Objective:To explore the correlation of stress, coping style and the concentration of dehydroepiandrosterone sulfate in umbilical blood between preterm birth and normal birth.Methods:46 with preterm birth and 42 normal birth controls were assessed with Life Event Scale (LES) and Defense Style Questionnaire (DSQ) at 28th-week of pregnancy. The concentrations of DHEA-s in umbilical blood were determined by ELISA for the two groups. Nineteen cases with incomplete paraplegia obtained an average improvement of 1.26 grades by Frankel neurological classification. The major complications in this group included one case with temporary deterioration of neurological function. Another case with pulmonary atelectasis on the operation side, but cured with appropriate treatment. Conclusion One stage surgical treatment for multiple-level tuberculosis of the upper thoracic spine by combined anterior and posterior spine fusion with posterior instrumentation is feasible and effective without major complications. Early surgical intervention should be emphasized for the treatment of progressive tuberculosis of the upper thoracic spine.
3.Study on expression of stem cell marker SALL4 gene in cervical cancer

Objective To research the expression and clinical significance of SALL 4 gene in cervical cancer .Methods The ex-pression of SALL4 in 56 samples of cervical cancer and 35 samples of normal cervical tissues was detected by immunohistochemistry and RT-PCR ,and its relationship with the clinicopathological characteristics of cervical cancer was analyzed .Results The expres-sion of SALL4 mRNA was 2 .56 ± 0 .22 in cervical cancer tissues ,which was significantly higher than 0 .38 ± 0 .03 in the normal cer-vical tissues .the difference between them had statistical significance(t=58 .1 ,P<0 .01);the positive expression rate of SALL4 pro-tein was 80 .4% (45/56) in cervical cancer ,which was significantly higher than 11 .4% (4/35) in the normal cervical tissues (χ2 =41 .177 ,P<0 .01) .The positive expression of SALL4 in the cervical cancer tissues was correlated with the differentiation status of tumor ,which in the middle and high differentiation groups was lower than that in the low differentiation group (χ2 =4 .226 ,P=0 .039) ,but had no correlation with age ,International Federation of Gynecology and Obstetrics (FIGO) stage ,tumor size ,pathologi-cal type and lymph node metastasis(P>0 .05) .Conclusion SALL4 is highly expressed in the cervical cancer tissues and correlated with the tumor differentiation ,which might play an important role in the occurrence and development of cervical cancer .

5.Expression of Nucleostemin gene in tumor cells derived from human embryomic bone marrow mesenchymal stem cells

Objective To study the tumorigenesis mechanism in bone marrow mesenchyme stem cells (MSC). Methods The bone marrow MSC could be induced into turnout (F6 cells) in vitro. The difference between gene expression of F6 cells and MSC was distinguished by fluorescent differential display (FDD). Verification of the result was detected by Real time RT-PCR and Western blotting, and immunocytochemistry. Results FDD analysis confirmed that Nucleostemin (NS) was positively up-regulated in F6 cells compared with MSC. Similar results were obtained by PCR and Western blotting. The NS gene expression levels in MSC, F6, 176-4, F6-6 and F6-7 were significantly different(F =160, P <0.05). The NS gene expression level in F6 (0.0372±0.0019) was 18 folds higher than those of MSC(0.0021±0.0002,P <0.05). Expression levels in F6-4, F6-6 and F6-7 tissue were 0.0504±0.0083, 0.0995±0.0026 and 0.0614±0.0036, and were significantly higher than that in MSC(P <0.05). The expression of NS increased significantly with the accreting volume of turnour, and high-level protein expression of NS was confirmed by Western blotting and immunocytochemistry. Conclusion The expression level of NS might be one of the factors playing important roles during turnour genesis, especially in MSC mutation.
6.Application of SWO technique in IMRT plan of post-operative cervical cancer

