1.Establishment and identification of L783V/3T3 cell line infected with leukemia viruses

L783 murine lymphocytic leukemia was established in 1978 by Dept. of Pathophysiology of Shanghai Medical University. L783V/3T3 cell line was established by infecting NIH-3T3 cells with the cell-free extracts of blood, spleen respectively of L783 mice. The plaque assay of L783V/3T3 cells was positive, typical A-type and C-type virus particles could be observed under electronmicroscope. Lymphocytic leukemia could be induced by inoculation of cell-free extracts from L783V/3T3 cells into Kunming strain newborn mice.
2.Preliminary study on the mechanism of 2:1 atrioventricular block during atrioventricular node reentrant tachycardia

Objective The purpose of this study was to clarify the mechanism of 2:1 atrioventricular block (AVB) during AV node reentrant tachycardia (AVNRT) induced during electrophysioloic study.Methods In consecutive patients with AVNRT referred for electrophysiologic study, the data of 2 : 1 AVB during induced AVNRT was retrospectively analysed. Results The data of 4 patients was excluded from analyzing because of the unsatisfactory recording of His bundle potential during AVNRT. A His bundle deflection was present in the blocked beats in three of the remaining 5 patients and absent in the other two. At the beginning of AVNRT induced in those patients whose His bundle deflection was present in the blocked beats, H-V Wenckebach sequence with a QRS pattern of RBBB or LBBB was seen preceding and following the 2 : 1 AVB. A pattern of H-V Wenckebach phenomenon occurred once during AVNRT with 2:1 AVB in one of the two patients whose His bundle deflection was absent in the blocked beats.Conclusion The induced 2:1 AVB during AVNRT is due to functional block in the His-Purkinje system regardless of the presence or absence of a His bundle deflection in blocked beats.
3.Effect of Shen Wu Guanxin Granules for Angina Pectoris Due to Coronary Heart Disease:An Observation of 60 Cases

Objective To observe the therapeutic effect of Shen Wu Guanxin Granules(SWGG),a prescription with the actions of tonifying kidney,strengthening Qi and activating blood,for angina pectoris(AP) due to coronary heart disease(CHD) with kidney deficiency and blood stasis.Methods Ninety patients with AP due to CHD with kidney deficiency and blood stasis were randomized into two groups according to the double-blind and double-dummy principles.The two groups received routine western medicine,and the treatment group(N=60) received SWGG and the control group(N=30) received Di'ao Xinxuekang Capsules additionally.The treatment lasted 4 weeks.After treatment,the therapeutic effect on AP and electrocardiogram(ECG) was evaluated,and the changes of main symptoms as well as the laboratory parameters were observed before and after treatment.Results In the treatment group,the total therapeutic effect on AP and ECG was 91.7% and 85.0% respectively,higher than 73.3% and 66.7% in the control group(P
4.Effects of Water-extracting and Concentration Process on Active Ingredients ofGanmao Tuire Granules

Objective To observe the changes in transfer rates of epigoitrin, forsythoside A and forsythin in Ganmao Tuire Granules prescription medicinal materials by water decoction extraction, vacuum concentration and atmospheric concentration.Methods Prescription medicinal materials fromGanmao Tuire Granules were decocted in water and the decoction was condensed by vacuum and atmospheric concentration process. The contents of epigoitrin, forsythoside A and forsythin at different states were measured by HPLC for transfer rates.Results The transfer rates of epigoitrin, forsythoside A and forsythin were 72.97%, 29.27%, 72.84%, respectively after decocted in water. The RSD of epigoitrin, forsythoside A and forsythin were less than 2% in the process of vacuum concentration. The transfer rates of epigoitrin, forsythoside A and forsythin by atmospheric concentration decreased by 13.51%, 7.38% and 4.24% respectively compared with vacuum concentration when concentrated to the same state.Conclusion The method for simultaneous determination of epigoitrin, forsythoside A and forsythin by HPLC has been established. A more suitable extraction and concentration process should be chosen for the preparation ofGanmao Tuire Granules.
5.Determination of Thermal Physical Parameters and Related Model of Water Extracts of Salvia Miltiorrhiza Radix et Rhizoma

