1.Effects of crocetin on doxorubicin-induced damage in rats with myocardial mitochondria

AIM: To study the protective effects of crocetin on the doxorubicin-induced damage in rats with myocardial mitochondria. METHODS: Rats were given intraperitoneal injection of doxorubicin to induce cardiotoxicity. After continuous oral administration of crocetin, the rats were sacrificed, and myocardial mitochondria were isolated. The mitochondrial membrane potential (MMP), O~-_2. production, the activity of respiratory chain complex IV and glutathione peroxidase (GSH-PX), and DNA fragmentation were determined. The expression level of COII gene was determined through RT-PCR. RESULTS: Crocetin increased the activity of respiratory chain complex IV and GSH-PX, MMP and the expression level of COII and decreased DNA fragmentation and superoxide anion radical O~-_2. production in rats with myocardial mitochondria. CONCLUSION: Crocetin may decrease the damage in rat myocardial mitochondria induced by doxorubicin.

6.Preparation and evaluation of biodegradable molecularly imprinted polymers

BACKGROUND:The cross-linking reagent is the main unit to support the skeleton of molecularly imprinted polymers, which is closely related to the bio-friendly adaption of polymers. The biocompatible and biodegradable capacities of common crosslinking agents are unclear.
OBJECTIVE:To prepare a novel biodegradable molecularly imprinted polymer and to evaluate the adsorption and biodegradability properties of the polymer.
METHODS:The biodegradable molecularly imprinted polymer was synthesized with the acrylated polyε-caprolactone as the cross-linking reagent under the UV polymerization. The adsorption of the polymer was evaluated by adsorption isotherm, Scatchard analysis and kinetic curves. The biodegradation of the polymer was observed in the simulated physiological environment system in vitro.
RESULTS AND CONCLUSION:Both the molecularly imprinted polymer and non-molecularly imprinted polymer showed an adsorption effect on theophyl ine, the template. But the adsorption amount of the molecularly imprinted polymer was significantly more than that of the non-molecularly imprinted polymer. The drug loading and encapsulation efficiency of the molecularly imprinted polymer were 1.54%and 12.48%, respectively. The degradation rates of molecularly imprinted polymer and poly(ε-caprolactone) diol were 6.60%and 1.33%, respectively, within the observation time in vitro. The obtained molecularly imprinted polymer not only exhibited specific adsorption to certain molecules but also showed good biodegradable properties in the simulated physiological environment system, which is necessary to be a potent drug carrier.
7.Effects of hypoxia-inducible factor-1α on radiation-induced autophagic cell death in breast cancer cells

Objective To study the effects of hypoxia-inducible factor-1α (HIF-1α) on radiationinduced autophagic cell death in breast cancer cells.Methods MCF-7 cells were divided into four groups:control (normoxia,21% Oxygen),irradiation (8 Gy X-rays),hypoxia (Cobalt chloride,CoCl2)and irradiation with hypoxia (CoCl2).150 μmol/L CoCl2 was utilized to induce hypoxic conditions.Western blot was applied to detect the expression of HIF-1α and MAPLC3.MDC and Hoechst staining were used to detect autophagy and apoptosis.Radiosensitivity was detected by cloning formation.The short hairpin interfering RNA (shRNA) retroviral transduction particles targeting HIF-1α was transfected into MCF-7 cells to establish HIF-1 α knockdown cells,then the radiosensibility,autophagy and apoptosis were detected.Results Compared with control group and irradiation group,the protein level of HIF-1 increased obviously in the normaxia,irradiation,hypoxia and irradiation with hypoxia groups,and the values were 0,0,1.00,1.89,respectively.The expression levels of MAPLC3 were markedly up-regulated in irradiation,hypoxia and irradiation with hypoxia groups as compared with control,and the ratios of LC3Ⅱ/LC3Ⅰ were 1.15,1.73,2.38 and 3.60,respectively.The radiosensitivity of MCF-7 cells decreased in the following order:normoxia with 3MA > normoxia > hypoxia with 3MA > hypoxia.HIF-1α knockdown cell (pSUPER-HIF-1α Ri) and vector control were constructed.After treatment with CoCl2,survival fraction of MCF-7-pSUPER was significantly higher than that of control (t =3.080,6.946,6.658,6.380,P <0.05),and radiosensitivity was down-regulated after irradiation,but there was no significant difference between normoxia and hypoxia in survival fraction of MCF-7-pSUPER-HIF-1α Ri.After treatment of irradiation or hypoxia,the autophagic fractions in MCF-7-pSUPER-HIF-1α Ri significantly decreased,reduced by 21.1%,25.5%,15.5%,respectively(t =4.635,4.738,6.354,P <0.05) as compared with MCF-7-pSUPER,but there was no change in apoptosis.Conclusions HIF-1α may increase radiationinduced autophagy and decrease radiosensitivity,but have no influence on apoptosis.
8.Effect evaluation of nursing intervention on coping with fatigue in patients with cervical spine fracture and high paraplegia