Objective To investigate the impact of segment weight optimization(SWO) technique on the intensity modulated radiation therapy(IMRT) plan for post-operative cervical cancer regarding the number of segments,monitor units (MU),the target homogeneity index (HI),conformal index (CI) and dose distribution of target volume and normal tissues.Methods Ten patients with stage Ⅰ and Ⅱ cervical cancer after radical resection were randomly selected for this study.The initial IMRT treatment plans were generated using ELEKTA XIO 4.62 system based on the step and shoot method (S-IMRT plan).With the same directions of fields and optimization parameters,the SWO tool was introduced to optimize the IMRT plans further (SWO-IMRT plan).Then the number of segments and MU were compared between the S-IMRT plan and SWO-IMRT plan.By using the dose-volume histogram (DVH),the target homogeneity index (HI),conformal index (CI) and dose distribution in the volumes of target and normal tissues were also analyzed.Results Compared with S-IMRT plan,the average number of segments in SWO-IMRT plan was decreased from 96 ± 4 to 87 ± 4 (t =10.049,P < 0.05),and MU was increased from(638.79±35.02)) cGy to (672.03 ±39.07) cGy (t =3.5952,P <0.05).The maximum and mean doses of the planning target volume (PTV) decreased (t =2.262,2.323,P < 0.05).A reduction of the maximum dose in the spinal cord was also observed [from (3856.00 ± 112.14) cGy to (3750.00 ± 141.38) cGy,t =3.976,P < 0.05].The values of V30,V40,V50 in bladders,rectal V30 and L-femoral V50 were reduced in the SWO-IMRTplan (t=4.223,5.801,7.534,2.451,2.269,3.976,P <0.05).However,there was no significant difference in target homogeneity index(HI),conformal index (CI),rectal V40,V50,L-femoral V30,V40,V50,R-femoral V40 and V50.Conclusions The application of SWO technique in the IMRT planning for cervical post-operative cancer could reduce the total number of segments,doses in the spinal cord and bladder,but increase the total number of MU.As a result,the spinal cord and bladder toxicity can be reduced which enables an opportunity for dose escalation of gross tumor volume (GTV).SWO technique provides clinicians with an optional optimization solution in IMRT plan for post-operative cervical cancer patients.
7.Comparison of spectral CT imaging and 64 slice CT in assessment of carotid artery atherosclerotic plaque

Objective To compare the efficacy of spectral CT imaging and 64 slice CT in assessment of carotid artery atherosclerotic plaque.Methods Thirty-eight patients with carotid plaque were detected retrospectively in this study (plaque,n=46) by using spectral CT.The composition of atherosclerotic plaque was measured by iodine-based images,lipid-based images with spectral CT imaging.Intraplaque hemorrhae,lipid components were also evaluated on spectral CT imaging and compared with pathology and specimen.Statistical comparison was performed with the Kappa value,independent-sample t test and exact test.Results The degree of carotid artery stenosis was (63.3 ±3.1)% on spectral CT imaging whereas (61.6 ± 3.8) % on 64 slice CT.Two imaging modalities were in good consistency in evaluation of the degree of stenosis (Kappa=0.993,P<0.01).There was no statistical difference in detecting fibrous cap rupture on spectral CT imaging and 64 slice CT (12 plaques vs 11 plaques,respectively,P>0.05).Furthermore,spectral CT imaging showed 11 plaques within intraplaque hemorrhage,however,no case was found on 64 slice CT (P<0.05).The plaque iodine concentration were (6.365 ± 1.937) and (1.573 ±0.776) mg/cm3 with or without internal hemorrhage(t=16.39,P<0.05).Spectral CT imaging showed 9 plaques within intraplaque liquid whereas only 2 plaques showed intraplaque liquid on 64 slice CT (P<0.05).Twenty-eight plaques within calcification in the plaque were found on spectrl CT imaging and 27 on 64 slice CT(P>0.05).Conclusion Spectral CT imaging is a reliable tool in detecting intraplaque hemor-rhage and liquid component.
8.Ultrasonic differential diagnosis of patients with liver alveolar hydatid disease and hepatic malignant tumor

