1.Characteristics of emergency injury incidents in sentinel hospitals of Fuyang District

Objective:
To investigate the epidemiological characteristics of injury cases admitted to department of emergency in sentinel hospitals in Fuyang District, Hangzhou City from 2015 to 2020, so as to provide insights into injury prevention and intervention in rural areas.
Methods:
Data of injury cases were collected from the department of emergency of two sentinel hospitals in Fuyang District from 2015 to 2020, and the incidence of injury was standardized by the data from the China's seventh national population census in Fuyang District. The demographics, causes, severity and outcomes of injury were descriptively analyzed.
Results:
Totally 84 360 injury cases were recorded in the department of emergency of two sentinel hospitals in Fuyang District from 2015 to 2020, and the annual average standardized incidence of injury was 15.85%. The injury cases included 48 330 men (57.29%) and 36 030 women (42.71%), and there were 10 653 cases at ages of <19 years (12.63%), 25 398 cases at ages of 19 to <46 years (30.11%), 39 951 cases at ages of 46 to <71 years (47.36%), and 8 359 cases at ages of 71 years and older (9.91%). Taking a private car (motor vehicles and non-motor vehicles) was the predominant way to seek medical care (75 246 cases, 89.20%). Blunt force injury and puncture wound/incision wound (23 668 cases, 28.06%), and fall injury (22 855 cases, 27.09%) were predominant causes of injury, and there were 45 961 cases with mild injury (54.48%), 28 043 cases with moderate injury (33.24%), and 356 cases with severe injury (0.42%). Returning home after hospital treatments was the predominant outcome of injury (74 401 cases, 88.19%). Mild injury was predominant among patients at ages of <19 years (81.48%), with a high rate of transfer to other hospitals (2.43%), and moderate to severe injury was predominant among patients at ages of 71 years and older (43.08% and 0.77%), with a high rate of hospital stay (18.71%) and a high mortality rate (0.14%).
Conclusions
Mild injury was predominant among patients admitted to the department of emergency of two sentinel hospitals in Fuyang District from 2015 to 2020, and blunt force injury, puncture wound/incision wound and fall injury were predominant causes of injury. There were age-specific epidemiological characteristics of injury cases.
2.The feasibility study of radiation absorbed dose measurement using oligonucleotide dually labeled with Alexa Fluor 488 and BHQ1 probe

Objective To study the feasibility of measuring radiation absorbed dose with the fluorescent probe Alexa Fluor 488-DNA-BHQ1.Methods An oligonucleotide dually labeled at its 5'-and 3'-end with fluorescent molecular Alexa Fluor 488 and specific fluorescence inhibitors BHQ1 was prepared.The Alexa Fluor 488-DNA-BHQ1 aqueous solution was exposed with X-ray and its fluorescence intensity was measured.Results When the concentration of Alexa Fluor 488-DNA-BHQ1 was between 0.5 and 1 μmol/L,the fluorescence intensity of its aqueous solution had excellent linear dose response from 0.1 to 30 Gy (R2 =0.99) and it was stably maintained after 40-80 min of irradiation especially at 4℃.Conclusions In the dose range of 0.1-30 Gy,the Alexa Fluor 488-DNA-BHQ1 fluorescent probe can be used to measure radiation absorbed dose.
3.Radiosensitizing effects of miR-101 on HeLa cancer cells and underlying mechanism

Objective To study the effects of microRNAl01 (miR-101)on radiosensitization of human uterine cervix cancer HeLa cells and underlying mechanism.Methods HeLa cells were divided into three groups including blank control,miRNA negative control and miR-101 transfection group.The cells were irradiated by 160 kVp X-ray generated from a linear accelerator at a dose rate of 1.15 Gy/min.Real-time quantitative PCR (qRT-PCR) was used to detect the expression of miR-101.The clonogenic survival assay was applied to evaluate the effect of miR-101 on radiosensitization of HeLa cells.γ-H2AX immunofluorescence and Western blot assays were performed to observe DNA double-strand breaks and the protein expressions of ATM and DNA-PKcs of HeLa cells,respectively.Results Compared with the negative control group,the expression of miR-101 was significantly increased in the HeLa cells at 48 h after transfection with miR-101 mimic,and the survival of HeLa cells over expression of miR-101 was significantly reduced(t =10.75,P < 0.05).The miR-101 had remarkable radiosensitive effect on HeLa cells(F =7.72,P <0.05) with a SERD0 of 1.29.Moreover,over-expression of miR-101 could inhibit the repair of DNA damage induced by irradiation.Compared with the control group,the protein expressions of ATM and DNA-PKcs were significantly decreased in the HeLa cells over expression of miR-101.Conclusions Over-expressions of miR-101 could inhibit cell growth and enhance radiosensitivity of HeLa cells by inhibiting the repair of radiation-induced DNA damage.
4.Evaluation of Different Surgical Procedures for the Treatment of Lumbar Spondylolisthesis: A Clinical Analysis of 202 Cases

