Effects of Sini decoctiou (SD ) on ischemic (anoxic ) electrocardiogram (ECG ) and possible action mechanism of SD were studied.Results indicate that SD significantly improves the pituitrin induced ischemic ECG of rabbits, significantly prevents S-T scgment from desconding and suppresses the elevation of T wave; SD can also lengthen significantly cardioe lectric activity time of anoxic mice. The protective cffects of SD on ischemic (anoxic ) myocardium may be related to the significant increase of myocardial nutritional blood flow induced by administrating SD.

BACKGROUND:Biocompatibility is a key parameter in drug delivery systems. In general, to obtain a proper microparticle carrier, the materials themselves should have excellent biocompatibility. Besides, spherical degree and surface smoothness both have significant influence on biocompatibility.OBJECTIVE: To obtain spherical and smooth poly (lactide-co-glycolide)microparticles, so as to improve the property of biocompatibility.DESIGN: Open experiment.SETTING: Research Laboratory of Biomaterials, Jinan University.MATERIALS: The experiment was conducted in the Research Laboratory of Biomaterials, Jinan University, between June 2004 and January 2005.The materials included poly(lactide-co-glycolide), lysozyme and poly (vinyl alcohol). The other reagents were analytical pure. The instruments included a homogenizer, a mechanical stirrer, an ultrasonic cleaning instrument, a scanning electron microscope and an atomic force microscope.METHODS: ① Preparation of microparticles: Lysozyme was selected as a model protein for encapsulation into poly(lactide-co-glycolide) using a dou-ble emulsion solvent extraction/evaporation method. Three separation methods, namely direct vacuum freeze-drying, filtration and centrifugation,were investigated and compared. ② Observation under the scanning electron microscope: We observed the effect of three separation methods on the shape of microparticles. All the samples were attached to copper mounts and coated with gold, and then were observed with an electron microscope.③ Observation under the atomic force microscope: The morphological structure of the surface was analyzed with atomic force microscope.RESULTS: ① Observation results of scanning electron microscope: Compared with direct vacuum freeze-drying and filtration, centrifugation method was more efficacious in obtaining spherical and smooth microparticles based on the scanning electron microscope pictures. But results also warned us to be more careful when we prepared scanning electron microscope samples using ultrasonic to separate the aggregates. ② Atomic force microscope results indicated that the surface was smooth with the average roughness of 48.55 nm.CONCLUSION: By investigating the influence of different downstream processes, we can obtain spherical and smooth products. Besides, a new one-step method is put forward in constructing some microparticle-combined polymer based scaffolds because the combined scaffolds and microparticles are formed synchronously.

Objective To explore the genotype distribution of lipoprotein lipase gene polymorphism at Ser447Stop locus and its possible association with metabolic syndrome and dietary intakes. Method Ser447Stop polymorphism was determined by PCR-PFLP method in 222 adults with MS and 222 normal adults as control. Their physical examination,dietary investigation and levels of biochemical profile,including BG,TG,TC and HDL-C were analyzed. Results (1) The genotype frequencies of Ser447Stop SS,SX and XX were 85.6%,13.3% and 1.1% respectively,which were in agreement with Hardy-Weinberg equilibrium. There was no significant difference in frequencies of genotypes or allele between MS and the control,and between male and female. (2) After adjusting age and gender,the levels of serum TG were significantly different among three genotype groups,the highest in SS genotype and the lowest in XX genotype. (3) After adjusting age,gender and body mass index,the intakes of protein and carbohydrate were significantly different among three genotype groups. (4) There was significantly different in negative correlation between the intakes of protein and serum TG levels after adjusting age,gender and body mass index. Conclusion Lipoprotein lipase gene polymorphism influenced serum TG levels,while associated with protein intakes. It might contribute to the predisposition in metabolic syndrome response to dietary intervention.

