1.The Interventional Effects of Yufeining on Lung Function in Patients with Chronic Obstructive Pulmonary Disease

Objective:To evaluate the interventional effects of Yufeining on lung function in patients with chronic obstructive pulmonary disease (COPD). Methods:38 patients with COPD were divided into therapeutic group (20 cases) and control group (18 cases) randomly. Therapeutic group was treated with oral Yufeining pill half a year and then was followed up 6 months without drug. Control group was not given any medicine during the observed one year. Routine lung function was measured at before 、6 months and 12 months after treatment. Total symptom score (TSS) of traditional Chinese medicine、the scores of quality of life (QOL) were recorded simultaneously. Blood gas analysis was measured only at before and 6 months after. 2.Biomechanics research on ATP6i gene-interference treating steroid-induced necrosis of the femoral heads in rats

Objective To explore the biomechanics mechanism of ATP6i gene-interference treating steroid-induced necrosis of the femoral heads(SINFH)in rats.Methods Forty SD rats were randomly assigned to four groups:blank control group,experimental control group,ATP6i SiRNA(+)group and ATP6i SiRNA(-)group,10 rats in each group.We established rat models of SINFH by injecting Escherichia Coli Endotoxin and prednisolone for 12 weeks,then cut along the inner side of the patella for about 1 cm and turn the patellar outside of the both knees of the experimental control group,the ATP6i SiRNA(+)group and the ATP6i SiRNA(-)group,and drill from the intercondylar until the femoral neck.Rats were treated by transfecting ATP6i small interference RNA(ATP6i SiRNA)into the osteoclasts of the femoral heads to interference the expression of ATP6i SiRNA(+),or by the ATP6i SiRNA(-)and same amount of saline.One month later,test the load,stress,modulus of elasticity,and axial stiffness of the right femoral heads of all four groups,and at the same time take the left femoral heads of all four groups for pathological study.Results The load,stress,modulus of elasticity,and axial stiffness of the ATP6i SiRNA(+)group were(307.01±27.34)N,(60.44±5.38)MPa,(125.29±11.16)MPa,(162.42±14.45)N/mm,respectively,which were much greater than the ATP6i SiRNA(-)group and the experimental control group(P<0.05).In the pathological studies,we found that the trabecula of the femoral heads of the ATP6i SiRNA(+)group was much thicker than the ATP6i SiRNA(-)group and the experimental control group,but the degeneration,necrosis and breakage of the osteoepiphysis in each group had no statistically significant differences.Conclusion ATP6i gene-interference can inhibit the acid-secreting function of osteoclasts and improve the biomechanical property of the femoral heads of rats in treating the SINFH.
3.Effects of IL-12p35 small interference RNA on immune function of dendritic cells in rats

Objective:To investigate the effects of IL-12p35 small interference RNA on immune functions of Dendritic cells in rats.Methods:DCs were generated by culturing bone marrow progenitor cells of rats with GM-CSF, IL-4 and LPS in vitro. The IL-12p35 siRNA was synthesized and transfected into DCs. The expressions of CD80 and MHCⅡwere examined by flow cytometry. Reverse transcriptase PCR analysis was used to detect IL-12p35 mRNA transcription and ELISA was used to detect IL-12 and IL-10 protein levels, respectively. The antigen presenting by DCs was evaluated by mixed lymphocyte responses.Results:IL-12p35 siRNA silenced DCs expressed lower IL-12, higher IL-10,and exhibited weak activity in stimulating the proliferation of allogenic T cells, but no changes on CD80 and MHCⅡexpressions.Conclusion:IL-12p35 siRNA interference exerts no effects on maturation of DC, but negative effect on immume function of DCs.
4.cDNA cloning and sequencing of MH2 domain of Smad1 from human dental papilla cells

Objective:To clone and sequence cDNA of MH2 domain of Smad1 gene from human dental papilla cells. Methods:Total RNA was isolated from primarily cultured human dental papilla cells and reversely transcribed into single stranded cDNA.The desired DNA product was obtained by PCR with two gene specific primers.The segment was inserted into PUC19 vector and the plasmid was transformed into E.coli JM109.The double stranded cDNA of positive clone was sequenced.Results:The sequence of MHz domain of Smad1 cloned from human dental papilla cells was consistent with that reported by Hoodless et al. Conclusion:Smad1 exists in human dental papilla cells,BMP signaling may be mediated by smad1 in human dental papilla cells.

