1.Research advances in cardiac stem cells
Chinese Journal of Tissue Engineering Research 2011;15(49):9299-9302
BACKGROUND: A large amount of studies have demonstrated that stem cells in the heart of mammal participate in heart self-renewal and endogenous repair.OBJECTIVE: To summarize the source, classification, features of cardiac stem cells and application in heart disease. METHODS: A computer-based online search of PubMed database was performed for articles published between January 2000 and December 2010 related to the source, classification, features of cardiac stem cells and its effects on myocardial regeneration with key words cardiac stem cell. Clinical studies and basic studies were all included. Repetitive studies and Meta analysis were excluded. Finally, 32 articles were included.RESULTS AND CONCLUSION: Cardiac stem cell is a type of stem cells in the heart, with properties of self-renewal and cloning proliferation. It can differentiate into cardiomyocyte and endothelial cell and plays a role in heart injury repair to improve heart function. Cardiac stem cells have been in vitro isolated, cultured and amplified to transplant into animal heart, which lays a foundation for clinical application. However, some questions remain poorly understood as to the homeostasis and dynamic changes, processes involved in their differentiation into heart functioning cells, as well as influential factors and regulation.

Long non-coding RNAs (lncRNAs) are non-coding RNAs that have transcript lengths exceeding 200 bp and do not have the capacity for protein coding because of having no open reading frame. In the human genome, lncRNAs play important regulato-ry roles in the process of epigenetic, transcription and post-transcription, so they have become the focus of research followed by mi-croRNAs. The abnormal expression of lncRNAs in cancer usually represents different functions, such as the function of oncogenes or tumor suppressor genes, which promote or inhibit tumor growth. Lung cancer is a common malignant tumor with a five-year survival rate of 17%. Previous literature shows that MALAT1, H19, lincRNA p21, UCA1, and BC200 are closely related to the development of lung cancer. They could promote cancer growth, invasion and metastasis, apoptosis and induce drug resistance, and so on. This review aims to provide assistance in the diagnosis, treatment, and prognosis of lung cancer by clarifying the functions and mechanisms of ln-cRNAs.

Objective Detecting the changes of the expression of vascular endothelial growth factor(VEGF) during rat liver regeneration so as to study the role of VEGF in the course of liver regeneration. Methods Three hundred adult SD rats were randomly divided into three groups:control group, sham operated group and test group. After taking out liver tissue and isolating hepatocytes at different phases after 2/3 rat hepatectomy, the change of VEGF expression was detected by both immunocytochemistry and Western blotting. Results The results of immunocytochemistry showed that VEGF positive cells were little in normal liver. The amounts of immunoreaction positive cells began to increase after 24 h of partial hepatectomy(P

Background and purpose:As a new candidate tumor suppressor gene, p33ING1b has many biological functions. This study was done to investigate its role in the regulation of the proliferation of human colon cancer cell line SW480. Methods:The pcDNA3.1(+)/p33ING1b/SW480 cells were identified by Western blot and S-P immunohistochemical method. In order to elucidate the effect of expression of exogenous p33ING1b gene on the colorectal cancer cell SW480, the proliferation rates were analyzed by growth curves and colony formation assay in soft agar for cells including SW480, pcDNA3.1(+)/p33ING1b/SW480 and pcDNA3.1(+)/SW480. At same time, the apoptotic rate of cells and the cell cycle analysis were also tested by flow cytometry.Furthermore, using western blot analysis,we detected the expression level of the protein p53, p21WAF1,Bax and Bcl-2 in those three group cells, which initially indicate the molecule mechanism of inducing apoptosis by gene p33ING1b. Results:The cell growth rates of SW480 cells transfected with pcDNA3.1 (+)/p33ING1b were slower than those transfected with pcDNA3.1(+) or untransfected. The colony formation efficiency of pcDNA3.1(+)/p33ING1b/SW480 were decreased and the apoptotic rates were increased compared with pcDNA3.1(+)/SW480 and SW480(P0.05). Conclusion:After overexpression of exogenous p33ING1b protein in SW480 cell, there were inhibition of the proliferation rate and induction of apoptosis, the molecular mechanism might be associated with up- regulated expression of Bax and down-regulated expression of Bcl-2 by p33ING1b gene.

Objective To investigate the short-term efficacy and safety of gefitinib combined with dendritic cells-cytokine induced killer (DC-CIK) cells in the treatment of advanced lung adenocarcinoma. Methods Fifty patients with lung adenocarcinoma who in previous had underwent radiotherapy and first-line chemotherapy failure received DC-CIK in combination with gefitinib treatment, blood count changes, imaging data, carcinoembryonic antigen and changes in the quality of life before and after treatment were compared and evaluated. Results DC-CIK could improve effectively and relieve bone marrow suppression response after chemotherapy and significantly increase the WBC content of blood (P < 0.01).After treatment, the tumor carcinoembryonic antigen (CEA) was significantly lower in patients (P < 0.01).Fifty cases of patients in complete remission (CR) were 0 cases, partial remission (PR) were 24 cases, stable disease (SD) were 17 cases and progression (PD) were 9 cases, and the response rates (RR) were 48%;The quality of life of patients was significantly improved (P < 0.05).Common adverse reactions were rash, diarrhea, chill and fever, but mild symptoms could be relieved after symptomatic treatment. Conclusions Gefitinib with autologous DC-CIK cell infusion second-line treatment of advanced lung cancer have a certain short-term efficacy, without significant adverse reactions.Gefitinib with autologous DC-CIK cell therapy can mitigate the response of bone marrow suppression in patients with advanced lung cancer and improve the quality of life of patients.Long-term effect remains to be investigated.

Objective To identify a myeloid differentiation factor 88(MyD88)in Oncomelania hupensis, and characterize the role of MyD88 against Schistosoma japonicum infection. Methods The complete cDNA of MyD88 in O. hupensis was ob-tained by using rapid amplification of cDNA ends(RACE),and homologues sequences and conserved domains were aligned and the structure of MyD88 was predicted either. A phylogenetic tree of MyD88 was further constructed with other species. In ad-dition,the mRNA expression level of O. hupensis MyD88 before and after S. japonicum infection was investigated by real-time quantitative PCR(RT-qPCR). Results The cDNA of O. hupensis MyD88 consisted of 1406 bp open reading frame(ORF),en-coding 468 amino acid residues, which contained death domain and Toll/interlrukin-1 receptor(TIR)domain, the typical fea-tures of MyD88 family proteins. The predicted amino acid sequence of O. hupensis MyD88 shared 38%-52%identity with other mollusc. O. hupensis MyD88 was phylogenetically closeted to Biomphalaria glabrata MyD88. The O. hupensis MyD88 existed in all selected tissues and expressed highly in hemocyte,up-regulated after S. japonicum infection in all selected tissues except cephalopodium, especially higher in whole snail and hemocyte. Conclusion MyD88-dependent signaling pathway is present in O. hupensis and plays an important role in innate immune response against S. japonicum infection.
