Objective To establish the enzyme‐linked immuno sorbent assay(ELISA) method for a large‐scale screening of IgA deficiency in blood donors .Methods The indirect ELISA was adopted .The goat anti‐human IgA antibody was coated in microwell plates and labeled by horse radish peroxidase (HRP) as enzyme‐labelled secondary antibody .Results The sensitivity of established ELISA detection method was 0 .1 μg/mL .The intraassay coefficients of variation (CV) for IgA concentrations of 0 .1 ,100 μg/mL were 1 .74% to 3 .49% .The median interassay CV was 3 .48% (range:1 .83% -6 .96% ) .The assay process was 80 min . Conclusion TheELISA detection method is successfully established with high sensitivity ,strong specificity ,timesaving and easy operating and can be used for a large scale screening of Ig A deficiency and establishment of blood donors bank of Ig A deficiency .

objective To investigate the clinicopathological features of mini-cancer of the stomach. Method In this study,out of 296 early gastric cancer cases there were 34 cases of early gastric cancer in which tumor diameter was≤10 mm,among those there were 5 cases with tumor size≤2 mm and 29 cases of the size 2～10 mm. Result Mini-cancer accunted for 2% of all early gastric cancers in this series:All these mini-cancers were of intramucosal cancer(100%),while that took up to 45%in control group in which tumors were between≥2 mm and≤10 mm:Tumors were high or moderately differentiated pathologically in 100%of mini-cancers and 55%in control group.None of mini-cancer patients had lymph node metastasis,however,1 of 29 patients in control group had lymph node metastasis.Both groups had no blood vessel and lymphatic vessel invasion:The differentiation concordance rate between superficial lesions and invasive fronts in mini-cancer was 100%,higher than 86%in control group. Conclusion Gastric mini-cancer is usually of high differentiation,low tumor invasion and low rate of lympy node metastasis than control group.Endoscopic therapy is applicable for most gastric mini-cancers.

Objective To ascertain the role of NF?B and relationship among TGF?1,caspase-3 and apoptosis in CsA-induced nephrotoxicity.Methods Sixty-four wistar male rats were randomly divided into 4 groups:Vehicle,PDTC,CsA and CsA + PDTC group.The animals were daily subcutaneously injected either with CsA(15 mg/kg)or vehicle/PDTC(100 mg/kg).Four weeks later,BUN,serum creatinine,NAG and urine routine were examined.The kidneys were processed for light microscopy,immunohistochemistry and TENUL.Results A rat model of CsA-induced nephrotoxicity was established.CsA group showed significant increase in BUN,SCr and urine NAG compared with the controls,whereas CsA + PDTC group did not.Apoptosis is observered by TUNEL and the expressions of NF-?B,TGF-?1,caspase-3 increased significantly in CsA group compared with conrtol group.Whereas,both the apoptosis and the expressions of these proteins significantly decreased in rats treated with PDTC.Conclusion Apoptosis is induced in the CsA-induced chronic nephrotoxicity.Inhibition of NF-?B activation blockaded apoptosis and ameliorated CsA-induced chronic nephrotoxicity.The results suggest that NF-?B plays important roles in the progression of chronic CsA nephrotoxicity.

ObjectiveTo investigate the effects of edaravone pretreatment on pulmonary inflammatory response during selective lobar blockade in patients undergoing thoracotomy,MethodsSixty ASA Ⅰ or Ⅱ patients of both sexes,aged 40-64 yr,weighing 50-80 kg,undergoing esophagectomy were randomly divided into 4 groups (n =15 each):the one-lung ventilation group (group OLV),the edaravone pretreatment + one-lung ventilation group (group E + OLV),the Univent tube selective lobar blockade group (group U) and the edaravone pretreatment + Univent tube selective lobar blockade group (group E + U).Padents in groups OLV and U were intubated with the double lumen endobronchial tube and Univent tube,respectively.Edaravone at a dose of 1 mg/kg was infused into patients at 5 min before one-lung ventilation in group E + OLV and before selective lobar blockade in group E + U,respectively.The patients in groups OLV and U received the equal volume of normal saline.The blood samples (5 ml ) were then drawn from the radial artery after anesthesia induction (T0),at 60 min after onelung ventilation (T1),at the end of operation (T2 ) and 120 min after operation (T3) for detecting the plasma concentrations of TNF-α,IL-6 and IL- 10 by enzyme-linked immunosorbent aesay.ResultsCompared with the group OLV and U,the plasma concentrations of TNF-α and IL-6 decreased at T2 and T3 in group E + OLV and group E + U (P ＜ 0.05).CondusionEdaravone pretreatment can reduce pulmonary inflammatory response during selective lobar blockade in patients undergoing thoracotomy.

N-(2-hydroxypropyl) methacrylamide (HPMA) copolymer-Mitoxantrone conjugates was successfully synthesized and characterized as an anti-cancer drug for solid tumors. The biodistribution and pharmacokinetics of the conjugates were examined in mice bearing Ehrlich solid tumor. The biodistribution of the HPMA copolymer-bound Mitoxantrone in tumor-bearing mice was significantly different from that of the free drug. It appeared that the circulation life times of the conjugates were three times more than those of the drugs in the free form. The concentrations of HPMA copolymer-bound Mitoxantrone in tumor reached maximum levels 24 h post injection. AUC is three times higher than that of free drug. The P-Mitoxantrone level in heart was five times lower than that of free drug. This reduces the possibility of toxicity to heart. In particular, HPMA copolymer-bound Mitoxantrone accumulated at higher levels in tumor tissues. This may be explained by "Enhanced Permeability and Retention effect" (EPR effect). These results show the possibility of targeting anticancer drug-Mitoxantrone with secondary amino residue to the tumor tissue by HPMA copolymer as carrier.

