OBJECTIVE: To study the in vitro anti-tumor activity of tungstophosphate compound(K6[P2W18O62]?14H2O) with Dawson structure.METHODS: MCF-7 cells were treated with different concentration of K6[P2W18O62]?14H2O,then its IC50 was measured by MTT assay;flow cytometry was applied to analyze cell cycle and apoptosis by computing the ratio of MCF-7 cells in different phase and its apoptotic ratio.Cell growth and viability were measured by trypan blue staining and cell counting by computing the ratio of living cells.The synergetic anti-tumor activity of tungstophosphate compound(K6[P2W18O62]?14H2O) and pharmorubicin was studied.RESULTS: K6[P2W18O62]?14H2O inhibited MCF-7 cell proliferation with IC50 of(33.7?3.2) ?mol?L-1,it reduced the proportions of MCF-7 cells at S phase and G1 phase,with the apoptotic rate at 3.7%～29.2% within 48 h.The percentage of living cells was 95.37%～76.78% at 48 h.As compared with pharmorubicin alone,the addition of K6[P2W18O62]?14H2O to pharmorubicin showed a significantly higher inhibitory action on proliferation of MCF-7 cells(P

Objective To investigate the effects of deguelin on the proliferation inhibition and apoptosis induction in RPMI-8226 cells in vitro,as well as the regulation of steroid receptor coactivator-3(SRC-3)to explore the relationship between them.Methods The effect of deguelin on the growth of RPMI-8226 cells was studied by MTT assay.Apoptosis was detected through Hoechst 33258 staining and Annexin-V FITC/PI double-labeled cytometry.RT-PCR Technology was applied to assessment of the mRNA expression of SRC-3,whereas,SRC-3 protein expression and localization were determined by using immunohistochemistry method.Results Deguelin presented striking proliferation inhibition potency on RPMI-8226 cells in vitro and apoptosis induction activity in a time-and dose-dependent manner.The IC50 value for 24 h was(54.55?0.40)nmol/L,(17.04?0.73)% RPMI-8226 cells went apoptotic when treated with 25 nmol/L deguelin for 24 h,with the dose of deguelin increasing to 100 nmol/L,(63.57?1.36)% cells were apoptotic.Over-expression of SRC-3 was found in RPMI-8226 cells,whereas the mRNA and protein expression levels of SRC-3 were significantly downregulated in RPMI-8226 cells induced by deguelin in a dose-dependent manner.The disposition of SRC-3 was situated mainly at the nuclear,occasionally in the cytoplasm.Conclusion Deguelin exhibited potent proliferation inhibition to the human myeloma cell line RPMI-8226 cells,furthermore,deguelin might induce RPMI-8226 cells apoptosis in a dose-dependent manner,which might correspond to the regulation of the expression of SRC-3.

Objective To evaluate the clinical effect of itraconazole oral solutions on pulmonary candidiasis albicans in ICU patients.Methods A total of 71 patients with candida ablicans in their sputum clutures were randomized into two groups.The itraconazole group(n= 36)received itraconazole 200 mg,p.o.(including nasal feeding),q12h for 14 days.The fluconazole group(n=35)received fluconazole 400 mg for the first dose,then 200 mg,i.v.drip,qd for 14 days.Results There were 31 patients in the itraconazole group and 29 paitents in the fluconazole group who completed their treatment.In the itraconazole and fluconazole group,the clearance of candida ablicans was 64.52%(20/31)vs.65.55%(19/29),cleaning time(10.12?2.57)vs(8.87?1.95)days,effective rate 58.06%(18/31)vs 62.07%(18/29),recovery rate is 19.35%(6/31)vs 17.24%(5/29),occurrence of hepatic dysfunction is 25.81%(8/31)vs 24.14%(7/29),and withdrawal for inefficacy was 3.23%(1/31)vs 6.90%(2/29),respectively.These parameters did not have statistical significance.Conclusion Itraconazole oral solutions can be used effectively and safely in the treatment of pulmonary candidiasis albicans in ICU.Patients with renal dysfunction do not need to adjust the dosage.

Its concept and characteristics, and the status quo of hospital information system construction were described. A green computing framework model was established for hospital information system construction, which plays an active role in implementing the idea of green environment protection, low carbon and energy conservation, and sustainable development, thus laying a foundation for studying the mechanisms underlying energy conservation in hospital information system construction and for assessing and optimizing its strategic system.

Objective To explore the changes and comparisons of the four biochemical indicator detection for the early renal im‐pairment in multiple myeloma patients .Methods The 54 patients with multiple myeloma of early onset(MM group) and 54 healthy peoples(control group) were retrospectively analyzed .The levels of serum Cystatin C(Cys C) ,creatinine(Cr) ,β2‐microglobulin(β2‐MG) ,and lactate dehydrogenase(LDH) were detected .And the results for statistical analysis .Results The levels of serum Cys C , Cr ,β2‐MG and LDH were higher significantly in the MM group than in the control group ,the difference was statistically significant (P<0 .05) .In the MM group ,the positive rate of Cys C ,Cr ,β2‐MG ,LDH ,combined detection of Cys C and Cr ,combined detection of β2‐MG and LDH ,combined detection of Cys C ,Cr ,β2‐MG and LDH was 63 .0% ,42 .6% ,74 .1% ,61 .1% ,66 .7% ,85 .2% and 96 .3% respectively .The positive rate of Cys C was higher than Cr ,the difference was statistically significant(P<0 .05) .Between the positive rate of combined detection of β2‐MG ,LDH and Cys C ,Cr ,the difference was statistically significant(P<0 .05) .The positive rate of combined detection of the four biochemical tests was the most highest .Conclusion In the early stage of the renal in‐jury in the patients with MM ,the combined detection of the four biochemical tests is most sensitive to discovery the early stage of the renal glomerular function injury .

