1.Determination of Aluminum in Drinking Water by Graphite Furnace Atomic Absorption Spectrometry

Objective To establish a method for determination of aluminum in water by graphite furnace atomic absorption spectrometry (GFAAS). Methods Aluminum in water was directly determined by GFAAS at wavelength of 394.4 nm without any matrix modifiers. Results A good linearity (r=0.999 5) was obtained, the linear range was 0-0.600 mg/L. RSD of samples ranged from 2.1% to 6.4%, the average rates of recovery were between 91.0%-108.0%. The detection limit of the method was 0.6 ?g/L, the quantitative limit was 1.9 ?g/L. There was no significant difference between the results determined at wavelength of 394.4 nm and 309.3 nm. Conclusion This method is simple, rapid, accurate and has a wide linear range. It can be applied in the determination of aluminum in drinking water.
2.Determination of trans-resveratrol in rat plasma after oral administration of Polygonum cuspidatum extract at a single dose by RP-HPLC

AIM: To develop a method for the determination of trans-resveratrol in rat plasma. METHODS: Plasma samples were performed on a reversed-phase column by gradient elution after liquid-liquid extraction. RESULTS: Good linearity (r= 0.9998) was obtained in the range of 0.0035- 1.400 mg?L -1 for trans-resveratrol, and the limit of quantitation (LOQ) was 0.035 mg?L -1. The average recovery was 103.0% and the RSD of intra-day and inter-day was less than 10%. This method was applied to the determination of resveratrol in rat plasma and the pharmacokinetical parameters were calculated as follows: t 1/2= 11.5 min, t max= 10.0 min,and C max= 1.93 mg?L -1. CONCLUSION: A reliable RP-HPLC method for resveratol determination is developed, and appropriate for the determination of resveratrol in rat plasma.
3.The absorption and metabolism of indocyanine green in human retinal epithelial cells

Objective To investigate the ingestion, metabolism and subcellular localization of indocyanine green (ICG) in human retinal epithelial (RPE) cells. Methods RPE cells were incubated with 0.25 mg/ml ICG under the condition of 37℃ in the camera. The ICG granule and ultrastructure of RPE cells were observed under the electron microscopy after 1-, 4-, and 24-hour incubation, and the ICG autofluorescence was detected by fluorescence microscopy after the incubation for 1, 2, 4, 8, 12, 24, and 48 hours, respectively. The absorbency (A value) of ICG solution was measured at 805 nm with ultraviolet/visible specrtrometer. The standard curve of concentration of ICG was drawn and the related equation of concentration of ICG and the A value was calculated. After being incubated for 1, 2, 4, 8, 12, 24, 48, and 72 hours, respectively, the A value of supernatant fluid was calculated according to the equation. After incubated with ICG for 24 hours, one sample was observed under electron microscope and fluorescence microscope per week to evaluate the metabolizable period of ICG. Results ICG granules were distributed evenly after entering the RPE cells. After incubated with 0.25 mg/ml ICG for 24 hours, no significant change of the ultrastructure of the RPE cells was found. ICG granules accumulated in the cells as the time goes by and reached the peak after 24 hours, and then they decreased because of the slowdown of the metabolism. Few ICG was still remained in the cells 1 week later. Conclusions RPE cells may take in ICG actively. ICG metabolizable period in RPE cells is long, which may be one of the mechanisms of the toxicity of ICG to the retina in the vitreous operation.
4.Effect of Fumai Decoction on ET, CGRP, TXB_2 and 6-keto-PGF_(1?) of Type 2 Diabetes Mellitus Patients Combined with Acute Cerebral Infarction

Objective To evaluate the mechanism of Fumai decoction in treating type 2 diabetes mellitus combined with acute cerebral infarction. Methods 68 cases of type 2 diabetes mellitus with acute cerebral infarction patients were divided into treatment group (34 cases) and western medicine control group (34 cases) at random. The serum levels of Endothelin (ET), Calcitonin gene related-pepitide (CGRP), Thromboxane-B2 (TXB2), 6-ketone-prostagla-ndins F1? (6-keto-PGF1?) and cerebral blood flow variation, clinical therapeutic effect were observed. Result Compared 2 groups, there was significant difference in improving neurological impairment, cerebral artery stenosis, cerebral circulation insufficiency and reducing levels of ET and TXB2, enhancing levels of CGRP and 6-keto-PGF1? (P0.05). Conclusion Fumai decoction can adjust the serum levels of ET, CGRP, TXB2 and 6-keto-PGF1?, and improve cerebrovascular function. Probably this is one of the mechanisms in treating type 2 diabetes mellitus with acute cerebral infarction.
5.Malignant mesothelioma of the tunica vaginalis testis: one case report

Objective To study the clinical and pathological features, diagnosis and treatment of malig-nant mesothelioma of the tunica vaginalis testis. Methods One case of malignant mesothelioma of the tuni-ca vaginalis testis was reported. The patient was a 73 years old man, presented with a history of aggravated hydrocele for 1 year. B-ultrasound showed 7.5 cm×4.1 cm liquid anechoic area around the testis. The Pubmed and CBM databases were searched for related articles, and the clinical data were reviewed. Results An operation was performed to incise the tunica vaginalis. The pathology showed fecal caneeration in the tunica vaginalis testis and the immunohistology showed CK5/6, Calretinin and E-cadlberin positive. Right or-chiectomy was performed after three weeks. Local tumor recurrence and inguinal lymph nodes metastasis were found 11 months after operation, and local tumor and lymph nodes were resected. The tumor was diagnosed as malignant mesothelioma of the tunica vaginalis testis and CK5/6 and Calretinin fecal positive. Conclu-sions Malignant mesothelioma of the tunica vaginalis is a rare disease of the testis with potentially aggres-sive behavior. Due to the lack of tumor markers and characteristic symptoms, correct preoperative diagnosis is extremely difficult. Radical surgical treatment is of major importance to the survival of the patients. Life-long follow-up should be offered to patients with malignant mesothelioma of the tunica vaginalis.
6.Epoxyeicosatrienoic acid: The signal transduction