Purpose: Through the analysis of 202 cases of spondylolisthesis treated with surgery, the different types of surgical procedures were reevaluated. Material and Method: Patients with spondylolisthesis were divided into 3 groups based upon their pathology and different surgical procedures were used accordingly. The results were evaluated after follow-up of more than one year. Group Ⅰ consisted of 38 patients with degenerative spondylolisthesis with back pain and isthmic spondylolisthesis of less than 33%; they were treated with anterior intervertebral body fusion. Group Ⅱ consisted of 99 cases of mild isthmic spondylolisthesis with unilateral sciatica, and were treated with semi-laminectomy and decompression of symptomatic side; interlaminar and interspinous process fusion of the opposite side was performed for 99 cases of mild isthmic spondylolisthesis with unilateral sciatica. Group Ⅲ consisted of 65 cases of spondylolisthesis more than 33% with sciatica and they were treated with decompression, reduction of sliding vertebra and posterior intervertebral body fusion. Result: In group Ⅰ, solid fusion was in 20 cases giving a fusion rate of 91%. In group Ⅱ, the satisfactory rate was as high as 91.5%. In group Ⅲ, the satisfactory rate and fusion rate reached 91.8% and 95.9% respectively. Conclusion: Reduction of spondylolisthesis could increase the fusion contact area, restore normal mechanics and saggital curve of the lumbar spine, relieve neural compression and improve the extemal appearance and function. For the surgical treatment of spondylolisthesis, the major goal should toward a solid fusion between the sliding vertebra and its adjacent vertebrae and laminectomy alone is not a procedure of choice.
5.Effects of AuNPs@PEG-AS1411 nanoparticles on radiosensitization of HeLa cancer cells

Objective To study the effects of AuNPs@PEG-AS1411 nanoparticles on radiosensitization of human uterine cervix cancer HeLa cells.Methods AuNPs were synthesized by citrate reduction method and then functioned with PEG and PEG-AS1411, respectively.CCK-8 assay and colon forming assay were used to detect the acute and chronic toxicity effects of AuNPs on HeLa cells, respectively.At the same time, clonogenic survival assay was applied to measure the cell survival rate of HeLa cells after exposure to AuNPs@PEG and AuNPs@PEG-AS1411 combined with X-ray radiation.The intracellular uptake of AuNPs@PEG and AuNPs@PEG-AS1411 in HeLa cells were detected by ICP-MS.Results The CCK-8 assay showed that AuNPs@PEG and AuNPs@PEG-AS1411 were not toxical on HeLa cells(P >0.05).But the clonogenic survival assay showed that AuNPs@PEG and AuNPs@PEG-AS1411 had toxicity on HeLa cells significantly after 10 d(t =4.38-11.60, P < 0.05).AuNPs functioned with AS1411 could increase the cellular uptake of AuNPs.AuNPs@PEG and AuNPs@PEG-AS1411 both had significant radiosensitive effect on HeLa cells (F =7.90,48.23, P < 0.05).The values of SERDo for AuNPs@PEG and AuNPs@PEG-AS1411 were 1.12 and 1.20, respectively, when the concentration of Au was 10 mg/L.Conclusions AuNPs@PEG and AuNPs@PEG-AS1411 could cause chronic toxicity on HeLa cells instead of acute effect.PEGylated AuNPs functioned with AS1411 could enhance the radiosensitivity of HeLa cells in vitro.
6.Intumescent abdomen,diminish strength and pericarditis

