3.The study of disseminated tumor cell(DTC)and p53 gene in mouse model of laryngocarcinoma
Journal of Chongqing Medical University 2003;0(05):-
Objective:We established nude mouse model of laryngocarcinoma and detect the disseminated tumor cell of peripheral blood and mutant p53 in tumour tissue,to research the correlation of DTC with variant p53 gene and their value of predicting cancer metastasis and prognosis in laryngocarcinoma. Methods:There are 40 nude mice in the experimental group which accepted hypodermic injection of laryngocarcinoma cell,10 in control.By the reverse transcription polymerase chain reaction and nested polymerase chain reaction(Nest-PCR),we tested CK19mRNA in nude mouse's peripheral blood,as a marker of disseminated cancer cell at the 10th weeks after tumor-planting.We also tested the expression of p53 gene in tumour tissue by RT-PCR and immunohistology. After the nude dead,we dissect lung, liver and lymph node and use continuous pathological dyeing confirmed whether the transfer. Results:The positive rate of DTC was 50.0%(20/40),but negative in the control group, the difference was significant(P
4.Clinical characteristics and logistic regression analysis of risk factors for short-term prognosis in elderly patients with acute decompensated heart failure
Chinese Journal of Geriatrics 2016;35(1):8-12
Objective To investigate the clinical characteristics and the associated risk factors for short-term prognosis in elderly patients with acute decompensated heart failure, in order to provide the evidence for improving clinical survival. Methods Clinical data of 200 elderly patients with acute decompensated heart failure in our hospital from June 2010 to December 2014 were retrospectively analyzed. The associated risk factors for prognosis were analyzed by using multivariate logistic regression analysis. All patients were followed up for six months, and cardiovascular events during the same follow-up period were recorded. All patients were divided into non-event group (n=158, no readmission during the follow-up period) and event group (n=42, readmission or death during follow-up).Results All the 200 elderly patients with acute decompensated heart failure had completed the 6 months follow-up, 23 (11.5%) patients died from cardiovascular events, and 19 (9.5%) patients had readmission due to heart failure.New York Heart Association (NYHA) classification, age, heart rate, systolic blood pressure, left ventricular ejection fraction (LVEF), left ventricular end-systolic diameter (LVESD), and left ventricular end-diastolic dimension (LVEDD) had significant differences between the two groups (x2=5.791, t=2.514, 2.552, 2.500, 2.582, 3.870, 2.275, all P<0.05).Levels of NT-terminal pro-brain natriuretic peptide (NT-proBNP), high sensitivity C reactive protein (hs-CRP), red cell distribution width (RDW), creatinine, and uric acid were higher in the event group than in the non-event group (t=54.948, 9.932, 2.815, 8.888, 2.368, all P<0.05).The hemoglobin level was lower in the event group than in the non-event group (t-2.455, P<0.05).Multivariate unconditional logistic regression analysis showed that NYHA classification, LVEF, and NT-proBNP were the independent risk factors for the prognosis of acute decompensated heart failure in elderly patients (x2 =10.438, 61.943, 6.976, all P<0.05).The mortality rate was 11.5% within the six months follow-up.The area under the receiver operating characteristic (ROC) curve for NT-proBNP to predict short-term mortality was 0.860 (SE=0.034,P=0.000, 95% CI: 0.794-0.926).The mortality within 6 months was lower in NT-proBNP ≤205 ng/L group than in NT proBNP> 205 ng / L group (x2=5.385, P=0.020).Conclusions NYHA classification, LVEF, and NT proBNP are the risk factors for the prognosis of acutely decompensated heart failure in the elderly. The elderly patients with acute decompensated heart failure have a poor prognosis.
5.Preoperative blood neutrophil to lymphocyte ratio as an independent prognostic predictor for epithelial ovarian cancer
Chinese Journal of Clinical Oncology 2014;(10):634-638
Objective:This study aims to evaluate the effect of an elevated preoperative neutrophil to lymphocyte ratio (NLR) on outcome after comprehensive staging laparotomy or optimal tumor debulking surgery for epithelial ovarian cancer (EOC) and determine the value of the NLR as an independent prognostic prediction marker. Methods:A total of 80 women with primary EOC and with complete clinical and pathological information documented at the time of surgery were selected for this study. The optimum cut-off value of the preoperative NLR was identified through receiver operator characteristic (ROC) curve, and the patients were then classified into two groups: low and high NLR group. Univariate and multivariate analyses were performed to assess the prognostic effect of the preoperative NLR patients who underwent comprehensive staging laparotomy or optimal tumor debulking surgery. The levels of expression of CD68 were measured through immunohistochemistry. Results:The optimal cut-off value of the NLR was 3.8. The preoperative NLR differed significantly in the FIGO stage between the low NLR group (NLR ≤3.8) and the high NLR group (NLR>3.8), but no discrimination was observed in other parameters. The mean follow-up time was 45 months, and the post-operative 1-and 3-year survival rates were 93.7%and 60.0%, respectively. The preoperative NLR>3.8 and stageⅢ/Ⅳwere all risk factors for poor overall and disease-free survival. Multivariate analysis revealed the patients with high NLR (P<0.05) and stage Ⅲ/Ⅳ (P<0.05) had prognostic significance for poor overall survival. The number of CD68-positive tumor-associated macrophages was significantly higher in the high NLR group than in the low NLR group (54.65 ± 8.78 and 41.78 ± 9.10, respectively; P<0.001). Conclusion: An elevated blood preoperative NLR indicates poor prognosis in patients with EOC. Preoperative NLR may function as an important independent prognostic factor for patients with EOC.

10.The Early Diagnosis of Epidemic Hemorrhagic Fever With Immunoenzymatic Technique
Journal of Xi'an Jiaotong University(Medical Sciences) 1982;0(04):-
This paper recommends the early diagnosis of epidemic hemorrhagicfever with immunoenzymatic direct staining (IDS) to study the patient'speripheral white blood cells. The samples of blood smears from 63 patientswere examined under the microscope with this metbod. In 54 patients, the results were positive (85. 71%). We systemically observed 10 blood sam-ples from the patients in different courses of the disease, and found thatduring the period of recovery the results were negative. This method wasalso used to observe the blood smears from both 22 patients infected withother viruses and 20 healthy persons, and a negative result was obtained. It was demonstrated that IDS had stronger specificity. The advantages ofthis method are that blood samples are easily taken, carried and preserved, and that the experimental method is simple. Thus, IDS is of practical signifi-cance in preventing and treating the disease.