Cytochrome P450 monoxygenase converts arachionic acid to four epoxyeicosatrienoic acid regiosomes: 5,6-EET(epoxyeicosatrienoic acid);8,9-EET; 11,12-EET and 14,15-EET.Recent studies show that EETs are involved in signal transduction. EETs open Ca 2+ -sensitive K + channel and inhibit Na + channel,Ca 2+ -sensitive Cl - channel and so on. What is more ,EETs have been demonstrated to activate PP60 c-src and initiate a tyrosine kinase cascade that mediates mitogenic effects.
7.The dose-response relationship between propofol and isoflurane in balanced anaesthesia

Objective To study the dose-response relationship between propofol and isoflurane in balanced anaesthesia. Methods 30 patients, age ranged from 46 to 68 years and body weight from 48 to 72 kilograms, were se-lected. Anaesthesia was induced with propofol 1.5mg/kg,vecuroniura 0.12 mg/kg, fentanyl 2~4μg/kg,and main-rained with vecuronium 1.2μg/kg,fentanyl 2μg/kg. Isoflurane was inhaled at first so that BIS could be maintained on the level of 50~60. Then, minishing the isoflurane concentration by 0.3%~0.5 % and adding the dosage of propofol synchronously, establishing 4~6 balanced level spots of anaesthesia, which could maintain BIS on the level of 50~60 all the time. Recording the isoflurane concentration, the dosage of propofol, MAP, HR, etc, and making the scatterplot graph in virtue of SPSS software. Results The relationship between propofol and isoflurane was accordance with ex-ponential curve model,of which the equation is :y= 102.991 x exp (-1.4456x), R2= 0.784 ; and the ratio of their dosages is 54/1. When isoflurane was inhaled alone at first, MAP was decreased significantly ( P<0.05 ). As the isoflurane concentration minished and the dosage of propofol added, MAP and HR were increasing gradually, but when isoflurane concentration was minished to zero, there is no significant difference as compared with the preanesthesia baseline ( P>0.05 ). Conclusion When BIS was on the balanced level of 50~60, exponential curve relationship ex-ists between the dosage of propofol and isoflurane concentration. If applied reasonabhly, propofol could be good for re-ducing circulation aide-effect of isoflurane.
8.Transrectal 12-core biopsy of the prostate guided by transrectal ultrasonography

Objective To evaluate the clinical value of transrectal ultrasound guided systematic 12-sample needle biopsy in patients with elevated serum prostate-specific antigen and/or abnormal digital rectal examination findings. Methods The data of 220 patients who underwent 12-sample transrectal ultrasound guided needle biopsy (comprising conventional 6 biopsies and 3 biopsies in each lateral peripheral zone) of the prostate were retrospectively analyzed. Results Of the 220 patients,73 (33.2%) had prostate cancers on biopsy;the clinical stages were as follows:4 cases of stage T 1,21 of T 2,15 of T 3 and 33 of T 4.If the conventional 6-core biopsy alone was performed, the detection rate would be 31.4%,4 cases (3 of stage T 1 and 1 of stage T 2,all with tumor volume less than 0.5 ml) would be missed. The improvement by 12-core biopsy was marked in patients with stage T 1-T 2 prostate disease (16%,4/25).No serious complications were observed in patients with transrectal ultrasound guided systematic 12-core biopsy of the prostate. Conclusions The 12-sample procedure can improve the cancer detection rate of the early stage prostate cancer and tumor with volume less than 0.5 ml.Therefore,more attention should be paid to the lateral peripheral zone biopsy.
9.THE RELATION BETWEEN SERUM COPPER VALUE AND PLACENTAL FUNCTION IN PREGNANT WOMEN

we examined the serum copper level in 125 pregnant women (83) normal pregnant women of 28~40 weeks of pregnancy; 12 patients with se-vere edema-proteinurine-hyperterision syndromeduring 37~40 weeks of pregnancy; 30 patientswith prolonged pregnancy) and compared it withurine E/C value and Apgar score in order to deter-mine the reliability of serum copper value in diag-nosing placental insufficiency. The results con-firmed that the serum copper value was increasingwith the growth of weeks of pregnancy, but it wasdecreasing after the fortieth week of pregnancy.Comparison of the serum copper value with theurine E/C value showed that when the E/C or = 10. And when the Apgar score7. These results indicatethat the serum copper value can serve as an indexof placental function in the third trimester of preg-nancy.
10.The inhibitive effect of adenovirus mediated CD gene and 5-FC on proliferative human retinal pigment epithelialcells

Objective To study the inhibitive effect of adenovirus mediated CD gene and 5-FC on proliferative human retinal pigment epithelial (HRPE) cells, and to search for an effective method to take precautions against proliferative vitroretinopathy (PVR). Method Different concentrations of CD and 5-FC were added respectively to the cultured third-growth-generation HRPE cells.Transferance rate was detected by positive HRPE cells marked by X-gal and LacZ. The number of HRPE cells were counted and evaluated by methylthiazol-tetrazollium (MTT) method. Results The adenovirus mediated CD gene could be transfered into HRPE cells with a dose-dependent manner. Positive HRPE cells with CD gene could transform 5-FC to 5-Fu,which could inhibit the increase of HRPE cells effectively. No obvious bystander effect on the growth of HRPE cells was detected. Conclusions The adenovirus may introduce a foreign gene into cultured HRPE cells efficiently. It could be a good method to treat and prevent PVR by medication.
