neoadjuvant chemotherapy. The relationships between the pretreatment variables and the response to neoadjuvant chemotherapy were assessed in univariate and multivariate settings. The overall three-year survival rates were analyzed using the log-rank test and Cox regression mod-el. Results:The response to neoadjuvant chemotherapy was associated with PLR. At the threshold of 123.0, the PLR was 88.5%sensi-tive and 52.2%specific. Multivariate analysis showed that the low independent PLR predicted the response to neoadjuvant chemothera-py well. Based on the log-rank test, the three-year survival rate was lower in patients with PLR >123.0 than those with PLR <123.0 (59.8%vs. 82.4%), but no statistically significant differences were observed between them (P=0.116). Mono-factorial analysis showed that vascular invasion (a tumor that is>4 cm in diameter) and lymph node metastasis influenced the three-year survival rate. In the Cox regression model, only the lymph node metastasis was identified as an independent risk factor for poor prognosis (RR:5.375;95%CI:1.351-21.379; P=0.017). Conclusion: Pretreatment PLR is an easily measured, reproducible, and inexpensive marker of systemic in-flammation and thus shows a prognostic and independent predictive value for the response to neoadjuvant chemotherapy in cervical can-cer. However, pretreatment PLR is not a clinically significant factor for the assessment of cervical cancer prognosis.
4.Quality change on Alisma plantago-aquatica at various growing-seedling stages, transplanting stages, and collecting stages

Objective To establish the quality standard of Alisma plantago-aquatica through comparing systematically the changes of HPLC fingerprint of 23-alisol B and 24-alisol A, and other corresponding components as well. Methods The gradient elution mode was applied in chromatographic separation and data were analyzed by "Computer Aided Similarity Evaluation" software and DPS statistic software. Results Total quality of A. plantago-aquatica was the best when grow-seedling was at 25th, June, transplanting at 10th, September, and collecting at 22nd, December in the same year. Conclusion Total quality will drop along with the postponement of grow-seedling stage, transplanting stage, and collecting stage.
5.Expression of CD44V6 in Human Epidermal Tumours and Malignant Melanoma

Objective To study the expression of CD44V6 in human epidermal tumours and malignant melanoma. Methods The expression of CD44V6 was detected by an immunohistochemical technique in squamous cell carcinoma(SCC), basal cell carcinoma(BCC) and malignant melanoma(MM). Results CD44V6 was expressed on the membrane of tumour cells of 10 patients with SCC, and there was downregulation of CD44V6 expression which along with the decrease of differentiation of SCC tumour cells. Tumour cells of 8 patients with MM and 10 patients with BCC did not express CD44V6. Conclusion Our findings suggest that CD44V6 expression is correlated with the types of skin tumours.
6.Effects of 2,3-dimethyl-2-butylamine derivatives on potassium channels in cultured cortical neurons from rats

Aim To investigate the effects of newly synthesized 2, 3-dimethyl-2-butylamine derivatives on potassium channels in cortical neurons from rats.Method On cultured cortical neurons, the influences of those novel compounds on I A and I K currents were evaluated using whole-cell patch clamp technique. Result On cortical neurons, these compounds (5),(6),(7),(9) showed no effects on outward potassium currents, while the compounds (4),(8) exhibited inhibition of I K currents. But on a rterial smooth muscle cells(SMCs), all the six compounds had potassium channels opening activities. While the other three compounds had no effects on potassium channels of SMCs, the compound (3) decreased the I K currents evoked on neu rons.Conclusion The novel 2,3-dimethyl-2-butylamine derivati ves had the different modulation of potassium channels on cortical neurons as SM Cs.
7.Selective actions of iptakalim on the subtypes of K_(ATP) channels