There was a 1 1 -year -old school -aged girl complaining of abdomen intumescing and declining physical fitness for 1 6 months,and hydropericardium for 5 months.The child had a intumescent abdomen and strength diminished strength suddenly.After the strenuous exercise she was more tired than before and lost her appetite.The girl was found cardiac enlargement and calcification of pericardium during security check at the airport.The thoracoabdomi-nal computed tomography(CT)suggested hydropericardium,hydrothorax effusion in the right,and seroperitoneum,pel-vic effusion.The girl had no response to pericardiocentesises,anti -inflammation and antituberculosis therapies.The in-flammatory markers and the findings of autoimmunity were normal after her admission.The purified protein derivatives (PPD)test was (++),but the antituberculosis therapy was invalid,so the diagnosis was unclear.The she had peri-cardiectomy.The pericardium visceral and parietale′s pathology showed hyperplasia,hyalinosis and organization of fi-brous connective tissue,congestion of the blood capillaries,infiltration of inflammatory cells.Terminally,she was diag-nosed as constrictive pericarditis.Symptoms disappeared after treatments with cardiotonic,diuretic and potassium sup-ply.The comprehensive analysis is important clinically,the possible causes should be removed gradually,and pathologi-cal examination must be emphasized during the diagnosis of constrictive pericarditis.
7.Inhibitory effects of autophagy inhibitor 3-MA on high glucose-induced proliferation of human retinal pigment epithelium cells

Background Diabetic retinopathy (DR) is one of the common complications of diabetes.Retinal pigment epithelium (RPE) cells,as important constituent cells of the retina,play important roles in the development and progression of DR.Objective This study was to investigate the suppressive effects of autophagy inhibitor 3-MA on the proliferation of human retinal pigment epithelium cells (hRPECs) following high glucose culture.Methods HRPECs were divided into control group,high-glucose group and 3-MA+high glucose group.The cells were cultured by the DMEM/F12 with 5 mmol/L glucose in the control group,and by the DMEM/F12 with 5 mmol/L glucose in the high glucose group and by the DMEM/F12 with 10 mmol/L 3-MA (for 1 hour firstly) and 30 mmol/L glucose in the 3-MA+high glucose group.The cells were inoculated into 24-well plate with the content of 1 ×105/well,and then the cells were consecutively cultured for 24 hours with DMEM/F12 containing 0.5% fetal bovine serum after achieved attached 80% confluence.The morphology and uhrastructure of the cells were examined by optics microscope and transmission electronic microscope.The proliferative rate of the cells was assayed by CCK-8 kit.The expression of autophagy-related gene microtubule associated protein light chain 3B (LC3B) in the cells was detected by Western blot and LC3B-Ⅱ/LC3B-Ⅰ value was quantitatively evaluated among the groups.Results The cells grew well with uniform size and regulatory arrangement in the control group.The cells in the high glucose group enlarged and the number of cells evidently increased.In the 3-MA+high glucose group,the cells decreased with a disorder arrangement.Under the transmission electron microscope,the cells were normal with the round-or oval-like nucleus and normal organelles in the control group,and autolysosome could be seen in the cells in the high glucose group.In the 3-MA+high glucose group,some autophagic bodies were found.The proliferative rate of the cells was (100.0±2.0) %,(116.9-±5.2)% and (103.7 ±4.7)% in the control group,high glucose group and 3-MA+high glucose group respectively,showing a significant difference among the groups (F =13.526,P =0.006).The proliferative rate was considerably raised in the high glucose group compared with the control group and 3-MA+high glucose group (both at P<0.05),but there was no significant difference in the proliferative rate of the cells between the control group and 3-MA+high glucose group (P>0.05).Compared with the control group,the expressing intensity of LC3B-Ⅰ protein was weakened and that of LC3-Ⅱ protein was enhanced,and the expression intensity of LC3B-Ⅰ and LC3B-Ⅱ proteins in the 3-MA+high glucose group was similar to that in the control group.The LC3-Ⅱ/LC3-Ⅰ ratio was 0.131 ±0.065,2.504±0.097 and 0.274±0.007 in the control group,high glucose group and 3-MA+high glucose group,respectively,with significant differences among the groups (F =1 694.676,P =0.000),and the LC3-Ⅱ/LC3-Ⅰ ratio was increased in the high glucose group in comparison with the control group and the 3-MA+high glucose group (all at P<0.05).No significant difference was found in the LC3B-Ⅱ/LC3B-Ⅰ ratio between the control group and 3-MA + high glucose group (P > 0.05).Conclusions High glucose culture of hRPECs can activate autophagy process and promote cell proliferation.3-MA,an autophagy inhibitor,suppresses the high glucoseinduced growth of HRPECs by inhibiting autophagy process.
8.Transfection of calcitonin gene-related peptide mediated by lentivirus vec-tor in vitro and its effects on cardiac stem cell viability