Objective To evaluate the change of left ventricular diastolic function and investigate the relation between left ventricular diastolic function and disease activity in rheumatoid arthritis (RA) without clinical manifestations of heart diseases. Methods Seventy consecutive active RA in-patients without clinical manifestations of heart disease were enrolled, while the control group was recruited from outpatient health physical check-up center and consisted of 60 age- and sex-matched healthy subjects. Cardiac related parame-ters were determined by echocardiography and the correlation between left ventricular diastolic function and the disease activity indexes were evaluated. Chi-square test, t test, Pearson or Spearman′s correlation test and Stepwise backward linear regression analysis were used for statistical analysis. Results RA patients had lower mitral inflow E/A ratio (1.2±0.4, 1.5±0.4, P<0.01), higher E/Em ratio (9.6±3.7, 7.8±2.0, P<0.01), longer isovolumetric relaxation time(IVRT)[(64±16) ms,(58±16) ms, P<0.05] than control group. Whilst, RA patients had higher pulmonary venous inflow A wave velocity-time integral (ArVTI) and A wave duration (DAr)[3.2±0.7,(2.8±0.6) cm; 117±11,(102±9) ms, P<0.05]. Moreover, the E/Em was positively corre-lated with C-reactive protein(CRP)(r=0.581, P<0.01), DAS28(r=0.456, P<0.01). Anti-CCP level was also associated with Em and early diastolic pulmonary venous inflow peak velocity(PVD)(r=-0.359, P<0.05;r=-0.305, P<0.05). In addition, multivariate analysis also revealed that there was linear regression relation-ship between E/Em and CRP, DAS28(t=3.266, P=0.002; t=2.949, P=0.005). Conclusion The study has revealed that left ventricular diastolic function is impaired in RA patients and the left ventricular diastolic function parameters is associated with the disease activity indexes. These results suggest that the decline of left ventricular diastolic function is associated with the inflammation activity in RA patients without clinical manifestations of heart disease.

Objective To reveal the influence of environmental factors on the active components in Scutellaria baicalensis through analyzing the data sampled from the herbal plants along the altitudinal gradient in mountainous region of Western Beijing.Methods In total 85 individuals of S.baicalensis herbs were collected and the surface soils under each of the plant were also sampled from eight different habitats,with an interval of about 100—150 m vertically ranging from 800 to 1 800 m in August through September,2007.The contents of active components in the herbal samples,namely baicalin,wogonoside,baicalein,wogonin,and oroxylin A were determined by HPLC.The organic matter and the total nitrogen contents in the soil samples were analyzed through standard measurement process.The various habitat conditions represented by temperature were estimated through a vertical lapse rate of-0.56 ℃/100 m,and those by precipitation and solar radiation calculated according to the isoline maps.Results The signi-ficant positive correlations were observed only between the contents of baicalin,wogonoside and the elevation.Both those two contents showed the significant negative correlations with the annual mean temperature and precipitation,while showed positive with annual mean solar radiation.No coorelation with any statistical significance was observed between the contents of active components in S.baicalensis and the soil parameters in this study.Conclusion It seems that the plants of S.baicalensis growing in a habitat with annual mean temperature within a range of 2—6 ℃ could concentrate relatively rich contents of active components.Within the natural distribution areas of S.baicalensis,the habitats with an annual tem-perature averaged at 3—4 ℃ tend to be the most fitting sites for growing best quality S.baicalensis rich in active components,and those sites are almost coincident with the environment for trueborn production area of traditional S.baicalensis herbs.

Objective To understand the infection situation of human T lymphocyte virus(HTLV) among blood donors in Zhongshan area.Methods Blood samples from 40 874 blood donors in Zhongshan from March to December 2016 were screened for HTLV antibody by using,enzyme linked immunosorbent assay(ELISA).The positive samples were reexamined two times,and specimens with positive results of reexamination were detected by using immunohistochemical method(CLIA).Then the positive samples were confirmed by Western blot(WB),and confirmed positive samples were judged as infection.Results Of all 40 874 cases of voluntary blood donors,21 cases were positive with HTLV antibody detected by ELISA,the positive rate of ELISA was 0.05%.Five cases were positive detected by CLIA method.One case was confirmed by WB,and the infection rate was 0.002 4%.Conclusion In order to ensure the safety of blood transfusion and reduce blood transfusion infection of HTLV,it might be necessary to perform HTLV screening in first-time blood donors in Zhongshan area.

Objective To investigate the Huatan Huoxue Jiangqi prescription in treating AECOPD clinical curative effect. Methods 123 patients with AECOPD were randomly divided into the treatment group and the control group. The two groups were treated with western medicine, and the treatment group added with Chinese herbal medicine treatment with Huatan Huoxue Jiangqi prescription. The syndrome effect, clinical symptom score, CAT score, mMRC classification, lung function, the serum FIB and CRP levels were obtained to evaluate the efficacy of the two groups before and after treatment. Results The syndrome effect of the treatment group was significantly better than that of the control group. Compared with the control group, there was significant difference between traditional Chinese medicine syndrome score, CAT score (P < 0.05); Lung function (including FEV1, FEV1%, FEV1/FVC) was significantly improved in the treatment group after treatment (P < 0.05). The serum FIB and CRP levels decreased significantly in the treatment group compared with the control group (P < 0.05). Conclusion Huatan Huoxue Jiangqi prescription has reliable effets on treating AECOPD.
