1.Detection of Potential Microcystin-Producing Cyanobacteria in Waters With a mcyD Molecular Biomarker

Objective To develop a new method to screen the potential microcystin-producing cyanobacteria in natural waters. Methods Cyanobacteria were cultured and waters samples were analyzed by polymerase chain reaction with special primers for gene of the operon mcyD which encodes a microcystin synthetase combining conserved gene 16S rRNA of alga, subsequently, whose microcystins were detected by ELISA. Results There was a unique amplified product of approximately 870 bp in microcystin-producing blue-green alga and water samples, but no target band in non-toxic strains and samples. Conclusion It is feasible to use the molecular biological method used mcyD as the microcystin molecular biomarker to screen microcystin-producing cyanobacteria in natural water.

The article is attempted to introduce PBL teaching in clinical biochemistry. The PBL teaching has improved some aspects of students' abilities in the self-study, accessing to information, solving the problem, teamwork and developing thinking. But in the course of PBL teaching, there are three problems: shortage of qualified teachers, teaching system and students' inability. This article puts forward the countermeasures to resolve these problems.
3.Studies on B cell differentiation from cord blood CD34~+CD19~- hematopoietic stem/progenitor cells in vitro

Objective:To investigate the condition of B cell differentiation from cord blood CD34+CD19-hematopoietic stem/progenitor cells in vitro.Methods:CD34+CD19-cells from cord blood were isolated and purified by using immunomagnetic beads separation system.CD34+CD19-cells supported by murine S-17 stromal cells were stimulated in co-culture with T3 and cytokines.Differentiated B cells were analyzed by flow cytometry.Results:The amplification of the B cells derived from CD34+CD19-hematopoietic stem/progenitor cells in co-culture with T3 and IL-7 reached 198-fold of increase,most of the induced cells expressed CD10 and CD19.Conclusion:In the experimental conditions selected,co-culture of CD34+CD19-cells with T3,IL-7 and murine S-17 stromal cells could stimulate differentiate toward to B cells in vitro.
4.Application of case teaching method in specialized courses of laboratory medicine

Case teaching method was used to undergraduates’specialized courses of clinical laboratory medicine such as the clinical laboratory hemotology and basic clinical laboratory medicine. This teaching method achieved satisfactory effect in clinical ability culture and obtained the positive opinion of students.
5.Expression of survivin and its splice variants in gastric cancer.

Survivin variants specific real time quantitative RT-PCR was developed to analyze their expression in 53 paired cancer and para-cancerous tissues, and the expression of the wild-type survivin protein was detected by immunohistochemistry. The results showed that survivin mRNA and protein were expressed in gastric cancer and para-cancerous tissues. The survivin-2B was dominantly expressed in para-cancerous tissues, whereas the survivin-DeltaEx3 was more frequently detected in cancer tissues. The positive rate of survivin-2a was 100% in both cancer and para-cancerous tissues, but its relative transcript expression level was not significantly increased in cancer tissues in comparison with para-cancerous tissues. The correlation analysis revealed that the expression of survivin-2a mRNA was significantly associated with that of total survivin (r (s)=0.4178, P=0.0018), whereas inversely to that of survivin-DeltaEX3 (r (s)=-0.4506, P=0.0007). It was suggested that survivin-2a may act as an antagonist of survivin-DeltaEX3. The balance between antiapoptotic survivin iso-forms and nonantiapoptotic ones may play an important role in tumorigenesis and tumor progression. Promising value is hinted to analyze survivin and its variants in tumor early diagnosis and distinguishing malignant tumors from benign ones.

The clinical ability of employees in laboratory medicine refers to their ability to give useful information and helps for clinical diagnosis and treatment.In laboratory medicine education,the cultivation of the undergraduates'clinical ability should be strengthened.Case teaching and clinical signs analysis is practical to the cultivation of clinical ability.In addition,we should carry out the reform of the teaching materials in order to adapt to the requirements of the clinical ability training. Improving the clinical ability should be the students' a lifelong career planning.

8.Treatment of primary rhegmatogenous retinaldetachment with cryopexy in combination with intravitravitreal injection of expending gas

Objective To demonstrate the visual results and complications of an cryopexy in combination with intravitreal injection of expending gas in the therapy of primary rhegmatogenous retinaldetachment (RRD). Methods Thirty-two cases (32 eyes) were retrospectively reviewed in this study. The RRD diagnosis was confirmed by best corrected visual acuity,slit-lamp microscope,indirected ophthalmoscope and Goldman three-mirror contact lens. All patients had undergone cryopexy with intravitreal gas injection and assisted by correct body position. Patients were followed for 6 to 24 months. Post-operative BCVA,final anatomical outcome, complications and failed cases were analyzed. Results The reattachment rate of cryopexy with intravitreal gas injection was 81%(26/32 eyes). Four eyes required additional scleral buckling. Two eyes needed additional vitrectomy with intravitreal injection of expending gas (SF6).Final retinal reattachment was achieved in all 32 subjects (100%). Postoperative BCVA was significantly improved (P < 0.01). Conclusion Cryopexy with intravitreal gas injection is a simple,less trauma, lower cost and effective surgery for primary rhegmatogenous retinaldetachment.

AIM: To explore the mechanism of Nao-re-qing oral liquid (NRQ) decreasing endotoxin (ET)-induced fever in rabbits. METHODS: (1) The ET-induced fever model was established in rabbits. Febrile response of rabbits was observed. 10. Research progress of the clinical applications of tumor liquid biopsy

In recent years, researches about liquid biopsies which contain circulating tumor cells (CTC), circulating tumor DNA (ctDNA) and exosomes in the body fluids of patients with solid tumor have caused great concern. Compared with traditional biopsies, liquid biopsies have many advantages as they are real time, noninvasive and comprehensive. With the progress in the detecting techniques of CTC, ctDNA and exosomes, liquid biopsies can be applied to the early screening, diagnosis, treatment, prognostic evaluation of tumor. This review focuses on the recent advances in detection methods and clinical applications of CTC, ctDNA and exosomes.(