This article reviews the historical development and up-to-date state of thermometric technologies for measuring human body temperature (BT) from two aspects: measurement methodology and signifi cance interpretation. Since the fi rst systematic and comprehensive study on BT and its relation to human diseases was conducted by Wunderlich in the late 19th century, BT has served as one of the most fundamental vital signs for clinical diagnosis and daily healthcare. The physiological implication of BT set point and thermoregulatory mechanisms are briefl y outlined. Infl uential determinants of BT measurement are investigated thoroughly. Three types of BT measurement, i.e., core body temperature, surface body temperature and basal body temperature, are categorized according to its measurement position and activity level. With the comparison of temperature measurement in industrial fi elds, specialties in technological and biological aspects in BT measurement are mentioned. Methodologies used in BT measurement are grouped into instrumental methods and mathematical methods. Instrumental methods utilize results of BT measurements directly from temperature-sensitive transducers and electronic instrumentations by the combination of actual and predictive measurement, invasive and noninvasive measurement. Mathematical methods use several numerical models, such as multiple regression model, autoregressive model, thermoregulatory mechanism-based model and the Kalman fi lter-based method to estimate BT indirectly from some relevant vital signs and environmental factors. Thermometry modalities are summarized on the dichotomies into invasive and noninvasive, contact and noncontact, direct and indirect, free and restrained, 1-D and n-D. Comprehensive interpretation of BT has an equal importance as the measurement of BT. Two modes to apply BT are classifi ed into real-time applications and long-term applications. With rapid advancement in IoT infrastructure, big data analytics and AI platforms, prospects for future development in thermometry and interpretation of BT are discussed.

7.Simultaneous Determination of Ornidazole and Dexamethasone Sodium Phosphate in Compound Ornidazole Film by HPLC

OBJECTIVE:To establish a method for the simultaneous determination of ornidazole and dexamethasone sodium phosphate in Compound ornidazole film. METHODS:HPLC was performed on the column of Inertsil ODS-3 with mobile phase of methanol-20 mmol/L phosphate buffer(pH was adjusted to 7.40 with glacial acetic acid)(55∶45,V/V)at a flow rate of 1.0 ml/min, column temperature was 30 ℃,detection wavelength was 242 nm and volume injection was 10 μl. RESULTS:The linear range were 1-100μg/ml for both ornidazole(r=0.999 7)and dexamethasone sodium phosphate(r=0.999 9);RSDs of precision,stabili-ty and reproducibility tests were lower than 2.0%;recoveries were 96.50%-99.80%(RSD=1.02%,n=9) and 96.50%-99.60%(RSD=0.99%,n=9). CONCLUSIONS:The method is specific with good precision and stability and high accuracy,and can be used for the simultaneous determination of ornidazole and dexamethasone sodium phosphate in Compound ornidazole film.
8.Efficacy of dexmedetomidine and dezocine used to supplement awake tracheal intubation assisted by fiberoptic bronchoscope in elderly patients

