1.The Changes of Oxygen Free Radical of the Rat Liver Sub-cellular Organelle at the Early Stage of Ischemia-reperfusion Injury

Objective To study the change principles of the oxygen free radical on the sub-cellular level in the process of ischemia-reperfusion injury. Methods The activity of superoxide dismutase(SOD) and the content of malondialdehyde(MDA) in the mitochondria and cytoplasm of rat liver with local ischemia-reperfusion were assayed. The histological changes of the liver were observed at the same time. Results The activity of SOD and the content of MDA kept rising and the histological changes of the liver cells kept aggravating in the process of ischemia-reperfusion. Conclusion The ischemia-reperfusion injury of rat liver occurred at the beginning of reperfusion and mitochondria was damaged first, however the injury action could be delayed by self defense.
2.Association of ER22/23EK polymorphism in glucocorticoid receptor gene with natural longevity in Xinjiang Uygur nationality and comparison with Han nationality

Objective To investigate association of the ER22/23EK polymorphism of the glucocorticoid receptor gene(GR) with natural longevity in the XinJiang Uygur nationality people and the race difference.Methods One hundred and nintyone healthy individuals over 90 years old from Uighur people were recruited as the longevity group at the same time,82 Han nationality people aged between 65~70 who immigrated in Xinjiang Hetian for more than 30 years were randomly selected and investigated.Genotyping was performed by PCR-SSP,PCR-RELP and PCR-sequencing.ResultsThe frequencies of ER22/23EK alleles and genotypes showed no significant difference between the longevity group and the controls in Uygur,but the carriers of ER22/23EK of GR gene in Xinjiang Han old folkswere significantly more than those in Uygurs,the frequencies of WM, MM genotypes and M allele were significantly higher in Han nationality,while the frequencies of WW genotype and W allele were significant lower.Conclusion There is possibly no association between ER22/23EK polymorphism and XinJiang Uygur natural longevity,but there are significant differences between the two ethnic groups.
3.Comparison of the radiographic results of total knee arthroplasty with or without using electromagnetic navigation

Objective To compare the lower limb alignment and prosthesis position after total knee arthroplasty (TKA) with or without using electromagnetic navigation.Methods Sixty-four patients (100 knees) underwent TKA under electromagnetic navigation,while 62 patients (100 knees) underwent conventional TKA.Three months after operation,the mechanical axial line angle and prosthesis position (angels α,β,γ) were measured via the full-length radiograph of both lower limbs and anteriorposterior and lateral Xrays of the knee.Results The average mechanical axial line angle and angle α were 1.20°±1.92°and 89.33°±1.64° in navigation group,respectively,and 2.31°±2.25° and 88.68°±2.57° in conventional group.And the differences were significant with regard to the above two indexes between two groups.The average angle β and angle γ were 89.64°±1.47° and 90.86°±2.37° in navigation group,respectively,and 89.26°±2.05° and 90.59°±3.44° in conventional group.However,the differences were not significant with regard to the above two indexes between two groups.After operation,mechanical axial line angle error was within ±3°in 86% of patients in navigation group,while in 79% of patients in conventional group; there was no significant difference between them.The angle α error was within 90°±3° in 92% of patients in navigation group,while in 77% of patients in conventional group; there was significant difference between them.However,there were no significant differences in percentage of patients whose errors of angle β and angle γ were within 90°±3° between two groups.Conclusion With using electromagnetic navigation in TKA,more precise prosthesis position and the mechanical axis can be achieved compared with the conventional technique.And its advantages mainly contribute to the coronal plane of the femoral side rather than the tibial side.
4.Separation of Indigo Naturalis by foam flotation technique in a continuous mode

