Objective To investigate the significance of enzyme-linked immunosorbent assay (ELISA) in cross-match test of donor-recipient before organ transplantation. Methods HLA glycoproteins were prepared by solubilizing the lymphocytes of donor with a non-ionic detergent, and bond to the monoclonal antibody specific for HLA class Ⅰ or Ⅱ immobilized in the ELISA tray. ELISA with addition of recipients' serum and complement-dependent cytotoxicity (CDC) were compared. Results Cross-match test was performed by two methods for lymphocytes of 40 donors and sera of 72 recipients. All samples were successfully tested. The results of one pair of donor-recipient cross-match test by two methods were different. Conclusion ELISA for cross-match test is simple, convenient and time-saving, but more sensitive and specific than CDC.

Objective To investigate the antitumor mechanism of sodium arsenite on human gastric carcinoma cell line SGC-7901 in vitro. Methods MTT assay, light microscopy, electron microscopy, flow cytometry, and immunocytochemical staining were used to analyze the effect of sodium arsenite on biologic behavior of SGC-7901 cells. Results Sodium arsenite (2.50 ~ 40.00 ?mol/L) could inhibit the growth of gastric carcinoma cells, it depended on the duxation and concentration, and its 50% inhibitory concentration(IC50) was 8.69 ?mol/L after 72 hours' treatment. SGC-7901 cells were arrested significantly in G2/M phase treated with sodium arsenite for 48 and 72 hours. SGC-7901 cells presented typical morphologic feature of apoptosis and necrosis after exposure to sodium arsenite. Sodium arsenite up-regulated Caspase-3 protein expression in SGC-7901. Conclusion Sodium arsenite could obviously inhibit the proliferation of SGC-7901 cells, induce cell cycle arrest and apoptosis and necrosis of the cells. its mechanism is possibly associated with inhibition of elimination of ROS and the up-regulated expression of Caspase-3 protein.

Chinese Journal of Organ Transplantation 2003;0(05):-
Objective To explore the surgical reconstructive procedures of adult-to-adult living donor liver transplantation (LDLT).Methods Two patients with end-stage liver disease were successfully subjected to adult-to-adult LDLT using dual grafts in our division. One patient’s donors were left lobe and left lobe from his two old sisters, respectively. The other graft was right lobe from his 56 years-old mother and left lobe separated from a cadaveric organ donor. Results Both recipients and three donors display good graft function and normal triangular shape regeneration of their liver grafts after liver transplantation. There was neither mortality nor serious complications in the donors. Conclusion The critical issue of LDLT is donor morbidity. Dual grafts from two living donors can help to alleviate the problem of small-for-size grafts and yet secure the safety of the donor. But the complicated surgical techniques give a great challenge for liver transplant surgeons.