Objective To evaluate the association between methylenetetrahydrofolate reductase (MTHFR) C677T gene polymorphism and depression risk.Methods Available studies containing genotype frequencies of MTHFR C677T were chosen,and pooled odds ratio (OR) with 95％ confidence interval (CI) was used to assess the strength of the association.Results Twenty-five eligible studies including 4 048 cases and 12 827 controls were identified.Meta-analysis showed a significant effect in the co-dominant model (T vs.C:OR 0 =1.25; Z=3.82,P=0.000;TTvs.CC:OR1=1.55,Z=3.79,P=0.000 ; except CT vs.CC:OR2=1.13; Z=1.78,P=0.076).Stratified analysis denoted that there was significantly higher risk in depression onset before 60 years old than depression onset after 60 years old.In subgroup analysis stratified by regressive meta-analysis,MTHFR C677T variant was statistically significantly relevant to depression risk in Chinese populations (OR 0 =1.58,Z=3.95,P=0.000; OR 1 =2.55,Z=3.92,P=0.000; OR 2 =1.43,Z=2.72,P=0.007),but not in no-Chinese populations (OR 0 =1.12,Z=2.10,P=0.035 ; OR 1 =1.23,Z=2.23,P=0.025 ; OR 2 =1.06,Z=0.74,P=0.456).Evidentiary strength rated by GRADE showed that 1 study moderate,3 studies low and 7 stdudies very low.Conclusion Our study provides evidence for a hereditary association between the MTHFR C677T variant and depression susceptibility,whereas there is an obvious variety in different regions,ethnicities and onset age.

Objective: To observe the protective effects of the alcohol extract, ethyl acetate extract and 1-butanol extract of Euonymus hederaceuson on H2O2-induced oxidative damage of vascular endothelial cells ECV-304.Methods: ECV-304 cells were cultured in vitro and H2O2-induced oxidative damage in ECV-304 cells was used as the model.After the treatment with ethanol extract, ethyl acetate extract or 1-butanol extract of Euonymus hederaceus (the final concentration of 400, 200, 100, 50 and 25 μg·ml-1) for 24h, the cell viability was detected by MTT assay.Hoechst staining was used to observe the cell apoptosis, and then the supernatant was collected, and the contents of SOD, NOS and NO were used to determine the effects of the extracts on the oxidative stress of vascular endothelial cells.Results: The ethanol extract, ethyl acetate extract and 1-butanol extract of Euonymus hederaceus within the concentration range of 25-400 μg·ml-1 showed no cytotoxicity on ECV-304 cells, and the ethyl acetate extract at the concentration of 400μg·ml-1 had certain proliferation effect on the cells (P<0.05).The ethyl acetate extract at the concentration of 50-400 μg·ml-1 and the 1-butanol extract at the concentration of 100-400 μg·ml-1of Euonymus hederaceus could increase the survival rate of ECV-304 cells with oxidative injury significantly, and the OD value showed significant difference when compared with that of the model group (P<0.05 or P<0.01).The ethanol extract, ethyl acetate extract and 1-butanol extract of Euonymus hederaceus could significantly improve the SOD and NO levels in ECV-304 cells with oxidative damage (P<0.05 or P<0.01), and had no notable influence on NOS level.The ethanol extract, ethyl acetate extract and 1-butanol extract of Euonymus hederaceus could inhibit the cell apoptosis with oxidative damage and improve the nuclear morphology, and the apoptosis exhibited significant difference when compared with that of the model group (P<0.01).Conclusion: The extracts of Euonymus hederaceus have protective effects on vascular endothelial cells with oxidative damage induced by H2O2.

AIM: To construct E. coli expression plasmid of recombinant human NDPK-A with a 6?His tag, optimize the expression condition and identify the activity of the product. METHODS: nm23-H1 was subcloned from plasmid pBVNMH1 to pQE40 which contain 6?His purification tag. The expression condition was modulated in grades to get the optimal expression. We purified protein with the Ni+-NTA affinity chromatography column, identified the immunogenicity of the product with Western blot, and measured the kinases activity with HPLC. In addition, angiogenesis inhibition activity of rhNDPK was identified by CAM. RESULTS: The sequence of nm23-H1 subclone in pQE40 was exactly correct. The expression rate of rhNDPK-A was 49 6%. Purified rhNDPK-A specially recognized the antiserum of NDPK-A. It also inhibited angiogenesis. CONCLUSION: PQE-nm23H1 containing 6?His can express target protein at high level. This purification method is simple than other methods, and the product has the same activity as natural human NDPK-A.

