1.Inhibition of HBV Release by BST-2.

Bone marrow stromal antigen 2 (BST-2) is a kind of host restriction factor. Since it was discovered to be responsible for the defect in virion release of HIV-1 mutants lacking the accessory gene vpu in 2008, it was thought to mainly restrict the viruses by directly tethering viral particles at the plasma membrane. Recent reports suggest that BST-2 also can inhibit the the release of HBV particles, which are budding in the intracellular vesicles, expanding the antiviral spectrum of BST-2. Futhermore, the machanism that BST-2 used to restrict HBV release in multivesicular bodies (MVBs) is similar to that used to restrict HIV at the plasma membrane. However, HBV have evolved strategies to antagonize the antiviral action of BST-2. There are two different opinions about the antagonist. One is HBV inactivated BST-2 by HBx requiring a hepatocyte-specific environment. Another thought envelope protein HBs counteract the antiviral action of BST-2. In this review, we focus on the current advances in the anti-HBV activity of BST-2.

2.A randomized and double-blind comparison with reboxetine and citalopram in patients with retardant depressive disorder

Objective To study the efficacy and safety of reboxetine and citalopram in treating retardant depression.Methods A total of 59 patients with retardant depression were randomly divided into reboxetine group(n=29)and citalopram group(n=30)for the treatment of 6 weeks.The patients were assessed with the Hamilton depression scale(HAMD)and treatment emergent symptom scale(TESS).Results At the end of 6 weeks,the total scores of HAMD in reboxetine(18.48±6.38)group and citalopram(11.60±4.46)group both significantly decreased(P<0.001).Between two groups,the efficacy(82.8% and 56.7%)had significant differences(P<0.05)but side effects had no statistical significance.Conclusion Reboxetine has significant curative efficacy,milder adverse effects and higher safety for treating patients with retardant depression.
3.Preliminary analysis of miRNA expression profile of gastric cancer tissues by microarray

Objective To analyze the expression profile of specific miRNA of gastric cancer tissues.Methods miRNA were isolated from five paired gastric cancer tissues and corresponding paracancerous normal tissues.To identify the profile of gastric cancer,miRNA hybridization and cluster analysis were processed by significance analysis of microarrays (SAM,version 2.1) and cluster software.The relative expression of hsa-miR-30c,hsa-miR-196a and hsa-miR-193b were validated by fluorescence Real-Time PCR using taqman probe.Further hsa-miR-30c taget gene was predictived by online software.Results Revealed by miRNA microarray detection,there were 16 miRNA with differential expression in gastric cancer tissues and para-cancerous normal tissues,among which HS_100,hsa-miR-27a,hsa-miR-214*,solexa-555-1991 and hsa-miR-196 were significantly up-regulated,and hsa-miR-502-3p,hsa-miR-29c*,hsa-miR-64,hsa-miR-193b,hsa-miR-551b,hsa-miR-30c,hsa-miR-582-5p,hsa-miR-625*,hsa-miR-660,hsa-miR-363 and hsa-miR-486-5p were down-regulation.The relative expression of hsa-miR-30c,hsa-miR-196a and hsa-miR-193b was in high concordance with microarray.Conclusion There is specific miRNA expression profile in gastric cancer tissues,among which differentially-expressed miRNAs may be involved in the regulation of gastric cancer progress and serve as new potential biomarkers.
4.Investigation on Medical Students' Ethical Cognition of Assisted Reproductive Technology

Objective:To investigate the medical students' cognition of ethical issues in assisted reproductive technology (ART) and discuss the importance of ethical education in medical colleges and universities.Methods:A questionnaire survey about the ethical cognition of assisted reproductive technology was conducted among 632 medical students.The questionnaire included the donation and management of gametes and embryos,and offspring problems.Results:Medical students agreed with most of the current ART-related laws and regulations.however,44.8% of medical students considered that criteria of donating eggs should be relaxed,such as allowing women who do not receive ART treatment be paid for eggs.In addition,a total of 39.0% of medical students supported the legalization of surrogacy technology in china.Conclusion:Medical students exists a certain misunderstanding in egg donation and surrogacy technology.We should give more training to medical students in ethical knowledge.In addition,our country can supplement and perfect the current laws and regulations to a certain extent.
5.Imaging features of Von Hippel-Lindau syndrome type Ⅰ

