Journal of Chinese Physician 2008;10(10):1333-1336
Objective To investigate the relationship between C-reactive protein (CRP) and carotid intima-media thickness in pa-tients with impaired glucose tolerance (IGT). Methods Senan CRP was measured with immunoturhidimetry and the carotid intima-media thickness (CAIMT) was measured using color Doppler in 108 patients with impaired glucose tolerance (IGT) and 80 subjects with normal glucose tolerance(NGT).Then we observed all IGT patients for 3 years using prospective follow-up method, Oral Glucose tolerance test (OGGT) and every index were measured in follow-up 1.5 year and 3 year. Results 2 objects were lost to follow-up. IGT group showed a significant higher CAIMT and CRP compared with NGT group. After follow-up 1.5 year and 3 year, the patients with impaired glucose toler-ance (IGT) were divided into type 2 diabetes (T2DM) group and IGT group based on the level of blood glucose. Both T2DM group and IGT group showed a significant higher CAIMT and CRP, compared with NGT group. The level of serum CRP of T2DM group was higher than that of IGT group, and the level of serum CRP of IGT group was higher than that of NGT group. There were great differences between each group.Linear correlation showed that the level of blood glucose was positively correlated with CRP and CAIMT in T2DM group after follow-up 3 year. CAIMT was positively correlated with the level of blood glucose and CRP. Mulfivariant stepwise regression showed that CRP was signifi-canfly correlated with the level of blood glucose and CAIMT. Conclusion Inflammation played an important role in the development of dia-betes, and it had great vessels complication. The patients with impaired glucose tolerance, who have high level of CRP, were facilitated to be diabetes, and they were at risk of getting great vessels complication during the phase of impaired glucose tolerance. So it would be helpful to prevent IGT patients with high CRP or CAIMT with anti-inflammatory therapy.

Aim: To study the synthesis and anti-inflammatory activity of p-(sulfamyl) benzylidene-linked hetero-cyclic ketone derivatives. Methods: A series of p-(sulfamyl) benzylidene-linked heterocyclic ketone derivatives were synthesized. Anti-inflammatory activity was evaluated against xylene-induced ear oedema in mice and against carrageenan-induced paw oedema in rats. Gastrointestinal side effects in the rats were also examined after continu-ous introgastric administration of these compounds once daily for 7 days. Results: Twelve compounds( LHZ-101-LHZ-112) were synthesized and their structures were confirmed by IR, ~1H NMR, MS and elemental analysis. LHZ-105, LHZ-106 and LHZ-111 exhibited marked anti-inflammatory activity in xylene-induced mice ear swelling model. LHZ-106 and LHZ-111 showed significant anti-inflammatory activity in carrageenan-induced rat paw ede-ma model. LHZ-105, LHZ-106 and LHZ-111 had less gastrointestinal side effects than diclofenac sodium and CI-1004. Conclusion: These results suggest that some of these compounds have the potential for anti-inflammatory activity with few gastrointestinal side effects.

Objective To investigate the effects of CD4+CD25high regulatory T cells(Treg)and the imbalance of helper T lymphocyte subsets(Th1/Th2)on the immunological mechanism of IgA nephropathy(IgAN)patients. Methods The percentage of Treg and helper T cells subpopulation (Th1/Th2)in the peripheral blood of IgAN patients and healthy controls was examined by flow cytometry. The FOXP3 expression was detected through intracellular staining. The correlation of Treg or Th1/Th2 with clinical parameters of IgAN was analyzed by Spearman or Pearson rank correlation test. Results The percentages of Treg and Th2 cells were significantly higher in peripheral blood of IgAN patients compared to that of healthy controls[Treg (2.14±0.82)%vs[1.59±0.53)%,Th2(2.57±0.72)%vs(1.81±1.10)%,all P<0.05].Th1/Th2 ratio was significantly reduced in IgAN patients(5.75±1.89 vs 12.73±9.79,P<0.05).The percentage of circulating Treg cells was positively correlated with serunl IgA concentration(r=0.397,P<0.05),and was negatively correlated with eGFR(r=-0.376,P<0.05).The percentage of circulating Th2 cells was positively correlated with serum IgA(r=0.468,P<0.05). Conclusions There is a disorder of T lymphocyte population in the peripheral blood of IgAN patients. The increased Treg and Th2 cells may play an important role in the pathogenesis of IgAN.

Journal of Clinical Hepatology 2019;35(1):119-122
ObjectiveTo investigate the expression features and clinical significance of the microRNA-888 (miRNA-888) gene family members in hepatocellular carcinoma (HCC). MethodsA total of 72 patients with pathologically confirmed HCC who were treated in The First Affiliated Hospital of Hunan University of Chinese Medicine from January 2012 to December 2017 were enrolled, and 72 liver tissue samples collected from healthy volunteers were enrolled as normal control group. Quantitative real-time PCR and in situ hybridization were used to measure the expression of miRNA-888 family members (miRNA-888, miRNA-891a, miRNA-891b, miRNA-892a, and miRNA-892b) in HCC tissue, and the association of the expression of miRNA-888, miRNA-891b, and miRNA-892a with the clinicopathological features of HCC was analyzed. The t-test was used for comparison of continuous data between two groups, and the chi-square test was used for comparison of categorical data between two groups. ResultsHCC tissue had significantly higher expression of miRNA-888, miRNA-891b, and miRNA-892a than normal liver tissue (miRNA-888: 2.53±0.75 vs 0.46±0.08, t=14.02, P<0.001; miRNA-891b: 2.26±0.38 vs 1.19±0.21, t=7.75, P<0.001; miRNA-892a: 5.44±1.01 vs 1.06±0.30, t=35.27, P<0001). According to the results of in situ hybridization, the miRNAs of miRNA-888, miRNA-891b, and miRNA-892a were mainly located in the cell nucleus of HCC tissue, and there were significant differences in the expression of these miRNAs between HCC tissue and normal tissue (miRNA-888: 3.91±0.92 vs 1.21±0.42, t=22.65, P<0.001; miRNA-891b: 2.92±0.76 vs 0.83±0.21, t=2292, P<0001; miRNA-892a: 3.81±0.99 vs 1.30±0.32, t=20.47, P<0001). There were significant differences in the expression of miRNA-888 and miRNA-891b between the patients with different histological grades and clinical stages (χ2=6.25, 4.44, 4.76, and 6.05, all P<0.05), and there was a significantly higher expression of miRNA-892a in stage III/IV tumors (χ2=8.50, P<0.001). ConclusionThere are significant increases in the expression of miRNA-888, miRNA-891b, and miRNA-892a in HCC tissue, and such increases are closely associated with the malignancy of HCC.