Objective To study on the effect of nttrsing intervention on coping with fatigue in patients with cervical spine fracture and high paraplegia.Methods 46 patients who suffered cervical spine fracture with paraplegia were divided into the control group and the intervention group with 23 patients in each group randomly.The control group received routine care in general,while the intervention group was given additional systematic nursing intervention by full-time nurses.The brief fatigue inventory(FSI),as fatigue assessment tools,was used to assess the fatigue of patients in the two groups before invention and 20 days after invention respectively.Results As a result,we found that the difference of fatigue between the two groups had no prominent significance before intervention,but after invention the difference had prominent significance.Conclusions The nursing intervention is very important for patients with cervical spine fracture and paraplegia and has significant advantages to alleviate or eliminate patients' fatigue and improve their quality of life.
9.Correlation between autophagy and polarization of macrophages in atherosclerosis plaque in arteriosclerosis obliterans amputees.

This study was designed to investigate the correlation between autophagy and polarization of macrophages in atherosclerosis (AS) plaque in arteriosclerosis obliterans amputees. Femoral artery specimens from arteriosclerosis obliterans amputees were performed hematoxylin and eosin (HE) staining, oil red O and immunofluorescence staining to observe the morphology of atherosclerotic plaque, phenotype of macrophages and autophagy in plaque; using real-time quantitative RT-PCR technology to detect the mRNA level of M1 and M2 type markers in arterial tissue; to analyze polarized signal pathway and autophagy protein levels in macrophages by Western blotting. Arterial specimens staining showed obvious lipid deposition and obvious infiltration of amount of foam cells and inflammatory cells. Macrophages were mainly expression M1 type in percentage in fibrous plaque. Although both M1 and M2 macrophages were upregulated in atheromatous plaque, the increase was dominant in M2 type in percentage. The level of autophagy was significantly higher in the atheromatous plaque than that of fibrous plaque. The expression of tumor necrosis factor- α (TNF-α), monocyte chemotactic protein-1 (MCP-1), inducible nitric oxide synthase (iNOS), interleukin-6 (IL-6) and interleukin-12 (IL-12) mRNA was significantly higher in fibrous plaque than that of atheromatous plaque (P < 0.01 or 0.05), and arginase-1 (Arg-1), transforming growth factor-β (TGF-β), CD163 and interleukin-10 (IL-10) mRNA was significantly lower than that in atheromatous plaque (P < 0.01). The levels of p-STAT1 and NF-κB were significantly increased in fibrous plaque (P < 0.01), while p-STAT6 expression was significantly increased in atheromatous plaque (P < 0.01). The level of LC3-II was significantly higher in atheromatous plaque than that in fibrous plaque (P < 0.01). Macrophages in early atherosclerotic plaque were induced to M1 type through p-STAT1/NF-κB pathway and expressed moderate levels of autophagy; while macrophages in advanced plaques were induced to polarization of M2 type through p-STAT6 pathway. M2 macrophages expressed a higher level of autophagy than M1 macrophages.