Objective To analyze the imaging characteristics of Von Hippel-Lindau Syndrome (VHL)typeⅠ.Methods 12 patients diag-nosed as VHL type Ⅰ were collected and all the patients had underwent 3.0T MRI and 64-slice spiral CT plain and enhanced scan. Imaging data was analyzed by two senior radiologists to summarize its imaging featueres.Results Multiple hemangioblastomas (HGBLs)in cerebellum,spinal cord or brainstem were seen in all the patients.Flow void effect with varied degree was seen in the lesions.Dilated feeding and drainage blood vessels around lesions was seen in 6 cases.Multiple kidney and pancreas cysts were seen in all the patients.There were 4 cases with syringomyelia,7 cases with ocular retinal angioma,5 cases with renal cell carcinoma,2 cases with ground-glass-like lesions and micronodulars in lung.Conclusion VHL type Ⅰ has complex imaging performance.Ima-ging examination is important for early detection of VHL disease.
6.Effects of different degrees of intermittent hypoxia on NF-κB, IL-10 and visfatin in 3T3-L1 adipocytes

Objective To determine levels of nuclear factor (NF)-κB, interleukin (IL)-10, and visfatin in adipocytes treated by different degrees of intermittent hypoxia (IH), and to investigate the mechanism of IH leading to insulin resistance (IR). Methods The cell model of intermittent hypoxia/re-oxygenation (IH/ROX) in obstructive sleep apnea (OSA) was established. Differentiation mature 3T3-L1 adipocytes, were randomly divided into 10 groups including four different-frequency intermittent hypoxia groups(IH1-4, fixed intermittent hypoxia scheme for 1.5%O2 45 s and then re-oxygen 21%O2 for 2 min 15 s, 4 min 15 s, 5 min 45 s and 8 min 45 s, 60 times circulation), and their normal oxygen control groups (SC1-4, instead each IH group 1.5%O2 to 21%O2, the rest groups were treated as same as IH group), continuous hypoxia group (CH, 10%O2 for 6 h) and normal oxygen control group (CC, 21%O2 for 6 h). ELISA method was used to determine the levels of IL-10 and visfatin in the supematant of adipocytes. Western blot method was used to determine the protein levels of NF-κB p65 and visfatin. Real-time PCR method was used to determine the mRNA levels of IL-10 and visfatin. Results The protein and mRNA expressions of IL-10 were significantly lower in IH group and CH group than those of control groups (P<0.01). The levels of NF-κB p65 protein were significantly increased in IH group and CH group than those of control group. The protein and mRNA expressions of visfatin were significantly higher in IH1, IH2 and CH groups than those of control group (P<0.01). Conclusion As a prominent feature of OSA pathophysiology, IH may take part in insulin resistance of OSA patients by abnormally secreting NF-κB, IL-10 and visfatin in adipocytes.
7.Construction of a recombinant adenovirus vector expressing shRNA targeting interleukin-1beta gene in rats

BACKGROUND:Specific down-regulation of interleukin 1 beta (IL-1β) may al eviate the pain behaviors effectively after peripheral nervous injury. Compared with smal interference RNA (siRNA), short hairpin RNA (shRNA) could inhibit the expression of target gene more stably and efficiently. However, simple shRNA could not enter target cel s to down-regulate target gene efficiently. Adenovirus vectors have wide host range, high infection efficiency and stable expression in host cel s.
OBJECTIVE:To construct recombinant adenovirus vector expressing shRNA targeting IL-1βgene and detect its effect on the expression of target gene.
METHODS:Three siRNAs were designed on the basis of the nucleotide sequence of IL-1βobtained from NCBI and then three shRNAs (shRNA1, shRNA2 and shRNA3) were synthesized. The annealed shRNA product and adenovirus vector pHBAd/U6/GFP digested by BamH I and EcoR I were connected to construct the recombinant adenovirus vector shuttle plasmid expressing shRNA targeting IL-1β. After sequencing, HEK 293 cel s were co-transfected by the shuttle plasmid and skeleton vector, and three recombinant adenovirus vector expressing shRNA targeting IL-1β(rAd/shRNA1, rAd/shRNA2 and rAd/shRNA3) were packaged and amplified. Rats H9C2 cel s were infected by recombinant adenovirus vector expressing shRNA targeting IL-1βand fluorescence microscope was used to observe the infection efficiency. The effect of recombinant adenovirus vector expressing shRNA targeting IL-1βon the expression of target gene was detected by western blot assay.
RESULTS AND CONCLUSION:The sequencing results showed that the sequences of three shRNAs adenovirus vector shuttle plasmid were consistent with the sequences of three designed shRNAs. rAd/shRNA1, rAd/shRNA2 and rAd/shRNA3 were constructed successful y. rAd/shRNA1, rAd/shRNA2 and rAd/shRNA3 could down-regulate the expression of IL-1βin rat H9C2 cel s and the down-regulation effect of rAd/shRNA2 was the most significant.
8.Comparative analysis of Chinese oncology papers included by CBM and SCI in 2012

