1.Effect of Deproteinated Bone-BMP Complex on Bone Tunnel Enlargement after Anterior Cruciate Ligament Reconstruction

Objective To investigate the effect of deproteinated bone(DPB)-bone morphogenetic protein(BMP) complex on bone tunnel enlargement after anterior cruciate ligament reconstruction in rabbits.Methods A total of 96 adult New Zealand white rabbits were used in this study,and divided into 4 groups.In each group,we reconstructed the anterior cruciate ligaments of the animals using semitendinosus autograft.After the operation,DPB-BMP complex,DPB,or BMP was implanted into the femoral bone tunnel of the rabbits in three of the 4 groups.Another group that received neither DPB nor BMP served as a control.In 4,8,12,or 16 weeks after the implantation(6 rabbits in each group at each time point),we killed the animals to obtain the specimens of the femoral bone tunnel.Then the width of the tunnel was measured,and the percentage of its enlargement was calculated.Results In the DPB+BMP,DPB,BMP,and control groups,the bone tunnel was enlarged by(23.52?0.43)%,(34.83?0.52)%,(51.57?0.76)%,and(56.90?0.81)% in 4 weeks,(22.21?0.34)%,(35.35?0.46)%,(60.97?0.63)%,and(67.18?0.70)% in 8 weeks,(21.94?0.37)%,(33.01?0.41)%,(50.56?0.54)%,and(54.61?0.55)% in 12 weeks,and(20.96?0.35)%,(32.11?0.50)%,(49.29?0.66)%,and(53.31?0.59)% in 16 weeks,respectively.The rates of bone tunnel enlargement in the DPB+BMP group was the lowest at each time point(DPB+BMP group
2.Effects of Sericin Pretreatment on the Expression of ECM Associated Protein in the Kidney of Diabetic Nephropathy Rats

Objective To investigate the effects of sericin pretreatment on the expression of extracellular matrix(ECM) associated protein in diabetic nephropathy(DN) rats' kidney.Methods Sixty six male SD rats were randomly divided into 3 groups(n=12):normal control group,DN model group and sericine pretreatment group.DN rats model in model group and sericine pretreatment group were established by intraperitoneally injection of streptozotocin(STZ).Blood glucose≥16.7 mmol/L was taken as the standard of successful modelization.The rats in sericine pretreatment group were lavaged with sericine(2.4 g·kg~(-1)·d~(-1)) for 35 days before injecting STZ.The enzymic method was used to measure the blood glucose.Type Ⅳ collagen(cⅣ)and laminin(LN)content in the serum were detected by ELBA.The expression of transforming growth factor-β_1,(TGF-β_1)and tissue inhibitors of maprix metalloproteinase-1(TMP-1) protein in the kidney was observed by immunohistochemical staining.The expression of Smad 3 protein in the kidney was detected by Western blot.Results Compared with normal control rats,the blood glucose,cⅣ and LN content in the serum,TGF-β_1,TIMP-1 and Smad 3 expression in the kidney of the model group rats increased obviously(P<0.01).The blood glucose,cⅣ and LN content in the serum,TGF-β_1,TMP-1 and Smad3 expression in the kidney of rats in sericine pretreatment group were significantly lower than those of the rats in model group(P<0.01).Conclusion Sericin pretreatment can inhibit the activation of TGF-β/Smad 3 signal pathway in the kidney of DN rats,and prevent the decrease of MMPs activity induced by up-regulation of TIMP-1.So sericin can prevent accumulation of ECM and glomerulosclerosis during DN,and has satisfactory apotropaic effects on the development of DN.
3.Simultaneous Determination of 34 Pesticide Residues in Vegetable Oil by QuEChERS-on-line Gel Permeation Chromatography-Gas Chromatography-Mass Spectrometry

A method for the simultaneous determination of 34 pesticides in sunflower oil, soybean oil and corn oil was developed. The samples were extracted and purified by a modified QuEChERS method, and then the supernatant was analyzed by on-line gel permeation chromatography-gas chromatography-mass spectrometry ( GPC-GC-MS ) . The linear range was from 0 . 01 to 0 . 2 mg/L with a good correlation coefficients ( r≥0. 9913). The average recoveries of 31 pesticides (except p,p′-DDE, p,p′-DDD, p,p′-DDT. For detail, please reference to section 3 . 6 ) ranged from 70 . 3% to 115 . 4%, 69 . 5% to 112 . 6% and 70 . 2% to 116 . 1%spiked at 0. 05 μg/g and 0. 1 μg/g with the relative standard deviations (RSDs, n=6) less than 13. 3%, 13. 5% and 12. 1% in sunflower oil, soybean oil and corn oil samples, respectively. The LODs of this method ranged from 0. 0692 to 2. 28, 0. 0559 to 2. 01 and 0. 0584 to 2. 14μg/kg (S/N=3) in sunflower oil, soybean oil and corn oil samples respectively. The convenient operation and versatility of this method are suitable for the fast screening and detection of 34 pesticide residues in sunflower oil, soybean oil and corn oil.
4.Prognosis of children with septic shock assessed by lactate and lactate clearance rate

