Objectlve To explore the effect of platelet-rich plasma (PRP) on cellular proliferation and intracytoplasm calcium ion concentration of osteoblasts and tenocytes during the process of tendon-bone healing. Methods We established a kind of indirect co-culture system using transwell chambers to co-culture osteoblasts and tenocytes.The cellular proliferation was carried out in following manners:single culture of osteoblasts versus single culture of osteoblasts plus PRP and co-culture of osteoblasts versus co-culture of osteoblasts plus PRP; single culture of tenocytes versus single culture of tenocytes plus PRP and co-culture of tenocytes versus co-culture of tenocytes plus PRP.The cellular proliferation rates were measured by CCK-8 test.The intracytoplasm calcium ion concentration of was measured by laser confocal microscopy and fluo-3/AM. Results The proliferation rates and fluorescence intensities in the co-culture groups without PRP were significantly lower than other groups ( P ＜ 0.05).The proliferation rates and fluorescence intensities in the single culture groups without PRP were significantly higher than in the co-culture groups without PRP but significantly lower than in the groups with PRP ( P ＜ 0.05).The proliferation rates and fluorescence intensities in the groups with PRP were significantly higher than other groups ( P ＜ 0.05) but there was no significant difference between groups of the same cells ( P ＞ 0.05).The intracytoplasm calcium ion concentrations were proportional to the proliferation rates. Conclusions During the process of tendon-bone healing,PRP has a potential not only to wipe off the depression effect of osteoblasts and tenocytes co-cultured indirectly but also to enhance the cellular proliferation rate to a higher level.At the same time the calcium ion concentrations will be elevated.

OBJECTIVE: To elaborate cartilage tissue engineering in the gene transfer technology and its application, in addition, to make a prospects for its further application.METHODS: The database of Science Direct database (2003-01/2009-04) and CNKI (2003-01/2009-04) were retrieved with key words of "cartilage tissue engineering, gene transfer". The literature was limited to English and Chinese languages. Literatures concerning cartilage tissue engineering in the gene transfer technology were selected, including clinical research and basic research. Other unrelated papers were excluded. Chondrocyte differentiation and gene expression were observed.RESULTS: A total of 90 literatures were searched by computer, according to inclusive and exclusive criteria, the papers regarding cartilage tissue engineering in the gene transfection and gene types and options were analyzed. Gene transfer technology in the field of cartilage tissue engineering has broad application prospects. How to select genes associated with cartilage repair as the transfacted gene need urgent solution. Currently, the used target gene can be divided into following categories, including stimulated cartilage cell proliferation and differentiation, matrix formation, inhibit chondrocyta hypertrophy and osteoblast differentiation, anti-inflammatory response, inhibit senescence and inhibit apoptosis.CONCLUSION: It has a special significance to select the appropriate target genes, and to use a safe gene transfer method to repair cartilage. The clinical application of gene transfer technology is depended on the construction of safe and effective carriers,target genes, as well as transfection systems.

BACKGROUND: Adipose-derived mesenchymal stem cells have similar morphological and biological characteristics to bone marrow-derived mesenchymal stem cells, which can be used as sources of seed cells for tissue engineering.OBJECTIVE: To understand the biological characteristics of adipose-derived mesenchymal stem cells, and to explore its clinical application and prospects in tissue engineering field.METHODS: The databases of PubMed and CNKI were searched with key words of "adipose tissue-derived stem cell, tissue engineering, and stem cells". Literature search was limited to English and Chinese languages. The ossification potential of adipose-derived mesenchymal stern calls and the outcomes combined adipose-derived mesenchymal stem cells with gene transfection to treat diseases were served as evaluative indicators. The in vitro study of comparison between bone marrow-derived mesenchymal stem cells and adipose-derived mesenchymal stern cells in ossification was included, and irrelative or repetitive papers were excluded.RESULTS AND CONCLUSION: Adipose tissue-derived stem cells can be obtained in large numbers from adipose tissue, and stably proliferate and differentiated in vitro, which possess the similar characteristics to bone marrow-derived menchymal stem call in morphology and biology. Under certain induction, the adipose tissue-derived stem cells can directional differentiated into all three germ layers of cells. Combined adipose tissue-derived stem cells with tissue engineering scaffold could be used to repair bone and articular cartilage defects, but the quality and the surrounding cartilage connecting cartilage, bio-mechanical strength, and future normal cartilage degradation have a certain gap to normal cartilage. With the understanding of adult mesenchymal stem cell research, it found that the self-amplification and differentiation potential of mesenchymal stem cells can effective disused the import of "exogenous gene", thus, it is easy to in vitro genetic modification. Therefore, the adipose-derived stem calls can be combined with genetic engineering, and applied to gene therapy. However, in the present research, the remaining potential carcinogenicity in the gene vector and the negative impact after transfection has not been clarified.

Assay"'of Epstein-Barr virus (EBV) DNA was conducted in the tissues of 60 patients with nasopharyngeal carcinoma (NPC), 30 patients with chronic inflamation of nasopharyngeal mucous membrane and 40 patients with other malignant epithelial tumor respectively using DNA in situ hybridization method. The positive rate of EBV-DNA in patients of NPC was 71.6%, while it was negative in pat ents with other malignant tumors a small number of EB-VDNA positive cells were also discovered in the epithelial cells of paratumours and chronic inflamation of nasopharyngeal mucous membrane. Significant correlation was found in the study of serum EBV-antiby in patients of NPC and EBV-DNA in cancer cells. The results suggest that EBV may be of etiologic significance in the pathogenesis of NPC.