Objective To evaluate the efficacy of dexmedetomidine and dezocine used to supplement awake tracheal intubation assisted by fiberoptic bronchoscope (FOB) in elderly patients.Methods Sixty elderly patients aged 65-77 yr,of ASA physical status Ⅱ or Ⅲ (Mallampati grade Ⅰ or Ⅱ),scheduled for elective surgery under general anesthesia,were randomly divided into 3 groups (n =20 each) using a random number table:dezocine group (group DEZ),dexmedetomidine group (group DEX) and dezocine combined with dexmedetomidine group (group DEZ+DEX).Dezocine 0.1 mg/kg was injected intravenously in group DEZ.Dexmedetomidine 0.4 μg/kg was infused intravenously over 10-15 min in group DEX.In group DEZ+DEX,dexmedetomidine 0.4 μg/kg was infused intravenously over 10-15 min,and dezocine 0.1 mg/kg was injected simultaneously.Laryngeal mucous membrane was sprayed with 2% lidocaine for topical anesthesia during infusion in all the three groups.In addition,1% tetracaine 3 ml was injected into trachea through cricothyroid membrane.Awake tracheal intubation was performed and assisted by FOB after the end of administration in all the three groups.Cardiovascular response (MAP or HR>30% of baseline values) and respiratory depression (SpO2<90% and RR<8 bpm) were recorded during the period between induction of anesthesia and 3 min after intubation was completed.The intubation time was recorded.The tolerance of tracheal tube was assessed in the patients.At the time of topical anesthesia,when epiglottis came into view,immediately after tracheal tube was successfully inserted into trachea,and at 3 min after successful intubation,perfusion index and Ramsay sedation score,and patients' satisfaction with the sedation (Ramsay sedation score 2-4) were recorded.Results Compared with group DEZ or DEX,the tolerance of tracheal tube was significantly enhanced,intubation time was shortened,the rate of satisfactory sedation was increased,perfusion index and the incidence of cardiovascular response were decreased in DEZ+DEX group.There was no significant difference in respiratory depression among the three groups.Conclusion Dexmedetomidine and dezocine can provide better condition for awake tracheal intubation assisted by FOB than dexmedetomidine or dezocine alone in elderly patients.
9.Diagnosis and treatment of intrahepatic biliary cystadenoma: a report of 7 cases

To investigate the clinical characteristics,diagnosis and treatment of intrahepatic biliary cystadenoma,a total of 7 patients with intrahepatic biliary cystadenoma admitted in our department from January 2008 to June 2013 were enrolled and the clinical data (i.e.clinical manifestations,imaging features and surgical treatment) were analyzed retrospectively.All the 7 patients were female and diagnosed as intrahepatic biliary cystadenoma.Five cases complained right upper abdominal pain of various severities.Color Doppler ultrasound,CT and MRI confirmed the diagnosis in all patients.The surgery included irregular fight hepatectomy (n =2),left lateral lobe hepatectomy (n =3),and left hepatectomy (n =2).Patients with intrahepatic biliary cystadenoma usually manifest nonspecific clinical symptoms,and thus are prone to be misdiagnosed as simple hepatic cysts.For such patients,surgery is of first choice,and complete resection is the key to avoiding the recurrence and malignancy.
10.Effects of dexmedetomidine on cellular immune function during analgesia with morphine after radical resection for esophageal cancer

Objective To evaluate the effects of dexmedetomidine on the cellular immune function during analgesia with morphine after radical resection for esophageal cancer in the patients.Methods Sixty patients of both sexes,of ASA physical status Ⅰ or Ⅱ,after radical resection for esophageal cancer under general anesthesia,were randomly divided into 2 groups (n =30 each) using a random number table:control group (group C) and dexmedetomidine group (group Dex).Patient-controlled intravenous analgesia (PCIA) was performed immediately after operation in the two groups.In group C,the PCIA solution (150 ml) contained morphine 0.48 mg/kg.In group Dex,the PCIA solution (150 ml) contained morphine 0.48 mg/kg and dexmedetomidine 1 μg/kg.The postoperative visual analogue scale (VAS) scores were maintained ≤ 3.The consumption of morphine was recorded within 24,48 and 72 h after operation.The adverse effects such as nausea,vomiting,pruritus,bradycardia,hypotension,oversedation and respiratory depression were also recorded after operation.Before induction of anesthesia (T0),immediately after extubation (T1),and at 24,48 and 72 h after operation (T2-4),venous blood samples were obtained for determination of the levels of T-lymphocyte subsets (CD3+,CD4+,CD8+) and natural killer (NK) cells by flow cytometry.CD4+/CD8+ ratio was calculated.Results Compared with group C,the consumption of morphine within 24,48 and 72 h after operation and incidence of nausea,vomiting and pruritus after operation were significantly decreased in group Dex.The levels of CD3+,CD4+,CD4+/CD8+ ratio and NK cells were significantly lower at T1-4 than at T0 in the two groups.The levels of CD3+,CD4+,CD4+/CD8+ ratio and NK cells were significantly higher at T1-4 in group Dex than in group C.Conclusion Dexmedetomidine can improve the cellular immune function during analgesia with morphine after radical resection for esophageal cancer in the patients.