AIM:To investigate the effect of calcitonin gene-related peptide (CGRP)transfection into c-kitpos cardiac stem cells (c-kit+CSCs) on the cell viability.METHODS: Under the sterile condition, the auricles of SD rats were taken out , and then c-kit +CSCs were collected through enzyme digestion and immunomagnetic bead separation (MACS).The cells were identified by flow cytometry .c-kit +CSCs were transfected with enhanced green fluorescent pro-tein CGRP lentiviral vector ( Lv-EGFP-CGRP) or enhanced green fluorescent protein lentiviral vector ( Lv-EGFP).The cells were randomly divided into Lv-EGFP-CGRP-CSCs group , Lv-EGFP-CSCs group and CSCs group .The transfection was observed under the fluorescence microscope .The transfection efficiency was detected by flow cytometry .The CGRP protein secretion in the cell culture supernatants was detected by ELISA .The viability of c-kit+CSCs transfected with Lv-EGFP-CGRP or Lv-EGFP was measured by CCK-8 assay.RESULTS:c-kit+CSCs were isolated and cultured successfully .The expression positive rate of c-kit was 91.0%and the expression positive rates of CD 45 and CD34 were 4.5% and 4.0%, respectively.After transfected with lentivirus for 48 h, the stable fluorescence in c-kit +CSCs was observed under fluores-cence microscope .The transfection efficiency were 80%when MOI was 20.The level of CGRP was significantly increased in Lv-ECFP-CGRP-CSCs group compared with Lv-EGFP-CSCs group and CSCs group (P<0.05).Meanwhile, transfection with lentiviral vector in each group did not affect the viability of c-kit+CSCs.CONCLUSION:Transfection of Lv-EGFP-CGRP into c-kit+CSCs was successful .The secretion of CGRP was found in the transfected c-kit+CSCs and the viability was not changed after transfection .CGRP-modified c-kit+CSCs may play a role in treating myocardial infarction .
9.Research and investigation of professional values among nursing students and registered nurses

Objective To investigate the nursing professional values of different nursing groups and probe into the influencing factors.Methods By cluster and stratified sampling,a total of 583 participants including nursing students and registered nurses were investigated by Nursing Professional Values Scale.Results Nursing students and registered nurses hold a higher level of nursing professional values.Nursing professional value scores of nursing students were significantly higher than registered nurses.There was statistical difference between different nursing groups.Working years and monthly income were important influencing factors.Conclusions Nursing educators and administers could choose effective methods according to the condition to improve nursing professional values.
10.Clinical observation on effects of Qingjiefang on early diabetic nephropathy and insulin resistance

Objective To observe the effects of Qingjiefang on insulin resistance and reduction of microalbuminuria in early diabetic nephropathy.Methods A total of 64 patients with early diabetic nephropathy were randomly recruited into a control group and a treatment group.The control group(32 cases)was treated with conventional western medicine,while the treatment group(32 cases)was treated with Qingjiefang based on the control group.The changes of the fasting plasma glucose(FPG),2hBG,glycosylated hemoglobin(HbAlC),fasting Insulin(FINS),24 hours urinary albumin excretion rate (UAER).body mass index(BMI)and insulin resistance(IR)were observed before and after the treatment.Results After the treatment,FINS,IR and UAER were decreased in the treatment group,the comparison with the control group showed significant difference (P<0.05).Conclusion Qingjiefang can effectively improve insulin resistance and prevent early diabetic nephropathy.