Objective To determine the thermal conductivity under different temperatures and concentrations, and specific heat capacity under different concentrations of water extracts of Salvia Miltiorrhiza Radix et Rhizoma; To establish the mathematical model of thermal conductivity-temperature, thermal conductivity-concentration, thermal conductivity-temperature-concentration and specific heat capacity-concentration of water extracts of Salvia Miltiorrhiza Radix et Rhizoma. Methods Thermal conductivity and specific heat capacity were measured by the instantaneous double hot wire thermal conductivity meter and the electrothermal specific heat capacity meter. Excel, 1stOpt and MATLAB were used to analyze the experimental data. Results The method of using brix to facilitate and accurately characterize the concentration was established. The relationship between the thermal conductivity and the temperature and the concentration of water extracts of Salvia Miltiorrhiza Radix et Rhizoma were all linearly negative (λ=a-bT, λ=a-bC), and the influence of temperature and concentration on the thermal conductivity had a certain interaction. λ=a-bC-cT-dCT could be used as the temperature and the concentration on the thermal conductivity of the integrated role model. And the specific heat capacity of the extract was negatively correlated with the concentration (cp=a-bC). Conclusion The thermal conductivity and the specific heat capacity of the extract of water extracts of Salvia Miltiorrhiza Radix et Rhizoma are different at different temperatures and concentrations, and the model can be used to characterize the changing law of thermodynamics of the extracts. It can provide guidance significance for the thermal characteristics analysis in TCM pharmaceutical process and TCM production equipment selection and design, and production process control.
6.Content Determination of Total Iridoid Compounds and Baldrinal and 11-ethoxyviburtinal from Valerianae Jatamansi Rhizoma et Radix

ObjectiveTo establish a method for content determination of total iridoid compounds and baldrinal and 11-ethoxyviburtinal from Valerianae Jatamansi Rhizoma et Radix; To determine the contents of total iridoid compounds and baldrinal and 11-ethoxyviburtinal in Valerianae Jatamansi Rhizoma et Radix from three medicinal origins.Methods UV spectrophotometry was applied, 11-ethoxyviburtinal (cyclopentane-pyran-7-formaldehyde, 4-ethoxy methyl) was set as the reference substance, and the content of total iridoid compounds was determined at 288 nm. HPLC method was used to simultaneously determine the contents of baldrinal and 11-ethoxyviburtinal. The HPLC analysis was performed on a Phenomenex Luna C18 column (250 mm×4.6 mm, 5μm). The mobile phase was composed of acetonitrile-water in gradient elution at a flow rate of 0.95 mL/min. The detection wavelength was 288 nm and the column temperature was 30℃.Results The total iridoid compounds, baldrinal and 11-ethoxyviburtinal were in good linearity within the ranges of 2.088–14.616μg/μL, 74.88–224.64μg, and 41.6–249.6μg, respectively. This method was precise, and with good repeatability, stability and recovery rate.Conclusion The method is accurate, simple, rapid, which can be used for the quality control of Valerianae Jatamansi Rhizoma et Radix.
7.Determination of thermal physical parameters and model establishment for Ganmao Tuire Granule extract

Objective To determine the thermal conductivity coefficient (λ)under different temperatures (T)and different concentrations (C)and specific heat capacity (cp),so as to establish the mathemati-cal models of λ-T,λ-C,λ-T-C and cp-C of Ganmao Tuire Granules extract.Methods λand cp were determined by using instantaneous double line method and equivalent specific heat capacity meter respec-tively.Then the experimental data were analyzed using software Excel,1stOpt and MATLAB.Results There was a negative linear correlation between λand T,λand C(λ=a -bT,λ=a -bC),further-more,T and C had certain interactions on influence of λ.λ =a -bC -cT -dCT can be used as a com-bination model of temperature and concentration on the coefficient of thermal conductivity.In addition,cp had linear negative correlation with C (cp =a -bC).Conclusion Taking Ganmao Tuire granules ex-tract as subject,this study proved the great difference of λand cp of Chinese medicine extract under dif-ferent temperature and concentration.This study provided data support and theoretical guidance of ther-mal-physical parameters,of selection and design the medicine production equipment,of production process control,and of the key technology of production.
8.Study on the doctor-patient interest demands satisfaction of the payment system reform in China's new rural cooperative medical care scheme