Objective To investigate the value of ultrasound contrast in differential diagnosis of liver alveolar hydatid disease and hepatic malignant tumor.Method Totally 36 cases of liver alveolar hydatid disease and 31 cases of hepatic malignant tumor were retrospectively analyzed,who visited the Ultrasound Department of Qinghai Red Cross Hospital from March 2010 to September 2013,the performance characteristics of gray-scale ultrasound and contrast-enhanced ultrasound (CEUS) on hepatic malignant tumor and liver alveolar hydatid disease were compared.Results Gray-scale sonography of 36 liver alveolar hydatid diseases showed clear boundary,heterogeneous hypoechoic nodules,and the maximum area was (5.06 ± 2.46) cm2 in 49 lesions;gray-scale sonography of 31 hepatic malignant tumors showed unclear boundary,hypoechoic nodules,and the maximum area was (7.29 ± 5.83) cm2.CEUS of liver alveolar hydatid disease showed non-enhancement in three phases,and rim enhancement was seen synchronized with the liver parenchyma.CEUS of hepatic malignant tumor showed hyperenhancement in arterial phase,equal-enhancement or hypo-enhancement in portal phase and delayed phase,and abnormal perfusion areas were clear.Conclusion Liver alveolar hydatid disease is different in the performance of CEUS from hepatic malignant tumor,CEUS has a great value in the differential diagnosis of liver alveolar hydatid disease and hepatic malignant tumor.
9.Expression of survivin and its splice variants in gastric cancer.

Survivin variants specific real time quantitative RT-PCR was developed to analyze their expression in 53 paired cancer and para-cancerous tissues, and the expression of the wild-type survivin protein was detected by immunohistochemistry. The results showed that survivin mRNA and protein were expressed in gastric cancer and para-cancerous tissues. The survivin-2B was dominantly expressed in para-cancerous tissues, whereas the survivin-DeltaEx3 was more frequently detected in cancer tissues. The positive rate of survivin-2a was 100% in both cancer and para-cancerous tissues, but its relative transcript expression level was not significantly increased in cancer tissues in comparison with para-cancerous tissues. The correlation analysis revealed that the expression of survivin-2a mRNA was significantly associated with that of total survivin (r (s)=0.4178, P=0.0018), whereas inversely to that of survivin-DeltaEX3 (r (s)=-0.4506, P=0.0007). It was suggested that survivin-2a may act as an antagonist of survivin-DeltaEX3. The balance between antiapoptotic survivin iso-forms and nonantiapoptotic ones may play an important role in tumorigenesis and tumor progression. Promising value is hinted to analyze survivin and its variants in tumor early diagnosis and distinguishing malignant tumors from benign ones.

10.Effects on the expression of lipopolysaccharide-induced inflammatory cytokines mediated by bovine bactericidal/permeability-increasing protein.

Bactericidal/permeability-increasing protein (BPI) can bind to and specifically neutralize lipopolysaccharide (LPS) from the outer membrane of Gram-negative bacteria. In order to evaluate potent LPS-neutralizing activity of bovine BPI, the full-length coding sequence (1 449bp) or 714 bp N-terminal coding sequence (BPI714) of bovine BPI was transfected into mHEK293 cells and the expression of LPS-induced inflammatory cytokines was studied. First, we constructed the lentiviral expression vectors and generated mHEK293 cells stably expressing recombinant bovine BPI or BPI714. Then, we detected the expression of IL-8, IL-1β, TNF-α, NF-κB-1 and NF-κB-2 genes by real-time PCR at 0, 1, 3, 6, 12, 24, 36 and 48 h post of LPS induction in cells with or without recombinant bovine BPI or BPI714 ectopic expression, respectively. In response to LPS, the robust abundance of inflammatory cytokines including IL-8, IL-1β, TNF-α and NF-κB-2 was observed in wild type mHEK293 cells at eachtime point. On the contrary, mRNA abundance of IL-8, TNF-α and NF-κB-2 in transfected mHEK293 cells showed no significant changes at each indicated time point. Our results demonstrated that recombinant bovine full length BPI or BPI714 down-regulated the expression of inflammatory cytokines and revealed that either of bovine BPI or BPI714 was able to inhibit the immune respond stimulated by LPS. This study provides evidence for further investigating the mechanisms and application of BPI/LPS-neutralizing activity and also documents a reliable approach for analysis of the efficacy of antibacterial proteins.