Objective To evaluate the relationship between lactate,lactate clearance rate and prognosis of children with septic shock.Methods A total of 42 children with septic shock admitted in the PICU from Oct 2009 to Jun 2011 were enrolled in the study.All the children were given goal-directed therapy early according to the treatment routine of septic shock.The blood lactate levels before resuscitation and at 1h,2h,6 h after resuscitation were detected,the lactate clearance rates at 1 h,2 h,6 h after resuscitation were calculated.All the patients were divided into the survival group ( n =20 ) and the non-survival group ( n =22 ) by prognosis.The relationship between the early lactate level,lactate clearance rate and prognosis of the children with septic shock was evaluated.Results ( 1 ) The lactate levels before resuscitation and at 1 h,2 h,6 h after resuscitation in the survival group were significantly lower than those in the non-survival group [ (3.92 ± 2.58) mmol/L vs (6.91 ± 4.16) mmol/L,( 2.79 ± 1.89 ) mmol/L vs ( 7.93 ± 4.39 ) mmol/L,( 2.20 ±1.83) mmol/L vs (9.20 ±4.97) mmol/L,( 1.32 ±0.51 ) mmol/L vs (9.94 ±5.02) mmol/L,P <0.05].(2) The lactate clearance rates at 1 h,2 h,6 h after resuscitation in the survival group were significantly higher than those in the non-survival group [ (26.31 ± 20.82) % vs ( - 24.28 ± 53.39 ) %,(43.46 ± 17.85 ) % vs (-34.31±58.98)%,(61.04±16.71)% vs ( -45.33 ±83.51)%,P<0.05].(3) The area under curve of the lactate and the lactate clearance rate at 6 h after resuscitation were 99.4% and 96.7% according to the receiver operating characteristic curve.The children had high mortality if the lactate value > 2.20 mmol/L and lactate clearance rate < 18.65% at 6 h after resuscitation.Conclusion It is important to dynamicly monitor the lactate of the children with septic shock,the lactate and the lactate clearance rate at 6 h after resuscitation can be used to evaluate the prognosis.

Olfactory neuroblastoma is a rare malignant tumor. Although multiple therapeutic modalities including surgery, radio-therapy, or chemotherapy could be used in patients with olfactory neuroblastoma, no standardized treatment has been achieved. This re-view introduces a case of adult olfactory neuroblastoma treated by a multiple disciplinary team in Tianjin Medical University Cancer In-stitute and Hospital. This review also aims to explore a complete set of diagnostic and treatment practices for the benefit of future pa-tients.

Adenovirus is a common cause of respiratory infection in children and can cause severe pneumonia.Adenovirus can also cause damage to immune system.It can cause decrease in T and B cell number and function, imbalance of Th1/Th2 cell proportion, but also can cause imbalance of proinflammatory cytokines and anti-inflammatory cytokines, further leading to dysfunction of immune system, with severe complications such as severe sepsis and multiple organ and system dysfunction.In this paper we will discuss immune pathogenesis of adenovirus infection and provide suggestion for treatment of adenovirus pneumonia.

8.Effect of ulinastatin on programmed necrosis in hippocampal neurons in a rat model of global cerebral ischemia-reperfusion

Objective To evaluate the effect of ulinastatin on programmed necrosis in hippocampal neurons in a rat model of global cerebral ischemia-reperfusion (I/R).Methods Forty-eight clean-grade healthy adult male Sprague-Dawley rats,aged 8 weeks,weighing 280-320 g,were divided into 3 groups (n=16 each) using a random number table method:sham operation group (Sham group),global cerebral I/R group (I/R group) and ulinastatin group (UT group).Global cerebral I/R was produced by 4-vessel occlusion method in chloral hydrate-anesthetized rats in I/R and UT groups.Ulinastatin 100 000 U/kg was injected via the tail vein at the onset of ischemia in group UT,and the equal volume of normal saline was given instead in Sham and I/R groups.Neurological deficit score (NDS) was estimated at 6,12 and 24 h of reperfusion.Animals were sacrificed at 24 h of reperfusion,brains were removed and the hippocampi were obtained for examination of pathological changes (with a light microscope) and for determination of the malondialdehyde (MDA) content and superoxide dismutase (SOD) activity (by spectrophotometry),and expression of receptor-interacting protein kinase 1 (RIPK1),RIPK3,and mixed lineage kinase domain-like protein (MLKL) in hippocampal tissues (by Western blot).Results Compared with Sham group,the NDS was significantly increased at each time point,the MDA content was increased,the SOD activity was decreased,and the expression of RIPK1,RIPK3 and MLKL was up-regulated in I/R and UT groups (P< 0.05).Compared with I/R group,the NDS was significantly decreased at each time point,the MDA content was decreased,the SOD activity was increased,and the expression of RIPK1,RIPK3 and MLKL was down-regulated in UT group (P<0.05).The pathological changes of hippocampi were significantly attenuated in UT group when compared with I/R group.Conclusion The mechanism by which ulinastatin ameliorates global cerebral I/R injury is related to inhibiting programmed necrosis in hippocampal neurons of rats.
9.Research progress of non-coding RNA in cardiovascular disease

Cardiovascular disease is the leading cause of human deaths in the world.Studies have found that non-coding RNA plays an important role in cardiovascular disease.Non-coding RNA is not only a potential specific biomarker for cardiovascular disease diagnosis, but also a therapeutic target for the disease.In this paper, the research progress of the diagnostic value and regulatory role of non-coding RNA in the pathogenesis of cardiovascular disease was reviewed in order to provide new ideas for the diagnosis and treatment of cardiovascular diseases.