Objective To study doctor-patient interest demands satisfaction and its influencing factors of the payment system reform of the new rural cooperative medical care scheme to provide reference for the reform. Methods Cross-sectional survey was conducted from September 2016 to February 2017. Multi-stage stratified random sampling was used in six counties of three provinces in the eastern, middle and western regions of China, and mathematical statistics was applied to analyze the data. Results The doctor-patient overall interest demands satisfaction was high, but the satisfaction was lower both with the income and ability improvement of medical staff and with the benefits of farmers. The influencing factors of the satisfaction of managers in medical institutions included the type of payment, educational level and work unit (P<0.05). The influencing factors of medical staff's satisfaction included the type of payment, work unit, and working years among others(P<0.05). The influencing factors of farmers'satisfaction included the type of payment and the average annual income, etc(P<0.05). Conclusions The core interest demands of both doctors and patients should be valued to enhance their satisfaction. Diseases related groups should be promoted and applied scientifically, and appropriately integrated with other methods of payment. Both doctors and patients'understanding of the payment reform should be improved by propaganda and training, to get their support and cooperation.
9.Empirical analysis on the synergistic development of the compound system for rural public health service

Objective To measure and analyze the order degree of the internal subsystems and the synergetic degree of the compound system of rural public health service in county areas based on the synergy model of the compound system. Methods According to the distribution of eastern, central and western regions in China, Xiangshan county in Zhejiang province, Wuzhi county in Henan province and Bin county in Shaanxi province were sampled. Based on the index system of synergetic degree measurement including the 52 level-2 evaluation indicators, a questionnaire survey was used to measure the synergetic degree of the compound system of rural public health service in county areas. Results The highest synergetic degrees of the compound system of rural public health service in Xiangshan county, Wuzhi county and Bin county were only 0. 18153, 0. 18068 and 0. 21312 respectively. This indicated the synergy at a low degree. And the synergy of the supply-demand system was an important influential factor for synergy of this compound system, as their development trends were consistent basically. Conclusions The synergy model of the compound system can play a useful role in rural public health service system in county areas. The demand of rural residents deserves more attention and the difference between the service supply and demand should be reduced.
10.A clinical study of clinical cure after the addition of interferon therapy in chronic hepatitis B patients with low-level HBsAg previously treated with nucleos(t)ide analogues

ObjectiveTo investigate the population with an advantage of clinical cure previously treated with nucleos(t)ide analogues (NAs), and to provide more methods for clinicians in pursuing the clinical cure of hepatitis B. MethodsA total of 42 chronic hepatitis B patients with low-level HBsAg who received NAs treatment in Hebi Third People’s Hospital from October 2017 to October 2019 were enrolled as subjects and divided into combination treatment group (group A) and NA monotherapy group (group B). The 22 subjects in group A were treated with NAs combined with PEG-IFN antiviral therapy for 48 weeks, and some patients withdrew from PEG-IFN after 24 weeks and continued to receive NA monotherapy, while the 20 subjects in group B received NA antiviral therapy alone. Both groups were observed till week 48, and the five makers for hepatitis B were measured to evaluate clinical outcome. The t-test was used for comparison of continuous data between two groups, and the Fisher’s exact test was used for comparison of categorical data between two groups; a multivariate logistic regression analysis was used to perform a multivariate analysis. ResultsCompared with group B at the 48-week treatment endpoint, group A had significantly higher HBsAg clearance rate (45.5% vs 0, P<0.01) and HBsAg seroconversion rate (31.8% vs 0, P<0.01). The population with HBsAg <1000 IU/ml, <500 IU/ml, <100 IU/ml, and <10 IU/ml had an HBsAg clearance rate of 52.6%, 61.5%, 66.7%, and 100%, respectively, and the population with an HBsAg level of 500-1000 IU/ml, 100-500 IU/ml, 10-100 IU/ml, and <10 IU/ml had an HBsAg clearance rate of 33.3%, 50%, 40%, and 100%, respectively. The 4 patients with baseline HBsAg <10 IU/ml (accounting for 18.2% in group A) achieved clinical cure at week 12 of combined treatment, and after observation to week 48, 2 patients had an anti-HBs level of >100 IU/ml and 2 had an anti-HBs level of >1000 IU/ml. The multivariate logistic regression analysis of HBsAg clearance showed that age at the initiation of combined treatment affected HBsAg clearance (odds ratio [OR]=0.877, 95% confidence interval [CI]: 0.781-0.985, P=0.026), and most of the patients with HBsAg clearance had an age of 36-49 (44.20±4.49) years; baseline HBsAg level also had an impact on HBsAg clearance (OR=0.996, 95% CI: 0.992-1.000, P=0.050). ConclusionThe addition of interferon therapy in chronic hepatitis B patients with low-level HBsAg previously treated with NAs can significantly improve the clinical cure rate. The younger the age and the lower the HBsAg level, the shorter the duration of combined treatment. Age and baseline HBsAg level are more important than the duration and type of NA medication.