Aim To establish a cell model heterologously expressing Kir6.x and SUR subunits of K_(ATP)channels and to study the selectivity of iptakalim on different subtypes of K_(ATP) channels.Methods Cell were transfected with the pcDNA vector containing the coding sequenced of Kir6.x and SUR using liposome and the pEGFP-N1 vector encoding for green fluorescent protein was added for easy identification of transfected cells.Using immunocytochemical technique,we detected the expression of Kir6.x and SUR protein in[FL(2K2]transfected cells.The electrophysiological experiment was performed after transfection 48-72 h.In whole cell configuration,the effects of K_(ATP) channel openers iptakalim,pinacidil and diazoxide on the clone currents in transfected HEK-293 cells and the antagonism of K_(ATP) channel inhibitor glibenclamide were evaluated.Results Immunocytochemical study identified the expression of Kir6.x and SUR protein in transfected cells.The electrophysiological experiment showed that in cells with recombinant expression of the Kir6.1/SUR2B channel complex,iptakalim(100 ?m),pinacidil(100 ?m)and diazoxide(200 ?m)significantly increased the clone current from(-88.0?30.8) pA to(-2042.6?127.3) pA,(-1431.9?142.4) pA and(-1104.7?228.9) pA,respectively,at-100 mV,and the actions were inhibited by glibenclamide(10 ?m).In cells expressed with Kir6.2/SUR2A channel,both iptakalim and pinacidil activated the currents,which was sensitive to glibenclamide.While diazoxide had no effects on the clone currents.In contrast,in cells with Kir6.2/SUR1channel,diazoxide increased the activity of recombinant K_(ATP) channels,while iptakalim and pinacidil had little effects.Conclusion From these observations,the effects of K_(ATP) channels openers with different chemical structures on the subtypes of K_(ATP) channels were diverse.Iptakalim showed selective action on Kir6.1/SUR2B and Kir6.2/SUR2A,but not Kir6.2/SUR1 K_(ATP) channels
8.Discussion on the application of 3D SPACE sequence to display normal brachial plexus

Objective: To investigate the value of enhanced 3D SPACE sequence in displaying brachial plexus. Methods:35 healthy volunteers with no history of brachial plexus injury of brachial plexus MRI examinations by DWIBS scan, and SPACE scanned. Analysis and comparison of DWIBS sequences, sequences of SPACE provides a clear display of the brachial plexus, and two sets of image sequence average signal-to-noise ratio (SNR) and the contrast to noise ratio (CNR). Results:A total of 35 cases 70 lateral brachial plexus after coronary DWIBS is clearly displayed on the sequence;the partial medial cord of brachial plexus, beams and lateral beams can also be displayed in the SPACE sequence. SPACE sequence of image signal-to-noise ratio and contrast noise ratio than the DWIBS sequence, and the difference is statistically significant. Conclusion:SPACE sequence can clearly show the brachial plexus, and DWIBS sequences compared to the normal image has a higher resolution of the brachial plexus.
9.Effects of pioglitazone on the expressions of HIF-1α and VEGF in renal tissues of diabetic rats

The effects of pioglitazone on the expressions of hypoxia-inducible factor-1 α (HIF-1 α) and vascular endothelial growth factor (VEGF) in renal tissues of diabetic rats were observed. Diabetic rat model was established by feeding high-carbonhydrate-fat diet and injecting streptozotocin. After the treatment with pioglitazone, the kidney index, 24 h urinary albumin, blood urea nitrogen, serum creatinine, fasting blood glucose, fasting insulin, homeostasis model assessment for insulin resistance (HOMA-IR), total cholesterol,triglycerides, and low-density lipoprotein-cholesterol of diabetic rats were significantly lower than those of untreated ones, high-density lipoprotein-cholesterol was increased, the expressions of HIF-1α and VEGF in renal tissue were decreased ( all P<0. 01 ). It suggested that pioglitazone may improve renal function and the balance of glucose-lipid metabolism in diabetic rats via down-regulating HIF-1/VEGF pathway.
10.An in vitro study on rabbit osteoarthritis gene therapy with GGCX gene

Objective To research the effect of GGCX gene on MMP13 in rabbit osteoarthritis cartilage cells and investigate its effect on osteoarthritis cartilage degeneration.Methods Six Japanese big ear rabbits weighted (2.0±0.2)kg were randomly di vided into three groups,each group seted a rabbit as control.Anterior cruciate ligament transection method was used to build osteoarthritis cartilage degeneration model at second,forth,sixth week.Articular cartilage was separated successfully after the model were built,cartilage cells were divided from articular cartilage and cultured in 6-well cell culture plate.Cartilage cells were divided into blank group,negative control group and transfection group.PCR method and Western blot were conducted to detect GGCX and MMP13 expressed at the level of mRNA and protein.Results Compare with blank group and negative control group,the expressin level of GGCX incresed,while the MMP13 expression level dcresed(P<0.05).Conclusion Over expression of GGCX gene can ob viously decrease the expression of MMP13,it provide experimental basis for osteoarthritis of the in vitro gene therapy.