OBJECTIVE:To investigate the effects and safety of rosuvastatin on blood lipid and thyroid nodules in elderly pa-tients with dyslipidemia. METHODS:70 elderly patients with dyslipidemia were selected and randomly divided into control group (33 cases) and treatment group (37 cases). Control group was treated with routine treatment as abstain from tobacco and drink, low fat diet,Aspirin enteric-coated tablets 100 mg at bed time;treatment group was additionally treated with Rosuvastatin calcium tablet 10 mg at bed time,on the basis of control group. Both groups received the treatment for consecutive 12 weeks. BMI,blood lipid and the size of thyroid nodules were compared between 2 groups before and after treatment,and the occurrence of ADR was also observed. RESULTS:There was no significant difference in BMI between 2 groups before and after treatment (P>0.05). There was no significant difference in blood lipid level and the size of thyroid nodules between 2 groups before and after treatment (P>0.05). The blood lipid level and the size of thyroid nodule of 2 groups were improved significantly after treatment,and the treatment group was significantly better than the control group,with statistical significance(P<0.05). 2 pationts of creatment suf-fered from the increase of ALT but recovered withont angtreatment. CONCLUSIONS:Rosuvastatin can significantly improve blood lipid and decrease thyroid nodules in elderly patients with dyslipidemia with good safety.

Salvia miltiorrhiza is a traditional Chinese medicine for the treatment of cardiovascular diseases .Recently, increasing evidence demonstrates that the water-soluble compounds isolated from Salvia miltiorrhiza,including tanshinol and salvianolic acid B, exert a regulatory influence on bone metabolism .The under-lying mechanism of these compounds involves various pathways , such as Wnt/β-catenin, ERK, BMP, OPG/RANKL/RANK and FoxO mediated oxidative stress pathway .This paper reviews pre-vious effects and mechanism of polyphenolic acids in Salvia milt-iorrhiza , which may provide the base for the research and devel-opment of the new agents to treat osteoporosis .

OBJECTIVE To comprehend the main pathogens and their drug resistance of multiple organ dysfunction syndrome(MODS) patients in ICU.METHODS We retrospectively analyzed all the bacteria isolated from 40 MODS patients in ICU.RESULTS The number of bacteria strains isolated was 173,92 G-bacteria strains made up 53.18%,60 G+ bacteria strains made up 34.68%,and 21 fungi strains made up 12.14%.The top six were Staphylococcus aureus(23.70%,MRSA was 13.87%),Pseudomonas aeruginosa(14.45%),Acinetobacter baumannii(11.56%),Stenotrophomonas maltophilia(8.67%),Candida tropicalis(8.09%),and Enterococcus faecalis(7.51%).The susceptive rate of S.aureus and Enterococcus to vancomycin was all 100%,the susceptive rate of A.baumannii and Klebsiella pneumoniae to carbapenems was high.64% patients had the multiplicity of infection(MOI) which always linked with long period in ICU,respiratory failure and mechanical ventilation.CONCLUSIONS MODS patients have a high morbility of G+ bacteria,fungi and MOI,most pathogens show multi-resistance to commonly used antibiotics.Strengthening the monitoring of infection and reasonable using antibiotics should be taken.

Objective To detect the A20 gene deletion,investigate the impacts of A20 gene deletion on clinicopathological features and prognosis of DLBCL,and relationship between activation of NF-κB pathway and relative molecular pathogenesis.Methods A20 gene deletion was detected by fluorescence in situ hybridization (FISH).The expression of A20,Survivin,P65 and Ki-67 were detected by immunohistochemistry stain.Apoptosis was assayed by TUNEL.Follow-up and statistical analysis were done.Results The deletion rate of A20 gene was 21.7％.The deletion rate of A20 gene was obviously higher in ABC-like DLBCL than that in GCB-like DLBCL (30.6％ vs.8.3％,P＜0.05).It was observed that there was a negative correlation between A20 protein expression and A20 gene deletion (r=-0.259,P =0.023).The expression of P65 and Survivin protein was positively correlated with the A20 gene deletion (r=0.280,P =0.015;r =0.313,P =0.007).Apoptosis rate was significantly reduced in DLBCL patients with A20 gene deletion.The apoptosis rate was higher in cases with positive expression of A20 protein,while that was lower in cases with positive expression of p65 and Survivin protein than those with negative expression of corresponding protein.There was no statistically significant difference in apoptosis rate between ABC-like and GCB-like DLBCL patients (P＞0.05).COX regression analysis indicated that age,A20 gene deletion,types of DLBCL and Ki67 expression were independent factors associated with survival status.Log-rank test showed that there was a statistical difference in survival status between the cases with and without A20 gene deletion (P=0.015).Conclusion A20 gene deletion may associate with the attenuation of A20 protein expression.The latter weakens negative feedback regulation of A20 protein for NF-κB pathway.An up-regulated expression of Survivin and abnormal proliferation and apoptosis may be result from the abnormal activation of NF-κB.A20 gene deletion brings certain influence on clinical course and prognosis of DLBCL.