Objective To study the process of foam flotation in separation of Indigo Naturalis in a continuous mode and to optimize the operational conditions.Methods Taking content and rate of recovery rate of indigo as index to investigate the single factors,such as the height of collecting region,the height of froth layer,flushing water rate,delivery rate,air flow rate,and aerating velocity,to study the effect of continuous foam floatation on Indigo Naturalis,and to optimize the process conditions finally.Results The flotation performance is good when the height of collecting region is 1.5 m,the height of froth layer is 30 cm,the delivery rate is 0.1 cm/s,the flushing water rate is 0.01 cm/s,the aerating velocity is 1.5 cm/s.The recovery rate of indigo is more than 75% and indgo content is over 5.0%.Conclusion Foam flotation technique is stable and can be used to the separation of Indigo Naturalis in a continuous mode.And this study is the foundation of semi-works production of Indigo Naturalis.
5.Comparison of electromagnetically navigated mechanical axis and component position with radiographic measurements in total knee arthroplasty

Objective To compare the discrepancy and consistency in mechanical axis and component position measured by electromagnetic navigation and radiograph in total knee arthroplasty (TKA)to assess whether the navigation system can be used as a substitute for radiograph.Methods A perspective study was performed on 40 cases (61 knees) undergone primary TKA under electromagnetic navigation from July 2006 to December 2006.There were 4 males and 36 females,at a mean age of (66.9 ±8.1) years (range,58-79 years).Mechanical axis angle,distal femoral and proximal tibial cut slope in coronal view (angles cα,β) were recorded both pre-and post-operatively with an intraoperative navigation system and compared against the mechanical axis angle,coronal femoral and tibial slope (angles α,β)measured via full-length radiograph of the lower limb preoperatively and at postoperative 3 months.Consistency in measurement of the same parameters with the two methods was assessed using intraclass coefficiency correlation (ICC).Results Mechanical axis determined by navigation and radiograph showed a mean valgus angle of 9.60° and 9.99° preoperatively and of 1.23° and 1.64° postoperatively,but the two pair parameters revealed no significant differences in the non-parametric test.Mean angle α determined by navigation and radiograph was 89.98° and 88.96° respectively (P < 0.05),and mean angleβ was 90.21 ° and 89.59°respectively (P < 0.05).With deviation value ≤3°,ICC for pre-and post-operative mechanical axis angles,angle α and angleβ was 0.887,0.754,0.632,0.640 respectively.Conclusions Within the acceptable range of deviation,intraoperative navigation data can reflect the pre-and post-operative mechanical axis and prosthesis position evaluated by radiograph.However,the advantages over the consistency of the two measurement methods rest with the evaluation of pre-and post-operative mechanical
6.Research on flow characteristics in a non-blade centrifugal blood pump based on CFD technology.

The problem of thrombus and hemolysis in blood pump has always been an important topic to study in the development of the blood pump. Numbers of research results show that it is the complicated flow and the high shear stress of the mechanical movement that result in the thrombus and hemolysis. In this study, with the cooperation of Shanghai Children's Medical Center, we have used computational fluid dynamics (CFD) commercial software FLUENT to compute and analyze the flow characteristics in a non-blade centrifugal blood pump. The results figure out that this pump has a reasonable flow distribution and the shear stress distribution is under the critical broken state of red blood cell; meanwhile, there is less thrombus and hemolysis in this pump. So it is in the foreground for clinical use.

7.Vascular and biliary complications after liver transplantation: interventional treatment.

OBJECTIVETo evaluate the value of angiography and cholangiography on the diagnosis and interventional treatment of vascular and biliary complications after liver transplantation.
METHODSSixteen of 46 patients (15 men and 1 woman, 17 - 60 years old) after orthotopic liver transplantation received angiography due to abnormal ultrasonography or edema of lower limbs, or cholangiography due to progressing jaundice. Percutaneous transluminal angioplasty or drainage was performed in some patients.
RESULTSFifteen patients experienced vascular complications and 4 patients had biliary complications. Three of them appeared to have both vascular and biliary complications. Hepatic artery complications were the most common complications (9/16), including hepatic artery thrombosis or stenosis (6/9), bleeding (2/9) and hepatic artery-dissecting aneurysm (1/9). One patient with hepatic artery thrombosis received transcatheter thrombolysis and two patients with bleeding received coil embolization. Inferior vena cava and portal vein stenosis were observed in 6 and 2 patients, respectively. After balloon angioplasty or stent placement, clinical symptoms were alleviated. Biliary complications, including biliary stricture and anastomotic bile leak, occurred in 4 patients. Jaundice decreased after percutaneous transhepatic cholangiography and drainage.
CONCLUSIONSBesides diagnosis, interventional methods include mini-invasive treatment for patients with vascular and biliary complications after liver transplantation. Balloon angiography and stent placement for venous stenosis are useful procedure for the treatment of these problems.

