1.Progression of lobaplatin for cancer
Journal of International Oncology 2012;(12):907-909
Lobaplatin is the third generation platinum drug. Many studies on pharmacodynamics have confirmed that lobaplatin can block the cell cycle and induce cell apoptosis, and thus inhibits tumor cell proliferation effectively.Lobaplatin is not cross-resistant to cisplatin.Besides,it has good curative effect to various kinds of cancer patients with mild adverse reactions, and the patients are well tolerable to it.

AIM: To explore the role of PI3K/Akt/nNOS in Zhouluotong extract resisting diabetic peripheral neuropathy.METHODS:The Schwann cells were divided into normal group ( D-glucose 25 mmol/L) , model group ( D-glucose 100 mmol/L) , Zhouluotong extract Z-6 +high glucose group, Zhouluotong +high glucose group, mecobalamine+high glucose group. The viability, nitric oxide content and the Ca2+-ATPase activity in Schwann cells were determined by Cell Counting Kit-8 , nitric oxide assay kit and Ca2+-ATPase assay kit, respectively. The apoptosis of Schwann cells were analyzed by flow cytometry. The expression of Bcl-2, Bcl-xL, Bax, Bak and caspase-3, and the phosphorylation levels of nNOS and Akt were determined by Western blotting. The signal pathway of PI3K/Akt was explored by dominant negative PI3K and Akt (δp85 and DN-Akt) transient transfection assay.RESULTS:Under high-glucose culture, the cell viability, nitric oxide content in culture supernatant, the expression of Bcl-2 and Bcl-xL, and the phosphorylation levels of Akt and nNOS in the Schwann cells were significantly increased. The cell apoptosis, the expression of Bax, Bak and caspase 3 in the Schwann cells were significantly decreased by Zhouluotong extract Z-6, compared with model group.In-creased nitric oxide content and the up-regulation of nNOS were observed. However, the effects of blocking PI3K/Akt, the upstream pathway of nNOS , by transfection with DN-δp85 on Akt phosphorylation in the Schwann cells was still unclear. CONCLUSION:Zhouluotong extract Z-6 changes the phosphorylation of nNOS, and the expression of anti-apoptotic fac-tors , caspase-3 and pro-apoptotic factors in Schwann cells under high-glucose culture, thus reducing apoptosis and elevating viability. The relationship to PI3K/Akt/nNOS pathway needs further investigation.

Chinese Journal of Neurology 2020;53(8):587-593
Objective:To summarize and review a Chinese family with cerebrotendinous xanthomatosis (CTX) so as to improve understanding of the disease. Methods:The proband was admitted to the Department of Neurology, the Second Hospital of Hebei Medical University on May 30, 2019. The medical history, neuro-imaging, pathology, CYP27A1 gene of the proband and CYP27A1 gene of her family were analyzed. Clinical features of similar cases from published literatures were retrieved and systematically summarized. Results:The proband was a 39-year-old female who was admitted to the Second Hospital of Hebei Medical University due to weakness of both lower limbs lasted for more than five years and aggravated for one year with speech slurred. The proband manifested with mental retardation, bilateral pyramidal tract impairment and cerebellar lesions, and had cholesterol crystal in xanthomas and compound heterozygous mutations of c.435G>A and c.1263+1G>A in CYP27A1 gene. The proband′s sister had the same mutation as the proband′s. The proband′s mother was the carrier of c.435G>A mutation, and father was the carrier of c.1263+1G>A mutation. Seventeen related cases concerning CTX with detailed clinical data were searched with major domestic databases. Combined with this case, clinical features with the frequency more than 50% were pyramidal sign, mental decline, ataxia, dysarthria, achilles tendon neoplasm, cataract, high arch foot. Conclusions:The onset of CTX is hidden, which can be diagnosed by its pathology and CYP27A1 gene detection. The possibility of CTX should be considered when there are unexplained clinical manifestations in common diseases such as pyramidal sign, mental decline, ataxia, dysarthria, achilles tendon neoplasm, cataract, high arch foot and so on.

Objective To explore the effect and possible mechanism of electroacupuncture on learning and memory impairment in rats with cerebral ischemia reperfusion. Methods 90 SD rats used for stepdown test were divided into normal control group, simple cerebral ischemia reperfusion of 7 d,14 d,30 d,60 d group (I/R) and cerebral ischemia reperfusion of 7 d,14 d,30 d,60 d and electroacupuncture intervention group (I/R+EA),through simple random sampling method (10 rats in each group);70 SD rats used for molecular biology experiments were randomly divided into normal control group, simple cerebral ischemia and reperfusion of 3 h,6 h,12 h group (I/R) and cerebral ischemia reperfusion of 3 h,6 h,12 h and electroacupuncture intervention group (I/R+EA),n=10.Use suture method to make model of cerebral ischemia reperfusion in rats.Take the rats' "baihui" acupoint and "shuigou" acupoint to acupuncture. The final strength was 5 mA and the duration was 30 minutes.Step down test was used to observe the restoration of learning and memory in rats.Western blot was used to detect the expression of procaspase-3,procaspase-12,PARP and ICAD in rat cerebral cortex and striatum.Immunofluorescence histochemical method was used to detect the number of GRP94 positive cells. Results The results of the platform showed that the learning and memory abilities of the rats were obviously improved after electroacupuncture treatment.Electric shock incubation period of rats in electroacupuncture of 7 d and 14 d group was (90.60± 17.01) and (128.80±8.85) respectively. It was significantly higher than that of cerebral ischemia reperfusion of 7 d group(51.20±5.09) and 14 d group(105.00± 10.39).The number of mistakes in single foot jump of rats in electroacupuncture of 7 d and 14 d group was (3.90±0.99) and (2.70±0.67) respectively. It was significantly lower than that of 7 d group (9.20± 1.61) and 14 d group (6.10± 0.99).Through SPSS13.0 statistical software one-way ANOVA,the difference was statistically significant (P< 0.05).Western blot results showed that the expression of procaspase-3,procaspase-12,PARP protein precursor and ICAD protein, upregulation increased after electroacupuncture(P<0.05).The results of immunofluorescence histochemistry showed that the number of positive cells of GRP94 protein was significantly decreased after 3 h,6 h and 12 h after electroacupuncture intervention compared with the corresponding I/R group. Conclusion Electroacupuncture can effectively improve the learning and memory ability of rats with ischemia-reperfusion,which may be related to the apoptosis of cerebral cells after cerebral ischemia reperfusion injury by blocking the apoptosis pathway mediated by endoplasmic reticulum and inflammatory factors.