Objective To explore the expression level of Tim-3,the marker of activated T_H 1 cells.in T lymphocytes in different sites from recipients with acute rejection.Methods The model of cervical heterotopic heart transplantation was established in mice Two groups were get up:the isograft group(C57BL/6→C57BL/6) and the allograft group (Balb/c→C57BL/6).Lymphocytes were isolated from peripheral blood,spleens,draining lymph nodes and grafts 3 or 6 days after transplantation.The expression of TIM-3 in CD4~+ and CD8~+ T subsets was detected by flow cytometry.Results There was no significant difference in Tim-3~+/CD4~+ and Tim-3~+/CD8~+ ratio in peripheral blood or spleens between two groups.As compared with the isograft group,the proportion of Tirn-3~+/CD4~+ cells was slightly elevated in draining lymph node(P<0.05),but the percentage of Tim-3~+/CD4~+ cells had no significant change between 3 days and 6 days in the allograft group(P>0.05).The expression of Tim-3 in CD4~+ and CD8~+ of graft infiltrating T cells was obviously increased in allograft group(P<0.01),and it was significantly (P<0.01) up-regulated on the 6th day as compared with that on the 3rd day.Conclusion The dynamic changes of Tim-3 expression in T lymphocytes in draining lymph node and graft were correlated with the progresston oi acute rejection in mice.

To monitor genetic variation of porcine reproductive and respiratory syndrome virus (PRRSV),RT-PCR was used to identify a sample suspected of PRRSV infection.A PRRSV named SC-GY strain was obtained,and its Nsp2,ORF5 and ORF3 genes were used for sequence alignment and phylogenetic tree construction.The results showed that SC-GY strain is highly pathogenic PRRSV American variant strains with Nsp2 gene discontinuous deletion of 30 amino acids,ORF3 gene aa17 a serine (S) insert.Comparing to VR2332,CH-1a,JXA1,HUN4,NADC30,HENAN-XINX and SC2012,the Nsp2,ORF5 and ORF3 of SC-GY shared 70.3％-97.9％,82.4％-97.6％ and 83.1％-98.2％ of nucleotide similarity,and 62.3％-96.3％,78.0％-95.7％ and 81.6％-96.5％ of deduced amino acid similarity;and compared to LV they shared only 18.9％,60.8％ and 63.7％ of nucleotide similarity,and 14.0％,54.9％ and 57.2％ of deduced amino acid similarity.The phylogenetic tree revealed that the SC-GY formed independent small branches although it belonged to the same subgroup as highly pathogenic PRRSV strains.The results showed that in high frequency live vaccine immunization of currently PRRSV,the gene of PRRSV epidemic strain is still in constant variation.Vaccination of live PRRSV vaccines should be reduced and surveillance of PRRSV strains should be enhanced.

Objective To analyse the status quo,hotspots and trends of researches on feeding intolerance in preterm infants from 2012 to 2016 in China by co-word analysis,in order to propose directions for future researches.Methods Related studies pub lished from November 2012 to November 2016 were obtained through searching in databases,including CNKI,Wanfang,VIP andCBM,with the keywords "preterm infant","low birth weight infant" and "feeding intolerance".Studies were selected in strict accordance with the inclusion and exclusion criteria.The co-occurrence matrix of high frequency keywords was established via Microsoft Office Excel2007,and the visual network was drawn by Ucinet6.0 and Netdraw.Results A total of 536 studies were included,and 35 high-frequency keywords were extracted.The subjects of these studies mainly were very low birth weight infants,and there were few researches on extremely low birth weight infants.The reasons and treatments for feeding intolerance,clinical efficacy,clinical manifestations,growth and development,enteral/parenteral nutrition and necrotizing enterocolitis were the focus of attention of clinicians.Conclusion There have been in-depth researches on clinical features,diagnosis standard,prevention and treatment measurement of feeding intolerance in China,further researches are needed to investigate the etiology,exact pathogenesis and biological indicators.