8.The fingerprint and durability study on various regions of Polygonum cuspidatum

Objective To study the fingerprint of Polygonum cuspidatum from various regions by HPLC and provide technical support for the identification.Methods HPLC conditions :the chromatographic column was Agilent ZORBAX SB-C18 (4.6 mm × 250 mm,5 μm);the mobile phase was 0.1% formic acid in water and acetonitrile (A-B);gradient elution ;the de-tection wavelength was 306 nm ;the flow rate was 1 ml/min;the column temperature was 30 ℃ ;injection volume was 20 μl. Results The HPLC gradient conditions could effectively separate polydatin,resveratrol,emodin and emodin ether,etc.Con-clusion This method is stable,effective and durable,which is instructive to the herbs data identification of Polygonum cuspi-datum.
9.Clinical observation on therapeutic effect of the pressing plantar reflex area with wooden needle for treatment of patients with insomnia.

OBJECTIVETo investigate the therapeutic effect of wooden needle on the patients with insomnia.
METHODSOne hundred and twenty patients with insomnia were randomly divided into a wooden needle group and a western medicine group, 60 cases in each group. In the wooden needle group, the patients were treated with wooden needle to press the plantar reflex areas, such as cerebellar, throid and cerebral areas. In the western medicine group, Alprazolam was taken orally. Before and after treatment, Pittsburgh Sleep Quality Index (PSQI) was used to evaluate the therapeutic effect of both groups.
RESULTSThe total therapeutic effect was 100.0% in the wooden needle group, while it was 90.7% in the western medicine group, there was no significant difference between the two groups (P > 0.05). Compared with PSQI before and after treatment, there was difference in the both groups (All P < 0.01), but there was no difference between the two groups (P > 0.05).
CONCLUSIONThe therapeutic effect of wooden needle is similar to that of Alprazolam on the insomnia, indicats that wooden needle is a better therapy for treating insomnia.

10.Association between apolipoprotein E polymorphism and myocardial infarction in patients with type 2 diabetes mellitus

Objective:To investigate the relationship between the E2 and E4 alleles of apolipoprotein E (apoE) gene and myocardial infarction (MI) in type 2 diabetes Mellitus (T2DM) patients, and to explore the relationship between apoE polymorphism and blood lipid metabolism.Methods:This case control study was conducted from August 2016 to March 2020 in China-Japan Friendship Hospital, 3 459 inpatients with T2DM were included including 3 044 patients without MI (T2DM group) and 415 patients with MI (T2DM+MI group). Real time fluorescent quantitative PCR was used to detect apoE polymorphism. Automatic biochemical analyzer was used to detect lipid levels. Logistic regression analyses were performed to determine the association of apoE with risk of MI in patients with T2DM.Results:(1) The frequency of E4 allele in T2DM+MI group (12.29%, 102/830) was significantly higher than in T2DM group (9.13%,556/6 088), while the frequency of E2 allele in T2DM+MI group (7.35%,61/830) was significantly lower than that in T2DM group (8.21%,500/6 088), P=0.012. Logistic regression analyses showed that E4 allele carrier (E3/E4+E4/E4) faced a higher risk for MI in T2DM patients ( OR=1.48, 95% CI 1.14-1.92, P=0.003), while E2 allele carrier(E2/E3+E2/E2)did not face a higher risk of MI in T2DM patients ( OR=0.88, P=0.642). (2) The levels of apoE polymorphism and blood lipid: The levels of TC, LDL-C and apoB increased in the order of E4 allele, wild type and E2 allele ( P<0.05). The levels of HDL-C, apoA1 and apoE decreased in the order of E4 allele, Wild type and E2 allele ( P<0.05). Conclusion:The E4 allele is a risk factor for MI in T2DM patients, and apoE polymorphism can affect blood lipid level in this patent cohort.