Objective To analyze Chinese oncology papers included by Chinese biomedical literature database (CBM) and science citation index (SCI),and to comprehensively and objectively reflect the current situation of Chinese oncology literatures published in domestic and international journals.Methods Chinese oncology papers were retrieved by using CBM and SCI.A comparison analysis was made from the respects of subject distribution,district distribution,organization distribution and high citation situation.Results 67 071 papers of 2012 were retrieved from CBM database,while 9226 papers were indexed by SCI.In terms of district distribution,the first three districts were Jiangsu (6707),Guangdong (4897) and Beijing (2214) in CBM database,while in SCI database,they were Beijing (2214),Shanghai (2008) and Guangdong (1393).The institute with the most amount of literature is university,respectively 9725 and 5755.Moreover,the mostly funded project is National Natural Science Foundation (4710,35.38% ; 3290,35.68%).Conclusion The 2012 Chinese oncology literatures published in domestic journals has advantage with respect to quantity,compared with the international published journals.However,there exists a bigger difference in terms of quality.Through the comparative analysis,the medical researchers in our country will have some implications in the research orientation.
9.The radiation physics advantages of 4DCT on intensity-modulated radiotherapy of chest wall after radical mastectomy

Objective To invesigate the influence of breathing motion on intensity-modulated radiotherapy (IMRT) of chest wall after radical mastectomy,and explore clinical value of accurately determined target volume.Methods A total of 17 radical mastectomy patients underwent 3DCT simulation scans sequentially followed by 4DCT simulation scans during free breathing.The targets and normal organs was determined based on CT images respectively.Three sets of radiotherapy plan were designed for each patient:plan 3D,plan 4D and plan 3D-A.The Plan 3D and plan 4D was designed based on 3D and 4D targets respectively.Plan 3D was copied to 4D target with the same isocenter coordinates.The dose distribution was calculated separately to evaluate the dose-volume histograms parameters for PTV,ipsilateral lung and heart,respectively.Two planning parameters was compared with paired t-test or Wilcoxon sign-rank test.Results The average volume of PTV4D was (10.35 ± 4.80) % larger than PTV3D (P =0.000).Compared with plan 3D,the V100,V95,V90,D95,D90,Dmin of plan 3D-A were reduced,that were (0.78 -18.0)% (P=0.000),(0.01-3.90)% (P=0.000),(0-2.12)% (P=0.000),(13-222) cGy (P=0.000),(1-118) cGy (P=0.000),(6-1 910) cGy (P=0.000).However,the V20,V10,V5,Dmean of the ipsilateral lung and V30 of heart were same between 3D plan and 4D plan (P =0.288,0.407,0.435,0.758,0.575).Conclusions The respiratory motion may reduce the target dose and its coverage in chest wall treatments,so 4DCT plan could accurately define target volume without increasing the exposure dose of normal tissues.
10.A Study on CpG Island Methylator Phenotype involving Tumor Suppressor Genes on Chromosome 3p in Gastric Cancer

Background:CpG island methylator phenotype(CIMP)involving tumor suppressor gene( TSG)on short arm of chromosome 3(chromosome 3p)has been found in various types of cancers. However,its correlation with gastric cancer has not been clarified. Aims:To study the clinical significance of CIMP involving TSG on chromosome 3p in gastric cancer. Methods:Methylation specific PCR(MSP)was used to examine methylation profiles for hOGG1,VHL,RAR-B, hMLH1,SEMA3B,RASSF1A,BLU and FHIT harbored in chromosome 3p in 100 gastric cancer and paired paracancerous tissues. High CIMP( CIMP-H)was referred for those samples having four or more synchronously methylated genes. Relationship between CIMP-H and clinicopathological characteristics in gastric cancer was analyzed. Results:Positive methylation rates of VHL(P = 0. 030),hMLH1(P < 0. 001),SEMA3B(P = 0. 003),RASSF1A(P < 0. 001)and FHIT(P < 0. 001)were significantly higher in gastric cancer tissue than in paracancerous tissue. Incidence of CIMP-H rate in gastric cancer tissue was significantly higher than that in paracancerous tissue(44. 0% vs. 4. 0% ,P < 0. 001). CIMP-H was negatively correlated with degree of tumor differentiation(P = 0. 004),and positively correlated with lymph node metastasis(P = 0. 005),but not correlated with gender,age,tumor location,tumor size,depth of infiltration and TNM staging(P > 0. 05). Conclusions:CIMP on chromosome 3p may occur in early stage of oncogenesis of gastric cancer,and influencing tumor differentiation and lymph node metastasis.