OBJECTIVE:To improve automatic outpatient pharmacy drug delivery system in our hospital,and to promote the efficiency of automatic drug delivery. METHODS:The outpatient pharmacy automation system of our hospital was introduced in terms of main structure,working process and application. The work efficiency,safety and error were compared between automatic drug delivery mode and manual drug delivery mode. Drug delivery machine was improved to increase the rate of drug delivery. RE-SULTS:The outpatient pharmacy automation system of our hospital included 2 boxed drug delivery machine and 1 drug dispensing machine,etc. Compared with manual drug delivery mode,automatic drug delivery mode improved drug delivery efficiency,health security and use security,and reduced drug delivery error. Through adjusting the time setting and step of drug delivery of gearing in drug delivery machine,idle time of transmission equipment was utilized effectively,and the quantity of delivered prescriptions was increased,increasing from 220 to 320 each hour. CONCLUSIONS:The reasonable improvement of outpatient pharmacy auto-mation system makes outpatient pharmacy automation system play a bigger role,and further optimize resources.

Objective To study the physical stress reactions of soldiers undergone 100 kilometer march in battle gear,and explore the mechanism of biochemical changes in special military physical training.Methods Thirty four soldiers of armed-police force,averagely aged 18.6?1.6 years,169.3?4.2cm in height,weighted 65.4?4.5kg and with one-year of military physical training experience,were involved in present study.The soldiers were loaded about 25 kilogram of battle gear in a supervised 100 kilometer march in three days.The changes on biochemical index after the march were analyzed.Results After 100 kilometer march in battle gear,significant decline were found in the following biochemistry indexes in the soldiers involved: HB(P

AIM　The aim is to investigate the chemical constituents of Belamcanda chinensis. METHODS　The chemical constituents were isolated and purified by solvent extraction together with various chromatographic techniques. The stuctures was elucidated on the basis of chemical evidence and spectral data. RESULTS　Eight compounds were isolated from the ethanol extract of the rhizome of Belamcanda chinensis (L.)DC..Among them,six isoflavones were elucidated as irilone(Ⅰ)、genistein(Ⅱ)、tectorigenin(Ⅲ)、 irigenin(Ⅳ)、dimethyltectorigenin(Ⅴ)、irisflorentin(Ⅵ). CONCULSION　 ⅠandⅡ were isolated from this medicinal plant for the first time.

Objective To compare the short-term follow-up cu ra tive effect of primary total hip replacement (THR) in treating fresh and old fem oral neck fractures. Methods Double follow-ups with an in terval of 13 months were performed in patients with femoral neck fractures under going THR in our department from January 1997 to October 1999. Results After a follow-up for 37-62 months (mean 44 months), we obtained f ull data of 47 patients [41-77 years old, (61.8?5.22) years old] with preoperative Harris score of 28.40?10.14 and postoperative score of 85.20?7. 91. The Harris score of patients with fresh femoral neck fractures (FFNF) incr eased from 21.60?8.73 preoperatively to 82.50?9.81 postoperatively〔at age of (63.50?5.53) years〕 and that of patients with old femoral neck fracture (O FNF) from 37.70?13.50 to 90.63?5.79, [at age of (58.90?7.81) years]. Of 47 patients, only one case had dislocation once within six weeks postoperative ly during function exercise and recovered after manual reduction. Another one wi th excellent function evaluation was given again internal fixation three years a fter THR because of femoral shaft fracture and obtained satisfactory recovery. Objective To explore the etiology of and timing for operation of acalculous biliary acute pancreatitis(ABAP).MethodsWe retrospectively analyzed the etiology study and treatment experience of 38 cases of ABAP.ResultsThirteen cases underwent a surgery, including 6 early operations for complicated biliary tract infection without morbidity and mortality, 5 middle staged patients (5～14 days after onset of ABAP) underwent surgery because of intraabdominal indications with one postoperative death and 2 late operations for severe acute pancreatitis (SAP) resulted in one postoperative death of sepsis. The total postoperative mortality rate was 15%( 2/13). Four SAP cases were managed conservatively with one death of MOF. The remaining 21 cases of acute edematous pancreatitis (AEP) were cured by nonoperative methods.ConclusionWhereas an early exploration is indicated, when complicated by biliary tract infections, it is judicious to try a conservative therapy first for ABAP patients.

ResultsThree compounds were isolated from the EtoAC extracts of the rhizome of B. chinensis which were isorhamnetin (Ⅹ), hispidulin (Ⅺ), dichotomitin ( ⅩⅡ); four compounds were isolated from n-BuOH extracts, which were iridin ( ⅩⅢ), tectoridin (ⅩⅣ), daucosterol ( ⅩⅤ), vittadinoside or stigmasterol-3-O-glucoside ( ⅩⅥ). ConclusionCompound Ⅺ is isolated from this medicinal plant for the first time.
